Critical Adjuvant Influences on Preventive Anti-Metastasis Vaccine Using a Structural Epitope Derived from Membrane Type Protease PRSS14

  • We tested how adjuvants effect in a cancer vaccine model using an epitope derived from an autoactivation loop of membrane-type protease serine protease 14 (PRSS14; loop metavaccine) in mouse mammary tumor virus (MMTV)-polyoma middle tumor-antigen (PyMT) system and in 2 other orthotopic mouse systems. Earlier, we reported that loop metavaccine effectively prevented progression and metastasis regardless of adjuvant types and TH types of hosts in tail-vein injection systems. However, the loop metavaccine with Freund's complete adjuvant (CFA) reduced cancer progression and metastasis while that with alum, to our surprise, were adversely affected in 3 tumor bearing mouse models. The amounts of loop peptide specific antibodies inversely correlated with tumor burden and metastasis, meanwhile both TH1 and TH2 isotypes were present regardless of host type and adjuvant. Tumor infiltrating myeloid cells such as eosinophil, monocyte, and neutrophil were asymmetrically distributed among 2 adjuvant groups with loop metavaccine. Systemic expression profiling using the lymph nodes of the differentially immunized MMTV-PyMT mouse revealed that adjuvant types, as well as loop metavaccine can change the immune signatures. Specifically, loop metavaccine itself induces TH2 and TH17 responses but reduces TH1 and Treg responses regardless of adjuvant type, whereas CFA but not alum increased follicular TH response. Among the myeloid signatures, eosinophil was most distinct between CFA and alum. Survival analysis of breast cancer patients showed that eosinophil chemokines can be useful prognostic factors in PRSS14 positive patients. Based on these observations, we concluded that multiple immune parameters are to be considered when applying a vaccine strategy to cancer patients.

Mathematical Modelling of Phenol Desorption from Spent Activated Carbon by Acetone (활성탄에 흡착된 페놀의 아세톤 탈착 모델에 대한 연구)

  • This research was designed to investigate the mathematical model and kinetics of phenol desorption from spent activated carbon. elucidating the desorption characteristics of phenol in the case of using acetone. The Freundlich isotherm constant ($k_e$) is expressed as a function of temperature: $k_e(T)=0.1exp(797.297/T)$. The Freundlich isotherm constant(n) is a weak temperature function and is rarely affected by temperature below $50^{\circ}C$. whereas it is necessary to correct the n value with respect to temperature above $100^{\circ}C$ owing to significant deviation (~5%). Based on the assumption that the surface desorption reaction of phenol is rate limiting, the desorption model was developed. Desorption reaction constant($k_d$) was determined by means of fitting the theoretical results best to experimental ones. The Arrhenius relationships for $k_d$ was expressed by: $k_d(sec^{-1})=0.0479{\cdot}exp(-3037/T)$. The model was verified by comparing the experimental ones under different reaction conditions with the theoretical results determined by the previously estimated $k_d$. Since the difference between them is with 5%, it is expected that the desorption model of this research seems to be appropriate to explain the desorption of phenol from activated carbon by acetone. According to studies of the model. regeneration time and ratio was estimated as a function of temperature under present conditions as follows: (1) regeneration time : ${\tau}_{reg}(hr)=-0.08130T_c+8.4775$. (2) regeneration ratio : ${\eta}(%)=0.2210T_c+83.745$. The regeneration time at 15, 55, and $100^{\circ}C$. respectively. was 7, 4.2, and 0.35 hours, whereas the regeneration ratio was 87. 96. and 99%. respectively. Also. studies of the model would make it possible to determine the regeneration time and ratio under other specific conditions (temperature, applied acetone volume, amount of activated carbon, and initially adsorbed phenol amount).

