Selection of High Laccase-Producing Coriolopsis gallica Strain T906: Mutation Breeding, Strain Characterization, and Features of the Extracellular Laccases

  • Commercial application of laccase is often hampered by insufficient enzyme stocks, with very low yields obtained from natural sources. This study aimed to improve laccase production by mutation of a Coriolopsis gallica strain and to determine the biological properties of the mutant. The high-yield laccase strain C. gallica TCK was treated with N-methyl-N-nitro-N-nitrosoguanidine and ultraviolet light. Among the mutants isolated, T906 was found to be a high-production strain of laccases. The mutant strain T906 was stabilized via dozens of passages, and the selected ones were further processed for optimization of metallic ion, inducers, and nutritional requirements, which resulted in the optimized liquid fermentation medium MF9. The incubation temperature and pH were optimized to be 30℃ and 4.5, respectively. The mutant strain T906 showed 3-times higher laccase activity than the original strain TCK under optimized conditions, and the maximum laccase production (303 U/ml) was accomplished after 13 days. The extracellular laccase isoenzyme 1 was purified and characterized from the two strains, respectively, and their cDNA sequence was determined. Of note, the laccase isoenzyme 1 transcription levels were overtly increased in T906 mycelia compared with values obtained for strain TCK. These findings provide a basis for C. gallica modification for the production of high laccase amounts.

Wind and solar energy: a comparison of costs and environmental impacts

  • This study is concerned with the analysis of two renewable technologies for electric energy production: wind energy and photovoltaic energy. The two technologies were assessed and compared by economic point of view, by using selected indicators characterized by a clear calculation approach, requirement of information easy to be collected, clear, but even complete, interpretation of results. The used economic indicators are Levelized Cost of Energy, $CO_2$ abatement cost and fossil fuel saving specific cost; these last two specifically aimed at evaluating the different capabilities that renewable technologies have to cut down direct $CO_2$ emissions and to avoid fossil fuel extraction. The two technologies were compared also from the environmental point of view by applying Life Cycle Assessment approach and using the environmental impact categories from the Eco-indicator'95 method. The economic analysis was developed by taking into account different energy system sizes and different geographic areas in order to compare different European conditions (Italy, Germany and Denmark) in term of renewable resource availability and market trend. The environmental analysis was developed comparing two particular types of PV and wind plants, respectively residential and micro-wind turbine, located in Italy. According to the three calculated economic indicators, the wind energy emerged as more favorable than PV energy. From the environmental point of view, both the technologies are able to provide savings for almost all the considered environmental impact categories. The proposed approach, based on the use of economic and environmental indicators may be useful in supporting the policies and the decision making procedures concerned with the promotion and use of renewables, in reference to the specific geographic, economic and temporal conditions.

  • Simulations have been used to evaluate the efficiency of logistics or manufacturing systems and predict the outcomes of the systems. New simulation models are needed to evaluate new alternative plants and layouts during the resource design process. Although it is easy to handle minor changes in parameters by modifying a simulation model, it takes considerable time and effort for simulation modelers to alter the layout, which involves changes in many simulation sub-models. Therefore, this study proposes a method to transfer information in AutoCAD layout to the simulation model automatically. This study also defines a standard document as an Excel Form, and suggests a method to transfer information on the basic layouts, processes, resources, and workers to a simulation model automatically. A simulation tool, called Tecnomatix Plant Simulation 9.0, was used for this study. The proposed approach in this study was applied to a semiconductor wafer factory for a case study.

  • This study was carried out to construct accurate and scientific system of assessment indicators in selection of National Trust conservation areas, which was new concept of domestic environment movement and offer the raw data of new analytic method by introducing the fuzzy theory and weight for overcoming the uncertainty of ranking decision. To transform the Likert's scale granted to assessment indicators into the type of triangular fuzzy number(a, b, c), there was conversion to each minimum(a), median(b), and maximum(c) in applying membership function, and in using the center of gravity and eigenvalue, there was to decide the ranking. The rankings of converted values applied a mean importance and weight were confirmed that they were generally changed. Therefore, the ranking decision was better to accomplish objective and rational ranking decision by applying weight that was calculated in grouping of indicator than to judge the singular concept and to be useful in assessment of diverse National Trust site. In the future, because AHP, which was general method of calculating weight, was lacked, there was to understand the critical point to fix a pertinent weight, and to carry out the study applying engineering concept like fuzzy integral using $\lambda-measure$.

Tariff Reduction and Within-Plant Productivity: Micro-evidence from Korean Manufacturing (수입관세 인하가 기업 생산성에 미치는 효과 분석)

  • This paper empirically investigates the effects of import tariff on within-plant productivity growth in Korean manufacturing, using the detailed plant-level longitudinal data of the Korea Census of Manufacturers for the period of 1993-2003. Our main findings are as follows: First, the productivity changes of Korean manufacturing for the period under analysis were mostly induced by within-plant productivity gains, rather than within-industry and/or between-industry resource reallocations. Second, after controlling for firm-specific heterogeneity, the estimation results indicate that lowering tariff-barriers has a positive impact on within-plant TFP growth. We interpret the results in a way that trade liberalization through the removal of tariff and non-tariff barriers heightens the competitive pressure, which in turn creates incentives to reduce production and managerial inefficiency and to invest more on innovative activities. Third, we also find that plant productivity growth from reducing tariff barriers is particularly conspicuous within a year after tariff changes, which implies that plants are quickly adjusting to heightened import competition. On the other hand, our results show that the trade effect on employment creation proceeds relatively slow.

