Relationship Linking Dietary Quercetin and Roughage to Concentrate Ratio in Feed Utilization, Ruminal Fermentation Traits and Immune Responses in Korean Indigenous Goats

  • A total of nine Korean indigenous goats were used in a cross-over arrangement to give nine replicates per treatment, and they were housed individually assigned to 1 of 9 dietary treatments. Nine treatments were 0, 500, and 1000 ppm of quercetin supplementation in diets by mixing roughage and concentrate with different ratios (RC ratio) of 3:7 (RC 30), 5:5 (RC 50) and 7:3 (RC 70). Nutrient utilizations of dry matter, crude fat and NDF were not affected by neither RC ratio nor dietary quercetin (p>0.05), but the rate of crude protein and ADF increased in animals in RC 70 group regardless of quercetin supplementation (p<0.05). In addition, higher RC ratio increased (p<0.05) N retention and N retention rate. Total VFA, acetic acid, propionic acid, iso-butyric acid, butyric acid, iso-valeric acid and valeric acid contents were not affected (p>0.05) by dietary quercetin. Meanwhile, lower total cholesterol level exhibited in animals in RC 70 group compared to RC 30 or 50 groups, unrelated to dietary quercetin (p<0.05), however other plasma parameters were not influenced (p>0.05) by RC ratio and dietary quercetin. Our results indicated that both RC ratio and dietary quercetin may not directly affect the production indices and immune responses in Korean indigenous goat.

Effect of Ginsenoside Rc on the Pharmacokinetics of Mycophenolic Acid, a UGT1A9 Substrate, and its Glucuronide Metabolite in Rats

  • Previous in vitro studies have demonstrated that ginsenoside Rc inhibits UGT1A9, but there are no available data to indicate that ginsenoside Rc inhibits UGT1A9 in vivo. The effect of single and repeated intravenous injection of ginsenoside Rc was evaluated on the pharmacokinetics of mycophenolic acid. After injection of ginsenoside Rc (5 mg/kg for one day or 3 mg/kg for five days), 2-mg mycophenolic acid was intravenously injected, and the pharmacokinetics of mycophenolic acid and mycophenolic acid-β-glucuronide were determined. Concentrations of mycophenolic acid and its metabolite from rat plasma were analyzed using a liquid chromatography-triple quadrupole mass spectrometry. Single or repeated pretreatment with ginsenoside Rc had no significant effects on the pharmacokinetics of mycophenolic acid (P > 0.05): The mean difference in maximum plasma concentration (Cmax) and area under the concentration-time curve (AUCinf) were within 0.83- and 0.62-fold, respectively, compared with those in the absence of the ginsenoside Rc. These results indicate that ginsenoside Rc has a negligible effect on the disposition of mycophenolic acid in vivo despite in vitro findings indicating that ginsenoside Rc is a selective UGT1A9 inhibitor. As a result, ginsenoside Rc has little possibility of interacting with drugs that are metabolized by UGT1A9, including mycophenolic acid.

Inductive Effects of Ginseng Saponins on the Rat LDH A-gene and the Synthetic rate of Hepatocyte DNA in Regenerating Rat Liver Cells

  • The effects of ginseng saponins, G-Rbl and G-Rc on the rat liver LDH A-gene transcnptional activity was investigated during pro-replicative phase of rat liver after partial hepatectomy. Changes in LDH A-mRNA levels in regenerating rat liver after intraperitoneal administrations of G-Rbl of G-Rc were tested by slot blot hybridization methods. The results showed that G-Rbl (1 mg/100g B.W) and G-Rc (1 ma/100g B.W) caused marked increases of LDH A-mRNA contents by respectively 1.9- and 1.5-fold in rat liver at 5·hours after partial hepatectomy. Dose dependent effect of G-Rbl and G-Rc (1-25 mg/100g B.W) on the LDH A-mRNA levels on regenerating rat liver were also analyzed. The maximal in- creases of liver LDH A-mRNA levels were observed with the doses of 1 mg for G-Rbl and 5 mg for G-Rc However, when the administration doses of G-Kbl and G-Rc were increased to 20 mg, G-Rbl caused a marked decrease of LDH A-mRNA level to 61% of those in sham-operated rat liver In contrast, G-Rc slightly decreased the liver LDH A-mRNA contents by 30% as compared to those of the maximum value but still maintained 22% higher LDH A-mRNA levels then those of sham-operated rate liver. On the basis of these experimental results, we conclude that ginseng saponin, G-Rb 1 and G·Rc have stimulatory effect at the lower concentration (1 mg/100g B.W) and inhibitory effect at the higher concentration (20 moi loos 5.W) on the LDH A-gene transcription during regeneration of rat liver, Additionally we also investigated the stimulatory effects of ginsenosides on the protein and DNA synthetic activities in hepatocyte primary cell cultures isolated from regenerating rat liver. Both of G·Rc and -Re increased the synthetic rates of hepatocytes proteins and DNA at the administration doses of 50 ug and 100 ug/3 ml/dish respectively representing 1.3-1.6 fold increases. From these results we postulate that G-Rc and -Re may have a mitogen enhancer activity for the hepatocyte proliferation during rat liver regeneration period. Seismic Curvature Ductility of RC Bridge Piers with 2.5 Aspect Ratio (형상비 2.5의 RC 교각의 내진 곡률연성도)

