This study was conducted to determine the characteristics of fertilization process and embryo development of Astragalus membranaceus Bunge (Astragali Radix) to provide basic data needed in its breeding. A. membranaceus showed poor seed setting when self-pollination was induced. When artificial pollination was induced, it showed less than 5% bearing in late August, but more than 13% bearing from the beginning of September 4th. The flower size was about $17.0\;mm{\times}4.0\;mm$ and pistils and stamens had the same length of 15.0mm at flowering stage. When self-pollination or cross-pollination was induced, pollen tubes extended to an ovule. While pollen tube was extending to the ovule, reproductive cell split and formed two male generative nuclei and a vegetative nucleus. In the case of self-pollination, fertilized embryo was not observed, but was formed in the case of cross-pollination. A. membranaceus is noted to have zygote self-incompatibility. In the case of cross-pollination, fertilization was observed in 6 to 8 h after pollination, where apical cell derivatives split after fertilization. A spherical pro-embryo was then formed three days after fertilization. The seed attained full shape with a seed coat showing its distinctive contour 15 days after fertilization. Thus, A. membranaceus in Leguminosae family is found to have zygote selfincompatibility although its flower shape is shown to match the self-compatibility plant.
Objectives : The root ofCyanchum wifordii(CW) had been used as herbal medicine, 'Baeksuo', in Korea. However, the root ofCynanchum auriculatum(CA) had also been mis-used as 'Baeksuo' in herbal markets, due to its morphological similarity with CW. This study aimed to compare discriminative features and establish the identification-keys between two herbal medicines using microscopic methods.Methods : Microscopic preparations including fixation, rapid dehydration, paraffin infiltration, paraffin embedding, sectioning, mounting, and staining, were performed and the microscopic features between CW and CA samples were compared.Results : The starch grain of parenchymal cells, the amounts of calcium oxalate crystals, cell sizes, and the shape of phloem were not different between CW and CA samples. However, the clusters of stone cells which were conglomerated at outer cortex were apparently observed in CA sample (4~8 layers of stone cells), but not in CW sample (2~4 layers of stone cells, or not). Xylem of CW consisted of several vessels and fiber bundles which were formed as linear shapes and discontinuous radial lines, while that of CA, in which radial lines was not observed, were shaped as a wedge and scattered near cambium with comparatively few fiber bundles enclosed vessels. Especially, in the xylem of CA, fiber bundles located towards cambium were relatively more developed than those located outward cambium.Conclusions : The identification-keys established in this study would be helpful to discriminate microscopic features between CW and CA.

Objectives : Ijung-tang (IJT) is a traditional herbal formula and has been used to treat digestive diseases such as abdominal pain, vomiting, and diarrhea. IJT consists of four herbal medicines, Ginseng radix, Atractylodis rhizoma alba, Zingiberis rhizoma, and Glycyrrhizae radix et rhizoma, containing various bioactive compounds. Quality assesment of IJT preparations was performed by analytical method for determining marker compounds.Methods : Determination of seven marker compounds in IJT preparations was quantitatively conducted by high-performance liquid chromatography equipped with a diode-array detector. The marker compounds were separated on a reversed-phase C18 column and the analytical method was successfully validated. Chemometric analysis was performed to compare IJT water extracts and commercial IJT granules.Results : Limit of detection and limit of quantification values were in the ranges of 0.093-2.649 μg/mL and 0.283-8.027 μg/mL, respectively. Precisions were 0.30-3.87% within a day and 0.23-2.35% over three consecutive days. Recoveries of the marker compounds ranged from 87.35-107.05%, with relative standard deviation (RSD) values < 6.15%. Repeatabilities were < 1.20% and < 1.71% of RSD value for retention time and absolute peak area, respectively. The results from quantitative analysis showed that the quantities of seven marker compounds of IJT samples varied, as were found in principal component analysis and hierarchical clustering analysis.Conclusions : The analytical method developed in the present study was precise and reliable to simultaneously determine marker compounds of IJT. Therefore, it can be used for the quality assessment of IJT preparations.
