가토 모델에서 인체지방유래 줄기세포가 슬관절의 퇴행성 변화에 미치는 영향 (The Effects of Human Adipose Tissue-derived Stem Cells on Degenerative Change of Knee in Rabbit Model)

  • Purpose: The survival of bone marrow derived stem cell was reported several times. But the survival of adipose tissue derived stem cells(hASCs) was not mentioned on. We studied the adipose tissue derived stem cell's survival and effect on articular cartilage in rabbits. Methods: Osteoarthritis was induced in twenty New Zealand white rabbits by intraarticular injection of monosodium iodoacetate(MIA). After four weeks, hASCs were also injected into the knee joints space without any vehicle, but the control group received phosphate buffered saline only. The histologic grade of articular cartilage was measured in 4 and 8 weeks after the transplantation of hASC and the viability of injected stem cells measured by Fluorescent in situ Hybridization (FISH) examination. Results: After 4 and 8 weeks from hASCs transplantation, histologic grade was not significantly difference between two groups(p>0.05), and the Y chromosome of the transplanted hASCs was not detected in articular cartilage. Conclusion: We found that direct injection of hASC in joint space didn't work on damaged articular cartilage repair.

Tetrodotoxin 중독가토(中毒家兎)의 심전도학적(心電圖學的) 연구(硏究) (An Electrocardiographic Study on Tetrodotoxin Intoxicated Rabbits)

  • Tetrodotoxin (TTX) is the purified active principle responsible for tetrodon (Puffer-fish) poisoning which has long been known in the Orient. The pharmacological actions of TTX have been rather extensively investigated. Two of the most prominent effects of intravenousely administered TTX are severe hypotension and respiratory paralysis resulting from its depressant actions on tissues. This depressant actions of TTX in turn result from the selective inhibition of sodium-carrying mechanism which is essential to generation of the action potential. TTX differs from local anesthetics in that it does not affect potassium conductance. Although the mechanism of the hypotensive action of TTX remains a subject of controversy, most investigator agree that TTX-induced hypotension is caused by alteration in the blood vessels rather than the heart. Not only the study on the effects of TTX on cardiac function is meager but the results of reported works are often contradictory. The present study was undertaken to investigate the effect of TTX on the electrocardiogram of the rabbit and to compare them with well known electrocardiographical characteristics found in digitalis and quinidine intoxicated animals. The results obtained from the present study are summarized as follows. 1. No changes were found in P-R interval and QRS duration after i.v. administration of $1.0\;{\mu}g/kg\;to\;1.5\;{\mu}g/kg$ TTX to the animals. It is obvious that there were no conduction disturbance between atria and ventricles as well as in the ventricular tissue. 2. In $1.0\;{\mu}g/kg$ TTX group, S-T interval and T-P segment were not changed whereas marked changes were observed in $1.5\;{\mu}g/kg$ TTX group. 3. The first and second degree A-V blocks appeared in the $2.0\;{\mu}g/kg$ TTX group. 4. TTX differs from digitalis and quinidine in that it does not cause S-T interval depression and T-wave inversion. In contrast with digitalis, TTX caused Q-T interval prolongation.

HSP90에 대한 단일클론항체의 생성 및 특성 조사 (Production and Characterization of Monoclonal Antibodies against the 90-kDa Heat Shock Protein)

  • 90 kDa-heat shock protein (HSP90)은 여러 가지 스트레스에 의해서 유도되는 주요 스트레스단백질 가운데 하나이다. HSP90은 스트레스가 없는 정상적인 상황에서도 일정량 합성되는 단백질로, 세포 내에서 kinases나 스테로이드호르몬 수용체와 같은 여러 조절단백질 및 구조 단백질과 결합하는 것으로 알려져 있다.본 연구에서는 HSP90의 생화학적인 특성을 조사하는데 사용할 목적으로DEAE- cellulose chromatography, hydroxyapatite chromatography, Sephacryl S-300 gel filtration의 방법으로 닭근육조직으로부터 HSP90을 순수 분리하고, 이에 대한 단일클론항체를 murine hybridoma technique을 이용하여 생성하였다. 생성된 클론들에 대해서 ELISA와 Western blot을 실시한 결과 HSP90을 인지하는 A204, C112, C302, C410의 4가지 단클론을 얻었다. 특히 C112는 Western blot과 native immunoprecipitation 실험에서 인간, 토끼, 닭, 쥐, 어류의 HSP90을 인지하지만 초파리, E. coli의 HSP90은 인지하지 못하는 것으로 나타났다.