  • Due to the 1989 Loma Prieta, 1995 Hyogoken Nambu earthquakes, etc, a number of bridge columns  were collapsed in flexure-shear failures as a consequence of the premature termination of the column longitudinal reinforcement. Nevertheless, previous researches for the performance of bridge columns were concentrated on the flexural failure mode. It is well understood that the seismic behaviour of RC bridge piers was dependent on the performance of the plastic hinge of RC bridge piers, the ductility of which was desirable to be computed on the basis of the curvature. Experimental investigation was made to evaluate the variation of the curvature of the plastic hinge  region for the seismic performance of earthquake-damaged RC columns in flexure-shear failure mode. Seven test specimens in the aspect ratio of 2.5 were made with test parameters: confinement ratios, lap splices, and retrofitting FRP materials. They were damaged under series of artificial earthquakes that could be compatible in Korean peninsula. Directly after the pseudo-dynamic test, damaged columns were retested under inelastic reversal cyclic loading under a constant axial load, $P=0.1f_{ck}A_g$. Residual seismic capacity of damaged specimens was evaluated by analzying the moment-curvature hysteresis and the curvature ductility. Test results show that the biggest curvature was developed around 15cm above the footing, which induced the column failure. It was observed that RC bridge specimens with lap-spliced longitudinal steels appeared to fail at low curvature ductility but significant improvement was made in the curvature ductility of RC specimens with FRP straps wrapped around the plastic hinge region. Based on the experimental variation of the curvature of RC specimens, new equivalent length of the plastic hinge region was proposed by considering the lateral confinement in this study. The analytical and experimental relationship between the displacement and the curvature ductility were compared based on this proposal, which gave excellent result.

  • The German cockroach(Blattelia germanica) population~ were successIVely selected with ch\orpyrifos and permethrin during the six generations. The resulting resistant $R_{chtorpenfos}$(Rc) and $R_{permethnn}$(Rp) stra.ins were studied to investigate the esterase activity by spectrophotometer, filter parper test, and electrophoresis. Esterase-$\alpha$ activities by filter paper test showed 2.65 and LBZ times higher in the Rc and Rp strains than the susceptible strain, respectively. ln the spectrophoLometer method, the esterase activit18s to $\alpha$-and $\beta$-naphthyl acetate were increased 2.34 and 5.28 times in the Rc than susceptible strain, and 1.48 and 2.92 times in the Rp Limn susceptible stram, respectlvely. Zymogram patterns of eslerase isozyme by agarose gel electrophoresis showed totally five bands. The Rc and Rp strains showed two additive bands as, Est-2 and Est-3, which were not shown in the susceptible strain. but the Rp strain dId not show Est-5 bands which was COlumon in the Rc and susceptible strams.

Seismic Performance Analysis of RC Bridge Piers with 3.5 Aspect Ratio depending on Testing Methods (형상비 3.5 RC교각의 실험 방법에 의한 내진성능 분석)

  • This paper deals with the shaking table test(STT), the Quasi-Static Test(QST), and the Pseudo-Dynamic Test(PDT) to evaluate the seismic performance of RC bridge piers under near fault ground motion. Five scaled specimens were constructed the weight of the superstructure was applied through the prestressing strand at the centroid of the column section during the QST and PDT. However, the STT was simulated. The lateral inertia force of the superstructure by the mass frame which was linked with the pier because of the limited payload of shaking table. Particularly for the STT, friction underneath the mass frame was minimized by special details and it was verified by a series of pre-load test. Scale factor of the RC piers was 4.25.

  • Before the implementation of the 1992 seismic design provisions in Korea, longitudinal steels of RC bridge piers were practically lap-spliced in the plastic hinge region. Experimental investigation was made to evaluate the seismic performance of RC bridge pier specimens in a flexure/shear mode. Six circular test specimens in an aspect ratio of 2.5 (600mm in diameter) were made with test parameters confinement ratio, lap splices, and retrofit FRP materials. They were damaged under a series of artificial earthquakes with 0.22g PGA, being compatible in Korean peninsula, through the pseudo-dynamic test. Probable damages were assessed by the Park and Ang damage index. Approximate 0.1 and 0.3 damage indices were obtained for RC specimens without lap splice and with lap splice, respectively. Directly after the pseudo-dynamic test, damaged test columns were laterally actuated under inelastic reversal cyclic loadings simultaneously under a constant axial load. Through curvature measurements, residual seismic performance was evaluated for test specimens. Test results show that RC pier specimens with lap-spliced appeared to fail at low ductility, but significant improvement was obtained for the ductility of these specimens if externally wrapped with FRP.

Flexural behavior of RC beams made with basalt and polypropylene fibers: Experimental and numerical study

  • The effect of basalt and polypropylene fibers on the flexural behavior of reinforced concrete (RC) beams is investigated in this paper. The compressive and tensile behaviors of the basalt concrete and polypropylene concrete cylinders are also investigated. Eight beams and 28 cylinders were made with different percentages of basalt and polypropylene fibers. The dosages of fiber were selected as 0.6%, 1.3%, and 2.5% of the total cement weight. Each type of fiber was mixed solely with the concrete mix. Basalt and polypropylene fibers are modern and cheap materials that can be used to improve the structural behavior of RC members. This research is designed to find the optimum percentage of basalt and polypropylene fibers for enhancing the flexural behavior of RC beams. Test results showed that the addition of basalt and polypropylene fibers in any dosage (0.6%, 1.3%, and 2.5%) can increase the flexural strength and displacement ductility index of the beams where the maximum enhancement was measured with 1.3% fibers. The maximum increments in the flexural strength and the displacement ductility index were 30.39% and 260% for the basalt fiber case, while the maximum improvement for the polypropylene fibers case was 55.5% and 230% compared to the control specimen. Finite element (FE) models were then developed in ABAQUS to predict the numerical behaviour of the tested beams. The FE models were able to predict the experimental behaviour with reasonable accuracy. This research confirms the efficiency of basalt and polypropylene fibers in enhancing the flexural behavior of RC beams, and it also suggests the optimum dosage of fibers.
