감마선 조사에 의한 수산 자숙액의 생리활성에 대한 연구 (Study on the Physiological Activities of Gamma-irradiated Seafood Cooking Drips)

  • Cooking drips which were obtained as by-product after seafood processing in the food industries, still contain lots of proteins, carbohydrates, and other functional materials. This study was conducted to investigate the effect of gamma irradiation on the biological activities of seafood cooking drips. When the cooking drips of Hizikia fusiformis, Enteroctopus dofleini and Thunnus thynnus were irradiated, the antioxidant activities, whitening effect, and angiotensin I converting enzyme inhibition activity of the ethanol extract from seafood cooking drips were all increased by gamma irradiation. This was because of the increased extraction efficiency of available compounds by irradiation. These results suggested that the seafood cooking drips, wasted as by-products, can be used as functional compounds with gamma irradiation treatment.

감마선 조사 냉동비빔밥의 미생물 및 관능적 품질특성 (Microbiological and Sensory Characteristics of Gamma-irradiated Frozen Bibimbap)

  • This study was conducted to evaluate the effect of gamma irradiation on the microbiological and sensory characteristics of frozen Bibimbap. Frozen Bibimbap vegetables were contaminated by microbial levels of $2.70{\sim}6.67log\;CFU\;g^{-1}$ in total aerobic bacteria. Frozen Bibimbap was contaminated by microbial levels of $4.8log\;CFU\;g^{-1}$ in total aerobic bacteria, but microorganisms in gamma-irradiated samples at above 25 kGy were not found at detection limit less then $1log\;CFUg^{-1}$. Sensory characteristics of the gamma-irradiated Bibimbap were decreased depending upon the irradiation dose, and sensory scores of the samples irradiated with doses higher than 10 kGy were significantly decreased. Therefore, the application of combination of the various food processing technology should be considered for the sterilization of Bibimbap, without any deterioration of quality occurred by high dose irradiation.

고선양(高線量) 방사선(放射線)이 백혈구(白血球) 수명(壽命)에 미치는 영향(影響) (Effects of High Dose Irradiation on The Leukocyte Life Span)

  • As a part of studies on acute effects of high dose irradiation the present report was carried out to evaluate the changes of the leukocyte life span in the Newzealand white male rabbits by a single whole body exposure to gamma rays from $^{60}Co$ teleirradiation unit. The exposure was done in dose levels of 100, 300, 550 and 1,000 rad to each experimental group of 10 rabbits. The life span and apparent half survival time of leukocytes, and the elution rate of leukocytes in the circulating blood were measured by McMillan method using $^{51}Cr$. 1. As a critical indicator of radiation hazards of the Newzealand male rabbits, the LD 50/30 and LD 100/30 after whole body exposure was estimated as 550 and 1,000 rads respectively. 2. The life span of leukocyte in the circulation after irradiation was slightly shortened in the 100 rad irradiated group, as compared with the unirradiated control group, but markedly shortened in the 300, 550 and 1,000 rads irradiated group. 3. After irradiation, decrease of leukocyte half survival time in the circulation showed the same pattern as that of leukocytes life span. 4. As the irradiation doses increased, the elution rate of $^{51}Cr$ loss from $^{51}Cr$ tagged leukocytes in the circulation were markedly increased gradually. 5. The life span shortening of leukocytes in the circulation after irradiation seems to occure by two processes of senescence acceleration and early destruction.

