Stress-Reducing Effects of Brown Rice Koji

  • The primary objective of this study is to determine whether a diet supplemented with brown rice koji (BRK) results in a reduced stress response in rats and mice. BRK, which has been suggested as a candidate for use as a stress- and fatigue-fighting supplement, was compared with red ginseng extract (RG) for its stress-reducing potential. The animals in this study were divided into no-stress, stress, RG, and BRK groups of 8 to 10 animals each. Stress was induced by means of immobilization (being restrained in plastic tubes for 30 min and electroshock (0.5 mA in mice or 2 mA in rats for 5 min). The no-stress group was not exposed to stress. Rats in the RG group received oral doses of 200 mg RG extract/kg body weight daily. The BRK group was fed a 30% BRK diet and exposed to stress. Animals were given supplements for 7 days before being exposed to stress, and then were given supplements for 5 days with exposure to stress. When the stress exposure ended, the animals were observed for stress-related changes in behavior and their plasma corticosterone levels were measured. BRK supplementation was associated with a partial blockade of the effects of stress on locomotion and elevated plus-maze test results in rats and mice. It was also associated with a partial reduction in stress-induced behaviors such as freezing, burrowing, smelling, face-washing, and rearing. BRK supplementation did not have a significant effect on plasma corticosterone levels, which were increased in the animals exposed to stress (p<0.01). The mice in the RG group received RG in water (2 mg RG/ mL $H_2O$), and the BRK group received a 30% BRK diet (weight) for 7 days. Both groups were evaluated for signs of fatigue. BRK supplementation increased endurance, as indicated by time on the rota-rod, in cold water, and on the horizontal wire. These results suggest that BRK supplementation partially protects the animal from the effects of stress and may also contribute to resistance to fatigue on physical exertion.

Changes in ginsenoside compositions and antioxidant activities of hydroponic-cultured ginseng roots and leaves with heating temperature

  • Background: This study evaluated changes in ginsenoside compositions and antioxidant activities in hydroponic-cultured ginseng roots (HGR) and leaves (HGL) with heating temperature. Methods: Heat treatment was performed at temperatures of $90^{\circ}C$, $110^{\circ}C$, $130^{\circ}C$, and $150^{\circ}C$ for 2 hours Results: The ginsenoside content varied significantly with heating temperature. The levels of ginsenosides Rg1 and Re in HGR decreased with increasing heating temperature. Ginsenosides F2, F4, Rk3, Rh4, Rg3 (S form), Rg3 (R form), Rk1, and Rg5, which were absent in the raw ginseng, were formed after heat treatment. The levels of ginsenosides Rg1, Re, Rf, and Rb1 in HGL decreased with increasing heating temperature. Conversely, ginsenosides Rk3, Rh4, Rg3 (R form), Rk1, and Rg5 increased with increasing heating temperature. In addition, ginsenoside contents of heated HGL were slightly higher than those of HGR. The highest extraction yield was 14.39% at $130^{\circ}C$, whereas the lowest value was 10.30% at $150^{\circ}C$ After heating, polyphenol contents of HGR and HGL increased from 0.43 mg gallic acid equivalent/g (mg GAE eq/g) and 0.74 mg GAE eq/g to 6.16 mg GAE eq/g and 2.86 mg GAE eq/g, respectively. Conclusion: Antioxidant activities of HGR and HGL, measured by 1,1-diphenyl-2-picrylhydrazyl and 2,2-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid radical scavenging ability, increased with increasing heating temperature. These results may aid in improving the biological activity and quality of ginseng subjected to heat treatments.

홍삼의 생체 내 사이토카인 분비에 대한 면역조절효과 (Immunomodulating Effects of Red Ginseng on the Regulation of Cytokine Release in vivo)

