Analysis of Two-Dimensional Pollutant Transport in Meandering Streams (사행하천에서 오염물질의 2차원 거동특성 해석)

  • In this study, RMA2 and RMA4, the 2-D depth-averaged models, were employed to simulate the two-dimensional mixing characteristics of the pollutants in the natural streams. The velocity and depth were first calculated using RMA2, 2-D hydrodynamic model, and then the resulting flow field was inputted to RMA4, 2-D water quality model, to compute the concentration field. RMA models were verified using the velocity and concentration data measured in S-curved meandering channel. The results showed that the RMA2 model simulated well the phenomenon that the maximum velocity line is located at the Inner bank of meandering channel, and the RMA4 model was well adapted to reproduce the general mixing behavior and the separation of tracer clouds. Comparing model simulations with measured data in the field experiments, RMA2 model simulated well general flow field and tendency that the maximum velocity line skewed toward the outer bank which were found in field experiments. The simulations of RMA4 model showed that the center of the tracer cloud tends to follow the path in which the maximum velocity occurs. In this study, the dispersion coefficients are fine-tuned based on the measured coefficients calculated using field concentration data, and the results show reasonable agreement with predictive equations.

A Study on Mixing Behavior of Dredging Turbidity Plume Using Two-Dimensional Numerical Model (이차원 수치모형을 이용한 준설 탁도플륨의 혼합거동 연구)

  • The numerical simulations were performed to analyze the advection-diffusion processes of dredging-induced turbidity plume using RMA2 and RMA4 models in Bunam reservoir, Seosan, Chungnam. Field survey was also performed to measure the turbidity using the multi water quality monitoring system (YSI6600EDS). In the field survey, the vertical and horizontal distributions of the turbidity were measured during the dredging operation in Bunam reservoir. RMA2 model was used to simulate the velocity distributions in both the whole domain and the 2nd part of Bunam reservoir. RMA4 model was also used to simulate the concentration distribution in only the 2nd part of Bunam reservoir, where the dredging work were conducted. The comparison of the simulation results with the field data for the advection-diffusion of the turbidity plume using the concentration ratio concepts shows that the numerical model can be applied to analyze the environmental impact of dredging works.

  • 자연하천유역에 있어서 유사량 산정은 용수공급 및 하천계획을 결정하는데 있어 필수적으로 이루어져야 하는 정량적 분석인자이다. 자연하천유역에 있어 유사량 산정은 하도상의 수공구조물 존재여부, 유량조건 및 산정공식 등에 따라 다양하게 이루어져야 하며, 산정공식에 따라 유사량 및 하상변동에 대한 그 특성은 많은 차이를 나타내게 된다. 본 연구에서는 낙동강 본류의 진동 지점 8.44km구간과 낙동강 지류인 감천 유역의 김천 지점 8.66km 구간의 부유사농도 분포를 모의하기 위해 동수역학적 모형인 RMA-4 모형을 적용하였다. 먼저 지점의 흐름특성을 파악하고자 RMA-2모형을 적용한 결과 비교적 계획홍수위와 잘 일치하였으며, RMA-4 모형은 RMA-2 모형의 출력치를 기본자료로 사용하였으며 진동 지점과 김천지점의 부유사농도 분포를 모의해 본 결과 하류로 향할수록 부유사농도가 증가하는 것을 알 수 있었다. 앞으로 하천개수, 저수지설계 등의 하천공사에 유사가 중요한 인자이므로 유사의 정확한 이도분포를 파악하기 위해 실측자료를 보다 많이 관측하고 우리나라 하천특성에 적합한 유사농도 수치모형을 개발하기 위한 작업으로 이 분야에 많은 연구가 필요하다.

Two-dimensional Numerical Modeling of Water Quality Variation by Gates Operation in the Seonakdong River (수문운영에 따른 서낙동강 수질변화에 관한 이차원 수치모의)

  • This study has used RMA2 model and RMA4 model, which are depth-averaged two-dimensional flow and water-quality prediction models, to analyze the variation of the water-quality by the gates operation in the Seonakdong River. Sensitivity analysis is performed to get the Manning coefficient and the coefficient of eddy viscosity for RMA2 model, and to get the diffusion coefficient for RMA4 model. Since the numerical simulation using RMA2 and RMA4 models did not consider tributary pollutant load except for that of Joman River, it could make a little difference from the natural phenomenon. Nevertheless, the numerical simulation shows that the discharge of $30m^3/s$, which is the continuous inflow from the Daedong-gate, can make it possible to achieve the target water-quality (BOD 4.3mg/L) of Nakbon-N watershed about 10 days later if the Daejeo-gate could remain opened in connection with the Noksan-gate operation.