Fertility Evaluation of Upland Fields by Combination of Landscape and Soil Survey Data with Chemical Properties in Soil (토양 화학성과 지형 및 토양 조사자료를 활용한 밭 토양의 비옥도 평가)

  • Evaluation method of soil fertility by application of geographic information system (GIS) which includes landscape characteristics and soil map data was investigated from productivities of red pepper and tobacco grown on the fields with no fertilization. Total 131 fields experiments, 64 fields of red pepper and 67 fields of tobacco were conducted from 22 and 23 fields for red pepper and tobacco, respectively, located at Cheangweon and Eumseong counties in 1996, from 20 and 25 fields at Boeun and Goesan counties in 1997, and 22 and 19 fields at Jincheon and Chungju counties in 1998. All the experimental sites were selected on the basis of wide range of distribution in landscape and soil attributes. Dry weights and nutrients (N, P and K) uptakes by red pepper plant and tobacco leaves were considered as basic fertility of the soil (BFS). The BFS was estimated by twenty-five independent variables including 13 chemical properties and 12 GIS data. Twenty-five independent variables were classified by two groups, 15 quantitative variables and 10 qualitative variables, and were analyzed by multiple linear regression (MLR) of REG and GLM models of SAS. Dry weight of red pepper (DWRP) and dry weight of tobacco leaves (DWTL) every year showed high variations by five times in difference plots with minimum yield and maximum yield indicating the diverse soil fertility among the experimental fields. Evaluation for the BFS by the MLR including independent variables was better than that by simple regression showing gradual improvement by adding chemical properties, quantitative variables, and qualitative variables of the GIS. However the evaluation for the BFS by the MLR showed the better result for tobacco than red pepper. For example the variability in the DWTL by MLR was explained 34.2% by only chemical properties, 35.0% by adding quantitative variables, and 72.5% by adding both the quantitative and qualitative variables of the GIS compared with 21.7% by simple regression with $NO_3-N$ content in soil. Consequently, it is assumed that this approach by the MLR including both the quantitative and qualitative variables was available as an evaluation model of soil fertility for upland field.

  • In this study, a seismic behavior of electrical cabinet system in Nuclear Power Plants(NPPs) was evaluated by the shaking table test. A 480V Motor Control Centers(MCCs) was selected for the shaking table test, and a real MCC cabinet for the Korea Nuclear Power Plant site was rented by manufactured company. For the shaking table tests, three kinds of seismic input motions were used, which were a US NRC Reg. guide 1.60 design spectrum, a UHS spectrum and PAB 165' floor response spectrum(FRS). Especially, the UHS input motion was selected for an evaluation of structural seismic amplification effects, three directional accelerations were measured at three points outside on the cabinet system and also that of the incabinet response amplification, accelerations were measure at two points which were mounted in electrical equipment such as relay. Seismic amplification effect is determined at the outside and inside of a cabinet as input seismic motion, and compared to the results which are calculated by analytical method based on NUREG/CR-5203.

Breeding of Doritaenopsis 'Hwasu 5205' with Vivid Red and Large Flowers (선명한 적색 대륜계 호접란 '화수 5205' 육성)

  • A new Doritaenopsis cultivar 'Hwasu 5205' was bred by Kyungpook National University, Korea, which produces young plants through tissue culture techniques. The new cultivar 'Hwasu 5205', showing the phenotype of vivid red and large flower type characteristics, was derived from crossing between Phalaenopsis Happy Valentine and Doritaenopsis Happy Rose. An elite individual, number '02-05-205' later named as 'Hwasu 5205', was selected among about 300 individual progenies after more than 2 years of intensive selection covering vegetative and flowering distinctiveness. In year 2004-2005, 1st and 2nd characteristic analyses were carried out through performance and uniformity tests. 'Hwasu 5205' produces vivid red (RHS #PN78B) flowers of i ncurved type with large size, of 9.2 and 12.0 cm in flower height and width, respectively. Leaves of 'Hwasu 5205' grow horizontally and are about 24.3cm in length and 8.5cm in width, respectively. This cultivar possesses no genetic variation. It can be propagated rapidly in vitro and is easy to grow due to its vigorous growth habit. 'Hwasu 5205' was registered (Reg. #: 2915) to Korea Seed & Variety Service (KSVS) on 1st December, 2009 and the PBR(plant breeder's right)is currently controlled by Sangmiwon Orchid Company, Korea.

