Activation Mechanism of Protein Kinase B by DNA-dependent Protein Kinase Involved in the DNA Repair System

  • DNA-dependent protein kinase(DNA-PK) is involved in joining DNA double-strand breaks induced by ionizing radiation or V(D)J recombination and is activated by DNA ends and composed of a DNA binding subunit, Ku, and a catalytic subunit, DNA-PKcs. It has been suggested that DNA-PK might be $2^{nd}$ upstream kinase for protein kinase B(PKB). In this report, we showed that Ser473 phosphorylation in the hydrophobic-motif of PKB is blocked in DNA-PK knockout mouse embryonic fibroblast cells(MEFs) following insulin stimulation, while there is no effect on Ser473 phosphorylation in DNA-PK wild type MEF cells. The observation is further confirmed in human glioblastoma cells expressing a mutant form of DNA-PK(M059J) and a wild-type of DNA-PK(M059K), indicating that DNA-PK is indeed important for PKB activation. Furthermore, the treatment of cells with doxorubicin, DNA-damage inducing agent, leads to PKB phosphorylation on Ser473 in control MEF cells while there is no response in DNA-PK knockout MEF cells. Together, these results proposed that DNA-PK has a potential role in insulin signaling as well as DNA-repair signaling pathway.

Therapeutic Results of Radiotherapy in Nonsmall Cell Lung Cancers (비소세포성 폐암의 방사선치료 성적)

  • Total 55 patients with nonsmall cell lung cancer treated with radiation therapy at Department of Therapeutic Radiology, Yeungnam University Hospital, between May-1 1986 and April-30 1993 were retrospectively analyzed by clinical characteristics, failure patterns, follow up duration and survival ratio according to prognostic factors. Obtained results were as follows : 1. Male to female ratio was 17.3 2. Sixth and seventh decades were predominant age group. 3. The patients were 8 in stage I-II, 34 in stage IIIA, 13 in stage IIIb, respectively. 4. Forty five patients out of 55 were squamous cell carcinoma. 5. Primary tumor were originated from upper lobe bronchi predominantly. 6. The size of the primary tumor, lymph node involvement and the degree of differentiation were important in evaluation of prognosis. 7. In conclusion, for patients with poor prognostic factors systemic chemotherapy and multidisciplinary approach were recommended for better treatment outcome and improvement of survival.

Surgical Approaches to the Middle Cranial Base Tumors (중두개와저 종양에 대한 수술적 치료)

  • Objective : We analysed various surgical approaches and surgical results of 28 middle cranial base tumors for the purpose of selecting optimal surgical approach to the middle cranial base tumor. Methods : In this retrospective review, 28 patients, including 16 meningioma, 6 trigeminal neurinoma, 2 pituitary adenoma, 2 craniopharyngioma, 1 facial neurinoma, and 1 metastatic tumor, underwent surgical treatment using skull base technique. Of theses, 16 tumors were mainly confined to middle cranial fossae, 5 tumors with extension into both anterior and middle fossa, and 7 tumors with extension into both middle and posterior fossa. Tumors that confined to the middle cranial fossa or extended into the anterior cranial fossa were operated with modified pterional, orbitozygomatic or Dolen'c approach, and tumors that extended into the posterior cranial fossa were operated with anterior, posterior or combined transpetrosal approach. Completeness of tumor resection, surgical outcome, postoperative complication, and follow up result were studied. Results : Total tumor removal was achieved in 9 tumors of 10 tumors that did not extended to the cavernous sinus, and was achieved in 7 tumors of 8 tumors that extended to the lateral wall of the cavernous sinus. Of 10 tumors that extended to the venous channel of the cavernous sinus, only 2 were removed totally. Surgical outcome was excellent in 14 patients, good in 10, fair in 2 and poor in 2. There were no death in this series. Dumbell type tumor which extended into both middle and posterior fossae showed tendency of poor prognosis as compared with tumors that confined middle cranial fossa and extended into both anterior and middle cranial fossa. Postoperative dysfunctions were trieminal hypesthesia in 3, oculomotor nerve palsy in 2, abducens nerve palsy in 2, hemiparesis in 2, cerebellar sign in 1, facial palsy in 1 and hearing impairment in 1. Conclusion : Based on our findings and a review of the literature, we conclude that, when selecting the surgical approach to the middle cranial fossa tumors, the most important factors to be considered were exact location of the tumor mass and existence of the cavernous sinus invasion by tumor mass. We recommend modified pterional or orbitozygomatic approach in cases with tumors located anterior and middle cranial base, without cavernous sinus invasion. In cases with tumors invading into cavernous sinus, we recommend Dolen'c or orbitozygomatic approach. And in lateral wall mass and the cavernous sinus, it is preferred to approach the tumor extradurally. For the tumor involing with middle fossa and posterior fossa(dumbell type) a combined petrosal approach is necessary. In cases with cavernous sinus invasion and internal carotid artery encasement, we recommend subtotal resection of the tumor and radiation therapy to prevent permanent postoperative sequele.

