Anti-allergic and Anti-inflammatory Effect of Leonurus sibiricus Seed Ethyl Acetate Fractions (충울자 EtOAc 분획의 항알러지 및 항염증 효과)

  • In this study, we investigate anti-allergic and anti-inflammatory effects of Leonurus sibiricus seed (LSS) extract in basophilic leukemia RBL-2H3 cells. To identify anti-allergic actions of LSS, the degranulation was evaluated in IgE and DNP-BSA stimulated RBL-2H3 cells. At the concentration of $100{\mu}g/ml$ of methanol (MeOH) extract and Methylene chloride (MC) and Ethyl acetate (EtOAc) fractions, the degranulation was significantly inhibited 16.7%, 16.7% and 27.9% respectively. And then, to assess anti-inflammatory effects of LSS, IL-4 and IL-13 mRNA level were detected in PMA/ionomycin (PI)-induced RBL-2H3 cells and cell proliferation and IL-4 mRNA level in isolated splenocytes from Balb/c mice. LSS MeOH extract and MC and EtOAc fractions significantly decreased the level of IL-4 and IL-13 mRNA in PI-induced RBL-2H3 cells and showed inhibitory effects on cell proliferation and expression of IL-4 mRNA level in mouse splenocytes. Taken together, these results suggest that LSS has potential anti-allergic and anti-inflammatory effects and EtOAc fraction is the most effective in regulating immune responses.

  • Objectives : We studied on anti-allergic effects of Semen Perillae Herbal Acupuncture(SPHA) and Semen Perillae Herbal Acupuncture Solution(SP). Methods : In vivo, Animals were herbal-acupunctured SPHA at both ST36 three times for 5 days. Then, we investigated compound 48/80-induced active systemic anaphylaxis (ASA) using ICR mice and anti-DNP IgE-induced passive cutaneous anaphylaxis (PCA) using Sprague Dawley rat. In vitro, we measured cell viability, b-hexosaminidase release, IL-4 and TNF-a from RBL-2H3 cells after treatment of SP of various concentrations. Results : In vivo, SPHA pretreatments at both ST36 inhibited compound 48/80-induced ASA. PCA was inhibited by SPHA pretreatments at both ST36 and optional points. In vitro, SP treatments were not affect on cell viability and inhibited b-hexosaminidase release, IL-4 and TNF-a. Conclusions : These results suggest that SPHA and SP may be beneficial in the inhibition of allergic inflammatory response.

  • Object : We investigated the effect of Gamiokyaek-tang (GOYT) on immediate type hypersensitivity. Methods : We investigated anti-dinitrophenyl (DNP) IgE-mediated passive cutaneous anaphylaxis and acetic acid-induced vascular permeability in rodents. Also perfonned MTT assay and b-hexosaminidase activity is measured in RBL-2H3. Results : GOYT inhibited passive cutaneous anaphylaxis and acetic acid-induced vascular permeability by oral administration. All the concentrations of GOYT from 0.1 to 5 mg/ml didn't have an effect on cell viability and cytotoxicity. b-hexosaminidase release in RBL-2H3 was significantly reduced by 2 and 5 mg/ml of GOYT. Conclusion : These results indicate that GOYT have inhibition effects on immediate type hypersensitivity .

Comparison of Anti-allergenic Activities of Various Polyphenols in Cell Assays

  • The inhibitory effects of 25 polyphenols against in vitro allergic reactions were compared using biochemical and cell assays. Three polyphenols including curcumin, gallic acid, and quercetin suppressed the release of $\beta$-hexosaminidase from ionophore A23187-stimulated RBL-2H3 cells more effectively (>50% inhibition at $100{\mu}M$ concentration). They were found to have potencies in suppressing the release of histamine not only from ionophore A23187-, but also from immunoglobulin E (IgE)-stimulated RBL-2H3 cells. Moreover, such suppressive effects of the three polyphenols were also observed in A23187 plus PMA-costimulated rat peritoneal mast cells. The extent of inhibition were quantified as the respective polyphenol concentration that inhibit 50% ($IC_{50}$) of $\beta$-hexosaminidase or histamine release, showing an inhibition tendency with decreasing order of curcumin>gallic acid>quercetin. Down-regulation of $Ca^{2+}$ influx was suggested as the cause of the inhibition of $\beta$-hexosaminidase and histamine releases in these cells. The immune process inhibition was confirmed by the observed reduction in the gene expressions and release of pro-inflammatory cytokine tumor necrosis factor (TNF)-$\alpha$, interleukin (IL)-$1\beta$, and IL-4, due probably to antioxidant activity of the polyphenols. These findings illustrate that curcumin, gallic acid, and quercetin may be beneficial against allergic inflammatory diseases.

