The Anti-oxidative and Anti-inflammatory Activities of Malus melliana Ethanol Extract (Malus melliana 에탄올 추출물의 항산화 및 항염증 활성)

  • Malus melliana (Hand.-Mazz.) Rehder (M. melliana) is a Chinese plant that belongs to the Rosaceae family. There have been no previous reports regarding its bioactivity. In this study, the anti-oxidative and anti-inflammatory activities of M. melliana ethanol extract (MMEE) were evaluated using a 1,1-diphenyl-2-picryl-hydrazyl (DPPH) radical scavenging activity assay, reactive oxygen species (ROS) scavenging activity assay, nitric oxide (NO) inhibitory activity assay, and the analysis of related protein expressions through Western blot hybridization. MMEE showed potent scavenging activity against DPPH, similar to ascorbic acid, a well-known anti-oxidative agent, which was used as a positive control. MMEE also inhibited hydrogen peroxide-induced ROS in RAW 264.7 cells. Moreover, MMEE induced the expression of an anti-oxidative enzyme, heme oxygenase 1, and its upstream transcription factor, nuclear factor E2-related factor-2, in a dose-dependent manner. On the other hand, MMEE was associated with a reduction in NO production, which was induced by the lipopolysaccharide treatment of RAW 264.7 cells. The expression of inducible nitric oxide synthase, which is the upstream regulator of NO production, was also inhibited. Taken together, these results suggest that MMEE has anti-oxidative and anti-inflammatory properties, thus appearing to be a potential anti-oxidant and anti-inflammatory agent. The further identification of active compounds that confer the biological activities of MMEE may be necessary.

Antioxidative and Inhibitory Activities on Tyrosinase of Hippophae rhamnoides Leaf Extracts (비타민나무 잎 추출물의 항산화 및 타이로시네이즈 저해활성)

  • In this study, the antioxidative and inhibitory effects on tyrosinase and elastase of Hippophae rhamnoides (H. rhamnoides) leaf extracts were investigated. The ethyl acetate fraction of H. rhamnoides extracts showed more effective free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activity ($FSC_{50}$ = 4.68 ${\mu}g$/mL). Reactive oxygen species (ROS) scavenging activity ($OSC_{50}$) of the aglycone fraction in the luminol-dependent $Fe^{3+}$-EDTA/$H_2O_2$ system was 0.19 ${\mu}g$/mL. The aglycone fraction exhibited more prominent cellular protective effects (${\tau}_{50}$, 133.3 min at 10 ${\mu}g$/mL) in the $^1O_2$-induced photohemolysis of human erythrocytes. The inhibitory effect ($IC_{50}$) of the aglycone fraction on tyrosinase was 54.86 ${\mu}g$/mL, and more effective than arbutin known as whitening agent. These results indicate that fractions of Hippophae rhamnoides extract can be used as antioxidants in biological system, particulaly skin exposed to UV radiation by quenching and/or scavenging $^1O_2$ and other ROS, and protecting cellular membranes against ROS.

Antioxidant Activities and Protective Effects of Hot Water Extract from Curcuma longa L. on Oxidative Stress-Induced C2C12 Myoblasts (강황 열수 추출물의 항산화 활성 및 C2C12 Myoblasts의 산화적 손상에 대한 보호 효과)

  • The aim of this study was to investigate the antioxidant activities and protective effects of hot water extract from Curcuma longa L. (CLW) on oxidative stress-induced C2C12 myoblasts. Antioxidant activities of CLW were evaluated based on 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging activities. Protective effects of CLW on oxidative stress-induced C2C12 myoblasts were determined based on cytotoxicity, $H_2O_2$ protective activity, and intracellular reactive oxygen species (ROS) level. DPPH and ABTS radical scavenging activities represented by $SC_{50}$ were $188.5{\pm}3.0{\mu}g/mL$ and $92.0{\pm}0.9{\mu}g/mL$, respectively. Using C2C12 myoblasts, CLW treatment increased cell viability against oxidative stress-induced cell death. Further, CLW treatment reduced the intracellular ROS level in cells treated with $H_2O_2$. These results suggest that CLW might have the capability to protect oxidative stress-induced C2C12 myoblasts.