  • 홍삼의 6주간의 복강투여가 생체 내 면역계에 미치는 영향을 알아보기 위하여 면역계를 구성하는 주요 장기인 비장과 흉선의 무게변화와 성숙된 B세포와 T세포가 많이 분포되어 있는 비장세포를 배양하여 mitogen에 대한 비장세포의 세포증식능력을 연구하였다. 또한, 생체내 투여되는 홍삼이 면역반응의 매개역할을 하는 분비성 사이토카인의 조절 양상을 확인하기 위하여 생쥐의 혈청에서 분리한 사이토카인의 생성변화를 측정하였다. 연구결과 홍삼의 투여는 비장과 흉선의 무게를 증가시켰고, 비장세포내의 B세포와 T세포의 증식능력에도 유의적인 효과를 나타내었다. 혈청 내 분비된 T세포, B세포 및 대식세포가 분비하는 사이토카인의 농도변화에서도 홍삼 투여군은 면역계를 활성화시키는 IFN-${\gamma}$, TNF-${\alpha}$, IL-2, IL-6 및 IL-12의 분비량을 모두 증가시켰으며, 면역억제성 사이토카인으로 알려진 IL-10의 분비변화에는 영향을 미치지 않았다. 이와 같은 결과는 홍삼의 투여가 면역 반응계 전반을 조절하는 사이토카인의 생성에 영향을 미치며 모두 면역계를 활성화시키는 방향으로 작용시키는 것을 의미한다. 특히, 흉선과 비장의 무게지수가 증가되었다는 것은 세포증식 등의 변화에 영향을 미침으로써 면역계를 활성화시키는 것으로 생각되며, 특히 흉선의 무게 증가와 ConA에 대한 T세포 증식능력의 유의성 변화 및 T세포가 분비하는 IL-2, IFN-${\gamma}$ 등의 사이토카인의 증가는 홍삼의 생체 내 투여가 T세포의 활성화에 크게 영향을 미치는 것으로 사료된다.

전자혀를 이용한 수삼의 원산지 판별 (Discrimination of geographical origins of raw ginseng using the electronic tongue)

  • 전자혀 분석을 통해 국내산과 중국산 수삼(Panax ginseng C. A. Meyer)의 신속하고 정확한 판별법을 개발하고자 하였다. 전자혀를 이용하여 센서 감응도 값(raw data)을 분석한 뒤 taste screening하여 맛 스코어로 나타낸 결과, 국내산 수삼과 중국산 수삼은 감칠맛(UMS)에서 가장 큰 차이를 나타냈고 쓴맛(BRS)과 단맛(SWS)도 상대적으로 차이를 나타냈다. 국내산 수삼이 중국산 수삼에 비해 상대적으로 강한 감칠맛을 나타냈고, 쓴맛과 단맛은 그 반대로 나타났다. 판별함수분석 결과, DF1 (discriminant function first score) 상으로 국내산 수삼 그룹과 중국산 수삼 그룹이 구별되는 것을 볼 수 있었다. Discriminant power 값은 UMS 센서가 0.522으로 두 그룹에 가장 큰 차별성을 부여했다. CDA(canonical discriminant analysis) 분석 결과 두 수삼 그룹의 distance between centroids값은 2.7463으로 판별이 잘 된 것을 볼 수 있었다. 또한, 국내산 수삼 77점에서 67점이 국내산으로, 중국산 수삼은 77점 중 73점이 중국산으로 판별되어, 최종 판별정확도는 90.91%로 나타났다. 간단한 전처리 및 여러 가지 통계 처리를 통해 전자혀를 이용한 수삼의 원산지 판별이 가능하다고 판단되었으며, 신속하고 정확하게 판별을 수행할 수 있을 것으로 기대된다.

Effects of Ginseng Saponins in Energy Metabolism, Memory, and Anti-neurotoxicity

  • Ginseng has been used as a key constituent in traditional medicine prescriptions for centuries. Other than its well-known anti-stress and adaptogenic properties, ginseng has also been shown to be very effective in treating age-related deterioration in metabolic and memory functions. Although it is generally believed that the saponin (GS) fraction of the ginseng root accounts for the bioactivity of ginseng, a direct demonstration on which ginsenoside does what is still generally lacking. In the past decade, our laboratory has endeavored to identify the active GS components involved in energy metabolism, memory, and anti-neurotoxicity. To examine the ergogenic effects of GS in enhancing aerobic capacity, rats were subjected to either severe cold ($40^{\circ}C$ under helium-oxygen, two hours) or exercise workload $(70\%\;VO_{2}max,$ to exhaustion). Acute systemic injection (i.p.) of ginseng GS (5-20 mg/kg) significantly elevated both the total and maximum heat production in rats and improved their cold tolerance. However, pretreating the animal with the optimal dose (10 mg/kg) of GS devoid of $Rg_1\;and\;Rb_1$ failed to elicit any beneficial effects in improving cold tolerance. This indicates that either $Rb_1\;and/or\;Rg_1$ may be essential in exemplifying the thermogenic effect of GS. Further studies showed that only pretreating the animals with $Rb_1(2.5-5\;mg/kg),\;but\;not\;Rg_l,$ resulted in an increase in thermogenesis and cold tolerance. In contrast to the acute effect of GS on cold tolerance, enhancement of exercise performance in rats was only observed after chronic treatment (4 days). Further, we were able to demonstrate that both $Rb_1\;and\;Rg_1$ are effective in enhancing aerobic endurance by exercise. To illustrate the beneficial effects of GS in learning and memory, a passive avoidance paradigm (shock prod) was used. Our results indicated that the scopolamineinduced amnesia can be significantly reversed by chronically treating (4 days) the rats with either $Rb_1\;or\;Rg_1$ (1.25 - 2.5 mg/kg). To further examine its underlying mechanisms, the effects of various GS on ${\beta}-amyloid-modulated$ acetylcholine (ACh) release from the hippocampal slices were examined. It was found that inclusion of $Rb_1$ (0.1 ${\mu}M$), but not $Rg_1$, can attenuate ${\beta}-amyloid-suppressed$ ACh release from the hippocampal slices. Our results demonstrated that $Rb_1\;and\;Rg_1$ are the key components involved in various beneficial effects of GS but they may elicit their effects through different mechanisms.

