  • It is important to know the accurate radionuclide inventory of radioactive waste for the reliable management. However, estimation of radionuclide concentrations in drummed radioactive waste is difficult and unreliable because of difficulties of direct detection, high cost, and radiation exposure of sampling personnel. In order to overcome these difficulties, scaling factors (SFs) have been used to assess the activities of radionuclides that could not be directly analyzed. A radionuclide assay system has been operated at KORI site since 1996 and consolidated scaling factor method has played a dominant role in determination of radionuclides concentrations. However, some problems are still remained such as uncertainty of estimated scaling factor values, inaccuracy of analyzed sample values, and disparity between the actual and ideal correlation pairs and the others. Therefore, it needs to improve the accuracy of scaling factor values. The scope of this paper is focused on the improvement of accuracy and representativeness of calculated scaling factor values based on statistical techniques. For the selection of reliable activity determination method, the accuracy of estimated SF values for each activity determination method is compared. From the comparison of each activity determination methods, it is recommended that SF determination method should be changed from the arithmetic mean to the geometrical mean for more reliable estimation of radionuclide activity. Arithmetic mean method and geometric mean method are compared based on the data set in KORI system.

  • Regulations and guidelines for radioactive waste disposal require detailed information about the characteristics of radioactive waste drums prior to transport to the disposal sites. However, estimation of radionuclide concentrations in the drummed radioactive waste is difficult and unreliable. In order to overcome this difficulty, scaling factor (SF) method has been used to assess the activities of radionuclides, which could not be directly analyzed. A radioactive waste assay system has been operated at Korean nuclear power plant (KORI site) since 1996 and consolidated SF concept has played a dominant role in the determination of radionuclide concentrations. However, SFs are somewhat dispersive and limited in KORI site. Therefore establishment of the assay system using more improved SFs is planned and progressed. In this paper, the scope of research is briefly introduced. For the selection of more reliable activity determination method, the accuracy of predicted SF values for each activity determination method is compared. From the comparison of each activity determination method, it is recommended that SF determination method should be changed from the arithmetic mean to the geometrical mean for more reliable estimation of radionuclide activity. Arithmetic mean method and geometric mean method are compared based on the data set in KORI system. And, this change of SF determination method will prevent an inordinate over-estimation of radionuclide inventory in radwaste drum.

  • KHNP(Korea Hydro and Nuclear Power Ltd., Co.) is developing a HIC transport package which is satisfying domestic and IAEA regulations and NETEC(Nuclear Environment Technology Institute) is conducting a conceptual design. In this study, the shielding thickness was calculated using the data from radionuclide assay program which is currently using in nuclear sites and Micro Shield code. Considering the structural safety, carbon steel was chosen as shielding material and the shielding thickness was calculated for 500 R/hr and 100 R/hr at HIC surface, respectively. Through the shielding analysis, it was evaluated that the regulation limit is satisfied when the shielding thickness is 22 cm for 500 R/hr and 17 cm for 100/hr.

  • Intakes of radionuclides through both inhalation and ingestion pathways may occur particularly in an incident involving unsealed radionuclides. If one assume only one intake path in this case, which is usual in routine monitoring, a significant error in the evaluated committed effective dose($E_{50}$) may result. In order to demonstrate the potential errors, variations of the resulting committed effective doses were analyzed for different fractions of the inhaled activities to the total intake of $^{241}Am$. Simulated bioassav measurements for the lungs, urine and feces were generated based on the biokinetic model and data of the radionuclide, 5 ${\mu}m$ AMAD and absorption type M for inhalation, for various inhalation fractions. The potential errors in $E_{50}$ due to the assumption of one intake path were in the range from -100% to as large as +34,000% when the bioassays were made 3 days after the intakes. Larger errors are expected when only the feces assay is applied while inhalation intake exists. A strategy which employs two types of bioassay was proposed to reduce the error caused by a misjudgement of the intake path.

  • In order to compare the correlation of radioactivity ratio between the radwaste streams and the primary coolant of PWR NPPs, A RCS sampling kit was installed to primary coolant system for the collection of the radionuclides during the normal operation of NPPs. RCS samples were collected from PWR type of domestic NPPs through 2004 to 2005, and pretreated with acid microwave digestion or leaching method to assay quantitatively of several interesting radionuclides. The radioactivity ratios of $^{137}Cs\;to\;^{60}Co$ in a filter cartridge and a resin cartridge were 2.3E-2 and 7.3E-1, respectively. At a same period of the reactor operating cycle, the radioactivity ratios of $^{137}Cs\;to\;^{60}Co$ were 6.3E-1 for a evaporator bottom, 6.7E-1 for a spent resin, and 5.6E-2 for a dry active waste, so that these radwaste streams were identified as having similar characteristics with the corresponding RCS samples.

  • Purpose: Cancer specific killing can be achieved by therapeutic gene activated by cancer specific promotor. Expression of sodium iodide symporter (NIS) gene causes transportation and concentration of iodide into the cell, therefore radioiodine treatment after NIS gene transfer to cancer cell could be a form of radionuclide gene therapy. luciferase (Luc) gene transfected cancer cell can be monitored by in vivo optical imaging after D-luciferin injection. Aims of the study are to make vector with both therapeutic NIS gene driven by AFP promoter and reporter Luc gene driven by CMV promoter, to perform hepatocellular carcinoma specific radiodiodine gene therapy by the vector, and assessment of the therapy effect by optical imaging using luciferase expression. Materials and Methods: A Vector with AFP promoter driven NIS gene and CMV promoter driven Luc gene (AFP-NIS-CMV-Luc) was constructed. Liver cancer cell (HepG2, Huh-7) and non liver cancer cell (HCT-15) were transfected with the vector using liposome. Expression of the NIS gene at mRNA level was elucidated by RT-PCR. Radioiodide uptake, perchlorate blockade, and washout tests were performed and bioluminescence also measured by luminometer in these cells. In vitro clonogenic assay with 1-131 was performed. In vivo nuclear imaging was obtained with gamma camera after 1-131 intraperitoneal injection. Results: A Vector with AFP-NIS-CMV-Luc was constructed and successfully transfected into HepG2, Huh-7 and HCT-15 cells. HepG2 and Huh-7 cells with AFP-NIS-CMV-Luc gene showed higher iodide uptake than non transfected cells and the higher iodide uptake was totally blocked by addition of perchlorate. HCT-15 cell did not showed any change of iodide uptake by the gene transfection. Transfected cells had higher light output than control cells. In vitro clonogenic assay, transfected HepG2 and Huh-7 cells showed lower colony count than non transfected HepG2 and Huh-7 cells, but transfected HCT-15 cell did not showed any difference than non transfected HCT-15 cell. Number of Huh-7 cells with AFP-NIS-CMV-Luc gene transfection was positively correlated with radioidine accumulation and luciferase activity. In vivo nuclear imaging with 1-131 was successful in AFP-NIS-CMV-Luc gene transfected Huh-7 cell xenograft on nude mouse. Conclusion: A Vector with AFP promoter driven NIS and CMV promoter driven Luc gene was constructed. Transfection of the vector showed liver cancer cell specific enhancement of 1-131 cytotoxicity by AFP promoter, and the effect of the radioiodine therapy can be successfully assessed by non-invasive luminescence measurement.
