Effects of Dioscorea daemona Roxb. Stem Extract on the Inflammatory Responses, Antioxidant System and Lipid Levels in Vivo and the Production of Inflammatory Mediators in RAW264.7 Cells (Dioscorea daemona Roxb. (Stem) 추출물이 Rat의 염증반응 및 항산화 체계에 미치는 영향과 RAW264.7 세포주의 염증성 매개물 생성에 미치는 영향)

  • 본 연구에서는 다양한 동물 모델을 사용하여 Dioscorea daemona Roxb. 줄기 메탄을 추출물(DD)의 항염증 활성을 측정하였으며 DD가 생체내에서 항산화 체계의 변화를 유도할 수 있는지도 살펴보았다. DD를 200mg/kg용량으로 3주간 경구투여하였을 때 동물실험모델에서 항염증 및 type IV 알레르기 억제 효과를 나타내었으며 혈청의 Catalase 활성, 지질 과산화, TG 및 HDL cholesterol 수치가 영향을 받았다. DD와 이를 클로로포름과 부탄올로 순차적으로 분획하여 얻은 fraction이 lipopolysaccharide(LPS)로 유도한 RAW264.7 대식세포주의 nitric oxide(NO), prostaglandin $E_2(PGE_2)$, tumor necrosis $factor-\alpha(TNF-\alpha)$, interleukin 6(IL6)의 생성을 억제하는지도 연구하였다. DD와 그 분획물들은 $4\~100{\mu}g/mL$ 농도에서 세포 독성을 나타내지 않고 LPS가 유도한 RAW264.7 세포주의 NO, $TNF-\alpha$, IL-6 생성을 억제하였다. LPS가 유도한 $PGE_2$ 생성은 DD의 클로로포름 분획에서 유의적으로 감소하였다(p<0.05). 따라서 Dioscorea daemona 추출물은 대식세포의 염증성 매개물의 억제를 통하여 항염증 활성을 나타내는 것으로 사료된다.

Effect of Conjugated Linoleic Acid on Nuclear Factor-${\kappa}B$ Activation and Tumor Necrosis Factor-${\alpha}$ Production in RAW 264.7 Cells Exposed to High Concentration of Glucose (고농도의 당에 노출된 RAW 264.7 세포에서 conjugated linoleic acid의 TNF-${\alpha}$ 생산과 NF-${\kappa}B$의 활성 효과)

  • Diabetes-related complications in human and veterinary medicine have been shown to be associated with hyperglycemia-induced inflammation. It has been recently suggested that the onset of insulin resistance may be caused by over-production of inflammatory cytokines such as tumor necrosis factor (TNF)-${\alpha}$ from immune cells. Conjugated linoleic acid (CLA) regulates inflammatory response through modulation of TNF-${\alpha}$ expression. The objective of this study was to examine the effect of CLA on nuclear factor kappaB (NF-${\kappa}B$) p65 binding activity, inhibitory kappaB ($I{\kappa}B$)-${\alpha}$ expression, and TNF-${\alpha}$ production from high glucose-treated RAW 264.7 cells. CLA was added to RAW cells that had been previously cultured with low or high concentration of glucose. The levels of TNF-${\alpha}$ protein in the culture supernatant of RAW cells exposed to high concentrations of glucose were higher than those of cells exposed to low concentrations of glucose. The treatment with the high concentration of glucose in RAW cells increased levels of NF-${\kappa}B$ p65 binding activity and the decreased $I{\kappa}B-{\alpha}$ expression when compared with those of low glucose. The treatments in combination with CLA and glucose (low and high) glucose in RAW cells increased TNF-${\alpha}$ production when compared with that glucose alone. These treatments with CLA increased TNF-${\alpha}$ production in high glucose-treated RAW cells than those with low glucose. These treatments of CLA also showed higher NF-${\kappa}B$ p65 binding activity and lower $I{\kappa}B-{\alpha}$ expression in high glucose than those in low glucose condition. This suggests that CLA can increase NF-${\kappa}B$ p65 binding activity and TNF-${\alpha}$ production from high glucose-treated RAW 264.7 cells and is likely to promote hyperglycemia-induced inflammation.

  • The effects of the hormonal factor (CCK) a and neural factor(carbachol) on the exocrine function of the pancreas were studied in th is experiment. A superfusion technique was used for in vitro study of stimulus-secreti- o on coupling in isolated pancreatic acinarce 11s frQm the rats fed heated or raw soybean diet. Chymotrypsin secretion was higher in cells from the raw soybean group than in those from the heated soybean group with both kinds of stimulants(CCK and carbachol), whereas, amylase secretion was higher inthe h heated soybean group than in the raw soy­b bean group. This indicated that chymotrvpsin a and amylase secretion from the acinar cells are not parallel with CCK and carbachol st­i imulation.