The Study on Concentration of Serum Selenium in Patients with Hepatoma (간암환자의 혈청내 셀레늄 농도에 관한 연구)

  • Serum concentration of selenium were measured ; Studying 36 patients with Hepatoma. 19 with Hepatitis 15 with Liver cirrhosis and 67 Normal control to discover the correlation between serum selenum and cancer. And it have tested also the serum protein and albumin to compare the nutrition status by researching the same persons above The results of those two experiments are as follow : 1) The mean of each group of patients resulting from mearsuing the serum protein content showing 6.91$\pm$0.10g/dl in the patients with hepatoma 7.05$\pm$0.17g/dl in the ones with hepatitis 6.42$\pm$0.13g/dl in the ones with liver cirrhosis and 7.46$\pm$0.06g/dl in the normal control is all being within the normal range such as 6.0-0.8(g/dl) while the distribution is revealin 10.5%, 13.3% in the patients with hepatitis and liver cirrhosis which are below the normal range and 47.2%, 96.9%, 80.0% in the low range of normal one ; therefore in could see that patientsw compared with normal control are in low level in the serum protein content 2) The mean based on the consquence of serum albumin measurement is 3.74$\pm$0.11g/dl. 4.26$\pm$0.13g/dl 4.73$\pm$0.06g/dl in the patients with hepatoma hepatits and normal control which are in between the normal range such as 3.5-5.5(g/dl) except the patients with liver cirrhosis(3/05$\pm$0.12g/dl) while the distribution of the patients with hepatoma an liver cirrhosis is showing 27.8% 73.3% which are below the normal one and 55.5%, 52.6% and 26.7% in the patients with hepatoma hepatits and liver cirrhosis which are in the low area of normal range ; as a result It could find that patients. compared with the normal control are in low level in serum albumin measurement. 3) The mean consulting the serum concentration is 430.86$\pm$26.06$\mu\textrm{g}$/1, 192.37$\pm$18.15$\mu\textrm{g}$/1 186.73$\pm$23.99$\mu\textrm{g}$/1, 196.94$\pm$8.94$\mu\textrm{g}$/1 in the patients with hepatoma hepatitis liver cirrhosis and noramal control ; it shows 2.19 times increasing of serum selenium concentra-tion in the patients with hepatoma 'compared with the normal control and patients with hepatoma spread out at a still higher range in serum selenium concentration test. 4) The serum selenium concentration is not quite dependent on sexes and doesn't show any correlation with age, serum protein, serum albumin. The nutrition status of patients, rested on the results of those experiments, stays in low level, which is caused by that they are in low stage of serum protein and serum albu¬min. In serum selenium concentration, patients with hepatitis and liver cirrhosis, specially, show the condition as good as normal control's; while patients with hepatoma are indicating a significant increasing compared with the three groups, which can be reasoned into that there is a possibility for the patients to take selenium and chemotherapy or radiation treatment, and have high serum selenium concentration by poor excreting of seleno methionine, which is caused by low serum protein content And another thing can be reasoned is that the difference in origination mechanism hetween korean and western people. As a result, It showed that the correlation between serum selenium and seses, age, serum protein, and serum albumin neither exists nor affects on each other.