  • 본 연구에서는 하천에 유입된 오염물질의 거동 및 확산 특성을 파악하기 위하여 하천에서 추적자를 이용한 현장실험을 실시하였으며, 실험구간은 경상북도 경주시 에 위치한 하천으로 총 길이 2km, 평균폭 25m로 이루어진 대종천이다. 현장실험에서의 추적자 실험결과를 수치모형과 비교하기 위해 2차원 수심적분 모형인 RMA2, RMA4를 이용하였다. 2차원 동수역학적인 모형인 RMA2를 사용하여 흐름장을 모의한 후 계산결과를 2차원 수질모형인 RMA4에 입력하여 농도자료를 모의하였으며 상 하류단의 경계조건은 현장 실험시 실측한 상류단의 유량과 하류단의 수위를 적용하였다. 실측한 자료를 경계조건으로 모형에 적용하여 시간에 따른 농도값을 계산하였으며, 그 계산값을 추적자의 농도 관측값과 비교하였다. 계산결과는 서로 잘 일치하였다.

Study on Pollutant Diffusion in the Midstream of Youngsan River (영산강 중류부의 오염물질 이송특성 연구)

  • 본 연구에서는 영산강의 수질이 가장 심각한 광주하수처리장 방류구 직하류부에 대한 오염물질 확산에 시.공간적 분포양상을 파악하기 위하여 대상하천 영산강 본류의 25km구간에 RMA4 모델을 적용하여 오염 물질의 농도변화를 예측하였다. RMA4 모델의 적용을 위하여 RMA2 모델을 수행하여 구한 출력자료를 입력자료 사용하였으며, 수치모델의 적용을 위하여 연구 대상하천의 25km구간에 대하여 유한요소망은 2,304개의 절점과 7,635개의 요소로 구성하였다. 광주하수처리장 방류구 직하류부에 대한 오염물질 확산에 시.공간적 분포양상을 파악하기 위하여 광주하수종말처리장의 방류수에 의한 오염물질유입에 대한 4개의 시나리오를 작성하여 RMA4 모델을 수행하였으며, 광주하수종말처리장의 평균방류량 6.13cms와 BOD 수질기준 110mg/L(2004년 하수종말처리장 유입원수의 평균농도)의 농도를 유입 오염물질로 가정하여 영산강의 저수기의 유출량을 대상으로 오염물질 유입시간에 따라 4개의 시나리오에 대한 오염물질의 확산의 시.공간적 분포양상, 유하길이에 대한 오염물질의 최대 오염원농도 도달시간을 제시하였다.

Comparison of Normalization Methods for Defining Copy Number Variation Using Whole-genome SNP Genotyping Data

  • Precise and reliable identification of CNV is still important to fully understand the effect of CNV on genetic diversity and background of complex diseases. SNP marker has been used frequently to detect CNVs, but the analysis of SNP chip data for identifying CNV has not been well established. We compared various normalization methods for CNV analysis and suggest optimal normalization procedure for reliable CNV call. Four normal Koreans and NA10851 HapMap male samples were genotyped using Affymetrix Genome-Wide Human SNP array 5.0. We evaluated the effect of median and quantile normalization to find the optimal normalization for CNV detection based on SNP array data. We also explored the effect of Robust Multichip Average (RMA) background correction for each normalization process. In total, the following 4 combinations of normalization were tried: 1) Median normalization without RMA background correction, 2) Quantile normalization without RMA background correction, 3) Median normalization with RMA background correction, and 4) Quantile normalization with RMA background correction. CNV was called using SW-ARRAY algorithm. We applied 4 different combinations of normalization and compared the effect using intensity ratio profile, box plot, and MA plot. When we applied median and quantile normalizations without RMA background correction, both methods showed similar normalization effect and the final CNV calls were also similar in terms of number and size. In both median and quantile normalizations, RMA backgroundcorrection resulted in widening the range of intensity ratio distribution, which may suggest that RMA background correction may help to detect more CNVs compared to no correction.

