Red blood cells (RBCs) are customarily adhered to a bio-functionalised substrate to make them stationary in interferometric phase-imaging modalities. This can make them susceptible to receive alterations in innate morphology due to their own weight. Optical tweezers (OTs) often driven by Gaussian profile of a laser beam is an alternative modality to overcome contact-induced perturbation but at the same time a steeply focused laser beam might cause photo-damage. In order to address both the photo-damage and substrate adherence induced perturbations, we were motivated to stabilize the RBC in OTs by utilizing a laser beam of ‘arbitrary intensity profile’ generated by a source having cavity imperfections per se. Thus the immobilized RBC was investigated for phase-imaging with sinusoidal interferograms generated by a compact and robust Michelson interferometer which was designed from a cubic beam splitter having one surface coated with reflective material and another adjacent coplanar surface aligned against a mirror. Reflected interferograms from bilayers membrane of a trapped RBC were recorded and analyzed. Our phase-imaging set-up is limited to work in reflection configuration only because of the availability of an upright microscope. Due to RBC’s membrane being poorly reflective for visible wavelengths, quantitative information in the signal is weak and therefore, the quality of experimental results is limited in comparison to results obtained in transmission mode by various holographic techniques reported elsewhere.
Recently, the horse racing industry in Korea has developed rapidly with good betting turnover. One of the most important factors in enhancing the performance of racehorses is the study of equine exercise physiology. This study was performed to analyze the results of pre-race blood test on race day and evaluate its correlation with the race performance in Thoroughbred racehorses. Twenty-one, healthy 3- to 6-year-old Thoroughbred horses were used to collect blood samples and hematological and biochemical measurements were performed. Results were analyzed to identify its correlation with racing performance. Analysis of hematological profile of pre-race blood of the racehorses revealed that red blood cell indices including RBC, Hb, and PCV levels were elevated. Additionally, levels of AP, AST, and GGT were also elevated in pre-race blood of horses on race day. RBC, Hb, and PCV levels of high performance racehorses were significantly lower than those of low class racehorses. Moreover, the analysis of correlation between blood test results and racing ability-related categories have shown negative correlations for rating, placing strike rate, prize money per race, RBC, Hb, and PCV and positive correlations for G3F and PCV. Additionally, AST value was positively correlated to prize money per race. Thus, the results of this study show that red blood cell indices and AST are good parameters for evaluating racing performance of horses before the race.
Effects of supplemental charcoal powder and vitamin A on growth performance, serum profile and carcass characteristics with 24 Hanwoo steers were inves$\ell$ated. The Hanwoo steers were randomly assigned to one of the three dietary treatments: 1) Control(commercial feed), 2) Tl(commercial feed supplemented with 2% charcoal powder and 0.2 % vitamin A), 3) T2(commercial feed with 2 % charcoal powder). In the age 26 month, body weight of control Tl and T2 were 633.8 kg, 621.0 kg, 619.4 kg, respectively, with not significantly different(P> 0.05). The daily weight gain of control was the highest followed by Tl and T2. The contents of vitamin A(retinol) and total cholesterol in the blood of Tl was higher Tl(56.32 $\ell$ / dt, 163.83 mg/ dz) than the other treatment groups(P < 0.05). The average of WBC value in the blood was 6.86-" 9.83 k / ${\mu}\ell$, RBC value was 7.98 - 11.26 k / ${\mu}\ell$ and blood platelet was 236.7 - 287.0 k / ${\mu}\ell$. But The serum profile in blood was not significantly different between control and treatment groups. The carcass grade and characteristics of all treatments in Hanwoo steers were not significantly different except the texture.