Self-Efficacy, Self-Care Behavior, Posttraumatic Growth, and Quality of Life in Patients with Cancer according to Disease Characteristics (질병관련 특성에 따른 암환자의 자기효능감, 자가간호행위, 외상 후 성장, 삶의 질)

  • Purpose: This study examined self-efficacy, self-care behavior, posttraumatic growth, and quality of life in cancer patients and their levels by disease characteristics groups to identify patient groups that require psychosocial intervention. Methods: We surveyed 107 patients using a structured questionnaire about the four factors and analyzed the factors by stratifying the patients by the period after the cancer diagnosis, by stage and by current treatment status. Results: The mean score for self-efficacy was 37.78, and that for self-care behavior 49.96. Patients who were diagnosed less than one year ago scored higher on medication, a sub-category of self-care behavior, than the post-diagnosis period of 1~2 year group. The score was higher in the currently-treated group than the follow-up and distant metastasis groups. For posttraumatic growth, the mean was 56.17, and the factor was higher in the 1~2 year post-diagnosis group after than the less than one year group. The score was higher in the follow-up group than the currently-treated group. With regard to quality of life, the mean score was 25.79, and no significant correlation was found with disease characteristics. Conclusion: A shorter post-diagnosis period increased self-care behavior, and the greatest posttraumatic growth was reported by the 1~2 year post-diagnosis group. It may be necessary to provide cancer patients with an education program and other strategies less than one year after the diagnosis to improve self-efficacy and self-care behavior. To promote post-traumatic growth, it may be helpful to provide patients with psychosocial intervention within two years after the diagnosis.

Optimization of Hybrid Process of(Chemical Coagulation, Fenton Oxidation and Ceramic Membrane Filtration) for the Treatment of Reactive Dye Solutions (반응성 염료폐수 처리를 위한 화학응집, 펜톤산화, 세라믹 분리막 복합공정의 최적화)

  • This study investigated the effects of hybrid process(chemical coagulation, Fenton oxidation and ceramic UF(ultrafiltration)) on COD and color removals of commercial reactive dyestuffs. In the case of chemical coagulation, the optimal concentrations of $Fe^{3+}$ coagulant for COD and color removals of RB49(reactive blue 49) and RY84(reactive yellow 84) were determined according to the different coagulant dose at the optimal pH. They were 2.78 mM(pH 7) in RB49 and 1.85 mM(pH 6) in RY84, respectively. In the case of Fenton oxidation, the optimal concentrations of $Fe^{3+}\;and\;H_2O_2$ were obtained. Optimal $[Fe^{2+}]:[H_2O_2]$ molar ratio of COD and color removals of RB49 and RY84 were 4.41:5.73 mM and 1.15:0.81 mM, respectively. In the case of ceramic UF, the flux and rejection of supernatant after Fenton oxidation were investigated. After ceramic UF for 9 hr, the average flux of RB49 and RY84 solutions were $53.4L/m^2hr\;and\;67.4L/m^2hr$ at 1 bar, respectively. In addition, the permeate flux increased and the average flux recovery were 98.5-99.9%(RB49) and 91.0-97.3%(RY84) according to adopting off-line cleaning(5% $H_2SO_4$). Finally, COD and color removals were 91.6-95.7% and 99.8% by hybrid process, respectively.