Objectives This study was carried out to find out the Antioxidant and Anti-inflammatory Effects of Components of Mahwangbujaseshin-tang in LPS-Stimulated RAW264.7 Macrophages. Methods There are 5 experimental groups. ; normal, control, EH (Ephedrae Herba), ALRP (Aconiti Lateralis Radix Preparata) and AR (Asiasari Radix). The extract of EH, ALRP and AR ($100{\mu}g/ml$) was added to each group. We examined cytotoxicity, total phenolic contents, DPPH and ABTS free radical scavenging activity, Intracellular ROS (reactive oxygen species) production, NO (Total Nitric oxide), iNOS (inducible nitric oxide synthase), PGE2 (prostaglandin E2), COX-2 (cyclooxygenase-2), $IL-1{\beta}$ ($interleukin-1{\beta}$), IL-6 (interleukin-6), $TNF-{\alpha}$ (tumor necrosis factor-${\alpha}$), MMP-9 (matrix metalloproteinase-9), TIMP-1 (tissue inhibitor of metalloproteinase-1) and HO-1 (heme oxygenase-1) expression level. Results 1. Total phenolic contents of EH were in the highest level. 2. DPPH and ABTS free radical scavenging activity of EH was in the highest level. 3. ROS production was significantly decreased in AR. 4. NO production was significantly decreased in EH, ALRP, AR and iNOS expression was decreased in EH, AR. 5. PGE2 and COX-2 expression was decreased in EH, AR. 6. $IL-1{\beta}$ production was significantly decreased in EH, AR and IL-6 production was significantly decreased in AR. $TNF-{\alpha}$ production was significantly decreased in ALRP, AR. 7. MMP-9 and TIMP-1 production were significantly decreased in EH. 8. HO-1 expression was significantly increased in EH. 9. With simultaneous usage of SnPP which is expression inhibitor of HO-1, NO, $IL-1{\beta}$, IL-6 and $TNF-{\alpha}$ production were partially increased in EH, ALRP, AR. Conclusions According to this study, Components of Mahwangbujaseshin-tang have anti-oxidants and anti-inflammation effects in LPS-Stimulated RAW264.7 Macrophages.
In order to establish utilijzation of medicinal herb as vegetable by Hadroponcis, the present studies were conducted to inverstigate the experiment on year-round Production of medicinal herb. The results obtained are summaried as follows: Among Planting date, April 5 Planting date obtained the highest yield of Scutellariae Radix and Agastache rugosa Folium. Treatment of $10\times5cm$(200 Plant/$m^2$) among Planting densities showed the highest yield of Scutellariae Radix and Agastache rugosa Folium. Plant height was thick in spacious planting, and was thin dense planting. Stem diameter was thick in spacious planting, and was thin in dense planting. Quality and yield of fresh weight ware highest in optimum planting densities($10\times5cm$ : 200 plants/$m^2$).
Methanol and/or boiling water extraction of 201 natural products and subsequent MTT assay using MT-4 cell line was carried out to screen the anti-HIV-1 activity. Among 97 methanol extracts, 7 extracts from Chrysanthemi Indicium Flos, Magnoliae Cortex, Machili Cortex, Reynoutriae Rhizoma, Lithospermi Radix, Agastachis Herba, and Chaenomelis Fructus showed anti-HIV-1 activity and their SI value were 2.25 to 5.77. In addition, among 119 boiling water extracts, 10 extracts from Lonicerae Caulis et Foloium, Elsholtziae Herba, Leonuri Herba, Portulacae Herba, Schizonepetae Herba, Curcumae Rhizoma, Amomi Cardamomi Fructus, Cirsii Radix et Herba, Carpesii Herba, and Siegesbeckiae Herba showed anti-HIV-1 activity and their SI value were 1.30 to 7.64. Methanol extracts of above seven natural products were fractionated and the anti-HIV-1 activity of each fraction was examined. Extraction was carried out with hexane, chloroform, butanol, and water to trace active anti-HIV-1 componets. As a result, the water fraction of Magnoliae Cortex, Machili Cortex, Reynoutriae Rhizoma, Agastachis Herba, Chaenomelis Fructus and the butanol fraction of Chrysanthemi Indicium Flos, Reynoutriae Rhizoma showed anti-HIV-1 activity and their SI value were 1.40 to 8.02. We could reach a conclusion that studies to trace the anti-HIV-1 active component of each natural products in further fractionation and to identify its structure by Infrared spectroscopy, NMR spectroscopy and gel permeation chromatography were needed.
Dansam, the root of Salvia miltiorrhiza BGE. (Labiatae), has a bitter and a slightly 'cold' property. and is nontoxic. It has been used for the treatment of diseases such as tumors, bruises, gynecologic diseases, menoxenia, anemia and so forth. As an oriental medicine pathway, anal therapy has many kinds of treatments, Retention enema is one the most useful, advantageous methods of anal therapy. This experiment was performed in order to study the effect of an aqueous extract of Salviae radix root(SRRAE) on Immediate type allergic reaction by Retention enema. The results were obtained as follows, 1. SRRAE inhibited compound 48/80-induced Immediate type allergic reaction 100% with the dose of 0.1g/kg by anal treatment. However, SRRAE showed no significant inhibitory effect on the same reaction by oral treatment. When mice were pretreated with SRRAE at a concentration ranging from 0.001 to 1.0g/kg, by intra-anal treatment, the serum histamine levels were reduced in a dose-dependent manner. 2. SRRAE also inhibited by 79.8% local cutaneous allergic reaction activated by anti-dinitrophenyl (DNP) IgE. 3. SRRAE close-dependently inhibited the histamine release from rat peritoneal mast cells(RPMC) by anti-DNP IgE., but SRRAE not inhibited compound 48/80-induced histamine release. 4. SRRAE dose-dependently inhibited tumor necrosis $factor-\;{\alpha}$$(TNF-\;{\alpha}$) production from RPMC by anti-DNP IgE. Moreover, the level of cAMP in RPMC, when SRRAE was added, significantly increased compared with that of a normal control. According to the above results, anal therapy(Retention enema) of SRRAE may be beneficial in the treatment of systemic and local Immediate type allergic reactions.

