Effect of quercetin on impaired immune function in mice exposed to irradiation

  • Radiation used in cancer treatment may cause side effects such as inflammation. Quercetin is a polyphenol that reduces inflammation. This study evaluated the recovery efficacy of quercetin on impaired immune function in irradiation-induced inflammatory mice. Quercetin administered at two concentrations of 10 and 40 mg/kg body weight was initiated 2 weeks before irradiation and was continued 30 days after irradiation. The animals exposed/not exposed to radiation were sacrificed on radiation days 10 and 30. Splenocyte proliferation, which was diminished after irradiation, was enhanced significantly by quercetin supplementation after 30 days of irradiation. Cytokine secretion increased in the radiation group compared to that in the non-radiation control group. After 30 days of radiation, interleukin (IL)-$1{\beta}$ and IL-6 secretion decreased significantly in the radiation-quercetin groups. When quercetin was administered for 44 days, it showed a possible protective effect against irradiation-induced inflammation in mice. Quercetin could be beneficial in the recovery of irradiation-induced increases in cytokine secretion.

Development of Freeze-dried Bibimbap, Korean Cooked Rice with Red Pepper Paste, as a Space Food Sterilized by Irradiation (동결건조 비빔밥을 이용한 방사선 멸균 우주식품 개발)

  • This study evaluated the microbial population (total aerobic count, fungi) and organoleptic quality of freeze-dried bibimbap sterilized by gamma irradiation to develop the space food. An irradiation dose above 30 kGy was needed to sterilize the freeze-dried bibimbap, while organoleptic quality of the sample was significantly decreased by irradiation (p<0.05). However, it was observed that scores on overall acceptance of rehydrated bibimbap after irradiation were the highest, when vitamin C and paprika extract were added at 0.1%, respectively. Therefore, it was considered that the freeze-dried bibimbap could be developed as a space food, which meet microbial requirements and organoleptic quality through addition of vitamin C 0.1% and paprika extract 0.1% before gamma irradiation at 30 kGy.

Effects of ${\gamma}$-Irradiation on Immunological Activities of ${\beta}$-Glucan

  • This study evaluated the effects of $\gamma$-irradiation on immunomodulating properties and structural changes of ${\beta}$-glucan. ${\beta}$-Glucan solutions (10 mg/mL) were ${\gamma}$-irradiated at 10, 30, and 50 kGy. Splenocyte proliferation and cytokine (interferon-${\gamma}$ and interlukin-2) productions by ${\gamma}$-irradiated ${\beta}$-glucan were evaluated in in vivo and in vitro, and structural changes of ${\beta}$-glucan were also determined after ${\gamma}$-irradiation. ${\gamma}$-Irradiation on ${\beta}$-glucan at 50 kGy enhanced splenocyte proliferation and cytokine productions, (p<0.05) and cleft glycosidic bonds of ${\beta}$-glucan resulting in lower the molecular weight. These results indicate that the use of ${\gamma}$-irradiation on ${\beta}$-glucan may be useful for improving its immunological activity by lowering the molecular weight of ${\beta}$-glucan.

Protective Effect of Silkworm Hemolymph against Gamma Irradiation Induced Damage in the Liver of Mice (방사선 손상 Balb/C 마우스 모델에서 누에 체액(Silkworm Hemolymph)의 간조직 보호 효과)

  • This study was designed to evaluate the protective effect of silkworm hemolymph against ${\gamma}-irradiation$ induced damage in the liver of mice. Female Balb/C mice (6 weeks old) were exposed to ${\gamma}-irradiation$ (6 Gy) and administered orally to silkworm hemolymph ($5ml\;kg^{-1}$ BW) for 7 days post-irradiation. The body weight, spleen index, plasma aspartate transaminase (AST), plasma alanine transaminase (ALT), and liver malondialdehyde (MDA) levels were determined. Compared with irradiated control mice, the activity of plasma AST and the level of MDA were significantly decreased in mice treated silkworm hemolymph. These results show that silkworm hemolymph is found to have a protective effect against ${\gamma}-irradiation$ induced damage in mice.