Root Bark of Morus Alba Suppresses the YAP Activity through Activation of Classical Hippo Signaling Pathway (상근피의 Hippo 신호전달 경로 활성화를 통한 YAP 억제 효능)

  • This study aims to evaluate the effects of the root bark of Morus alba (RMA) on the regulation of the Hippo-YAP pathway. Hippo-YAP signaling is a critical regulator in controlling organ size and tissue homeostasis. Hippo, the serine/threonine kinase phosphorylate the LATS. Phosphorylated LATS then phosphorylates and inactivates the YAP and TAZ, which are two closely related transcriptional co-activator. Here we report RMA activates the Hippo signaling, thereby inhibits the YAP/TAZ activity. First, we examine the cytotoxic effects of RMA by MTT assay. RMA was cytotoxic at concentrations higher than $50{\mu}g/ml$ in HEK293A cells. The reporter gene assay was performed to measure the activity of TEAD, a key transcription factor that controls cell growth and proliferation. RMA significantly suppressed the luciferase activity. By phos-taq gel shift assay, and western blotting, we showed that RMA enhanced the phosphorylation of YAP in wild type cells, but not in LATS1/2 knock out cells, which means RMA activates classical Hippo pathway. RMA induced the cytoplasmic sequestration of YAP. RMA also suppressed the mRNA expression of CTGF and CYR61; the two major YAP dependent genes. Taken together, RMA is considered to be a good candidate for proliferative disease such as cancer, by facilitating cell death through activating the Hippo signaling pathway.

Numerical Simulation of Water Quality Enhancement by Removal of Contaminated Bed Material (하상오염물 제거에 의한 수질개선효과 수치모델링)

  • This study has an objective to estimate effect on water-quality enhancement by removal of contaminated river-bed material using a two-dimensional numerical modeling in the Seonakdong River, the Pyunggang River and the Maekdo River. RMA2 and RMA4 models were used for flow and contaminant transport simulation, respectively. After the analysis of the effects of flow restoration plan for the Seonakdong River system made by Lee et al (2008), simulation have been performed about scenarios which contains operations of the Daejeo Gate, the Noksan Gate, the Makdo Gate (on planning), and the Noksan Pumping Station. Because there is no option for elution from bed sediment in the RMA4 model, a simple technique has been used for initial condition modification for elution. The analyses revealed that the effect on water quality improvement due to dredging of bed sediment seemed to be less than 10 % of the total effect. The most efficient measure for the water quality improvement of the river system was the linked operation of water-gates and pumping station.

Prediction of Water-Quality Enhancement Effects of Gates Operation in the West-Nakdong River Using RMA2/RMA4 Models (RMA2/RMA4 모형을 이용한 서낙동간 수문연계운영의 수질개선 효과 예측)

  • An objective of this study is as follows: 1) performing sensitivity analysis and parameter estimation of RMA2 and RMA4 models for the West-Nakdong River, 2) drawing up alternatives of gates-operation for water-quality enhancement, and 3) quantitative evaluation of methodology of 'flow-restoration by gates-operation' among 'Comprehensive Plan Improving Water-Quality in the West-Nakdong River(WNR)' with the target water-quality(BOD at Nakbon-N point: below 4.3 mg/L). The parameters for the RMA2 (depth-averaged two-dimensional flow model) and RMA4 (depth-averaged two-dimensional water-quality model) were determined by sensitivity analysis. Result of parameter estimation for RMA2 and RMA4 models is $1,000\;Pa{\cdot}s$ of the eddy viscosity, 20 of the Peclet number, 0.025 of the Manning coefficient, and $1.0\;m^2/s$ of the diffusion coefficient. We have evaluated the effects of water-quality enhancement of the selected alternatives by numerical simulation technique with the models under the steady-state flow condition and the time-variant transport condition. Because of no-resuspension from river bottom and considering BOD as conservative matter, these simulation results slightly differ from real phenomena. In the case of $50\;m^3/s$ of Daejeo-gate inflow, two-dimensional flow pn results result represents that small velocity occurs in the Pyungkang Stream and no flow in the Maekdo River. In the WNR, there occurs the most rapid flow near timhae-bridge. In the WNR, changes of water-quality for the four selected simulation cases(6, 10, 30, $50\;m^3/s$ of the Daejeo-gate inflow) were predicted. Since the Daejeo-Gate and the Noksan-Gate can be opened up to 7 days, it would be found that sustainable inflow of $30\;m^3/s$ at the Daejeo-gate makes BOD in the WNR to be under the target of water-quality.