  • For environmental reasons and because of our limited energy resources, high-efficiency power generation technology will be necessary in the future. Ultra-supercritical (USC) power generation technology is the key to managing the greenhouse gas problems and energy resource problems discussed in the Kyoto Protocol to the United Nations Framework Convention on Climate Change. Other countries and manufacturers are trying to build commercial power plants. In this paper, an efficient method of achieving near-zero emission operation of a high-efficiency fossil power plant using USC power generation is discussed. Development of USC power generation in Korea has been supported by the Korean government in two phases: Phase I was USC key technology development from 2002 to 2008, and Phase II is USC development and technology optimization from 2010 to 2017.

  • The 2011 Japanese Society of Turfgrass Science was divided in oral presentation, sectional meeting and small symposium, and the total 55 subject was announced. The oral presentation was reported physiological ecology of turf, disease and weed; field management; pesticidal activity and spectrum of new turf pesticides. Above all, there was presentation that investigated control method of necrotic ring spot of new turf disease and Cyperus brevifolius, annual bluegrass and mouse-ear chickweed. In addition, the control effect of new registration fungicide (Furametpyr) and herbicides (S-metolachlor, Flucetosulfuron and Foramsulfuron) were announced. There was various information interchange through 4 kinds of subjects of a golf course, ground turf, a urban park greens and ground cover plants greening by the sectional meeting, and the genetic resource of turfgrass discussed by small symposium. Specially, park greens sectional meeting was announced about the radioactive material contaminated on mechanism and effectively removal method of the radioactivity contaminant from public park, school ground and domestic lawn.

Production of liquid fertilizer from broken eggs and evaluation of its effect on lettuce growth

  • Eggs are likely to be used in agriculture because they can provide enough nutrients for crop growth. Statistics show that a large number of eggs are lost due to breakage before reaching the final consumer. The purpose of this study was to make a natural liquid fertilizer as a substitute for chemical fertilizers using broken eggs as a resource and to evaluate the efficiency of the formulated fertilizer. To make the liquid fertilizer, the broken eggs and distilled water were mixed at ratios of 6 : 4 and 4 : 6. Then, effective micro-organisms (EM) and sugar were added, and the mixture was fermented. The temperature and electrical conductivity (EC) increased gradually with the fermentation while the pH decreased. When evaluated following the seed germination index method of the compost, it was found that the fertilizer matured 10 days after the beginning of the experiment. The growth experiment was conducted with lettuce in which the fermented liquid fertilizer was compared with a commercial liquid fertilizer. The 6 : 4 treatment produced plants with the densest fresh shoot and roots weighing 41.6 and 4.6 g, respectively. The number of leaves (12.3 per plant) was also the highest for the 6 : 4 treatment. Soil analysis showed that the soil pH was improved, and the soil organic matter was increased in the fermented liquid fertilizer treatment.

  • The most powerful energy resource in nature is solar energy which becomes directly converted to electric power in worldwide. Most of the photovoltaic power plants are commonly installed on sunny side of the ground. Thus the installation of photovoltaic power plant could produce an unexpected adverse effect by sacrificing the productivity from green field or forest. To avoid these adverse effect floating photovoltaic plant has been devised and installed on inland reservoir. The photovoltaic plant could utilize ignored water surface without sacrificing the productivity of the ground. Additionally the photovoltaic efficiency has been reenforced by the cooling effect induced by the circulating air flow from water surface. The floating photovoltaic plant could be furnished solar tracking ability by tilting the system operated with the aid of the ballast system. This report is provided to introduce the design of the floating structure with solar panel which furnished solar tracking ability with the aid of ballast system.

Development of Anti-Obesity Agent from Resource Plants

  • Obesity is a physical condition that results from excessive storage of fat in the body. The present study examined the anti-obesity effects of the selected natural medicine, Galla rhois extract (GRE) and solvent fractions on 3T3-L1 preadipocytes and in vivo studies. Here, we show that EtOAc fraction of Galla rhois inhibits the differentiation of the 3T3-L1 preadipocytes induced by differentiated medium in a dose-dependent manner. To investigate the effect of the GRE-EtOAc fraction on obesity in high fat diet-fed C57BL/6 mice, which included a normal diet (ND), high-fat diet (HFD) and HFD+GRE concentration-dependent, were fed to the mice for 6 weeks. The GRE-EtOAc fraction was inhibited the highest adipocyte differentiation in vitro, the GRE supplement significantly decreased body weight and visceral fat mass compared to the HFD group. The total cholesterol and triglyceride levels in the plasma were significantly decreased by GRE supplementation compared with those of the HFD group. Also, we aimed to determine the differentiation inhibition and the modulation of differentiation genes brought about by the Galla rhois in adipocyte. A cDNA microarray-based method was introduced for the high contents screening (HCS) of gene expressions. This technology has revolutionized gene expression studies by providing the means to measure mRNA levels in thousands of genes simultaneously in simple and complex biological samples. 13 genes were founded to be affected in their expression levels by more than 5-fold up-regulation after 4 days treatment with the EtOAc fraction from Galla rhois. Otherwise, 21 genes were founded to be affected in their expression levels by more than 5-fold down-regulation treated with the EtOAc fraction. Therefore, Galla rhois extract may be considered for use in a therapeutic agent to control obesity.