Antioxidant Activities of Green and Purple Kohlrabi Juices (녹색 및 자색 콜라비 착즙액의 항산화 활성)

  • In this study, we investigated the antioxidant activity of green kohlrabi juice (GKJ) and purple kohlrabi juice (PKJ) using various in vitro methods. The results of 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) radical scavenging activities, ferric ion reducing antioxidant power (FRAP), reducing power, and nitrite scavenging activities showed that GKJ possessed higher antioxidant activity than PKJ. Green kohlrabi powder (GKJP) and purple kohlrabi powder (PKJP) inhibited hydrogen peroxide-induced cell death in human dermal fibroblasts. In addition, GKJP and PKJP suppressed intracellular reactive oxygen species (ROS) production induced by hydrogen peroxide in human dermal fibroblasts. These results suggest that green and purple kohlrabi juices are potential natural sources of antioxidants.

Protective effects of Carthamus tinctorius L. seed on C6 glial cells treated with ethanol (홍화씨 추출물의 in vitro 항산화 및 ethanol로 손상을 유도한 C6 신경교세포 보호 효과)

  • Chronic alcohol is responsible for oxidative stress and neurodegenerative diseases such as dementia. In the present study, we investigated the antioxidant activity and protective effects of seed of Carthamus tinctorius L. on ethanol-induced C6 glial cells. Antioxidant effect of seed of C. tinctorius L. was measured by scavenging activity of 1,1-diphenyl-2-prcrylhydrazy (DPPH), hydroxyl radical (·OH), superoxide radical, and nitric oxide. The seed of C. tinctorius L. extract showed significant radical scavenging activities in a concentration-dependent manner. In particular, it revealed strong DPPH and ·OH scavenging activity, displaying more than 80% at 500 and 100 ㎍/mL, respectively. Treatment of 500 mM ethanol to C6 glial cell led to decline of cell viability and elevation of reactive oxygen species (ROS) generation. However, seed of C. tinctorius L.-treated groups significantly increased cell viability and decreased ROS levels, compared to ethanol-induced control group. These results suggest that seed of C. tinctorius L. would have protective effect against neuronal oxidative stress induced by alcohol.

Effects of the Young Branch of Prunus persica Methanol Extracts and Solvent Fractions on ROS, RNS and Oxidized Low-Density Lipoprotein (도지(桃枝) 메탄올 추출물과 용매별 분획물이 활성산소종, 활성질소종 및 저밀도 지단백의 산화에 미치는 효과)

  • This study was designed to analyze the effects of methanol extracts and solvent fractions of the young branch of Prunus persica on scavenging activity of ROS, RNS and inhibiting activity of oxidative modification in human LDL induced by copper ion or free radical generator. The scavenging capacities of the fractions on DPPH radical, superoxide radical, nitric oxide and peroxynitrite were exhibited the highest in ethylacetate fraction, followed by butanol fraction. In the copper-induced LDL oxidative modification system, the highest antioxidant activity was revealed in ethylacetate fraction, and butanol fraction exhibited a similar activity. However, solvent fractions of the young branch of Prunus persica showed a relatively low antioxidant activity in the AAPH-mediated LDL oxidation. In addition, ethylacetate and butanol fractions also inhibited the copper-mediated LDL oxidation in the REM assay, which was comparable to that of the positive controls, including EDTA, ascorbic acid and BHT. Futhermore, a content of total phenolics in these two fractions was higher than that of the other fractions. These results indicated that ethylacetate and butanol fractions of the young branch of Prunus persica were useful for the prevention of the free radical- or metal ion-induced oxidative damages.

Antioxidant potential of buffalo and cow milk Cheddar cheeses to tackle human colon adenocarcinoma (Caco-2) cells

  • Objective: The aim of present study was to assess the anti-oxidant potential of water-soluble peptides (WSPs) extract derived from buffalo and cow milk Cheddar cheeses at different stages of ripening. Methods: The antioxidant potential of WSPs extract was assessed through 2,2'-azinobis-3-ethylbenzothiazoline-6sulfonic acid (ABTS)-radical scavenging activity. In addition, impact of WSPs extract on cell viability and production of reactive oxygen species (ROS) in human colon adenocarcinoma Caco-2 (tert-butylhydroperoxide-induced) cell lines was also evaluated. Results: The ABTS-radical scavenging activity increased progressively with ripening period and dose-dependently in both cheeses. However, peptide extract from buffalo milk Cheddar cheese demonstrated relatively higher activity due to higher contents of water-soluble nitrogen. Intracellular ROS production in Caco-2 cells decreased significantly (p<0.05) till 150th day of cheese ripening and remained constant thereafter. Additionally, dose-dependent response of WSPs extract on antioxidant activity was noticed in the Caco-2 cell line. Conclusion: On the basis of current in vitro study, the Cheddar cheese WSPs extract can protect intestinal epithelium against oxidative stress due to their antioxidant activity.