Panax ginseng (Korea Red Ginseng) repairs diabetic sensorineural damage through promotion of the nerve growth factor pathway in diabetic zebrafish

  • Background: Diabetic sensorineural damage is a complication of the sensory neural system, resulting from long-term hyperglycemia. Red ginseng (RG) has shown efficacy for treatment of various diseases, including diabetes mellitus; however, there is little research about its benefit for treating sensorineural damage. Therefore, we aim to evaluate RG efficacy in alloxan-induced diabetic neuromast (AIDN) zebrafish. Methods: In this study, we developed and validated an AIDN zebrafish model. To assess RG effectiveness, we observed morphological changes in live neuromast zebrafish. Also, zebrafish has been observed to have an ultrastructure of hair-cell cilia under scanning electron microscopy. Thus, we recorded these physiological traits to assess hair cell function. Finally, we confirmed that RG promoted neuromast recovery via nerve growth factor signaling pathway markers. Results: First, we established an AIDN zebrafish model. Using this model, we showed via live neuromast imaging that RG fostered recovery of sensorineural damage. Damaged hair cell cilia were recovered in AIDN zebrafish. Furthermore, RG rescued damaged hair cell function through cell membrane ion balance. Conclusion: Our data suggest that RG potentially facilitates recovery in AIDN zebrafish, and its mechanism seems to be promotion of the nerve growth factor pathway through increased expression of topomyosin receptor kinase A, transient receptor potential channel vanilloid subfamily type 1, and mitogen-activated protein kinase phosphorylation.

Ginsenosides Rk1 and Rg5 inhibit transforming growth factor-β1-induced epithelial-mesenchymal transition and suppress migration, invasion, anoikis resistance, and development of stem-like features in lung cancer

  • Background: Lung cancer has a high incidence worldwide, and most lung cancer-associated deaths are attributable to cancer metastasis. Although several medicinal properties of Panax ginseng Meyer have been reported, the effect of ginsenosides Rk1 and Rg5 on epithelial-mesenchymal transition (EMT) stimulated by transforming growth factor beta 1 (TGF-β1) and self-renewal in A549 cells is relatively unknown. Methods: We treated TGF-β1 or alternatively Rk1 and Rg5 in A549 cells. We used western blot analysis, real-time polymerase chain reaction (qPCR), wound healing assay, Matrigel invasion assay, and anoikis assays to determine the effect of Rk1 and Rg5 on TGF-mediated EMT in lung cancer cell. In addition, we performed tumorsphere formation assays and real-time PCR to evaluate the stem-like properties. Results: EMT is induced by TGF-β1 in A549 cells causing the development of cancer stem-like features. Expression of E-cadherin, an epithelial marker, decreased and an increase in vimentin expression was noted. Cell mobility, invasiveness, and anoikis resistance were enhanced with TGF-β1 treatment. In addition, the expression of stem cell markers, CD44, and CD133, was also increased. Treatment with Rk1 and Rg5 suppressed EMT by TGF-β1 and the development of stemness in a dose-dependent manner. Additionally, Rk1 and Rg5 markedly suppressed TGF-β1-induced metalloproteinase-2/9 (MMP2/9) activity, and activation of Smad2/3 and nuclear factor kappa B/extra-cellular signal regulated kinases (NF-kB/ERK) pathways in lung cancer cells. Conclusions: Rk1 and Rg5 regulate the EMT inducing TGF-β1 by suppressing the Smad and NF-κB/ERK pathways (non-Smad pathway).