  • Histamine-releasing factors (HRFs) are soluble mediators that can release histamine and other mediators from basophils and mast cells and their activity can vary, depending on the type of IgE. The activity of HRFs is affected by the presence of IgE, although HRF is thought to bind to a specific receptor other than IgE. Until now, HRF signaling pathway including its receptor remains unclear in spite of numerous studies. Since there had been many reports about reactive oxygen species (ROS) as a signaling molecule rather than as a by-product of metabolism, we investigated the possibility of ROS as an intracellular messenger involved in HRF-mediated histamine degranulation. In RBL-2H3 cells, ROS was generated by HRF using $H_2O_2$-sensitive fluorescence of fluorescent 2', 7'-dichlorofluorescein ($H_2DCFDA$). These effects were blocked by anti-oxidant N-acetylcysteine (NAC). These results suggest that ROS generation could play a role as an intracellular messenger in histamine release by HRF.

Inhibitory Effect of Extracts from Rhododendron Brachycarpum and Abies Koreana E.H. Wilson on Degranulation and Cytokine Expression in RBL-2H3 Cells (만병초와 구상나무 추출물의 RBL-2H3 세포 탈과립, 싸이토카인 유전자 발현에 미치는 영향)

  • Ethanol extracts from Rhododendrom brachycarpum and Abies koreana were investigated for their anti-allergic activities using RBL-2H3 cell line. After treatment with ethanol extracts of various concentrations on the immune response induced mast cell by concanavalin A (Con A), the expressions of cytokine interleukin-4 (IL-4), interleukin-13 (IL-13) were determined by using RT-PCR and the degranulation of mast cells was determined by measuring ${\beta}$-hexosaminidase release. Expression level of IL-4 was decreased by the extract from Rhododendrom brachycarpum in $10^{-7}$, $10^{-5}$ and $10^{-3}%$ concentrations. Expression level of IL-13 was also decreased by both extracts. ${\beta}$-Hexosaminidase release by RBL-2H3 cells was inhibited at the $10^{-5}$ and $10^{-3}%$ concentration of extracts from Rhododendrom brachycarpum and Abies koreana, respectively. These results demonstrate that ethanol extracts of Rhododendron brachycarpum and Abies koreana exert anti-allergic effects by regulating the reduction of IL-4 and IL-13 genes expression and also the secretion of ${\beta}$-hexosaminidase.

Anti-Allergic Effect of 1,2,3,4,6-Penta-O-Galloyl-β-D-Glucose on RBL-2H3 Cells (RBL-2H3 세포에서 1,2,3,4,6-Penta-O-Galloyl-β-D-Glucose의 항알레르기 효과)

  • 1,2,3,4,6-Penta-O-galloyl-${\beta}$-D-glucose (PGG) is a gallotannin isolated from various plants such as Galla Rhois. In a previous study, it was reported that PGG has anti-allergic effects by inhibiting interleukin (IL)-4 signaling in B cells. However, the effect of PGG on basophilic cells remains unclear. Therefore, the aim of this study was to investigate the inhibitory effect of PGG on mitogen and calcium ionophore-induced allergic responses. PGG had no effect on proliferation and cytotoxicity of RBL-2H3 cells. PGG significantly suppressed cell degranulation (histamine and ${\beta}-hexosaminidase$) as well as inflammatory cytokine production such as IL-4 and tumor necrosis factor-${\alpha}$. The underlying mechanism of PGG on these anti-allergic actions was correlated with inhibition on translocation of nuclear factor-${\kappa}B$ from the cytosol to nucleus. These data suggest that PGG is a potentially effective functional compound for prevention of allergic diseases.