가공식품 중 새우의 검출을 위한 샌드위치 ELISA의 개발 (Development of Sandwich ELISA for the Detection of Shrimp in Processed Foods)

  • 가공식품 중 새우의 검출을 위한 샌드위치 ELISA(sELISA)의 조건을 확립하기 위해 홍다리 새우의 tropomyosin에 대한 특이항체를 이용하여 하였다. 이때, 새우의 검출범위는 1-100 ppm (${\mu}g/mL$)이며, 검출한계는 0.3 ppm이었다. 특이항체의 교차반응 결과, 홍다리새우, 흰다리 새우, 칵테일 새우, 바다가재, 꽃게에 대한 반응성은 각각 100, 73, 155, 18, 0%를 나타내었으며 새우에 대한 특이성이 매우 높았다. 열처리한 시료와 항체간의 반응성은 $100^{\circ}C$까지는 121-221%로 안정하였으나 $121^{\circ}C$에서는 반응성이 급격히 감소하였다(7.8%). 크림스프, 이유식, 소시지, 어묵, 소스에 대한 spike test에서 새우의 분석회수율은 각각 397, 639, 168, 234, 0%로 소스를 제외하고 매우 높게 나타났다. sELISA에 의하여 14점의 시판시료 중 새우의 함유 유무를 조사한 결과, 정성적으로 원료의 표시사항과 일치하는 비율은 79%이었다.

종 특이 프라이머를 이용한 동물성 식품원료의 진위 판별법 개발 (Development of Species-Specific PCR to Determine the Animal Raw Material)

  • 본 연구에서는 식품 중 동물성 사용원료의 진위 판별을 위하여 분자생물학적 기법을 이용한 시험법을 개발하였다. 동물성 식품원료의 종 판별을 위한 유전자로는 미토콘드리아 DNA에 존재하는 COI, Cytb, 및 16S rRNA 유전자를 대상으로 하였으며, 가공식품에 적용하기 위하여 PCR 산물의 크기는 200 bp 내외가 되도록 종 특이 프라이머를 설계하였다. 대상종으로는 가축류 2종, 가금류 6종, 민물어류 2종, 해양어류 13종 및 갑각류 1종, 총 24종을 선정하였으며 종 특이 프라이머를 이용하여 예상되는 PCR 산물의 생성 유무를 확인하였다. PCR을 수행한 결과 토끼, 여우, 꿩, 집비둘기, 멧비둘기, 메추리, 참새, 제비, 메기, 쏘가리, 날치, 열빙어, 청어, 까나리, 멸치, 참조기, 넙치, 조피볼락, 홍어, 가오리, 말쥐치, 농어, 성게 및 바닷가재에 대하여 각각 156, 204, 152, 160, 113, 163, 167, 152, 165, 121, 136, 151, 178, 178, 146, 188, 177, 166, 179, 218, 188, 185, 127 및 172 bp에서 PCR 증폭 산물을 확인하였다. 그리고 프라이머 별로 비교종에서는 비특이적 PCR 산물(non-specific PCR product)은 생성되지 않았다. 본 연구에서 개발된 유전자 분석법을 이용하여 동물성 식품원료가 사용된 식품 원료 및 가공식품의 진위 판별에 활용이 가능할 것이며, 불량식품 근절에 크게 기여할 것으로 기대된다.