Thoracic Irradiation Recruit M2 Macrophage into the Lung, Leading to Pneumonitis and Pulmonary Fibrosis

  • Background: Radiation-induced pneumonitis and pulmonary fibrosis are common dose-limiting complications in patients receiving radiotherapy for lung, breast, and lymphoid cancers. In this study, we investigated the characteristics of effective immune cells related to pneumonitis and fibrosis after irradiation. Materials and Methods: After anesthesia, the whole thorax of C57BL/6 mice was irradiated at 14 Gy. The lung tissue and bronchoalveolar lavage fluid were collected at defined time points post-irradiation for the determination of histological and immunohistochemical analysis and inflammatory cell population infiltrated into the lung. Results and Discussion: Whole thoracic irradiation increased the deposition of extracellular matrix (ECM), lung weight, and pleural effusions, which started to die from 4 months later. At 4 months after irradiation, the numbers of macrophages and lymphocytes as well as neutrophils were increased dramatically in the lung. Interestingly, the macrophages that were recruited into the lung after irradiation had an enlarged foamy morphology. In addition, the expressions of chemokines (CCL-2, CCL-3, CXCL-10) for the attraction of macrophages and T cells were higher in the lung of irradiated mice. The high expressions of these chemokines were sustained up to 6 months following irradiation. In thoracic irradiated mice, infiltrated macrophages into the lung had the high levels of Mac-3 antigens on their surface and upregulated the hallmarks of alternatively activated macrophages such as arginase-1 and CD206. Furthermore, the levels of IL-4 and IL-13 were higher in a BAL fluid of irradiated mice. Conclusion: All results show that thoracic irradiation induces to infiltrate various inflammation-related immune cells, especially alternatively activated macrophages, through enhancing the expression of chemokines, suggesting that alternatively activated macrophages are most likely important for leading to pulmonary fibrosis.

Effect of Gamma Irradiation on the Structural and Physiological Properties of Silk Fibroin

  • This study was conducted to examine the changes in the molecular structure and physiological activities of silk fibroin by gamma irradiation. The results of gel permeation chromatography and sodium dodecyl sulfate-polyacrylamide gel electrophoresis showed that the molecular weight of fibroin was increased depending upon the irradiation dose. Secondary structure of fibroin determined by using circular dichroism revealed that the ratio of $\alpha$-helix was increased up to 10 kGy and then decreased depending upon the irradiation dose. Whereas, the ratio of $\beta$-sheet, $\beta$-turn, and random coil were decreased and then increased with an alteration in the $\alpha$-helix secondary conformation. The 2.2-diphenyl-1-picryl-hydrazil (DPPH) radical scavenging activity of fibroin was increased by gamma irradiation at 5 kGy, but was decreased above 10 kGy depending upon the irradiation dose. Also, the inhibition activities of tyrosinase and melanin synthesis of fibroin were increased by gamma irradiation. These results indicated that gamma irradiation could be used as an efficient method to make fibroin more suitable for the development of functional foods and cosmetics.

Differential Expression of C4H and F5H Genes in Rice (Oryza sativa L.) after Gamma-irradiation

  • To reveal effects of gamma-irradiation with various doses on the expressions of C4H and F5H genes, the transcription levels of OsC4HL and OsF5HL were investigated in leaves and stems of two rice cultivars, Ilpoombyeo and IR-29, after the irradiation with 5, 10, 50, or 100 Gy for 4 h. In overall pattern of 24 h after the irradiation, the transcription levels of the two genes increased with the increasing doses of radiation in the leaves of both cultivars, except that of OsC4HL in IR-29. However, in the stems, the transcription level of OsF5HL increased in Ilpoombyeo and decreased in IR-29 dose-dependently, while that of OsC4HL decreased in Ilpoombyeo with the increasing doses of radiation and remained constant in IR-29. When the expressions of OsC4HL and OsF5HL were investigated in a time-course after the irradiation with 100 Gy, they reached their highest levels in the leaves of both cultivars 5 hand 72 h after the irradiation, respectively. Therefore, we suggest that the expressions of OsC4HL and OsF5HL, which involved in the same phenylpropanoid pathway, are differentially regulated during the post-irradiation period, showing different cultivar and tissue specificity. Furthermore, the dose dependency of the gene expressions is also discussed immediately after the irradiation.