말레이시아 무슬림의 인삼·홍삼제품 인식과 태도 및 구매행동 (Malaysian Muslim's Awareness, Attitude and Purchasing Behavior of Ginseng and Red Ginseng Products)

  • 본 연구는 말레이시아 무슬림의 인삼 및 홍삼제품에 대한 인식과 태도 및 이용실태를 이해하기 위하여 실시되었다. 웹 기반 설문조사방법을 이용하여 말레이시아에 거주하는 성인 남녀 무슬림(n=200)을 대상으로 인삼·홍삼제품의 인지수준, 선호도와 선호요인, 효능에 대한 인지도, 구매현황 및 만족도 등을 조사하였다. 연구결과 말레이시아 무슬림의 인삼·홍삼제품 인지도는 각각 50%, 40%정도로 나타났다. 특히, 인삼·홍삼제품에 대한 인지도는 말레이시아 여성 또는 기혼 소비자에서 상대적으로 높았다. 섭취 경험이 가장 높게 나타난 제품유형은 인삼커피, 캔디 및 초콜릿유형이었으며, 특히 인삼커피는 40~50대, 기혼자가 상대적으로 많았다. 전체 응답자의 75%는 인삼·홍삼제품의 효능에 대하여 알거나 들어본 적이 있었으며, 특히, 40-50대에 비해 20-30대가 섭취하는 이유는 건강증진, 기분전환, 주위권유 등이 주요하며, 방송이나 지인추천을 통하여 효능을 알았고, 피로개선, 면역력 증진, 고혈압개선 효능에 대한 인지도가 높게 나타났다. 인삼·홍삼제품의 건강증진효능, 구매편의성, 맛과 향에 대한 만족도는 높았지만 가격이나 포장규격은 개선할 부분으로 나타났다. 지인 추천의향(82.6%)과 지속구입의향(83.5%)은 모두 높은 편이었다. 결론적으로 할랄 인삼·홍삼제품에 대한 말레이시아 소비자의 관심과 수요를 확인할 수 있었으며 향후 인삼·홍삼제품의 인지도를 향상시킬 수 있는 전략적인 제품개발과 마케팅이 필요하다고 판단된다.

Effects of Ginsenosides $Rg_3$ and $Rh_2$ OH the Proliferation of Prostate Cancer Cells

  • Ginseng has an anti-cancer effect in several cancer models. This study was to characterize active constituents of ginseng and their effects on proliferation of prostate cancer cell lines, LNCaP and PC3. Cell proliferation was measured by $[^3H]$thymidine incorporation, the intracellular calcium concentration by a dual-wavelength spectrophotometer system, effects on mite-gen-activated protein (MAP) kinases by Western blotting, and cell attachment and morphologic changes were observed under a microscope. Among 11 ginsenosides tested, ginsenosides $Rg_3\;and\;Rh_2$ inhibited the proliferation of prostate cancer cells. $EC_{50}s\;of\;Rg_3\;and\;Rh_2$ on PC3 cells were $8.4{\mu}M\;and\;5.5{\mu}M$, respectively, and $14.1{\mu}M\;and\;4.4{\mu}M$ on LNCaP cells, respectively. Both ginsenosides induced cell detachment and modulated three modules of MAP kinases activities differently in LNCaP and PC3 cells. These results suggest that ginsenosides $Rg_3\;and\;Rh_2$-induced cell detachment and inhibition of the proliferation of prostate cancer cells may be associated with modulation of three modules of MAP kinases.

인삼 잎 줄기 추출물의 초단파 및 식초 처리에 의한 인삼 사포닌 성분 변화 (The Change of Ginsenoside Composition in Ginseng Leaf and Stem Extract by the Microwave and Vinegar Process)

  • The purpose of this study was to develop a new preparation process of ginseng extract with the high concentration of prosapogenin, the specific component in Red ginseng. Chemical transformation from the ginseng saponin glycosides to the prosapogenin was analyzed by the HPLC. The extracts of ginseng leaf and stem were processed at the several treatment conditions of the microwave and vinegar(about 14% acidity). MGLS-20 findings show that the ginseng leaf and stem extracts that had been processed with microwave and vinegar for 20 minutes peaked in the level of ginsenoside $Rg_3$(0.906%). MGLS-25 peaked in the level of ginsenoside $Rg_5$(0.329%) in the ginseng leaf and stem extract processed with microwave and vinegar for 25 minute. And the other kinds of ginseng prosapogenin did not show a higher content.
