Anticancer effect of joboksansam, Korean wild ginseng germinated from bird feces

  • Background: Joboksansam, Korean bird wild ginseng, is an artificially cultivated wild ginseng germinated from bird feces. Although numerous pharmacologic activities of wild ginsengs have been reported, the beneficial effect of joboksansam in cancer has not been elucidated. In this study, we investigated the in vivo and in vitro anticancer activities of joboksansam powder. Methods: To evaluate the in vivo anticancer activity of joboksansam, we established a xenograft mouse model bearing RMA cell-derived cancer. Direct cytotoxicity induced by joboksansam powder was also investigated in vitro using (3-4-5-dimethylthiazol-2-yl)-2-5-diphenyltetrazolium bromide (MTT) assay. The inhibitory activity of this powder on the activation of cell survival signaling involving Akt and Src was examined with immunoblot analysis. Results: Joboksansam powder displayed strong inhibitory activity against the increased tumor size, increased weight of total body and cancer tissues, and mortality of tumor-bearing mice. Joboksansam powder also suppressed the activation of survival regulatory enzymes Akt and Src, as assessed by phosphorylation levels in the immunoblot analysis of tumor tissues. Interestingly, the viability of RMA cells in vitro was directly decreased by joboksansam treatment. Conclusion: Overall, our results strongly suggest that joboksansam powder has the potential to protect against cancer generation by direct cytotoxic effects on cancer cells resulting from suppression of cell survival signaling.

Korean Red Ginseng water extract arrests growth of xenografted lymphoma cells

  • Background: Although numerous studies of the anticancer activities of Korean Red Ginseng (KRG) have been performed, the therapeutic effect of KRG on leukemia has not been fully elucidated. In this study, we investigated the antileukemia activities of KRG and its cellular and molecular mechanisms. Methods: An established leukemia tumor model induced by xenografted T cell lymphoma (RMA cells) was used to test the therapeutic activity of KRG water extract (KRG-WE). Direct cytotoxic activity of KRG-WE was confirmed by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay. The immunomodulatory activities of KRG-WE were verified by immunohistochemistry, nitric oxide production assay. The inhibitory effect of KRG-WE on cell survival signaling was also examined. Results: Orally administered KRG-WE reduced the sizes of tumor masses. Levels of apoptosis regulatory enzymes and cleaved forms of caspases-3 and -8 were increased by this extract. In addition, expression of matrix metalloproteinase-9, a metastasis regulatory enzyme, was decreased by KRG-WE treatment. The proportion of CD11c+ cells was remarkably increased in the KRG-treated group compared to the control group. However, KRG-WE did not show significant direct cytotoxicity against RMA cells. Conclusion: Our results strongly suggest that the KRG might have antileukemia activity through CD11c+ cell-mediated antitumor immunity.

쌍치 수위관측소의 수위-유량관계곡선 안정화를 위한 기초 연구 (A Basic Study of Stage-discharge Rating Stabilization at the Ssang-chi Gauging Station)

  • 쌍치 수위관측소는 과거 12년 동안 봉부자로 유량을 측정해왔다. 그러나 수위-유량관계곡선의 전이가 수용할 수 없을 만큼 너무 크다. 전이를 분석해 보기 위하여 주요인자들을 검토하였고, 특히 부자법에 의한 유량 산정을 위해서는 두 단면이 필요하나, 상 하단측선의 관측수위가 없어 수위관측소의 수위만을 사용하여 계산된 오류를 교정하였다. 이를 위해서 기준 수위-유량관계곡선을 작성하였다. 본 연구에서는 기준 수위-유량관계곡선에 대하여 RMA2모형으로 수면곡선을 모의하였다. 이를 바탕으로 기존유량을 재 산정하여 기존 수위-유량관계곡선을 재 작성하였다. 그 결과 전이의 변동 폭이 줄어들었으며 최근의 수위-유량관계곡선이 물리적으로 합당함을 확인하였다.

