Mangiferin is a kind of natural xanthone glycosides and is known to have various pharmacological activities. However, since the beneficial efficacy of this compound has not been reported in retinal pigment epithelial (RPE) cells, this study aimed to evaluate whether mangiferin could protect human RPE ARPE-19 cells from oxidative injury mimicked by hydrogen peroxide (H2O2). The results showed that mangiferin attenuated H2O2-induced cell viability reduction and DNA damage, while inhibiting reactive oxygen species (ROS) production and preserving diminished glutathione (GSH). Mangiferin also antagonized H2O2-induced inhibition of the expression and activity of antioxidant enzymes such as manganese superoxide dismutase and GSH peroxidase, which was associated with inhibition of mitochondrial ROS production. In addition, mangiferin protected ARPE-19 cells from H2O2-induced apoptosis by increasing the Bcl-2/Bax ratio, decreasing caspase-3 activation, and blocking poly(ADP-ribose) polymerase cleavage. Moreover, mangiferin suppressed the release of cytochrome c into the cytosol, which was achieved by interfering with mitochondrial membrane disruption. Furthermore, mangiferin increased the expression and activity of heme oxygenase-1 (HO-1) and nuclear factor-erythroid-2 related factor 2 (Nrf2). However, the inhibition of ROS production, cytoprotective and anti-apoptotic effects of mangiferin were significantly attenuated by the HO-1 inhibitor, indicating that mangiferin promoted Nrf2-mediated HO-1 activity to prevent ARPE-19 cells from oxidative injury. The results of this study suggest that mangiferin, as an Nrf2 activator, has potent ROS scavenging activity and may have the potential to protect oxidative stress-mediated ocular diseases.
본 연구에서는 대두가공부산물의 하나인 비지(SCR: soybean crud residue)를 이용하여 만든 메주를 항비만 또는 혈중지질 개선용의 식 의약 소재로써의 직접적인 이용 가능성을 검토할 목적으로 우리나라의 전통메주 발효의 주요 미생물의 하나인 Aspergillus oryzae var effuses KACC 44990로 발효시킨 메주(ASCR)와 홍국 제조용으로 사용되는 미생물의 하나인 Monascus pilosus IFO 4480으로 발효시킨 메주(MSCR) 및 이들을 1:1(w/w)로 상호 혼합한 메주(AMSCR)를 제조하였으며 이들 메주를 각 2%씩 함유하는 고지방식이로 8주간 마우스를 사육하였을 때의 체중, 혈액 및 간 조직 지질의 함량 변동과 간 조직 ROS 생성계 및 소거계 효소의 활성을 측정하였다. SCR, ASCR, MSCR 및 AMSCR은 고지방식이 또는 비만으로 유도된 간, 심장, 신장, 고환의 비대 현상과 부고환 주변 지방의 함량을 감소시키는 효과가 있으며, 그 효과는 AMSCR>ASCR>MSCR>SCR 순으로 ASCR과 AMSCR에서 높았다. SCR, ASCR, MSCR 및 AMSCR은 고지방식이 또는 비만으로 유도된 혈청과 간조직의 중성지방과 총콜레스테롤 함량을 감소시키는 효과가 있으며, 혈청의 HDL 콜레스테롤 함량을 높이면서 LDL 콜레스테롤 함량을 낮추는 효과가 있다. 특히 ASCR은 중성지방의 함량을, 그리고 MSCR은 콜레스테롤의 함량을 각각 감소시키는 효과가 높으며, ASCR과 MSCR을 혼합한 AMSCR에서 종합적인 지질개선 효과가 높았다. SCR, ASCR, MSCR 및 AMSCR은 고지방식이 또는 비만상태에서 나타나는 간 손상을 예방하는 효과가 있으며 그 효과는 AMSCR>ASCR>MSCR>SCR 순으로 컸다. SCR, ASCR, MSCR 및 AMSCR은 고지방식이 또는 비만상태에서 나타나는 ROS 생성계 효소인 XO의 활성을 억제시킴과 동시에 ROS 소거계 효소인 SOD, GPX 및 GST의 활성을 높이는 효과가 있으며 그 효과는 AMSCR>ASCR>MSCR>SCR 순으로 컸다. 결론적으로 본 실험을 통하여 발효비지는 항비만 효과가 있을 뿐만 아니라 지질을 개선할 수 있음을 확인하였다.