Anti-inflammatory Effect of the Robinia pseudoacacia L. High Temperature Extract (아까시 나무 고온추출물의 항염증 효과)

  • This study was conducted to compare anti-inflammatory effect of Robinia pseudoacacia L. using different extraction methods (water extraction, ethanol extraction and high temperature extraction). We investigated anti-inflammatory effect of Robinia pseudoacacia L. extract (RP1, water extract; RP2, ethanol extract; RP3, high temperature extract) on lipopolysaccharide (LPS)-stimulated inflammation using Raw 264.7 cell. Cells were treated with various concentrations (12.5, 25, 50, 100 or $200{\mu}g/m{\ell}$) of water extract, ethanol extract and high temperature extract. Cytotoxicity was not observed on Raw 264.7 cells, LPS-stimulated production of NO (nitric oxide), $PGE_2$ (prostaglandin $E_2$) and cytokines ($TNF-{\alpha}$, IL-6 and $IL-1{\beta}$) was reduced by RP3 treatment more than RP1 and RP2. In conclusion, these results indicated that inflammation on Raw 264.7 cells was improved by RP3. Treatment of RP3 could be used to natural medicine for improving inflammatory response. However, further experiment is required to observe how the high temperature extraction at $500^{\circ}C$ for 48 h influences on alteration of active ingredient in Robinia pseudoacacia L., and conducts the inflammation signal pathway on Raw 264.7 cells.

Anti-inflammatory effects of Rosa rugosa extracts in RAW264.7 cells exposed to particulate matter (PM10) (미세먼지 PM10에 노출된 RAW264.7 세포에 대한 해당화 추출물의 항염증 활성)

  • Airborne fine dust (FD) particles smaller than 10 ㎛ in diameter (PM10) are one of the major causes of air pollution in East Asia, including Korea, and have become a major contributor to respiratory and skin problems. FD inordinately promotes the production of reactive oxygen species and inflammatory response in macrophages, leading to cell damage and death. Rosa rugosa, a deciduous shrub of the Rosa genus, has been used in traditional East Asian herbal medicine to treat various illnesses. The present study investigated the anti-inflammatory effects of R. rugosa organ extracts on PM10-stimulated RAW264.7 macrophages. Compared to non-treated RAW264.7 cells, treatment with 100 ㎍.ml-1 PM10 resulted in increased nitric oxide (NO) production, similar to lipopolysaccharide treatment. Additionally, 100 ㎍/ml stem extract reduced NO production by more than 45% compared to mock treatment. Furthermore, PM10-induced expression of interleukin (IL)-1β, IL-6, inducible NO synthase, and cyclooxygenase-2 was significantly reduced by stem extract treatment, indicating that the anti-inflammatory effect of the stem extract is mediated by the inhibition of pro-inflammatory mediators in PM10-stimulated RAW 264.7 cells. These results indicate that the R. rugosa stem could be considered a natural remedy with a protective effect against inflammatory responses induced by harmful airborne dust.

The immune enhancement effect of Cheonggukjang Water Extract (CWE) via activation of NF-κB pathways in murine macrophage RAW 264.7 cells (RAW 264.7 대식세포에서 청국장 열수 추출물(Cheonggukjang Water Extract, CWE)의 면역 증강 효과)

  • Due to the COVID-19 pandemic, the immuneenhancing health functional food market that protects our bodies from pathogens such as viruses continues to grow. In this study, we aimed to prove the Cheonggukjang, a high-nutrient food with high protein, fat, and dietary fiber content, as an immuneenhancing nutraceutical. Cheonggukjang water extract (CWE) increased the production of nitric oxide, reactive oxygen species, and cytokines such interleukin (IL)-6, IL-1β, and tumor necrosis factor-α without affecting viability in RAW 264.7 cells. Furthermore, CWE significantly upregulated the expression of inducible nitric oxide synthase and cyclooxygenase-2 in RAW 264.7 cells. CWE enhanced the phosphorylation of I kappa B kinase α/β and I kappa B (IκB)α, as well as the degradation of IκBα. CWE also induced increased phosphorylation of nuclear factor-kappa B p65 and facilitated the redistribution of p65 from the cytoplasm to the nucleus in RAW 264.7 cells. These findings suggest that CWE has potential as a health functional food material that can enhance the innate immune response.