Catecholamines에 관(關)하여 -제삼편(第三編) 심장(心臟) Catecholamines에 관(關)한 실험적(實驗的) 연구(硏究)- (Experimental Studies on Cardiac Catecholamine Content)

  • During the last decade extensive studios on catecholamines have evolved new knowledge in the physiology and biochemistry of adrenergic mechanism. Cardiac muscle, receiving adrenergic fibres from the stellate, cervical and thoracic ganglia, has been repeatedly shown to have a specific capacity to uptake and to store catecholamines. The catecholamine stores in cardiac muscle have also been shown to be important sites for the action of numerous drugs. Under normal condition, a certain level of catecholamines is maintained in the stores and serves as the basis for studying the changes in the catecholamine content of the heart. Because myocardial catecholamines play such important role in the patho-physiology of the heart, it would be interesting to compare the normal level of myocardial catecholamines among various species of animals. An occasional study has dealt with myocardial catecholamines of several species add ages of animals but these have been insufficiently comprehensive to afford a basis for an understanding of the importance of these amines as related to species and ages. The present investigation was undertaken to determine whether or not there is any significance of myocardial catecholamines in the course of the evolution and development of animals. Seasonal changes, sex difference and regional and subcellular distribution of myocardial catecholamines were also examined. The concentration of cardiac catecholamines was determined by the spectrophotofluorometric procedure described by Shore and Olin. The results obtained were summarized as follows: 1. As animals phylogenetically progressed larger amounts of catecholamines were resent in their hearts. A negligibly small amount of catecholamine was present in the hearts of the clam, a non-vertebrate. Among the vertebrates, cold-blooded animals (snake, turtle, frog, eel and fish) had less myocardial catecholamines than warm-blooded animals, of which aves (fowl and duck) had less than mammalia (cat, dog, rabbit, rat, cow and pig). The ratio of norepinephrine to epinephrine also was greater as the animals progress phylogenetically. 2. Examination of the regional distribution of cardiac catecholamines in warm-blooded animals showed that the content of the auricle was generally higher than that of the septum and considerably than that of the ventricle, but the differences of contents among these regions were not so marked. 3. In the embryonic chick, cardiac catecholamines were firstly detected on the 4th day of incubation, the time before the cardiac innervation of sympathetic nerves. The concentrations of these catecholamines increased but not markedly on the 6th day of incubation, soon after the innervation of sympathetic nerves to the heart. The level of the cardiac catecholamines fluctuated throughout the remainder of embryonic development. 4. In newborn rat hearts, a considerable amount of catecholamines was present. With the development of the rats, the concentrations of myocardial catecholamines increased. The ratio of epinephrine and norepinephrine fluctuated within the range of 40 to 60 pervent. However, as development progressed, the percentage of norepinephrine continued to rise, attaining the adult value of $80{\sim}90%$ after $45{\sim}60$ days. In contrast, the total amount of epinephrine remained fairly constant throughout the animal's development. 5. No significant sexual differences were observed in the concentration of myocardial catecholamines in the developing rat. 6. The catecholamines in the rabbit hearts increased during the summer season (from May to August) and maintained a fairly constant level in the other seasons of the year. 7. The subcellular distribution of cardiac catecholamines was examined by differential centrifugation of homogenates of cardiac muscles in rabbits, cats and rats. The catecholamines were found to be present approximately 20% in particles of mitochondrial fraction, 45% in particles of microsomal fraction and 35% in soluble supernatant fraction. The particle containing catecholamines in cardiac muscle appears to be two different sizes.

제주지역(濟州地域)의 식품금기(食品禁忌)에 관(關)한 연구(硏究) (I)-임신기(妊娠期)를 중심(中心)으로- (A Study of Food Taboos on Jeju Island (I)-Focused on Pregnancy-)

