Differentially expressed genes of Acanthamoeba castellanii during encystation

  • To examine the expressed gene profile during encystation of Acanthamoeba castellanii Castellani, we used differentially expressed gene (DGE) screening by RT-PCR with 20 sets of random primers. From this analysis, we found that approximately 16 genes showed up regulation during encystation. We chose 6 genes, which had relatively higher expression levels, for further investigation. Based on homology search in database, DEG2 showed 55% of similarity with xylose isomerase, DEG9 showed 37% of similarity with Na P-type ATPase, and DEG14 showed 77% of similarity with subtilisin-like serine proteinase. DEG3 and DEG26 were identified as hypothetical proteins and DEG25 exhibited no significant similarity to any known protein. Encystation of Acanthamoeba has been suggested to be a process to resist adverse environmental or nutritional conditions. Further characterization studies of these genes may provide us with more information on the encystation mechanism of Acanthamoeba.

Real-Time Personalized Advertisement Techniques for Internet Shopping Mall (인터넷 상점에서의 실시간 개인화된 광고 제공 기법)

  • This paper describes a personalized advertisement technique as a part of intelligent customer services in Internet shopping malls. Based on customers' initial profile, purchase history, and behaviors in an Internet shopping mall, the technique displays appropriate advertisements on Internet web pages when customers' visit to the shopping mall. Customers preference scores for product groups which are main sources to select advertisements, are stored either a preference table or preference trees. Both of the two storage methods can support selection of advertisements on real time, and the preference tree method can reflect affinity among product groups. The suggested technique selects different advertisements to reflect changes of customers preferences as time goes by. An experiment has been performed to evaluate the effectiveness of the algorithm, which revealed that the algorithm selects more customer-oriented advertisements rather than random selection.

  • In this paper, a composite controller cosisted of bandpass feedback controller and LQG/LTR controller is applied to a quarter-car model moving on a randomly profiled road. The LQG/LTR controller is used to achieve a design transfer toop. A bandpass feedback controller is adopted to eliminate the response due to the disturbance, which generally can not be measured, confined within an interested frequence range. The random road profile considered as colored noise is shaped from white noise by use of shaping filter. The performance of the composite control system is compared with that of an LQG/LTR control system.

Effects of chassis flexibility and engine vibration in ride quality (차체의 턴성효과와 엔진의 진동이 승차감에 미치는 영향분석)

  • In this study, dynamic analysis of a passenger car is carried out to analyze ride quality over a random road profile. The front suspension of the car is a MacPherson strut type and the rear suspension is a multi- link type. The following five different models are constructed and compared to see the effects of engine vibration and chassis flexibility in the ride quality. (1) one rigid chassis model, (2) a rigid chassis and rigid engine model, (3) a rigid engine and flexible chassis model with one vibration mode, (4) one flexible chassis model with six engine vibration modes and one chassis vibration mode, (5) one flexible chassis model with seven vibration modes and four static correction modes. The result shows that engine vibration modes and the first bending mode of the chassis are important in the ride quality.

A Study on the Machined Surface Morphology of Laminate Composite (적층구조 복합재료의 절삭면 형상에 관한 연구)

  • Machined graphite/epoxy surfaces were studied by using SEM (Scanning Electron Microscopy), surface profilometry and its analysis to determine suitable surface describing parameters for machined unidirectional and multidirectional laminate composite. The surface roughness and profile are found to be highly depdndent on the fiber layup direction and the measurement direction. It was possible to machine 90 .deg. and -45 .deg. plies due to the adjacent plies, which were holding those plies. It was found that the microgeometrical variations in terms of roughness parameters $R_{a}$ without $D_{y}$(Maximum Damage Depth) region and $D_{y}$are better descriptors of the machined laminate composite surface than commonly used roughness parameters $R_{a}$and $R_{max}$ The characteristics of surface profiles in laminate composite are well represented in CPD (Cumulative Probability Distribution) plot and PPD (Percentage Probability Density) plot. Edge-trimmed multidirectional laminate surfaces are Gaussian and random for profiles measured along the tool movement direction, they are periodic and non-Gaussian in the direction perpendicular to the tool movement.t.ent.t.

  • Shear wave velocity (or shear modulus) is very important in the evaluation of seismic performance of a fill dam under an earthquake. A shear wave velocity profile can be determined by surface wave method such as HWAW and SASW methods but this profile has uncertainty caused by spatial variation of material property in a fill dam. This uncertainty in shear wave velocity profile could be considered using a coefficient of variation of material property in the reliability based analysis. In this paper, the possible 600 shear wave velocity profiles in the core and rockfill zone of fill dam were generated by the random shear wave velocity profile generation method, proposed by Hwang and Park, based on the field shear wave velocity profiles determined by the HWAW and SASW methods. And, through the statistical analysis of generated shear wave velocity profiles in the fill dam, the coefficient of variation (COV) of shear wave velocity with depth were evaluated for the core and rock filled zone of fill dam in Korea.