Gamma Irradiation Effects on Conchospores of Porphyra Species 2. The Effects of High Gamma Irradiation on Germination and Growth of Conchospores of Two Varieties (김의 각포자에 대한 r-선의 조사효과 2. 두 품종의 각포자의 발아생장에 미치는 고선량 r-선의 조사효과)

  • For the fundamental studies of radiation breeding in edible marine algae, the biological effects on conchospores of Porphyra species by gamma-irradiation were examined. Two varieties, Keun-cham-gim (Porphyra tenera Kjell. form tamatsuensis Miura) and Saga No.5, were chosen for this study, and their conchospores after r-irradiation($5.0{\sim}20.0$ KR) were cultured for 50 days. The results obtained were summarized as follows. 1. Gamma-irradiation in less than the dose of 20KR did not affect germination of conchospores, and almost all spores grew into two cells germ in 24 hours after irradiation, but withering germs were gradually increased in number according to higher exposure within 5 days old culture. 2. High irradiation caused the induction of giant cells, abnormal useless growth of hold-fast, lumpish thalli and callus-like lumpy tissues. 3. The liberation of neutral spores from young germs and carpospores from mature thalli were observed on the frond exposed at $10{\sim}20$ KR irradiation. All spores were normal in division and its size. 4. The best irradiation effect on growth of Keun-cham-gim was observed at 10 KR dose, whose growth-rates were $140\%$ in wet weight and $108\%$ in mean frond area, but only $48\%$ was recorded in wet weight at 20 KR exposure. Saga No.5 were in contrast with Keun-cham-gim, and their most growth-rate was $400\%$ in wet weight ($258\%$ in frond area) at 10 KR irradiation and the worst was $20\%$ at the dose of 20 KR. 5. The withering phenomenon to death by treatment of gamma-ray presented substantial difference between two varieties. Survival rate compared with control in Keun-cham-gim was $70.7\%$ at 20 KR, but that in Saga No.5 recorded $47.0\%$ at same dose. 6. Synthesizing the results of high and low r-irradiation, it was suggested tat high r-irradiation in more than 5.0 KR inhibited conspicuously the growth of germs derived from conchospores, and about half of them withered at 15.0 KR dose or more.

The Prognosis following Radiation Therapy or Surgical Resection for Stage 1 Non-Small Cell Lung Cancer (방사선치료 혹은 외과적 절제술을 받은 임상적 제 1기 비소세포폐암의 치료성적)

  • Background: Surgical resection is the treatment of choice for localized, operable non-small cell carcinoma of the lung. Curative radiotherapy, however, is considered an alternative to surgery in patients with poor performance state, poor cardiopulmonary function, or who refuse surgery. However, the difference in prognosis after surgery and radiotherapy is not well established in the patients with stage I non-small cell lung cancer. Method: To evaluate the difference in progonsis between surgery and radiotherapy in stage I non-small cell lung cancer, a retrospective study was done with 15 patients treated with curative radiotherapy and 24 patients treated with curative surgery. Results: The overall response rate was 80%, with 33% complete response, after radiotherapy. The median survival time of the patients with radiotherapy was 14.9 months, with 2-year and 5-year survival rates of 22% and 0%, respectively. The median survival time of the patients with surgery was 37.7months, with 2-year and 5-year survival rates of 65% and 41%, respectively. Conclusion: These results suggest that surgery is better than the radiotherpy in view of survival rate and it is necessary to recommend, more strongly, curative surgery to patients with stage I non-small cell lung cancer if the patients are able to receive operation. To compare, more accurately, the difference in prognosis by the modality of therapy, large multicenter study is needed.