감마선을 조사한 시판 도토리묵의 저장 중 품질 특성 (Quality Characteristics of Gamma Irradiated Commercial Arcon Starch Gel during Storage)

  • The physical properties and sensory characteristics of acorn starch gels (Dotori Mook), which were gamma-irradiated up to 3 kGy, were evaluated during storage at $4^{\circ}C$. Even at the dose of 2 kGy, the gamma irradiation decreased total bacteria in the Dotori Mook during 5 days of storage, to lower than the detection limit $(10^2CFU/g)$. The hardness of the control sample increased according to the days of storage, while the gamma irradiated samples had decreased hardness according to the irradiation dose. The sample irradiated at 3 kGy maintained the same hardness as the control at day 0 of storage. Irradiation did not affect the Hunter color values. No significant differences were observed in off-odor, color, springiness, and overall acceptability (p<0.05) at the irradiation dose of 2 kGy. It can be concluded that the irradiation of Dotori Mook, up to 2 kGy, does not affect the quality of the Mook during storage, with regard to texture and sensory characteristics. Moreover, the irradiated Mook was superior in maintaining hardness and had prolonged shelf-life time by sanitation.

Synergistic effect of ionizing radiation and $\beta$-lapachone against tumor in vitro and in vivo

  • ${\beta}$-lapachone(${\beta}$-Lap), a natural o-naphthoquinone, presents in the bark of the Lapacho tree. ${\beta}$-Lap is cytotoxic against a variety of human cancer cells and it potentiates the anti-tumor effect of Taxol. In addition, ${\beta}$-Lap has been reported to radiosensitize cancer cells by inhibiting the repair of radiation-induced DNA damage.In the present study, we investigated the cytotoxicity of ${\beta}$-Lap against RKO human colorectal cancer cells as well as the combined effect of ${\beta}$-LaP and ionizing radiation. An incubation of RKO cells with 5 ${\mu}$M of ${\beta}$-Lap for 4 h killed almost 90% of the clonogenic cells. An incubation of RKO cells with 5 ${\mu}$M of ${\beta}$-Lap for 4 h or longer also caused massive apoptosis. Unlike other cytotoxic agents, ${\beta}$-Lap did not increase the expression of p53 and p21 and it suppressed the NFkB expression. The expression of Caspase 9 and 3 was minimally altered by ${\beta}$-Lap. Radiation and ${\beta}$-Lap acted synergistically in inducing clonogenic cell death and apoptosis in RKO cells when ${\beta}$-Lap treatment was applied after but not before the radiation exposure of the cells. Interestingly, a 4 h treatment with 5 ${\mu}$M of ${\beta}$-Lap starting 5 h after irradiation was as effective as that starting immediately after irradiation. The mechanisms of ${\beta}$-Lap-induced cell killing is controversial but a recent hypothesis is that ${\beta}$-Lap is activated by NAD(P)H: quinone-onidoreductase (NQO1) in the cells followed by an elevation of cytosolic Ca$\^$2+/ level and activation of proteases leading to apoptosis. It has been reported that NQO1 level in cells is markedly up-regulated for longer than 10 h after irradiation. Indeed, using immunological staining of NQO1, we observed a significant elevation of NQO1 expression in RKO cells 5h after 2-4 Gy irradiation. Such a prolonged elevation of NQO1 level after irradiation may be the reasons why the ${\beta}$-Lap treatment applied S h after irradiation was as effective as that applied immediately after irradiation in killing the cells. In view of the fact that the repair of radiation-induced damage is usually completed within 1-2 h after irradiation, it is highly likely that the ${\beta}$-Lap treahment applied 5 h after irradiation could not inhibit the repair of radiation-induced damage. For in vivo study, RKO cells were injected S.C. into the hind-leg of Nu/Nu mice, and allowed to grow to 130 mm3 tumor. The mice were i.p. injected with ${\beta}$-lapachone or saline 2 h after irradiation of tumors with 10 Gy of X-rays. The radiation induced growth delay was increased by 2.4 $\mu\textrm{g}$/g of ${\beta}$-lapachone. Taken together, we may conclude that the synergistic interaction of radiation and ${\beta}$-Lap in killing cancer cells is not due to radiosensitization by ${\beta}$-Lap but to an enhancement of ${\beta}$-Lap cytotoxicity by radiation through an upregulation of NQO1. The fact that NQO1 is elevated in tumors and that radiation causes prolonged increase of the NQO1 expression may be exploited to preferentially kill tumor cells using ${\beta}$-Lap in combination with radiotherapy.