Insights into the Role of Follicular Helper T Cells in Autoimmunity

  • Follicular helper T ($T_{FH}$) cells are recently highlighted as their crucial role for humoral immunity to infection as well as their abnormal control to induce autoimmune disease. During an infection, na$\ddot{i}$ve T cells are differentiating into $T_{FH}$ cells which mediate memory B cells and long-lived plasma cells in germinal center (GC). $T_{FH}$ cells are characterized by their expression of master regulator, Bcl-6, and chemokine receptor, CXCR5, which are essential for the migration of T cells into the B cell follicle. Within the follicle, crosstalk occurs between B cells and $T_{FH}$ cells, leading to class switch recombination and affinity maturation. Various signaling molecules, including cytokines, surface molecules, and transcription factors are involved in $T_{FH}$ cell differentiation. IL-6 and IL-21 cytokine-mediated STAT signaling pathways, including STAT1 and STAT3, are crucial for inducing Bcl-6 expression and $T_{FH}$ cell differentiation. $T_{FH}$ cells express important surface molecules such as ICOS, PD-1, IL-21, BTLA, SAP and CD40L for mediating the interaction between T and B cells. Recently, two types of microRNA (miRNA) were found to be involved in the regulation of $T_{FH}$ cells. The miR-17-92 cluster induces Bcl-6 and $T_{FH}$ cell differentiation, whereas miR-10a negatively regulates Bcl-6 expression in T cells. In addition, follicular regulatory T ($T_{FR}$) cells are studied as thymus-derived $CXCR5^+PD-1^+Foxp3^+\;T_{reg}$ cells that play a significant role in limiting the GC response. Regulation of $T_{FH}$ cell differentiation and the GC reaction via miRNA and $T_{FR}$ cells could be important regulatory mechanisms for maintaining immune tolerance and preventing autoimmune diseases such as systemic lupus erythematosus (SLE) and rheumatoid arthritis (RA). Here, we review recent studies on the various factors that affect $T_{FH}$ cell differentiation, and the role of $T_{FH}$ cells in autoimmune diseases.

Antioxidant Effect of Korean Red Ginseng Extract on Aqueous Linoleic Acid and LDL (수용성계의 Linoleic Acid와 LDL에 대한 한국산 홍삼의 산화방지효과)

  • The antioxidant activities of Korean Red Ginseng Extract(RGE) and ${\alpha}-tocopherol$, as reference compound, were tested with HPLC and fluorometry which measure the MDA after reacting an aqueous 1% linoleic acid buffer solution, and LDL(1 mg protein/ml) buffer solution with $H_2O_2{\;}and{\;}FeCl_2$. The generation of conjugated-diene in LDL(0.25 mg protein/ml) was also measured by spectrometry. MDA determination showed the antioxidant effect on linoleic acid oxidation with oxidation inhibition ratio of 71.8% and 76.1%, respectively, by addition of 1000 ppm RGE and 100 ppm ${\alpha}-tocopherol$. LDL(1 mg protein/ml) oxidation was inhibited by 25.2% and 21.2%, respectively, by addition of 200 ppm REG and 100 ppm ${\alpha}-tocopherol$. The generation of conjugated diene in LDL(0.25 mg protein/ml) was also inhibited by 44.2%, by addition of 50 ppm RGE.

DEVELOPMENT OF AN AMPHIBIOUS ROBOT FOR VISUAL INSPECTION OF APR1400 NPP IRWST STRAINER ASSEMBLY

  • An amphibious inspection robot system (hereafter AIROS) is being developed to visually inspect the in-containment refueling storage water tank (hereafter IRWST) strainer in APR1400 instead of a human diver. Four IRWST strainers are located in the IRWST, which is filled with boric acid water. Each strainer has 108 sub-assembly strainer fin modules that should be inspected with the VT-3 method according to Reg. guide 1.82 and the operation manual. AIROS has 6 thrusters for submarine voyage and 4 legs for walking on the top of the strainer. An inverse kinematic algorithm was implemented in the robot controller for exact walking on the top of the IRWST strainer. The IRWST strainer has several top cross braces that are extruded on the top of the strainer, which can be obstacles of walking on the strainer, to maintain the frame of the strainer. Therefore, a robot leg should arrive at the position beside the top cross brace. For this reason, we used an image processing technique to find the top cross brace in the sole camera image. The sole camera image is processed to find the existence of the top cross brace using the cross edge detection algorithm in real time. A 5-DOF robot arm that has multiple camera modules for simultaneous inspection of both sides can penetrate narrow gaps. For intuitive presentation of inspection results and for management of inspection data, inspection images are stored in the control PC with camera angles and positions to synthesize and merge the images. The synthesized images are then mapped in a 3D CAD model of the IRWST strainer with the location information. An IRWST strainer mock-up was fabricated to teach the robot arm scanning and gaiting. It is important to arrive at the designated position for inserting the robot arm into all of the gaps. Exact position control without anchor under the water is not easy. Therefore, we designed the multi leg robot for the role of anchoring and positioning. Quadruped robot design of installing sole cameras was a new approach for the exact and stable position control on the IRWST strainer, unlike a traditional robot for underwater facility inspection. The developed robot will be practically used to enhance the efficiency and reliability of the inspection of nuclear power plant components.

