RV-23, a Melittin-Related Peptide with Cell-Selective Antibacterial Activity and High Hemocompatibility

  • RV-23 is a melittin-related antibacterial peptide (MRP) with lower cytotoxicity than either melittin or AR-23, another MRP. The aim of this study was to explore the mechanism of RV-23's antibacterial selectivity and its hemocompatibility. The results showed that all the peptides exhibited lytic activity against Staphylococcus aureus and Escherichia coli, with RV-23 showing the highest potency. Moreover, RV-23 had lower cytotoxicity than melittin or AR-23 at their minimal inhibitory concentration. In addition, CD experiments showed that melittin, RV-23, and AR-23 all had a typical α-helical structure, and RV-23 had the lowest α-helix content. The structural information showed that RV-23 has the lowest hydrophobicity and highest hydrophobic moment. Because hydrophobicity and α-helix content are believed to correlate with hemolysis, the results indicate that the selective lytic activity against bacteria of RV-23 may be due to its low hydrophobicity and α-helicity, which lead to low cytotoxicity without affecting antibacterial activity. Furthermore, RV-23 did not affect the structure and function of blood components such as red blood cells, platelets, albumin, and the blood coagulation system. In conclusion, RV-23 is a cell-selective antibacterial peptide with high hemocompatibility due to its unique structure.

A Design and Implementation of 32-bit RISC-V RV32IM Pipelined Processor in Embedded Systems (임베디드 환경에서의 32-bit RISC-V RV32IM 파이프라인 프로세서 설계 및 구현)

  • Recently, demand for embedded systems requiring low power and high specifications has been increasing, and RISC-V processors are being widely applied. RISC-V, a RISC-based open instruction set architecture (ISA), has been developed and researched by UC Berkeley and other researchers since 2010. RV32I ISA is sufficient to support integer operations such as addition and subtraction instructions, but M-extension should be defined for multiplication and division instructions. This paper proposes an RV32I, RV32IM processor, and indicates benchmark performance scores compared to an existing processor. Additionally, A non-stalling method was proposed to support a 2-stage pipelined DSP multiplier to the 5-stage pipelined RV32IM processor. Proposed RV32I and RV32IM processors satisfied a maximum operating frequency of 50 MHz on Artix-7 FPGA. The performance of the proposed processors was verified using benchmark programs from Dhrystone and Coremark. As a result, the Coremark benchmark results of the proposed processor showed that it outperformed the existing RV32IM processor by 23.91%.

Design Verification of APR1400 Reactor Vessel Through Re-engineering Approach

  • This paper describes verification of APR1400 reactor vessel by applying the system engineering approach, in which the design re-engineering method is used to check the design parameters of APR1400 RV (reactor vessel). The RV is classified as safety class 1 and therefore must adhere strictly to the rules of ASME BPVC section III, subsection NB and seismic category I. This study explores designing the RV by following the ASME guidelines and making a comparative study with the current design. To meet this objective we apply system engineering methodologies to structure the process and allow for verification and validation of the major RV design parameters such as thickness of RV. The structural thicknesses of various part of RV are determined as well as reinforcements on the RV major nozzles. A 3D virtual reality model was created based on the design parameters using CATIA V5 and animation using Dassault Composer V2016. A comparison of re-engineered ARP1400 RV and standard APR1400 RV was done to show which design parameters were taken more conservative approach.

Rv3168 Phosphotransferase Activity Mediates Kanamycin Resistance in Mycobacterium tuberculosis

  • Tuberculosis is a worldwide epidemic disease caused by Mycobacterium tuberculosis, with an estimated one-third of the human population currently affected. Treatment of this disease with aminoglycoside antibiotics has become less effective owing to antibiotic resistance. Recent determination of the crystal structure of the M. tuberculosis Rv3168 protein suggests a structure similar to that of Enterococcus faecalis APH(3')-IIIa, and that this protein may be an aminoglycoside phosphotransferase. To determine whether Rv3168 confers antibiotic resistance against kanamycin, we performed dose-response antibiotic resistance experiments using kanamycin. Expression of the Rv3168 protein in Escherichia coli conferred antibiotic resistance against $100{\mu}M$ kanamycin, a concentration that effected cell growth arrest in the parental E. coli strain and an E. coli strain expressing the $Rv3168^{D249A}$ mutant, in which the catalytic Asp249 residue was mutated to alanine. Furthermore, we detected phosphotransferase activity of Rv3168 against kanamycin as a substrate. Moreover, docking simulation of kanamycin into the Rv3168 structure suggests that kanamycin fits well into the substrate binding pocket of the protein, and that the phosphorylation-hydroxyl-group of kanamycin was located at a position similar to that in E. faecalis APH(3')-IIIa. On the basis of these results, we suggest that the Rv3168 mediates kanamycin resistance in M. tuberculosis, likely through phosphotransferase targeting of kanamycin.

  • In this study, the performance evaluation of the RC frame specimen (RV2) which was strengthened by a steel frame and a steel damper with the lateral deformation prevention details proceeded. The comparison objects are bare frame specimen (BF), RV2 and AWD, where AWD is a specimen reinforced with steel damper and aramid fiber sheets. In the evaluation of envelope curve, stiffness degradation, and energy dissipation capacity, RV2 was evaluated to have excellent capacity as a whole. To evaluate the strengthening effect of the steel frame based on the maximum strength and energy dissipation capacity, it was evaluated to have a 38% of the RV2's capacity.