Arthrospira platensis Mutants Containing High Lipid Content by Electron Beam Irradiation and Analysis of Its Fatty Acid Composition (전자빔 조사에 의해 지질 함량이 증대된 Arthrospira platensis 변이주 분리 및 지방산 분석)

  • Arthrospira platensis (A. platensis) is an economically important microalgae because it has carbohydrates, lipids, proteins and a number of phytochemicals. It is also a valuable source used in the production of biodiesel and functional foods. In this study, A. platensis was exposed to electron beam irradation (240 kGy) and induced random mutagenesis for strain improvement. Several mutants were obtained, and the resulting mutant was designated as EB29. The growth rate and chlorophyll content of EB29 was similar to those of wild type. However, the lipid content of EB29 was increased seven-fold compared to that of wild type when comparing the nile red fluorescent intensity. Semi-quantitative analysis of EB29 using the calibration plot of standard lipid, triolein, represented $78.6{\mu}g/mL$, which increased 2 times compared to wild type ($41.4{\mu}g/mL$). When analyzing the fatty acid profile of EB29, polyunsaturated fatty acids (PUFAs), such as gamma-linolenic acid (GLA) in EB29 increased about six-fold. Moreover, fatty acids affecting the quality of biodiesel increased compared to that of wild type. Thus, electron beam could be used for the strain improvement of microalgae in order to accumulate PUFAs and alteration of fatty acid profile for biodiesel.

Estimate of the Fluctuating Pressure Distribution of Tall Building under Hazard Fluctuating Wind Load (재난변동풍하중을 받는 고층건물의 변동풍압분포의 평가)

  • In this paper, used by the boundary layer wind tunnel test, have conducted a series of wind tunnel experiments, i.e. test the mean velocity profile regarding the surface roughness, turbulence intensity and power spectrum measured by augmentation device. After that, to provide data relevant for the preliminary design step of tall building hazard fluctuating wind loads may be obtained fluctuating pressure coefficients, fluctuating pressure spectrum, autocorrelation coefficients by the boundary layer wind tunnel test. From the results of experiments, this study can be obtained conclusions as follows. 1. We know the fact that the mean velocity profile and the turbulence intensity are well fitted natural wind flow in the boundary layer wind tunnel. 2. The satisfactory agreement of velocity spectrum can be obtained from the compare of fluctuating power spectrum and Von Karman spectrum. 3. We know the fact that the fluctuating pressure spectrums distributed peak at 0.01 Hz-0.1 Hz in the windward surfaces and at 0.1 Hz in the leeward surfaces. 4. We know the fact that the autocorrelation coefficients distributed stationary random processes with application time of hazard fluctuating wind loads.

Multiscale Clustering and Profile Visualization of Malocclusion in Korean Orthodontic Patients : Cluster Analysis of Malocclusion

  • Understanding the classification of malocclusion is a crucial issue in Orthodontics. It can also help us to diagnose, treat, and understand malocclusion to establish a standard for definite class of patients. Principal component analysis (PCA) and k-means algorithms have been emerging as data analytic methods for cephalometric measurements, due to their intuitive concepts and application potentials. This study analyzed the macro- and meso-scale classification structure and feature basis vectors of 1020 (415 male, 605 female; mean age, 25 years) orthodontic patients using statistical preprocessing, PCA, random matrix theory (RMT) and k-means algorithms. RMT results show that 7 principal components (PCs) are significant standard in the extraction of features. Using k-means algorithms, 3 and 6 clusters were identified and the axes of PC1~3 were determined to be significant for patient classification. Macro-scale classification denotes skeletal Class I, II, III and PC1 means anteroposterior discrepancy of the maxilla and mandible and mandibular position. PC2 and PC3 means vertical pattern and maxillary position respectively; they played significant roles in the meso-scale classification. In conclusion, the typical patient profile (TPP) of each class showed that the data-based classification corresponds with the clinical classification of orthodontic patients. This data-based study can provide insight into the development of new diagnostic classifications.

Diversity of the Streptococcal Strains Isolated from Diseased Olive Flounder (Paralichthys olivaceus) (넙치 (Paralichthys olivaceus) 병어에서 분리된 연쇄상구균의 다양성)

  • To evaluate the biological diversity of fish pathogenic streptococci, 35 strains isolated from diseased olive flounder (Paralichtys olivaceus), were analyzed using a random amplified polymorphic DNA (RAPD) technique with the oligonucleotide commercial primer 6 (Amersham Biosciences). Api 20 Strep test, drug resistance and artificial infection were carried out for further characterization of the isolates. RAPD fingerprints showed similar pattern in 25 strains (about $71.4\%$ of 35 isolates) and these strains were designed as RA group 1. Similarities greater than $44\%$ were obtained when the Dice coefficient was applied among the isolates of RA 1. On the other hand, the reference Streptococcus iniae showed a similar RAPD profile to the isolates with similarity levels of $40-93.3\%.$ Rh I was suggested to be the dominant group isolated from olive flounder suffering from streptococcosis. However, the isolates of Rh 1 group were not classified into the same species by the Api 20 Strep identification system. There was no peculiarity in drug resistance patterns of Rh I group isolates against 7 antibacterial agents. However, only 3 of 25 isolates $(0.12\%)$ showed oxytetracycline (OTC) resistance and OTC might be a useful chemotherapeutic agent in controlling the streptococcosis by strains of RA I group in olive flounder. Fish injected intraperitoneally with $10^5$ CFU of an isolate of Rh I and RA III group showed $60\%\;and\;50\%$ accumulative mortality for 20 days, respectively ($20\%$ in control or Rh II). However luther comparative studies about differences in virulence between isolates are needed.