본 연구에서는 참치 부산물을 활용한 항산화 신소재 개발의 가능성을 탐색하기 위해 부산물을 이용한 단백가수분해물 제조 및 항산화 활성을 평가하였다. 5종의 단백질 분해효소(alcalase, bromelain, flavourzyme, neutrase 및 papain)를 이용한 단백가수분해물 제조 시 12시간을 최적의 가수분해 시간으로 선정하였으며, 특히 alcalase, flavourzyme, neutrase의 가수분해도가 높게 나타났다. 이를 각 가수분해물의 수율과 비교하였을 때 유사한 경향임을 확인하였다. 각 효소제를 12시간 처리하여 제조한 단백가수분해물의 항산화 활성을 비교한 결과, TPF가 다른 가수분해물에 비해 유의적으로 높은 ROS(ABTS radical 및 H2O2) 소거 활성을 보이는 것으로 확인되었다. 또한 TPF는 hydroxyl radical에 의한 2-deoxy-D-ribose의 산화 및 linoleic acid의 과산화에 대해서도 우수한 억제 활성을 나타내었으며, 참치 가공부산물로부터 제조한 추출물, 밀웜 단백가수분해물 및 대두 단백가수분해물과의 ROS 소거 활성 비교 실험에서도 TPF가 상대적으로 우수한 활성을 보였다. 본 연구를 통해 참치 가공부산물을 이용한 항산화 소재개발 가능성을 확인할 수 있었으며, 본 연구 결과들은 수산가공부산물을 이용한 신규 생리활성 소재 개발용 기초연구자료로 활용될 수 있을 것으로 예상된다.
본 연구에서는 뽕잎차와 Monascus pilosus로 발효시킨 뽕잎차가 ICR mouse의 체중과 간 조직 항산화계 효소류의 활성에 미치는 영향을 조사하기 위하여 정상대조군(NC), 고지방 식이 대조군(HC), 뽕잎차 분말을 2% 혼합한 고지방 식이군(UM), 발효 뽕잎차 분말을 2% 혼합한 고지방 식이군(FM)으로 나누어 8주간 사육하였다. 체중증가량에 있어서는 HC군의 주당 체중증가량은 NC군에 비해 2.13배로 높았으나, UM군과 FM군은 NC군의 1.80 및 1.67배로 체중증가량이 크게 완화되었다. 장기 중량은 고지방 식이군에서 증가되었던 간의 중량은 UM과 FM군에서 회복되는 경향을 보였으며, 신장의 경우에도 NC군 수준으로 회복되는 경향을 나타내었다. 부고환 주변 지방의 함량은 UM과 FM군의 경우 모두 고지방 식이로 현저하게 증가된 지방함량을 유의적으로 감소시켰으며, 그 효과는 FM군에서 현저하였다. ALT 활성은 HC군이 NC군에 비하여 2.56배나 높은 수치를 나타내었으며, UM군과 HC 간의 유의차는 보이지 않으나 FM군은 HC군에 비하여 유의적으로 낮았다. TG, TC, LDL-cholesterol의 함량 및 동맥경화지수(AI)는 HC군은 NC군에 비하여 각각 18.42%, 39.59%, 212.56% 및 230.70%가 증가하였으며, HDL-cholesterol 함량은 17.84%가 감소하였다. UM군의 TG, TC, HDL- 및 LDL-cholesterol 함량은 HC군과 유의적인 차이가 없으나, FM군에서는 유의적인 차이를 보였으며, 동맥경화지수도 HC군 2.63, UM군 2.09, FM군 1.70으로 FM군에서 양호한 결과를 나타내었다. 간 조직 TG 함량은 UM 및 FM군이 HC군에 비하여 25.18~25.72%가 감소되었으며, 총 콜레스테롤의 함량에서도 TG와 유사한 경향을 보였다. Glutathione(GSH)의 함량은 FM군이 UM군에 비하여 유의적으로 증가하였으며, lipid peroxide(LPO)의 함량은 UM군과 FM군은 HC군에 비하여 각각 49.82% 및 44.52% 감소하였다. Xanthine oxidase(XO) 활성은 UM군과 FM군에서는 HC군에 비하여 각각 38.41% 및 53.62%가 감소하였다. SOD 활성은 UM과 FM군 모두 NC군과 유사한 활성을 나타내었다. Glutathione S-transferase(GST)활성은 UM군과 FM군은 고지방 식이에 의해 감소된 HC군의 활성을 14.81~23.46% 증가시켰다. Glutathione peroxidase(GPX) 활성(NADPH nmole/min/mg protein)은 고지방 식이 대조군인 HC군에서의 활성은 NC군의 65.59%에 불과하였으나, UM군과 FM군은 HC군에 비하여 각각 22.95% 및 38.52% 증가하였다. 간 조직을 검경한 결과, 고지방 식이 대조군인 HF군에서는 간 조직 세포가 지방 방울의 축적에 의해 확장되어 간굴모세혈관(hepatic sinusoids)이 폐쇄된 상태였다. 하지만, UM군과 FM군에서는 중심정맥 주위의 세포들이 정상적으로 잘 보존되어 있으며, 간 소엽의 간세포삭과 간 조직 세포 내에 지방방울의 축적이 보이긴 하나 구분이 가능하며, UM군에 비하여 FM군 쪽의 간 손상이 보다 경미한 것으로 나타났다. 이상의 실험결과, M. pilosus로 발효시킨 뽕잎차는 정상식이군에서는 간 조직 손상과 관련된 LPO의 함량을 경감시키는 효과와 함께 경미한 체중 감소 효과가 있는 것으로 사료되며, 고지방 식이에 의한 비정상 생리 상태에서는 비만 해소와 ROS 생성계효소의 저해와 소거계 항산화 효소의 활성화를 통한 간 기능 증진과 조직 손상을 예방 또는 경감시키는 효과가 있는 것으로 생각된다.
