Lipid-lowering and hepatoprotective effects of Vitis vinifera dried seeds on paracetamol-induced hepatotoxicity in rats

  • BACKGROUND/OBJECTIVES: Red grape seeds as functional food are a good source of important bioactive components such as phenolics and antioxidants, which decrease oxidative stress that contributes to the pathogenesis of hepatotoxicity. The current study was conducted in order to evaluate the protective effect of red grape dried seeds (RGDS) on antioxidant properties, lipid metabolism, and liver and kidney functions of rats with paracetamol (750 mg/kg) induced hepatotoxicity. MATERIALS/METHODS: RGDS was added to the basal diet at 5, 10, and 20%. Thirty five adult male rats were assigned to five groups (n = 7) for a six-week feeding period; group (1) normal control, group (2) induced control, groups (3, 4, and 5) fed a diet with RGPS at different levels, 5, 10, and 20%, respectively. At the end of the feeding period, animals' blood and tissues were collected for estimation of serum lipid profile, serum liver, and kidney biomarkers. The protection was measured by detecting lipid peroxidation (LPO), glutathione (GSH), superoxide dismutase (SOD), Catalase (CAT) (in liver tissues), and liver histological examination. RESULTS: The results showed a significant (P < 0.05) decrease in levels of serum cholesterol, triglycerides, low density lipoprotein (LDL-C), and very low density lipoprotein (VLDL-C), with a significant increase in level of high density lipoprotein (HDL-C) for RGDS groups compared to induced control. Rats administered a diet containing RGDS levels produced significant (P < 0.05) hepatoprotection by decreasing the activities of liver enzymes, kidney parameters, and lipid peroxidation, while levels of GSH, SOD, and CAT were increased significantly to near the normal levels. CONCLUSION: The RGDS 20% group was more effective than others against hepatotoxicity of paracetamol, which may be attributed to RGDS total phenols and antioxidant contents, which were 1.438 mg and 1.231 mg, respectively.

  • 폴리머 표면에 생체물질의 고정화 방법은 생체적합성을 자닌 재료의 개발에있어서 중요한 방법중의 하나이다. 광반응을 이용한 photolithography방법을 사용하여 재료 표면의 원하는 부위에 단백질을 고정할 수 있다. 본 연구에서는 파이브로넥틴의 세포부탁 리간드, GRGDS펩타이드를 N-hydroxysuccinimidyl phenol azide를 이용하여 미세한 선의 형태로 표면에 광반응으로써 고정하였다. 광반응 유도체가 고정된 표면은 형광물질을 사용하여 확인하였다. 또한 혈관내피 세포는 GRGDS펩타이드가 고정화된 표면에서만 부탁됨을 관찰하였다.

흰쥐에서 RGDS tetrapeptide가 소 심낭 이식절편의 피하이식 후 석회화에 미치는 영향 (The Effects of RGDS Tetrapeptide on the Calcification of the Bovine Pericardium Transplanted Subcutaneously in Rats)