Anti-oxidant Effect of Agastache rugosa on Oxidative Damage Induced by $H_2O_2$ in NIH 3T3 Cell

  • The plant Agastache rugosa Kuntze has various physiological and pharmacological activities. Especially, it has been regarded as a valuable source for the treatment of anti-inflammatory and oxidative stress-induced disorders. However, little has been known about the functional role of it on oxidative damage in mammalian cells by ROS. In this study, we investigated the DPPH radical, hydroxyl radical, hydrogen peroxide and intracellular ROS scavenging capacity, and $Fe^{2+}$ chelating activity of the extracts from Agastache rugosa. In addition, we evaluated whether the extract can be capable of reducing $H_2O_2$-induced DNA and cell damage in NIH 3T3 cells. These extracts showed a dose-dependent free radical scavenging capacity and a protective effect on DNA damage and the lipid peroxidation causing the cell damage by $H_2O_2$. Therefore, these results suggest that Agastache rugosa is useful as a herbal medicine for the chemoprevention against oxidative carcinogenesis.

Effect of Chilgitang Extract for Herb-Acupuncture on ROS, RNS, Lipid Peroxidation and LDL oxidation (칠기탕(七氣湯) 약침액(藥鍼液)이 활성산소종, 활성질소종, 지질과산화 및 LDL의 산화에 미치는 영향)

  • Objectives : This study was undertaken to determine the in vitro antioxidant activity of the extract of Chilgitang herb-acupuncture solution (CHAS). Methods : The radical scavenging capacity was tested by 2,2-diphenyl-1-picryl-hydrazyl (DPPH), hypoxanthine-xanthine oxidase system, DCFH-DA assay, nitric oxide and peroxynitrite generating system. In addition, antioxidant activity on copper and AAPH mediated human low-density lipoprotein (LDL) oxidation was measured by using TBARS assay and relative electrophoretic mobility assay. The amount of total phenolic compounds was assayed by the Folin-Ciocalteu method. Results : CHAS revealed a potent scavenging activity on DPPH radical(82%), superoxide anions(73%), hydroxyl radical(63%), nitric oxide (99%) and peroxynitrite (99%). Moreover, CHAS showed a strong inhibitory effect (59%) on $FeCl_2$-ascorbic acid induced lipid peroxidation of rat liver homogenate. CHAS also markedly inhibited copper(81%) and AAPH(56%)-mediated LDL oxidation, and effectively suppressed the electrophoretic mobility during exposure of human LDL to copper ions. CHAS (82 mg/g) contained higher concentration of total phenolic compounds than that of water extract (45 mg/g) obtained from Chilgitang. Conclusions : These results indicate that CHAS may protect against ROS- or RNS involved diseases, including cardiovascular diseases.

New anti-wrinkle cosmetics

  • In the aged skin especially in the face and eyelid, deep and slight wrinkles are one of the remarkable phenomena of aging and the cause of wrinkle is various. Among the cause of wrinkles an oxidative stress plays an important roles in wrinkle formation process. It caused the lipid peroxidation of cell membrane, the increase of the MMPs(MatrixMetalloProteinase) gene expression and cellular DNA damage. These ROS induced materials may cause the degradation of collagen matrix system in the dermis and cause the formation of skin wrinkle. So, it is very important for protecting skin wrinkle formation to regulate ROS activity. In this study, we developed one active ingredient having multi functional activities such as activation of collagen synthesis, inhibition of MMPs activity, lipid peroxidation and free radical scavenging activity and inhibition of free radical induced DNA damage in vitro. Pericarpium castaneae extracts showed collagen synthesis increase in Normal Human Fibroblast and the inhibition of elastase activity (IC$\_$50/ of Elastase: 43.9$\mu\textrm{g}$/㎖). It showed also anti-oxidative activity (IC$\_$50/ : 48$\mu\textrm{g}$/㎖) and free radical scavenging activity(IC$\_$50/: 7.6$\mu\textrm{g}$/㎖). Conclusively, Pericarpium castaneae extracts may be used as an ingredient for new anti-wrinkle cosmetics.