Telomere-Mitochondrion Links Contribute to Induction of Senescence in MCF-7 Cells after Carbon-Ion Irradiation

  • The effects of carbon-ion irradiation on cancer cell telomere function have not been comprehensively studied. In our previous report cancer cells with telomere dysfunction were more sensitive to carbon-ion irradiation, but the underlying mechanisms remained unclear. Here we found that telomerase activity was suppressed by carbon-ion irradiation via hTERT down-regulation. Inhibition of telomere activity by MST-312 further increased cancer cell radiosensitivity to carbon-ion radiation. hTERT suppression caused by either carbon-ion irradiation or MST-312 impaired mitochondrial function, as indicated by decreased membrane potential, mtDNA copy number, mitochondrial mass, total ATP levels and elevated reactive oxygen species (ROS). PGC-$1{\alpha}$ expression was repressed after carbion-ion irradiation, and hTERT inhibition by MST-312 could further exacerbate this effect. Lowering the mitochondrial ROS level by MitoTEMPO could partially counteract the induction of cellular senescence induced by carbon-ion radiation and MST-312 incubation. Taken together, the current data suggest that telomere-mitochondrion links play a role in the induction of senescence in MCF-7 cells after carbon-ion irradiation.

Physiological Responses of Rice by Acute and Chronic Gamma Irradiation (방사선 완·급조사에 따른 벼의 생리적 영향 평가)

  • We investigated the physiological responses by acute and chronic gamma-irradiation in rice. The rice, Oryza sativa L. cv Dongan, plants were irradiated with 100 and 400 Gy of gamma-rays for acute and chronic irradiation, and their morphological, chlorophyll content, MDA, proline and activities of antioxidant enzymes were examined. The plant height of chronic irradiation samples were decrease with increase of a does than the control, but the number of tiller and dry weight of shoot were increased 100 Gy. Carotenoid and chrolorphyll content were decreased of all irradiated plants than non-irradiated plants. But, MDA and proline content were increased in 400 Gy both acute and chronic gamma irradiation. And the activities of antioxidant enzymes were different as gamma-irradiation patterns.

Antibacterial Effect of PVP/PEG/Carrageenan/Silver Acetate Hydrogels by γ-ray (γ-ray를 이용하여 합성한 PVP/PEG/Carrageenan/Silver acetate 하이드로겔의 항균효과)

  • In recent day, there is much interest in the biocidal activity of silver since silver is known to be safe and effective as disinifectant and biocidal material against coliforms and viruses. In this study, hydrogels containing silver acetate as antibacterial agent have been prepared using gamma rays irradiation. The hydrogels are composed of poly(vinyl pyrrolidone) (PVP), poly(ethylene glycol) (PEG), carrageenan and silver acetate. The concentration of solution was 9 wt%. The ratio of PVP : PEG : carrageenan was 6 : 1 : 2. The concentration of the silver acetate were 0, 0.01, 0.03, 0.05 and 0.07% and Gamma irradiation dose was 25 kGy. The Gamma irradiation dose in hydrogels with 0.01% silver acetate were 20 kGy, 35 kGy, 50 kGy, 65 kGy, and 80 kGy. The results showed that 0.01% silver acetate concentration of hydrogels by 25 kGy irradiation dose showed the highest antibacterial activity against E. coli and Staphylococcus aureus. Moreover, antibacterial activity of various Gamma irradiation dose in hydrogels treated 0.01% silver acetate showed highest 35 kGy irradiation dose against Staphylococcus aureus.