The study was to compare the effect of dietary fatty acids on fatty acid profile in tissue and the status of tocopherol and lipid peroxidation, and superoxide dismutase and glutathione peroxidase activities at two fat levels. Male Sprague Dawley rats weighing average 350g(17 weeks) were fed either low fat(LF, 4.3% w/w, 10% kcal) or high fat(HF, 20.8%, w/w, 40% kcal)diet for 6 weeks. The fats used were beef tallow as a source of saturated fatty acid, corn oil for n-6 linoleic acid, perilla oil for n-3 $\alpha$-linolenic acid and fish oil for n-3 eiocosapentatenoic acid(EPA) and n-3 docosahexaenoic acid(DHA). Palsma tocopherol was significantly reduced by fish oil compared to beef tallow at body fat level. However, there was no significant effect on the levels of plasma MDA, RBC MDA and tocopherol, and RBC hempolysis by the type and amount of dietary fat. The peroxidizibility index of fatty acid profile in plasma and liver was increased and liver MDA level was significantly increased by fish oil when dietary fat level was increased. The activities of SOD and GSHPx tended to be increased by perilla oil and fish oil at both fat oil significantly reduced the incorpration of c20:4 and increased the incorporation of c20:5 into liver compared to corn oil. The incorporation of n-3 fatty acids into tissue by perilla oil rich in $\alpha$-linolenic acid was significantly higher tan corn oil and its effect was improved with higher amount of perilla oil in diet by high fat diet. Overall, the lipid peroxidation of tissue could be prevented by tocopherol supplementation when dietary fat level was low in diet. However, at high fat diet, tocopherol supplementation might not be enough to prevent the lipid peroxidation in tissue since the potential for lipid peroxidation was tended to be increased with higher incorporation of higher unsaturated n-3 fatty acids into tissue. Therefore, it could not be recommended to consume large amount of fish oil even with excess amount of tocopherol supplemented to the high fat diet.
The aim of this study was to evaluate the effect of Lactobacillus acidophilus probiotic (LAP) product supplementation on the blood profile, fecal noxious gas emission, and fecal shedding of lactic acid bacteria and coliform bacteria in healthy adult Beagle dogs. In total, 14 Beagle dogs with an average initial body weight of 10.19 ± 0.61 kg were randomly assigned into two dietary treatments,with and without LAP supplementation, for a 28-day feeding trial. At the end of the experiment, there was no significant (p > 0.05) difference in the concentration of serum total cholesterol, high-density lipoprotein cholesterol (HDL-C), low-density lipoprotein cholesterol (LDL-C), white blood cell (WBC), red blood cell (RBC), blood lymphocyte percentage, fecal hydrogen sulfide (H2S) and total mercaptans (R.SH) emission, and fecal coliforms counts. However, the serum concentrations of the triglyceride and fecal ammonia (NH3) emission of the LAP treatment were significantly (p < 0.05) decreased in the group compared with the CON dogs. Fecal total lactic acid bacteria counts were significantly (p < 0.05) increased in the LAP treatment. In conclusion, the supplementation of LAP in Beagle dog diets could decrease the blood triglyceride level and enhance the gut Lactobacillus count which may have positive effects on dogs.

This study reports the analgesic, anti-inflammatory and membrane-stabilizing property of alcoholic extract of Ophiorrhiza nicobarica (ON), a wild herb, used as an anti-infective ethnomedicine of Nicobarese and Shompen tribes of Great Nicobar Island, India. We for the first time investigated the analgesic and antiinflammatory potential of this herb in acute, subacute and chronic model of inflammation in Swiss albino mice and Wistar albino rats, along with sheep RBC-induced sensitivity and membrane stabilization. The acetic acid induced writhing, tail flick and tail immersion tests are used as a model for evaluating analgesic activity; while the carrageenin-induced paw oedema was used as the model for acute inflammation, dextran-induced oedema as sub-acute and cotton-pellateinduced granuloma as chronic inflammatory model. The probable mode by which ON mediate its effect on inflammatory conditions was studied on sheep RBC-induced sensitivity and membrane stabilization. The in vitro results revealed that the ON extract possesses significant (P < 0.05) dose dependent analgesic and antiinflammatory activity at 200 and 300 mg/kg and its fractions at 50 mg/kg, p.o. respectively, compared to the control groups. However, the extract failed to exhibit membrane-stabilizing property as it unable to reduce the level of haemolysis of RBC exposed to hypotonic solution. The acute toxicity studies of ON extract in rats and mice revealed that the extract was nontoxic even up to 3.0 g/kg body weight of the animals, with a high safety profile. We have isolated ursolic acid, ${\beta}$-sitosterol and harmaline respectively, from the bioactive part of the extract. The results indicated that the O. nicobarica is indeed beneficial in primary health care, and suggest that its anti-inflammatory activity may not be related to membrane-stabilization.