Etiological studies of diabetes and its complications showed that oxidative stress might play a major role, Therefore, many efforts have been tried to regulate free oxygen radicals for treating diabetes and its complications. Mori Fructus extract has been known to be effective for the antidiabetic, antihyperlipidemic and antiobesitic prescription, and composed of four crude herbs, The aim of this study was to investigate the effect of Mori Fructus extract in male ob/ob mouse with severe obesity, hyperinsulinemia, hyperglycemia, hyperlipidemia. Mice were grouped and treated for 5 weeks as follows. Both the lean (C57BL/6J black mice) and diabetic (ob/ob mice) control groups received standard chow. The experimental groups were fed with a diet of chow supplemented with 7.5, 15 and 30 mg Mori Fructus extract per 1 kg of body weight for 14 days. The effects of Mori Fructus extract on the ob/ob mice were observed by measuring the serum levels of glucose, insulin, lipid components, and the kidney levels of reactive oxygen species (ROS), MDA+HAE, GSH and also the enzyme activities involved in polyol pathway. Western blotting was performed using anti-AGE, anti-RAGE respectively. Mori Fructus extract lowered the levels of serum glucose and insulin in a dose dependent manner. Total cholesterol, triglyceride and free fatty acid levels were decreased, while the HDL-cholesterol level was increased, in Mori Fructus extract treated groups. Renal aldose reductase and sorbitol dehydrogenase activities were increased in the ob/ob mice, whereas those were inhibited in the Mori Fructus extract-administered groups. Mori Fructus extract inhibited the generation of ROS in the kidney. MDA+HAE level was increased and the GSH level was decreased in the ob/ob mice, whereas those were improved in the Mori Fructus extract-administered groups. Mori Fructus extract inhibited the expression of AGE, RAGE in the ob/ob mice. The results suggested that Mori Fructus exerted the antidiabetic and antihyperlipidemic activities by regulating the activities of polyol pathway enzymes, scavenging the ROS, decreasing the MDA+HAE level, increasing the GSH level and inhibiting the expression of AGE, RAGE in the ob/ob mice.