  • 배경: 모든 조직판막은 면역학적인 불활성화 및 내구성 강화를 위하여 이식 전 터치를 필요로 한다. 그러나 조직 판막은 이식 후 점차적인 석회화 과정을 보이며, 결국 판막의 기능을 상실하게 된다. 최근 석회화 과정의 병리기전에 관한 연구에서는 이식된 조직판막의 석회화 부위에서 정상 골성석회화에 관련하는 결합단백이 검출되기에 이르렀다. 본 실험은 이러한 결합단백의 공통 결합부인 RGD(Arg-Gly-Asp)를 갖는 RGDStetrapeptide를 이식 전 조직에 처치하여 이식 후 숙주의 골성석회화 관련 결합단백의 결합을 억제하는 것이 석회화과정에 영향을 줄 수 있는가를 확인하기 위하여 계획되었다. 대상 및 방법: 실험은 소 심낭을 0.6%글루타알데하이드에 처치한 후 이를 작은 절편으로 나누어 1군은 생리식염수에서 60분간 처리, 2 군은 RGDS와 같은 분자량을 갖는 0.5% GRSD tetrapeptide에서 60분간 처리, 3군은 0.5% RGDS에서 30분간 처리, 4군은 0.5% RGDS에서 60분간 처리, 5군은 0.5% RGDS에서 120분간 처리하였다. 이렇게 처리된 소 심낭절편을 흰쥐의 복부 피하에 이식하였으며, 30일 후 채취하여 방사선 검사와 생화학적 검사 및 조직학적 검사를 시행하여 이식절편의 석회화 정도를 판정하였다. 결과: 이식절편을 방사선 촬영 후, 측정한 음영도는 3, 4, 5군에서 48.00$\pm$3.57, 43.67$\pm$2.31, 42.58$\pm$2.47로 1, 2군의 68.42$\pm$3.06, 64.25+5.58과 통계학적인 차이를 보였다(p<7.05). 1군과 2군간의 유의한 차이는 없었다(p=0.105). 생화학적 검사에서 이식절편의 칼슘 총량은 1군 33.09$\pm$6.59mg, 2군 28.12+5.5mg,, 3군 25.42+7.67mg,, 4군 20.51$\pm$5.11mg, 5군 15.43$\pm$4.25mg을 보여, 5군이 1, 2, 3군에 비하여 통계적으로 유의하게 석회화 정도가 감소되어 있었다(<0.05). 1군과 2군간에 유의한 차이는 없었다(p=0.388). 조직학적 검사에서는 1, 2군에서 현저한 칼슘침착을 확인할 수 있었으며 3, 4군은 적은 양의 칼슘침착을 보여주는 반면, 5군에서는 뚜렷할 칼슘 침착 부를 확인할 수 없었다. 결론: 이상의 결과에서 이식 전 RGDS tetrapeptide를 처치하는 것이 흰쥐의 똑부 피하에 이식된 소 심낭절편의 석회화를 억제하는 효과가 있음을 확인할 수 있었다.

Cross-linkable and water-soluble phospholipid polymer as artificial extracellular matrix

  • The objective of this study is to prepare an artificial extracellular matrix (ECM) for cell culture by using polymer hydrogels. The polymer used is a cytocompatible water-soluble phospholipid polymer: poly[2-methacryloyloxyethyl phosphorylcholine (MPC)-n-butyl methacrylate-p-nitrophenyloxycarbonyl poly(ethylene oxide) methacrylate (MEONP)] (PMBN). The hydrogels are prepared using a cross-linking reaction between PMBN and diamine compounds, which can easily react to the MEONP moiety under mild conditions. The most favorable diamine is the bis(3-aminopropyl) poly(ethylene oxide) (APEO). The effects of cross-linking density and the chemical structure of cross-linking molecules on the mechanical properties of the hydrogel are evaluated. The storage modulus of the hydrogel is tailored by tuning the PMBN concentration and the MEONP/amino group ratio. The porous structure of the hydrogel networks depends not only on these parameters but also on the reaction temperature. We prepare a hydrogel with $40-50{\mu}m$ diameter pores and more than 90 wt% swelling. The permeation of proteins through the hydrogel increases dramatically with an increase in pore size. To induce cell adhesion, the cell-attaching oligopeptide, RGDS, is immobilized onto the hydrogel using MEONP residue. Bovine pulmonary artery endothelial cells (BPAECs) are cultured on the hydrogel matrix and are able to migrate into the artificial matrix. Hence, the RGDS-modified PMBN hydrogel matrix with cross-linked APEO functions as an artificial ECM for growing cells for applications in tissue engineering.

Evaluation of the platelet adhesiveness using a peptide-immobilized surface

  • The adhesion of stimulated and unstimulated platelet to fibrinogen requires the receptor binding site of GPIIb/IIIa. These recognition sites are existed in the Au chain(RGDS at positions 572-575 and RGDF at 95-98) and the carboxyterminal $\gamma$ chain (HHLGGAKQAGDV at 400-411) of fibrinogen. The unstimulated platelet does not adhered on the fragment E-coated surface containing RGDF sequence. In this study, we developed RGDF-immobilized surface to detect the functional state of platelet. RGDF-immobilized surface was prepared on the glass using photolithographic technology. Platelet adhesion to petide(RGDF)-immobilized surface was observed by the fluorescence microscope using mepacrine.