  • Painless treatment determines the quality of pediatric dental care. Although local anesthesia has been used to manage pain in dentistry, children often cite traditional aspirating syringes as a symbol of fear and pain. Adequate pain control during dental procedures may help alleviate fear and anxiety and instill positive oral health attitudes in children. Newer approaches such as intranasal spray, centbucridine, jet injectors, buzzy devices, and acupressure have been developed to help dentists provide near-painless injections while reducing dental anxiety. This review aims to summarize newer approaches to alleviate pain and anxiety in children.

Characteristics of Allergy Inducing Materials Isolated from Rhus verniciflua Stokes on BALB/c Mice (옻(Rhus verniciflua Stokes)으로 부터 분리한 알레르기 유발물질의 BALB/c mice에 대한 특성)

  • We purified crude urushiol from natural lacquer produced in Korea, China, and Japan and then isolated several urushiol congeners known to induce allergic reactions. The 3 major kinds of urushiol congeners that were isolated and purified were RV-l (C$_{21}$H$_{34}$O$_2$, M.W. 314.462), RV-2 ((C$_{21}$H$_{32}$O$_2$, M.W. 316.240) and RV-3 ((C$_{23}$H$_{34}$O$_2$, M.W. 342.515), occupying 80% of total crude urushiol. The content of RV-l was the highest in natural lacquer from China at 70.07%, and was the lowest in that from Japan at 62.38%. However, the content of RV-2 in natural lacquer from Japan was 9.25%, 2$\sim$3 times higher than those from Korea (4.28%) and China (3.09%). As an allergy inducing character, RV-l had strong inducing power and durability in the primary stage, showing slow recovery. RV-2 had weak power in the primary stage and also showed slow recovery. Although RV-3 had comparatively weak power at the primary stage, it induced the strongest allergy contact dermatitis after 48 hr. However, it recovered to nearly the same level as control group 72 hr after sensitization time. Accordingly, we found out that RV-1 is the most influential of urushiol congeners in inducing allergic reactions, natural lacquer from China having the most inducible strength and slowest recovery compared to those from Korea and Japan.

Rabies immune status in the stray and companion dogs in Korea

  • Rabies virus (family Rhabdoviridae, genus Lyssavirus, RV) is the causative agent of rabies in mammals. We conducted a sero-epidemiological survey for RV using sera from South Korean stray and companion dogs in the present study. A total of 533 canine serum samples were collected between February 2006 and December 2007 and were screened for rabies immunity with a neutralizing peroxidase linked assay. Both companion (49.1%) and stray (60.1%) dogs demonstrated RV seropositivity. Regional RV antibody prevalence was measured in the Jeju (87.5%), Gyeonggi (62%), Gyeongsang (59.1%), Jeonra (42%), Chungcheong (37.9%), and Gangwon (30.4%) provinces. Prevalence increased with age but did not exceed 80% in any age group. Stray and companion dogs had RV antibody prevalence values of 26.7% and 23.7%, respectively. Seroprevalence was significantly associated with age $({\chi}2\;=\;9.46;\;p\;=\;0.024)$ for companion dogs, although this association was not evident in stray dogs. There were no significant differences in age between stray and companion dogs and no gender differences in RV seroprevalence. Our results suggested that a widespread and reinforced vaccination program must be applied to Korean dogs.

Characterization of G9 genotype porcine rotavirus isolated in Korea (국내 양돈장에서 분리한 G9형 돼지로타바이러스의 특성)

  • Porcine rotaviruses are the most common causes of viral gastroenteritis in piglets around the world. The major G genotypes of porcine rotaviruses causing diarrhea were G4, G5 and G11 genotypes. Recently, G9 genotype rotaviruses were problemed at swine farms and frequently recognized from diarrheic piglets. In this study, a porcine rotavirus (PoRV-1) was isolated from piglet showing diarrhea using MA104 cells and confirmed as rotavirus by electron microscopy, genomic RNA electropherotyping and indirect immunofluorescence antibody tests. The nucleotide sequence of the VP7 gene of PoRV-1 was determined and compared with those of other genotype rotavirus strains from other parts of the world. Also, the nucleotide sequences of VP4, VP6 and NSP4 genes of PoRV-1 were determined and compared with those of other rotavirus strains from other countries. The results showed that the PoRV-1 isolate belonged to the G9 genotype and the P, I and E genotypes of PoRV-1 were P[23], I5 and E1, respectively. The Korean G9 PoRV-1 isolate and its nucleotide sequence data would be usefully used for the development of porcine rotavirus vaccines in near future.

Molecular characterization of avian rotavirus isolated in Korea

  • An avian rotavirus (AvRV-2) was isolated from feces of broilers suffering from acute gastroenteritis in 2011. It was the first avian rotavirus isolated in Korea. To investigate the molecular characteristics of AvRV-2, the VP4, VP6, VP7 and NSP4 gene nucleotide sequences were determined and compared with those of rotavirus strains available in the GenBank database. The phylogenetic tree of VP7 gene showed that AvRV-2 had a high degree of nucleotide sequence homology (93.4% to 94.7%) with those of rotaviruses belonging to genotype G19 cluster. The phylogenetic tree of the VP4 gene revealed a high degree of nucleotide sequence homology (95.8% to 95.9%) with genotype P[30] rotaviruses isolated from chickens. The VP6 and NSP4 gene nucleotide sequences showed the highest identities with those of avian strains with 95.3% to 96.4% and 90.3% to 92.2%, respectively. Genetic characterization of the VP4, VP6, VP7 and NSP4 showed that AvRV-2 strain was most closely related to chicken rotavirus strains from Germany and Japan. Comparative nucleotide sequences and phylogenetic analysis indicated that avian rotavirus isolated from broilers belonged to genotype G19P[30] and it was the first report on avian rotavirus infection in Korea.