고선량 감마선 조사가 즉석 취식용 양념 돼지 갈비 구이의 저장성 및 지질 산화에 미치는 영향 (Effects of High Dose Gamma Irradiation on Shelf Stability and Lipid Oxidation of Marinated and Precooked Pork Rib Steak)

  • 본 연구는 사막이나 우주 등 극한 환경에서도 취식이 가능한 즉석 양념 돼지 갈비 구이의 저장 안정성 확보를 위한 고선량 조사 적용을 평가하기 위해 실시하였다. 이를 위해 즉석 양념 돼지 갈비 구이를 제조하여 진공 포장한 후 10, 20, 30, 40 및 50 kGy로 조사하고 $35^{\circ}C$에서 가속 저장하면서 미생물 생육과 지질 산패도를 평가하였다. 가열처리에 의해 약 3 Log CFU/g의 미생물 감소 효과를 얻을 수 있었으나, 완전 사멸시키지는 못했다. 감마선 조사에 의해 저장 초기 미생물 생육은 관찰되지 않았으나, 저장 기간이 경과함에 따라 10, 20, 30 kGy조사구에서 저장4일, 7일, 14일에 미생물 생육이 관찰되었다. 오염 미생물의 최대성장률 예측 결과에서 비조사구, 10, 20, 30 kGy 감마선 조사구의 최대 성장률이 1.13, 0.60, 0.52, 0.18 Log CFU/g/day로 나타나, 감마선 조사에 의한 초기 미생물 감소가 저장성 증진에 크게 효과적인 것으로 판단되었다. 그러나 가속 저장 중 malondialdehyde 함량은 저장 기간이 경과함에 따라 모든 처리구에서 증가하였고, 특히 감마선 조사에 의해 심한 지질 산화가 발생되는 것으로 나타났다. 따라서, 고선량 조사 적용시 품질 변화 없이도 육가공품의 완전멸균을 위해서는 다양한 식품 가공 방법과의 병용 처리 연구가 수행되어야 할 것이다.

Irradiation Detection in Korean Traditional Soybean-Based Fermented Powdered Sauces: Data for Establishing a Database for Regulation of Irradiated Foods

  • To facilitate establishing regulations for irradiated foods, Korean traditional soybean-based fermented powdered doenjang (PD), kanjang (PK), kochujang (PKC) and chungkukjang (PC) were irradiated at 1, 3, 5 and 7 kGy, and subjected to irradiation detection analyses as part of establishing a database for detecting irradiated foods. Photostimulated luminescence (PSL) and electron spin resonance (ESR) were applied as the detection methods. Using PSL analysis, the irradiated PD, PK and PKC could be easily distinguished from the non-irradiated ones, while irradiation of the PC at 5 kGy or higher was detectable. The ESR spectra of the irradiated PD, PK and PKC exhibited symmetrical multiplet lines, which might be induced from the crystalline sugar, whereas, the PC showed a single signal at the paramagnetic centers. The signal intensity increased with incremental increases of irradiation doses distinguishing the irradiated samples from the control. In addition, the peak height also revealed that irradiation induced an increment in the intensity of single and/or multiplet lines of the ESR signals, resulting in clear confirmation of irradiation. Thus, the data from this study could be used as references for detecting irradiated soybean-based fermented powdered sauces.