Microplate법에 의한 된장유래의 항혈전 펩타이드 탐색 (Screening of Antithrombotic Peptides from Soybean Paste by the Microplate Method)

  • 전통식품인 된장으로부터 혈소판 응집 저해활성을 갖는 항혈전 펩타이드를 탐색하고자 하였다. 증류수로 추출한 된장액을 한외 여과하여 얻은 분자량 3,000 dalton 이하의 여액에 대하여, SD흰쥐의 세척혈소판 (washed platelet)을 이용하여 ADP 자극으로 유도되는 혈소판응집의 저해정도를 aggregometer에서의 탁도로 측정하였을 때, $96\;{\mu}g/ml$로 처리시 90% 가량의 응집 저해효과가 확인되었다. 추출물을 Dowex 50W X-2 강양이온 교환칼럼을 이용하여 pyridine-formic acid 완충액의 pH (2.8-5.0) 및 몰농도(0.1-3.0M)를 단계별로 증가시키면서 용출시켜 19개의 획분(No.B-18)으로 분리하였다. 이들 획분의 활성을 microplate를 이용한 방법으로 측정하였을 때, 전체적으로 앞의 방법보다는 감도가 낮게 나타났으나 모든 분획에서 응집억제 효과가 확인되었다. 이때 용량반응 곡선으로부터 구한 각 획분의 $IC_{50}$$10-1,000\;{\mu}g/ml)$의 범위를 보였으며, 대부분의 획분들은 양성대조구인 RGDS $(IC_{50},\;205\;{\mu}g/ml)$보다 높은 활성을 나타내었다. 또한, 후반부의 염기성 획분$(IC_{50},\;10-20\;{\mu}g/ml)$에서 된장추출물$(IC_{50},\;30\;{\mu}g/ml)$보다 활성이 높게 나타났다. 이들 중 활성이 가장 높은 16번 획분을 아미노산 분석한 결과, 펩타이드 부분에 histidine, arginine, alanine 등이 높은 비율로 존재하였다.

  • The adhesion of activated and normal platelets to fibrinogen requires the receptor binding site of GPIIb/IIIa. These recognition sites exists in the A ${\alpha}$ chain(RGDS at 572-575 and RGDF at 95-98) and the carboxy-terminal of ${\gamma}$ chain (HHLGGAKQAGDV at 400-411) of fibrinogen. In this study, we developed RGDF-immobilized surface to detect the unctional state of platelet. RGDF-immobilized surface was prepared on the glass using photolithographic technology. Platelet adhesion to RGDF-immobilized surface was observed by staining platelets with mepacrine using a fluorescence microscope using mepacrine. Using the RGDF peptide of fragment E, we observed that the platelets pretreated with PGE1 interacted incompletely with RGDF-immobilized surface, whereas ADP activated platelets interacted with the surface extensively. These results show that the distinct selectivity of RGDF-immobilized micro-patterned surface can be used to detect the unctional state of platelets.

Ginseng Intestinal Bacterial Metabolite IH901 as a New Anti-Metastatic Agent

  • Anti-metastatic activities of IH901, an intestinal bacterial metabolic derivative formed from Ginseng protopanaxadiol saponins, was determined in vitro and in vivo. Under in vitro conditions, IH901 inhibited the migration of bovine aortic endothelial cells 25 times stronger than suramin and suppressed the invasion of HT1080 human fibrosarcoma cells into reconstituted basement membrane components of Matrigel 1000 times stronger than RGDS peptide. IH901 also showed inhibitory effect on type-IV collagenase secretion from HT 1080 cells and platelet aggregation. When the anti-metastatic activity of IH901 was evaluated in comparison with that of 5-FU using a spontaneous lung metastatic model of Lewis lung carcinoma, the administration of IH901 (10 mg/kg p. o.) to tumor-bearing mice led to a significant decrease in lung metastasis (43% of untreated control), which was slightly more effective than that obtained with 5-FU (56% of control). Thus, IH901 seems to exhibit its anti-metastatic activity partly through the inhibition of tumor invasion which results from the blockade of type IV collagenase secretion and also through anti-platelet and anti-angiogenic activities.