The traditional Asian medicinal herb, roots of Astragalus (A.) membranaceus (Leguminosae), is used for many purposes, some of which are purported to stimulate the release of growth hormone in vivo. Extracts of A. membranaceus were tested to determine whether they stimulate the release of growth hormone in rat pituitary cell culture. A. membranaceus was extracted sequentially with 80% ethanol (fraction A), n-hexane (fraction B); the test compound from the herbal extraction was isolated using silica gel column chromatography and was identified with spectral data. Test compound was also extracted by traditional boiling water methods. Induction of growth hormone in pituitary cell culture was conducted with isolated compounds and extracted fractions of A. Radix (dried roots of A. membranaceus). The fraction A was not active in the rat pituitary cell culture, but the fraction B derived from the ethanol fraction stimulated the release of growth hormone in culture. Six compounds from fraction B (1-6) were isolated and identified previously. The compounds 1,2-benzendicarboxylic acid diisononylester (1), $\beta$-sitosterol (2), and 3-Ο-$\beta$-D-galactopyranosyl-$\beta$-sitosterol (5) did not induce growth hormone release in the culture. Formononetin (3), 9Z, 12Z-octadecadienoic acid (4), stigmast-4-en-6$\beta$-o1-3-one (6) and 98-E, a mixture of 1'-9, 12-octadecadienoic acid (Z,Z)-2',3'-dihydroxy-propylester (7) and 1'-hexadecanoic acid-2',3'-dihydroxy-propylester (8) stimulated the release of growth hormone in the rat pituitary cell culture significantly compared to the control. In conclusions, four compounds isolated from extracts of A. Radix induced growth hormone release in the rat pituitary cell culture. The 98-E isolate was the most active inducer of growth hormone release.
Achyranthis Bidentatae Radix(ABR) is important prescriptions that have been used in oriental medicine for stomatitis and wound healing. The study was done to evaluate the inhibitory effects of cytotoxicity, formation of superoxide on the macrophage and neutrophil, prostaglandins($PGE_2$), interleukins($IL-1{\beta}$), collagenase activity and synthesis of collagen and DNA. The results were obtained as follows: 1. ABR was not showed the proliferation difference of human fibroblast and monocyte in 0.01% and 0.001% concentrations to be experimented and in result, it was concluded that they have no cytotoxicity but showed cytotoxicity in 0.1% concentrations. 2. ABR inhibited the formation of superoxide to 48% at the concentration of 0.001% in the mouse monocyte. 3. ABR inhibited the formation of superoxide to 40% at 0.001%, 58% at 0.0001% as compared with control in the human monocyte. 4. ABR inhibited the formation of superoxide to 58% at 0.0001%, 40% at 0.001% in the human neutrophil. 5. ABR was not showed the proliferation difference of human monocyte in all concentrations to be experimented and in result, it was concluded that they inhibited the formation of prostaglandins($PGE_2$) in the human monocyte stimulated with E. coli. 6. ABR showed the all concentration of inhibiting the production of inter1eukins($IL-1{\beta}$) in the human monocyte stimulated with E. coli. 7. ABR didn't influence on collagen synthesis and total protein in fibroblasts. 8. ABR inhibited the collagenase activity to 84% at 0.1%, 69% at 0.2%, 76% at 0.5%, 91% at 0.001% respectively.
Objective : To observe Sagunjatanghapchangchuljiyutang in rectifying the digestive dysfunctional spleen deficiency syndrome caused by chronic diarrhea. Methods : Sagunjatanghapchangchuljiyutang was chosen to treat the digestive dysfunctional spleen deficiency syndrome caused by chronic diarrhea. The model was induced by Rhei Radix and castor oil. The effects on the gastric secretion ability(gastrin, secretin, volume of gastric juice, free acidity, total acidity, pepsin activity) and nutritional condition(protein, albumin, total cholesterol, triglyceride, total lipid, BUN, creatinine, RBC, WBC, hemoglobin, hematocrit) were observed in vivo. Results : The group induced chronic diarrhea by Rhei Radix and castor oil had lower gastric secretion ability and nutritional condition was lower than that of the normal group. The figures for group given Sagunjatanghapchangchuljiyutang showed improvement, especially gastrin, total lipid and hematocrit. Conclusion : Sagunjatanghapchangchuljiyutang have an effect on the digestive dysfunctional spleen deficiency syndrome caused by chronic diarrhea. And experimental study syndrome could be proceed.