Anti Allergic Effects of Cimicifuga Racemosa on Allergic Models (승마(升麻)추출물이 RBL-2H3 Cell과 Mouse에 미치는 항알레르기 효과)

  • Cimicifuga racemosa (Black cohosh) has been used as therapeutics for pain and inflammation in Korean folk medicine. The potential effects of cimicifuga racemosa extract on mast cell dependent allergy reaction, however, have not been well elucidated yet. In the present study, we investigated the effect of cimicifuga racemosa extract on the allergy reaction using mast cell dependent in vivo and in vitro models. The oral administration of cimicifuga racemosa extract showed inhibitory potential on the compound 48/80 induced active systemic anaphylactic shock. cimicifuga racemosa extract also significantly inhibited the anti DNP IgE induced passive cutaneous anaphylaxis (PCA) reaction and acetic acid induced vascular permeability. In addition, cimicifuga racemosa extract inhibited the beta hexosaminidase release and TNF alpha and IL 4 mRNA induction by DNP HSA in rat leukemia mast cells, RBL 2H3. but cimicifuga racemosa extract didn't affected to RBL 2H3 cell viability. These results demonstrated that cimicifuga racemosa extract has an anti allergic potential and it may be due to the inhibition of histamine release and cytokine gene expression in the mast cells.

Experimental Study about Pathway of Aconiti Ciliare Tuber on Allergic Reaction of Inflammation (초오의 항알레르기 염증반응 및 기전탐색에 관한 연구)

  • Objetives : The purpose of this study was to examine the pathway of anti-allergic effects of Aconiti Ciliare Tuber (ACT). Methods : We examined cell viability, ${\beta}$-hexosaminidase release, pro-inflammatory cytokines secretion and mRNA expressions, nuclear factor-kappa B (NF-${\kappa}B$) (p65) activation, inhibitor kappa B-alpha ($I{\kappa}B-{\alpha}$) degradation, and MAPKs activation from RBL-2H3 cells pre-treatment by ACT of 1.0 mg/ml, 2.0 mg/ml separately. Results : We observed that ACT reduced the secretion of ${\beta}$-hexosaminidase, TNF-${\alpha}$, IL-4 and the expression of COX-2 mRNA in RBL-2H3 cells. Futhermore, ACT inhibited the levels of activation of NF-${\kappa}B$ (p65) protein, ERK MAPK, and degradation of $I{\kappa}B-{\alpha}$ in RBL-2H3 cells. Conclusions : These results show that ACT has an anti-histamine effect and inhibitory effect of NF-${\kappa}B$ (p65) through regulation of $I{\kappa}B-{\alpha}$ degradation. This improves that ACT could be used as an anti-allergic medicine.

Suppression of Th2-associated Cytokines Expression by Schizandrin in Basophils and Mast Cells (Schizandrin에 의한 호염구와 비만세포의 Th2 관련 사이토카인 발현 억제)

  • Schizandrin (SZ), was isolated from the fruit of Schizandra chinensis, has been reported to have many biological properties, including anti-inflammatory and antitumor activities. However, its anti-allergic effects are not completely elucidated. We focused on the anti-allergic effects of SZ in PMA/ionomycin (PI)-induced rat basophilic leukemia (RBL-2H3) cells and P815 mast cells. Cytokines (IL-4, IL-13), synthesized by basophils and mast cells, are implicated in pathological conditions such as asthma and allergy. The production of IL-4 and IL-13 was quantified by ELISA and the mRNA expression was detected by using RT-PCR assay. In this study, we found that SZ did not show cytotoxic effect at up to 100 ${\mu}M$ on RBL-2H3 cells and mast cells. In addition, SZ inhibited the production of IL-4 and IL-13 and also decreased the level of mRNA in PI-induced RBL-2H3 cells and mast cells in a dose-dependent manner. Thus, we suggest that SZ may have the effect on preventing allergic disorders by inhibiting IL-4 and IL-13 cytokines.