Changes in Ion Balance and Individual Ionic Contributions to EC Reading at Different Renewal Intervals of Nutrient Solution under EC-based Nutrient Control in Closed-loop Soilless Culture for Sweet Peppers (Capsicum annum L. 'Fiesta') (EC 기준 파프리카 순환식 수경재배에서 양액 교체 주기에 따른 양액 중의 이온 균형 및 각 이온의 EC 기여도 변화)

  • Individual ion concentrations and ionic contributions to EC reading in the circulated nutrient solution are the important factors to be considered for stable EC-based closed-loop soilless culture. This study was conducted to determine appropriate ion-analysis intervals of the circulated nutrient solutions based on ion concentration, ion balance, and ion electrical conductivity under different renewal intervals in EC-based nutrient control systems for sweet peppers (Capsicum annum L. 'Fiesta') in early growth stage. Average node numbers of the plants were 13 and 18 when the experiment started and finished, respectively, and three plants were grown in each rockwool slab. Four different renewal intervals of circulated nutrient solutions such as 1, 2, 3, and 4 weeks were used as treatment. Nutrient solutions were supplied to the plants based on integrated radiation. Drainage was collected into drain tanks after irrigation ended in the day and then mixed with fresh water until the EC reaches 2.69 $dS{\cdot}m^{-1}$. The replenished nutrient solution was supplied to the plants in the next day. Ion concentrations of the individual ions periodically analyzed in the circulated nutrient solutions showed no significant differences among the treatments during the experimental period. Ion concentrations of $K^+$, $Ca^{2+}$, $Mg^{2+}$, $Na^+$, $NO_3{^-}$, ${SO_4}^{2-}$, ${PO_4}^{3-}$, and $Cl^-$ varied within 5-8, 11-14, 2.0-2.7, 0.5-0.6, 14-19, 4-5, 1-4, and 0.3-0.5 $meq{\cdot}L^{-1}$, respectively. Ion balance showed a consistent tendency over all the treatments and especially $K^+$ : $Ca^{2+}$ and ${SO_4}^{2-}$ : ${PO_4}^{3-}$ played great roles in the cation and anion balances in the nutrient solutions, respectively. Activity coefficients of ions such as $K^+$, $NO_3{^-}$, and $H_2PO_4{^-}$ varied within 0.8-0.9 and those of $Ca^{2+}$, $Mg^{2+}$, ${SO_4}^{2-}$ varied within 0.5-0.6, showing little changes with time. Ionic contributions of $K^+$ and $NO_3{^-}$ to EC reading were the greatest followed by $Ca^{2+}$, ${SO_4}^{2-}$, and $Mg^{2+}$ in the order. From the results, we thought that allowable ranges in ion concentration, ion balance, and subsequent individual ionic contributions to EC reading would be obtained within 4-week renewal interval of nutrient solution in EC-based closed-loop soilless culture for sweet pepper plants.

Analysis of Literatures Related to Crop Growth and Yield of Onion and Garlic Using Text-mining Approaches for Develop Productivity Prediction Models (양파·마늘 생산성 예측 모델 개발을 위한 텍스트마이닝 기법 활용 생육 및 수량 관련 문헌 분석)

  • Growth and yield of field vegetable crops would be affected by climate conditions, which cause a relatively large fluctuation in crop production and consumer price over years. The yield prediction system for these crops would support decision-making on policies to manage supply and demands. The objectives of this study were to compile literatures related to onion and garlic and to perform data-mining analysis, which would shed lights on the development of crop models for these major field vegetable crops in Korea. The literatures on crop growth and yield were collected from the databases operated by Research Information Sharing Service, National Science & Technology Information Service and SCOPUS. The keywords were chosen to retrieve research outcomes related to crop growth and yield of onion and garlic. These literatures were analyzed using text mining approaches including word cloud and semantic networks. It was found that the number of publications was considerably less for the field vegetable crops compared with rice. Still, specific patterns between previous research outcomes were identified using the text mining methods. For example, climate change and remote sensing were major topics of interest for growth and yield of onion and garlic. The impact of temperature and irrigation on crop growth was also assessed in the previous studies. It was also found that yield of onion and garlic would be affected by both environment and crop management conditions including sowing time, variety, seed treatment method, irrigation interval, fertilization amount and fertilizer composition. For meteorological conditions, temperature, precipitation, solar radiation and humidity were found to be the major factors in the literatures. These indicate that crop models need to take into account both environmental and crop management practices for reliable prediction of crop yield.