일체형 하천호안블럭의 개발 및 모형실험 적용 (Invention and Hydraulic Model Test of Combined Block System in River Bank Protection)

  • This research focused on analyzing and comparing between the results of hydraulic physical modeling and the results of numerical modeling of Grass Concrete which is newly developed in-situ block system. The physical model was built as a scale of 1:50 by Froude similitude measuring the water levels and the water velocities for before and after vegetation and the effects were analyzed after reviewing the results. In consequence, the water velocities were observed to decrease meanly 19.1%, and the water depth were determined to increase meanly 27.8% in case of the of design flood, $Q=200m^3/sec$. Moreover, the velocities were produced reduction effects of 27.2%, and the water levels were derived from addition effects of the highest 31.3% in case of the probability maximum flood(PMF), $Q=600m^3/sec$. To verifying the hydraulic physical modeling, the numerical modeling was conducted for a close examination of before and after vegetation. HEC-RAS model was for 1 dimensional numerical analysis and RMA-2 was for 2 dimensional numerical analysis. The results of the numerical simulation, under the condition of roughness coefficient calibration, shows similar results of the physical modeling. These satisfactory results show that the accomplished results of hydraulic modeling and the predicted results of numerical modeling corresponded reasonably each others.

수리조건을 이용한 생물서식처 적합도 지수 산정 -홍천강을 대상으로- (Estimation of Habitats Suitability Index based on Hydraulic Conditions)

  • 본 연구에서는 생물서식처에 적합한 조건을 가진 하천 구간을 선정 및 분석하기 위해 홍천강을 대상으로 생물 서식처 적합도 지수를 산정 하였다. 서식처 적합도 지수의 산정을 위해 수리특성으로 유속과 수심, 수질특성으로 수온, BOD, DO, TN. TP 총 7가지 항목에 대해 산정 하였고, 고려된 대상 생물상은 어류이다. 이를 위해 1차원 수리 모형(HEC-RAS), 2차원 하천 수리모형(RMA-2), 수질모형(QUAL2E), GIS를 이용하였다. 수리모형 입력 자료의 정확성을 높이기 위해 BASIN의 Delineation 기능을 적용해 대상유역을 분할한 후 비유량법으로 유량을 산정 하였으며, 산정된 유량을 입력하여 모형을 구축하고 검 보정 하였다. 산정결과 Reach 27에서 통합 서식처 적합도 지수의 평균이 0.782로 가장 높게 산정 되었고, 각 변수들의 평균은 유속 0.122 m/s, 수심 0.782 m, 수온 $14.3^{\circ}C$, BOD 0.68 mg/${\ell}$, DO 10.3 mg/${\ell}$, TN 2.4 mg/${\ell}$, TP 0.0121 mg/${\ell}$로 분석되었다.

고정보 단면형상에 따른 월류능력 분석 (Development of a culvert design model)

  • 최근 들어 국가하천의 주요구간에 대규모 보가 설치되고 있으며 설치된 고정보와 가동보는 형식에 따라 부분적으로 상이한 기능을 나타내나 기본적으로 상류의 수위가 일정하게 유지되도록 하여 하천 내 수자원을 확보하는데 기본적인 목적이 있다. 그러나 이로 인해 치수 및 하천환경에 많은 변화가 예상되므로 발생 가능한 하천관리의 문제점을 최소화하기 위해서는 하천정비에 따른 수리학적 영향을 검토할 필요가 있다. 본 연구에서는 4가지 형식의 고정보 본체의 형식별 월류능력을 수치해석모형인 Flow-3D 모형을 이용하여 검토하여 하천에서 고정보 설계시 단면형식 결정의 기초자료로 활용할 수 있도록 하였다. 이를 위해 선정된 고정보 단면의 형상에 따라 보 하류측 사면형태 및 경사별 월류량 검토를 수행하였으며 하류관리수위가 없을 때와 있을 때의 월류량과 하류부 흐름양상을 분석하였다. 고정보 단면 형식 선정을 위한 분석결과 적용된 4가지 유형중 선행한 월류능력별 수치해석 결과에서는 월류능력, 월류수맥의 수충, 구조체의 수축 팽창에 의한 불안정성 등을 종합적으로 고려할 때 오지형이 가장 적합한 구조인 것으로 검토되었다.