방사선 조사 마우스에서 학습기억 장애에 대한 홍삼의 효과 (Effect of Red Ginseng on Radiation-induced Learning and Memory Impairment in Mouse)

  • Previous studies suggest that even low-dose irradiation can lead to progressive cognitive decline and memory deficits, which implicates, in part, hippocampal dysfunction in both humans and experimental animals. In this study, whether red ginseng (RG) could attenuate memory impairment was investigated through a passive-avoidance and object recognition memory test, as well as the suppression of hippocampal neurogenesis, using the TUNEL assay and immunohistochemical detection with markers of neurogenesis (Ki-67 and doublecortin (DCX)) in adult mice treated with a relatively low-dose exposure to gamma radiation (0.5 or 2.0 Gy). RG was administered intraperitonially at a dosage of 50 mg/kg of body weight, at 36 and 12 h pre-irradiation and at 30 minutes post-irradiation, or orally at a dosage of 250 mg! kg of body weight/day for seven days before autopsy. In the passive-avoidance and object recognition memory test, the mice that were trained for one day after acute irradiation (2 Gy) showed significant memory deficits compared with the sham controls. The number of TUNEL-positive apoptotic nuclei in the dentate gyrus (DG) was increased 12 h after irradiation. In addition, the number of Ki-67- and DCX-positive cells was significantly decreased. RG treatment prior to irradiation attenuated the memory defect and blocked apoptotic death as well as a decrease in the Ki-67- and DCX-positive cells. RG may attenuate memory defect in a relatively low-dose exposure to radiation in adult mice, possibly by inhibiting the detrimental effect of irradiation on hippocampal neurogenesis.

Identification of Gamma Irradiation of Imported Spice

  • Photostimulated Luminescence (PSL), Electron Spin Resonance (ESR) and Thermoluminescence (TL) analysis were conducted to detect irradiation treatment of imported whole and ground spices. The screening by PSL detected no irradiation treatment, except un the ground thyme and bay leaves which exhibited photon counts in the intermediate level. Irradiation of the two spices was detected after irradiating them at 1.0, 3.0, 5.0 and 10.0 kGy, and then subjecting them to PSL analysis, which resulted in the significantly low photons of non-irradiated spices compared to that at 1.0 kGy, indicating that the photon counts varied depending on the amount of inorganic mineral debris in the spices. To confirm a successful detection by using PSL, ESR and TL methods, some spices were selected, irradiated at 5.0 and 10.0 kGy, and subjected to the detection methods. PSL identified the irradiated spices except the cassia, which showed very weak PSL sensitivity, but was identified by ESR analysis. Also, the ESR and TL exhibited the typical signals induced by irradiation treatment and were able to successfully detect all of the irradiated spices. In addition, we found a positive correlation between the intensity of ESR and TL signals and irradiation doses.

5-Fluorouracil 투여가 마우스 공장 소낭선세포의 방사선조사 효과에 미치는 영향 (The Effect of Combination of Radiation with 5-Fluorouracil on Mouse Jejunal Crypt Cells)

  • 방사선조사와 5-Fluorouracil(5-FU)과의 병용시 5-FU투여로 인한 마우스 공장 소낭선세포의 방사선 감수성에 미치는 영향과 방사선조사 효과 증강율을 측정하기 위하여 $C_3H$마우스 110마리를 대상으로 동물실험용 세시움 방사선 조사기를 이용하였다. 방사선조사 단독시행군은 $1,000{\sim}1,600rad$를, 5-FU와 병용군은 $800{\sim}1,400rad$의 방사선조사와 복강내 5-FU투여를 병용하였다. 방사선조사 단독시행 군은 조사 후 90시간 후에, 병용요법군은 120시간 후에 마우스 공장을 횡절단하여 마우스공장 소낭선 측정 법을 이용하여 평군치사선량과, 5-FU주입이 공장 소낭선세포 생존케 미치는 dose effect factor(DEF)를 측정하였으며, 결과는 다음과 같다. 1, 방사선조사 단독시행 군, 방사선조사 분전 5-FU주입군, 방사선조사 6시간 후에 5-FU주입군의 평균치사선량(Do)은 각각 135, 135, 114rad였다. 2. 방사선조사 단독시행 군에 비하여 방사선조사 15분전 5-FU주입군과, 방사선조사 6시간 후에 5-FU주입군의 DEF는 각각 1.13, 1.27이였다.