Injury caused by reactive oxygen species (ROS), known as oxidative stress, is one of the major damaging factors in plants exposed to environmental stress. Chloroplasts are specially sensitive to damage by ROS because electrons that escape from the photosynthetic electron transfer system are able to react with relatively high concentration of $O_2$ in chloroplasts. To cope with oxidative stress, plants have evolved an efficient ROS-scavenging enzymes such as superoxide dismutase (SOD) and ascorbate peroxidase (APX), and low molecular weight antioxidants including ascorbate, glutathione and phenolic compounds. To maintain the productivity of plants under the stress condition, it is possible to fortify the antioxidative mechanisms in the chloroplasts by manipulating the antioxidation genes. A powerful gene expression system with an appropriate promoter is key requisite for excellent stress-tolerant plants. We developed a strong oxidative stress-inducible peroxidase (SWPA2) promoter from cultured cells of sweetpotato (Ipomoea batatas) as an industrial platform technology to develop transgenic plants with enhanced tolerance to environmental stress. Recently, in order to develop transgenic sweetpotato (tv. Yulmi) and potato (Solanum tuberosum L. cv. Atlantic and Superior) plants with enhanced tolerance to multiple stress, the genes of both CuZnSOD and APX were expressed in chloroplasts under the control of an SWPA2 promoter (referred to SSA plants). As expected, SSA sweetpotato and potato plants showed enhanced tolerance to methyl viologen-mediated oxidative stress. In addition, SSA plants showed enhanced tolerance to multiple stresses such as temperature stress, drought and sulphur dioxide. Our results strongly suggested that the rational manipulation of antioxidative mechanism in chloroplasts will be applicable to the development of all plant species with enhanced tolerance to multiple environmental stresses to contribute in solving the global food and environmental problems in the 21st century.
The aim of this study was to investigate relationship among seed viability and enzymes activities involved in scavenging reactive oxygen species (ROS), especially, superoxide dismutase (SOD), glutathione reductase (GR), and catalase (CAT). In other respects, osmopriming has been demonstrated to reinvigorate aged seeds. Various viabilities of seeds that were ranged from 80 to 100% of germination rate could be produced using osmopriming and accelerated aging treatments. Priming treatment of Pinus thunbergii seeds for 3 days at $15^{\circ}C$ with a polyethylene glycol solution at -1.2 MPa improved their subsequent germination at $25^{\circ}C$. Accelerated aging (3, 6, 9, and 12 days at $41^{\circ}C$ and 100% relative humidity) decreased seed germination percentage depending on aging treatment duration. Electrolyte conductivities of seeds were measured as assay of membrane integrity. The conductivity from electrolyte leakage of P. thunbergii seed was also correlated with seed germinability. Conductivity for control seeds that had 95% of germination percentage was 3.48 ${\mu}S\;g^{-1}$, but jumped as doubled (7.98 ${\mu}S\;g^{-1}$) in 12-day-aged seed that had 80% of germination percentage. Our results demonstrate that aging of P. thunbergii seeds is associated with changes in the electrolyte leakage, lipid peroxidation, and antioxidant defense system. Priming of aged seeds progressively restored the initial germinative ability and resulted in a marked decrease in the levels of MDA and conductivity of seed leachate. These effects of priming were also well recovered of GR and CAT activities in aged seed. The improved seed quality by priming treatment appears at least partly attributable to reduced lipid peroxidation, resulting from enhanced antioxidative enzyme activities that are suggesting the antioxidant defense systems play a key role in seed vigor.