Powertrain에 의한 차량실내소음 예측을 위한 엔진 가진력 해석에 관한 연구 (Analysis of excitation forces for the prediction of the vehicle interior noise by the powertrain)

  • The objective of this paper is to get excitation forces of the engine. A powertrain geometry model is produced by CATIA and its FE model is made by MSC/Patran. A vibration mode analysis which makes us know the natural frequency and mode shape and a running mode analysis which measures the mode shape as a relative displacement about one reference point by measuring the acceleration of each bracket to take a place at the running vehicle are experimentally implemented. After getting a satisfied MAC value by doing a correlation about a measured mode analysis value and analyzed value through MSC/Nastran software, all components are assembled through MSC/ADAMS software which is a dynamic analysis tool. We can predict the vibration of brackets which is the last points to occur the force of the engine combustion by analyzing the combustion force produced by engine mechanism.

Utilizing Concept of Vegetation Freeboard Equivalence in River Restoration

  • The concept of vegetation freeboard equivalence (VFE) is presented from the comparison between the rise in stage with/without vegetation and the freeboard height under design discharge conditions. In South Korea, the freeboard height of large, medium and small rivers is defined as a function of river discharge. Two models are used for this analysis of flood stage with and without vegetation: the 1-D model HEC-RAS and the 2-D model RMA-2. Both models are applied to three river study sites of the Geum River in South Korea as representative sites for a large, a medium and a small river. The analysis shows that without vegetation, both models provide comparable results and the calculated results are in very good agreement with the design configuration. The vegetation effects on the medium river are less significant, and the freeboard is adequate to contain the rise in stage from the added floodplain vegetation in large rivers. The concept of vegetation freeboard equivalence is therefore useful for the analysis of flood river stages after the restoration of channels with increased floodplain vegetation.

한강수계 수질오염 자동측정망의 합리적인 측정지점 선정에 관한 조사연구 (Site Evaluation of Automated Monitoring Networks in Han River)

  • At present, automated monitoring of water quality in Han river has been operated at each water treatment plant. But the automated measurement sites must be choosen newly because water source in Seoul move to the upper stream of Chamshil weir. In this study, automated monitoring sites in Han river were reviewed, and the proper sites for automated monitoring of water quality have been selected by Qual2E model, RMA model, water sampling guidline, Sanders' method and topograpical characteristics of Han river in order to resonable operate. 8 sites have been selected as follows: (1) the site of immediately after Paldang drainage (2) the left site and a right site of $Gu{\check{u}}i$ water intake (3) the left site and a right site at Noryangjin(Han river bridge) (4) the site between Shingok weir and Anyangchun confluence point(Hangju bridge) (5) the site of Chungryangchun downstream(existence) (6) the site of Tanchun downstream(existence) (7) the site of Anyangchun downstream(existence) (8) the site of Wangsukchun downstream. The results proposed resonable operating management of network and economical system built up.

댐 건설이 하류하천 하상에 미치는 영향 분석 (Influence Analysis for Natural River Bed with Dam Construction)

  • 낙동강의 지류인 내성천의 회룡포는 섬 안의 섬으로 불리는 모래 백사장을 가지고 있는 매우 잘 알려진 관광명소이다. 그러나 상류 지역 댐의 건설이 계획되었다. 따라서 상류로 부터의 유사 유입이 차단되게 되었다. 이러한 상황에서 내성천의 지류로부터 유입되는 유사량 등을 통하여 회룡포의 모래 백사장에 미치는 영향을 1차원 및 2차원 모형을 사용하여 분석하였다. 유량-유사량은 댐의 유지유량과 지류에서 들어오는 유사량의 유역면적비에 의해서 산정된 값을 입력 자료로 활용 하였다. 1차원 모형은 HEC-6 모형을 사용하였고 2차원 모형은 SMS의 SED2D 모형을 사용하여 분석하였다. HEC-6 모형은 10년 전 하상에 데이터를 넣어 현재 하상과 가장 일치하는 공식인 Yang공식을 선정하여 현재 하상으로부터 20년 후를 모의하였다. SED-2D 모형은 현재 하상을 기준으로 20년 후를 모의하였고 두 모형 분석 결과 회룡포 구간에서는 동일한 양상이 나타나는 것으로 확인되었다.