Fertility Evaluation of Tobacco Field by Quantitative and Qualitative Characteristics of Soils (토양의 정량적 및 정성적 특성을 이용한 연초 경작지의 비옥도 평가)

  • Evaluation method of soil fertility by combination of soil color characteristics and survey data from soil map as well as chemical properties was investigated on total 35 field and pot experiments. Total 35 tobacco fields including 11 fields located at Cheonweon county in Chungnam Province, 9 fields located at Goesan county in Chungbuk Province, and 15 fields located at Youngcheon county in Kyongbuk Province were selected in 1984 to cover the wide range of distribution in landscape and soil attributes. Yields of tobacco grown on the plots of both the pot and field experiment which were not applied with any fertilizer were considered as basic fertility of the soil (BFS). The BFS was estimated by 32 independent variables including 15 chemical properties, 3 color characteristics, and 14 soil survey data from soil map. Twenty-four independent variables containing 16 quantitative variables selected from 24 quantitative variables by collinearity diagnostics and 8 qualitative variables, were classified and analyzed by multiple linear regression (MLR) of REG and GLM models of SAS. Tobacco yield of field experiment showed high variations by eight times in difference between minimum and maximum yield indicating the diverse soil fertility among the experimental fields. Evaluation for the BFS by the MLR including quantitative variables was still more confidential than that by a single index and that showed more improvement of coefficient of determination ($R^2$) in pot experiment than in field experiment. Evaluation for the BFS by MLR in field experiment was still improved by adding qualitative variables as well as quantitative variables. The variability in the BFS of field experiment was explained 43.2% by quantitative variables and 67.95% by adding both the quantitative and qualitative variables compared with 21.7% by simple regression with NO$_3$-N content in soil. The regression evaluation for the best evaluation of the BFS of field experiment by MLR included NO$_3$-N content, L value, and a value of soil color as quantitative variables and available soil depth and topography as qualitative variables. Consequently, it is assumed that this approach by the MLR including both the quantitative and qualitative variables was available as an evaluation model of soil fertility for tobacco field.

Predicting the Potential Distribution of Korean Pine (Pinus koraiensis) Using an Ensemble of Climate Scenarios (앙상블 기후 시나리오 자료를 활용한 우리나라 잣나무림 분포 적지 전망)

  • Preparations need to be made for Korean pine(Pinus koraiensis) in anticipation of climate change because Korean pine is an endemic species of South Korea and the source of timber and pine nut. Therefore, climate change adaptation policy has been established to conduct an impact assessment on the distribution of Korean pine. Our objective was to predict the distribution of Korean pine while taking into account uncertainty and afforestation conditions. We used the 5th forest types map, a forest site map and BIOCLIM variables. The climate scenarios are RCP 4.5 and RCP 8.5 for uncertainty and the climate models are 5 regional climate models (HadGEM3RA, RegCM4, SNURCM, GRIMs, WRF). The base period for this study is 1971 to 2000. The target periods are the mid-21st century (2021-2050) and the end of the 21st century (2071-2100). This study used the MaxEnt model, and 50% of the presences were randomly set as training data. The remaining 50% were used as test data, and 10 cross-validated replicates were run. The selected variables were the annual mean temperature (Bio1), the precipitation of the wettest month (Bio13) and the precipitation of the driest month (Bio14). The test data's ROC curve of Korean pine was 0.689. The distribution of Korean pine in the mid-21st century decreased from 11.9% to 37.8% on RCP 4.5 and RCP 8.5. The area of Korean pine at an artificial plantation occupied from 32.1% to 45.4% on both RCPs. The areas at the end of the 21st century declined by 53.9% on RCP 4.5 and by 86.0% on RCP 8.5. The area of Korean pine at an artificial plantation occupied 23.8% on RCP 4.5 and 7.2% on RCP 8.5. Private forests showed more of a decrease than national forests for all subsequent periods. Our results may contribute to the establishment of climate change adaptation policies for considering various adaptation options.