Protulaca oleracea, a widely distributed weed, has been reported to exhibit different health promoting effects. The objective of this study was to evaluate the anti-oxidative and anti-inflammatory effects of P. oleracea on LPS-stimulated AGS cells. The cytotoxicity of P. oleracea in AGS cells was examined by MTT assay. The anti-oxidative effects of P. oleracea were examined by DPPH assay. RT-PCR was carried out to examine the effect of P. oleracea in the mRNA expression of different inflammatory mediators. MTT assay revealed that P. oleracea have almost no cytotoxity in AGS cells. DPPH radical scavenging activities were better than butylated hydroxyl toluene (BHT). The mRNA expression of different endogenous anti-oxidative enzymes (SOD2, GPx3 and catalase) were preserved by P. oleracea in AGS cells. The nitric oxide production and expression of iNOS in LPS stimulated RAW264.7 were suppressed in P. oleracea treated groups. Based on these findings, P. oleracea has protective anti-oxidant and anti-inflammatory effects.
The medicinal mushroom Inonotus obliquus is a traditional and widely used multi-functional fungus. In this study, we have investigated whether Inonotus obliquus (Chaga mushroom) extracts exerts anti-oxidant effects on cisplatin-induced cytotoxicity in auditory cell line, HEI-OC1 cells. First of all, Chaga extracts has no harmful effects on viability of HEI-OC1 cells in the dose range of $65{\sim}125{\mu}g/m{\ell}$. Moreover, it shows cyto-protective effects on the cells treated with cisplatin-induced cytotoxicity in HEI-OC1 cells and the damage of hair cells arrays of the rat primary organ of Corti explants in the presence of cisplatin. Pretreatment with Chaga extracts inhibited the cell death, reactive oxygen species generation (ROS), lipid peroxidation induced by cisplatin. These effects were associated with the induction of antioxidant enzyme by Chaga extracts. We further investigated the effects of Chaga extracts on expression of antioxidant enzymes such as Cu, Zn superoxide dismutase (SOD 1) and Mn SOD (SOD 2) by RT-PCR. In addition, Chaga extracts shows SOD activity and SOD protein expression in cisplatin treated group induced similar to control group. Taken together, these results indicate that Chaga extracts can prevent cisplatin-induced cytotoxicity by radical-scavenging activity (SOD activity) in HEI-OC1 cells. It might be an effective as antioxidant and further studies on the chemo-preventive mechanisms of Inonotus obliquus are needed.
Wheat is highly susceptible to heat stress, which significantly reduces grain yield. In this study, we used RNA-seq technology to analyze the transcript expression at three different time-points after heat treatment in three cultivars differing in their susceptibility to heat stress: Jopum, Keumkang, and Olgeuru. A total of 11,751, 8850, and 14,711; 10,959,7946, and 14,205; and 22,895,13,060, and 19,408 differentially-expressed genes (log2 fold-change > 1 and FDR (padj) < 0.05) were identified in Jopum, Keumkang, and Olgeuru in the control vs. 6-h, in the control vs. 12-h, and in the 6-h vs. 12-h heat treatment, respectively. Functional enrichment analysis showed that the biological processes for DEGs, such as the cellular response to heat and oxidative stress-and including the removal of superoxide radicals and the positive regulation of superoxide dismutase activity-were significantly enriched among the three comparisons in all three cultivars. Furthermore, we investigated the differential expression patterns of reactive oxygen species (ROS)-scavenging enzymes, heat shock proteins, and heat-stress transcription factors using qRT-PCR to confirm the differences in gene expression among the three varieties under heat stress. This study contributes to a better understanding of the wheat heat-stress response at the early growth stage and the varietal differences in heat tolerancea.

① 서비스 이용은 당 사이트의 업무상 또는 기술상 특별한 지장이 없는 한 연중무휴, 1일 24시간 운영을

② NDSL에서 제공하는 콘텐츠를 상업적 및 기타 영리목적으로 복제/배포/전송할 경우 사전에 KISTI의 허락을
받아야 합니다.
③ NDSL에서 제공하는 콘텐츠를 보도, 비평, 교육, 연구 등을 위하여 정당한 범위 안에서 공정한 관행에
합치되게 인용할 수 있습니다.
④ NDSL에서 제공하는 콘텐츠를 무단 복제, 전송, 배포 기타 저작권법에 위반되는 방법으로 이용할 경우
저작권법 제136조에 따라 5년 이하의 징역 또는 5천만 원 이하의 벌금에 처해질 수 있습니다.
제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