  • Nutrition counselors in Korea often encounter difficulty in their attempt to change village women's attitudes regarding food taboos which are counter to good eating habits. There are a great many food superstitions which are not due to religious influence, but seem to be related to shape and composition of food. Many expectant mothers superstitiously avoid eating certain foods for fear that they may cause mental or physical abnormality in their babies. As was shown in a previous survey (Mo, 1966)of villages in all provinces except Jeju Island, such superstitions were common among pregnant and lactating mothers. Many food taboos and superstitions based on non-scientific and irrational ideas do exist even in modern society, and are a major obstacle to nutritionally adequate food consumption. A study of food taboos among women of Jeju Island was undertaken from November to December of 1976, these results to be compared as well with those of the previous study. There were 73 items found to be prohibited during pregnancy. Of these, 48.7% were of the deaf group, 17.4% fish, 5.5% eggs, 4.7% cereal, and only 2.2% fruit. Of 252% women respondents, 111 (45% ) abstained from eating chicken, duck, and shark because of the belief that they would cause their babies to be born with gooseflesh or shark skin. Many of them avoided rabbit meat for fear that their babies might be born with harelip. It was also feared that a baby would become disfigured if his mother ate duck, goat, dog meat, chicken or duck eggs, or soup made of bones. A common superstition was that highly spiced or salty foods would cause the fetus to be hairless. Squid and octopus were believed to cause babies to have weak bones, or none at all. Most of these food taboos were associated with fears concerning Physical structure and appearance of unborn babies. Other taboos were associated with fear of undesirable behavioral characteristics. For example, some mothers thought that a baby would pinch or bite the mother's breast during the weaning period, if crab meat were eaten during pregnancy. Unevenly sliced rice cake, loach, snake meat and eel were also believed to cause a baby to be ill-tempered. The findings of this study are remarkably similar to those of the previous study conducted by the authour in 1966. Most of the same food taboos, based on non-scientific and irrational reasons, were found on Jeju Island as on the peninsula, and thor were similarly wide-spread. The results of correlational analysis show that the most significant factors related to prevalence of food taboos, are level of education and religious background. Number of food taboos is correlated with level of education. Also, food taboos are least freqent among the Christian woman. Proper nutrition education should he undertaken in order to encourage intake of protein-rich food, particularly during pregnancy when nutritional needs of mother and fetus are great.

경상북도 안강 행주 기씨 묘 출토 직금단 치마와 저고리 보존처리 - 직금단 편금사의 금박 재접착을 중심으로 - (Conservation of Golden Decorative(Jikgeumdan) Jeogori and Chima of Costumes Excavated from Hangju Gi' Tomb of Angang, Kyungsangbukdo Province - Re-adhesion of Gold Leaves in Gilt Paper Strips -)

  • The purpose of this research is to conserve of golden decoration(Jikgeumdan) of Chima(skirt) and Jeogori(Korean jacket) of the costumes excavated from Hangju Gi' tomb(17th Century) of Joseon dynasty$(1392{\sim}1910)$ and to focus on the development of conservation skill to prevent flakings of gold leaves in gilt paper strips. Up to the present, in case of golden decorations of costumes excavated from tombs of Joseon dynasty, some of gold leaves in gilt paper strips of costumes were flaked away by deterioration of adhesives in tombs. However, most of gold leaves were flaked away and totally lost by wet cleaning for eliminating contaminants after excavation. In order to prevent flaking, preliminary experiments for re-adhesion of gold leaves have been carried out. Firstly, simulation was performed using gold leaf which was available in the market. Adhesives used in this research were water-soluble adhesives(hide glue(cow, rabbit), glue made from air bladders of sciaenoid fish and Primal AC-3444 of acrylic emulsion) and solvent-soluble adhesives(acrylic adhesive Paraloid B-67 and B-72). Because of difficulty in wetting and spreading of adhesive solutions into the interface between gold leaf and Korean paper, water-soluble adhesives were not proper for adhesion of gold leaf and Korean paper. Solvent-soluble adhesives were easily infiltrated into interface between gold leaf and Korean paper and the adhesive force was also satisfied. From this result, the researchers chose more flexible Paraloid B-72$(Tg\;40^{\circ}C)$ 1% solution than Paraloid B-67$(Tg\;50^{\circ}C)$ for adhesion of gold leaf and Korean paper. Secondly, using Paraloid B-72 1% solution, the estimations of re-adhesions of gold leaves in gilt paper strips of Jeogori were carried out. When Paraloid B-72 1% solution was injected three times into the interface between gold leaf and Korean paper, the re-adhesion was most effective. On the basis of the results in these preliminary experiments, re-adhesions of gold leaves in gilt paper strips of Chima and Jeogori were carried out on condition of three times injections of Paraloid B-72 1% solution before wet cleaning. After wet cleaning, the most of the gold leaves were survived, which was confirmed by both the examination with the naked eye and the microscopic examination.