This study was designed to compare the effect of different dietary fats on plasma lipids, the degree of lipid peroxidation and the activity of antioxidant enzymes in RBC and liver rats treated with or wighout 1, 2-dimethylhydrazing (DMH). Male Sprague Dawley rats, at 7 weeks-old, were divided into control and DMH-treated grous, and each group was again subdivided into four were perilla oil (PO), blend fat (BF) containing ten different kinds of dietary oil, beef tallow (BT), corn oil (CO). At the same time, each rat was injected intramusculary with saline(for control) or DMH twice a week for 6 weeks to give total dose of 180 mg/kg body weight. Compared with BT feeding, BF reduced plasma total choesterol level and PO and Co reduced plasma TG levels (p<0.05). DMH injection decreased plasma cholesterol in all dietary groups. However, PO decreased tocopherol levels and increased TBARS levels in RBC compared to BT. The degree of hemolysis in PO group was higher than that of BT group (p<0.05 only in control group. Fatty acid composition of hepatic microsome was reflected by dietary fatty acid profile. The peroxidizability index and TBARS level in hepatic micorsome were significantly increased but tocopherol level was lowered in PO group compared to BT group. Activites of superoxide dismutase and glutathione peroxidase in RBC and hepatic cytosol were not influenced y dietary fats and DMH treatment(p<0.05). Overall, perilla oil rich in $\omega$3 $\alpha$-linolenic acid could be a very important dietary source in reducing plasma lipids and blend fat was also good dietary oil mixture in reducing plasma cholesterol. However, the degree of lipid peroxidation was greater in tissue by perilla oil feeding and it is very difficult to use only perilla oil as oil source for meal preparation, so that it could be suggested to use more perilla oil and fish to give an equal effect of blend fat in order to reduce the risk factors against cardiovascular disease.

Tick-borne pathogens can cause serious problems in grazing cattle. However, little information is available on tick-mediated diseases in cattle grazing on mountains. Thus, this study aimed to understand the potential problems related to tick-borne diseases in grazing cattle through the investigation of prevalent tick-transmitted infections, and their associated hematological changes, in terms of season and grazing type in Korean indigenous cattle (=Hanwoo). Hanwoo cattle from 3 regions of the Republic of Korea (=Korea) were either maintained indoors or placed on grassy mountains from spring to fall of 2014 and 2015. Cattle that grazed in mountainous areas showed a greater prevalence of tick-borne infections with an increased Theileria orientalis infection rate (54.7%) compared to that in non-grazing cattle (16.3%) (P<0.001). Accordingly, the red blood cell (RBC) count and hematocrit (HCT) values of grazing cattle were significantly lower than those of non-grazing cattle throughout the season (P<0.05). Moreover, RBC, hemoglobin (Hb), and HCT of T. orientalis-positive group were significantly lower than those of T. orientalis-negative group (P<0.05). T. orientalis is a widespread tick-borne pathogen in Korea. Grazing of cattle in mountainous areas is closely associated with an increase in T. orientalis infection (RR=3.4, P<0.001), and with consequent decreases in RBC count and HCT. Thus, these findings suggest that the Hanwoo cattle in mountainous areas of Korea are at a high risk of infection by T. orientalis, which can lead to hematological alterations. This study highlights the necessity of preventive strategies that target T. orientalis infection.
In order to obtain velocity profile of blood flow with high spatial resolution, a micro PIV technique consisted of a fluorescent microscope, double-pulsed YAG laser, cooled CCD camera was applied to in-vitro blood flow experiment through a micro round tube of a diameter $100{\mu}m$. Velocity distributions of blood flow for rabbit were obtained. The viscosity profiles for shear rate were found at flowing condition. To provide hemorheological characteristics of blood flow, the viscosities for shear rate were evaluated. The viscosity of blood also steeply increase by decreasing shear rate resulting in Non-Newtonian flow, especially in low shear rate region caused by RBC rheological properties. The results show typical characteristics of Non-Newtonian characteristics from the results of velocity profile and viscosity for blood flow. From the inflection points, cell free layer and two-phase flow consisted with plasma and suspensions including RBCs can be separated.
In order to investigate flow characteristics of blood flow in a micro tube ($100{\mu}m$ in diameter) according to hematocrit, in-vitro experiments were carried out using a micro-PIV technique. The micro-PIV system consists of a microscope, a 2 head Nd:YAG laser, a 12 bit cooled CCD camera and a delay generator. Blood was supplied into the micro tube using a syringe pump. Hematocrit of blood was controlled to be 20%, 30% and 40%. The blood flow has a cell free layer near the tube wall and its thickness was changed with increasing the flow rate and hematocrit. The hemorheological characteristics such as shear rate and viscosity were evaluated using the velocity field data measured. As the flow rate increased, the blunt velocity profile in the tube center was sharpened. The viscosity of blood was rapidly increased with decreasing shear rate, especially in the region of low shear rate, changing RBC rheological properties. The variation of velocity profile and blood viscosity shows typical characteristics of Non-Newtonian fluids. On the basis of inflection points, the cell free layer and two-phase flow consisting of plasma and suspensions including RBCs were clearly discriminated.