제주도 한림 연안 정치망 어장의 환경특성과 어획량 변동에 관한 연구 III. 어획량변동과 환경요인 (Environmental Character and Catch Fluctuation of Set Net Ground in the Coastal Water of Hanlim in Cheju Island III. Environmental Character and Catch Fluctuation)

  • 제주도 서부 연안역인 한림 정치망어장의 해황과 어획량 변동과의 관계를 파악하기 위해 1995년$\~$1996년에 실시한 정선 및 정점조사의 수온 및 염분자료, 시계열분석, 단기변동, 해수유동상황과 1994년$\~$1996년의 한림 정치망의 어획량 변동에 관해 검토가 분석한 결과를 요약하면 다음과 같다. 1) 한림 정치망에 가장 많이 잡히는 어종은 전갱이 ($69.2\%$)이고 그 다음으로 고등어 ($18.4\%$), 갈치 ($5.6\%$), 오징어($2.7\%$), 독가시치 ($1.4\%$)의 순이며. 조업기간은 5월에서 12월까지로 6월과 10월에 어획량의 피크가 있고 이중 10월에 년중 최고 어획량이 나타난다. 2) 한림 정치망에서는 밀물인 북류성분과 썰물인 남류성분이 명료히 나타나 해수의 연직혼합이 원활하고 남류의 지속기간(3시간 이상)이 길어 외측의 중$\cdot$저층수가 강하게 영향을 미쳐 어장까지 공급되어, 일별 평균수온이 낮고 염분 상승내지는 고염분수가 출현할 때 좋은 어획량을 보이는 경향이 있다. 또 대조기인 망이나 삭보다 소조기인 상$\cdot$하현에 어획량이 많g으며, 남$\~$남동풍이 3$\~$6.5m/sec정도 볼 때 호어가 나타날 가능성이 높다.

가공식품 중 고등어의 검출을 위한 ELISA의 개발 (Development of Sandwich ELISA for the Detection of Mackerel in Processed Foods)

  • 기존에 생선 검출을 위한 ELISA 개발은 여러 편 보고된 바 있으나, 고등어만을 특이적으로 검출하는 방법에 관한 보고는 거의 없었다. 본 연구에서는 고등어 parvalbumin에 대한 토끼 항체를 이용하여 가공식품 중 고등어의 검출을 위한 ELISA를 개발 하였다. 암모늄침전법과 Sephadex G-50 column chromatography를 이용하여 열에 안정한 12 kDa의 고등어 parvalbumin을 분리, 정제하였다. 여기에서 얻어진 parvalbumin을 토끼에게 면역하여 생산한 항 parvalbumin 항체 및 특이항체-HRP 복합물을 이용하여 샌드위치 ELISA(sELISA)의 조건을 확립하였다. 이때 parvalbumin 의 검출한계는 3 ng/mL이고, 고등어(생살)의 검출범위는 5-5,000 ${mu}g/mL$이었다. 이어서. 광어, 우럭, 오징어, 꽃게, 꽁치, 대구, 새우, 연어, 바다가재, 고등어에 대한 교차반응을 조사한 결과 고등어에 대한 반응성은 월등히 높았으나 다른 수산식품에서는 반응성이 거의 없거나 매우 낮게 나타나 고등어에 대한 특이성이 매우 높은 항체임을 알 수 있었다. 또한 분석시 시료의 열 안정성은 $100^{\circ}C$까지 양호하였으며 크림스프, 이유식, 소시지, 소스에 대한 spike test(0.01-0.3%)에서 분석회수율은 각각 104, 101, 54, 0%로 나타났다. sELISA에 의하여 16점의 시판시료 중 고등어의 함유 유무를 조사한 결과, 정성적으로 표시와 일치하는 것은 75%이었다. 그러므로 본 연구에서 개발한 sELISA는 일부 한계가 있기는 하지만, 가공식품 중 고등어를 정성적으로 검출하는데는 효과적으로 활용할 수 있을 것이다.