Conditioned Medium of Soybean Extract Treated Osteoblasts Inhibits RANKL Induced Differentiation of Osteoclasts (대두추출물을 처리한 조골세포 조건배양액은 RANKL에 의해 유도된 파골세포 분화를 억제)

  • Soybean is of particular interest as a food supplement of isoflavones for inhibiting bone resorption in postmenopausal woman. These beneficial effects of isoflavones are caused by functioning as partial agonists or antagonists of estrogen, of which anti-resorptive effect is mediated indirectly through paracrine factors produced by osteoblasts that act on osteoclasts. In this study, the indirect effect of soybean on osteoclastic differentiation of RAW264.7 cells were investigated. The conditioned medium was collected from MC3T3-E1 osbeoblasts treated with 0.001 mg/mL~0.1 mg/mL soybean extracts for 6 days, mixed in 1:1 ratio with osteoclast medium, and then added into RAW264.7 cells with receptor activator of nuclear factor kappa B ligand (RANKL), a differentiation inducer for 3 days. Of paracrine factors in the conditioned medium, the protein expression of osteoprotegerin (OPG) with soybean extract was specifically higher in a dose dependent manner than with $10^{-9}$ M~$10^{-6}$ M of estrogen, genistein or daidzein standards. In RAW264.7 cells, the conditioned medium with soybean inhibited RANKL induced osteoclastic differentiation as total number of multinucleated tartrateresistant alkaline phosphatase (TRAP)-positive osteoclasts and protein expression of MMP-9 were significantly decreased. Coupled with the low expression of estrogen receptor $\alpha$ and $\beta$ proteins in RANKL treated RAW264.7 cells, we demonstrate that the conditioned medium of soybean treated osteoblasts inhibits RANKL induced differentiation of osteoclasts with the selective expression of OPG in osteoblasts.

Availability of Passiflora Caerulea Extract as Inner Beauty Material (이너뷰티 소재로서의 시계꽃 추출물의 활용 가능성)

  • This study attempted to investigate the physiological activities of Passiflora caerulea extract and provide basic data needed to verify its availability as an inner beauty material. To examine its anti-oxidant, anti-inflammatory and anti-aging effects, the followings were measured: total polyphenol contents, total flavonoid contents, DPPH radical scavenging activity, cytotoxicity and NO inhibition effects in RAW 264.7 macrophage cells and HDF cells and MMP-1 inhibitory effects in HDF cells. The results found the followings: First, polyphenol and flavonoid contents were great with 157 mg/g and 173.5 mg/g respectively at 10mg/mL, and DPPH radical scavenging activity was confirmed. In RAW 264.7 macrophage cells and HDF cells, no cytotoxicity was observed for Passiflora caerulea extract. In addition, 69.3% anti-inflammatory effects were found at 100 ㎍/mL through NO inhibition in RAW 264.7 macrophage cells. In addition, a significant inhibition of MMP-1 expression in HDF cell was confirmed. The above results confirmed the anti-oxidant and anti-inflammatory effects of Passiflora caerulea extract, low toxicity on skin cells and its anti-aging effects by inhibiting MMP-1 expression. Therefore, it appears that Passiflora caerulea extract would be available as diverse inner beauty materials.

Effect of Violae Herba Water Extract on the Proinflammatory Factors of LPS-Induced Macrophages (자화지정 추출물이 LPS로 유발된 대식세포의 염증인자에 미치는 영향)

  • The purpose of this study was to investigate the effects of Violae Herba Water Extract (VH) on the proinflammatory factors of lipopolysaccharide (LPS)-induced on the production of inflammatory mediators in RAW 264.7 mouse macrophages cells. We examined effect of Violae Herba Water Extract on the cell viability of RAW 264.7 mouse macrophages cells. Futhermore, After 24 hours treatment we investigated anti-inflammatory effect of Violae Herba Water Extract by the production of Bio-Plex cytokine assay, concentrations of various cytokines such NO, $interleukin(IL)-1{\beta}$, tumor necrosis factor ${\alpha}(TNF-{\alpha})$ and IL-6. The water extract of Violae Herba significantly inhibited the production of NO, $IL-1{\beta}$, $TNF-{\alpha}$ and IL-6 at the concentration of 25, 50, 100 and $200{\mu}g/mL$ in the LPS-induced RAW 264.7 mouse macrophages cells with no changes in the cell viability of them. These results suggest that water extract of Violae Herba has anti-inflammatory effect related with its inhibition of proinflammatory cytokines such as $IL-1{\beta}$, $TNF-{\alpha}$ and IL-6 in the LPS-induced RAW 264.7 mouse macrophages cells. Further research is needed to develop therapeutic agents for inflammatory diseases using Violae Herba.

Anti-inflammatory Effects of Achyranthes japonica Nakai and Aralia continentalis Kitagawa Complex Fermented Extracts on LPS-stimulated RAW264.7 Macrophage (LPS로 자극된 RAW264.7 대식세포에서 우슬 및 땅두릅 복합 발효추출물의 항염증 효과)

  • This study investigated the anti-inflammatory effects of mixed extracts of Achyranthes japonica Nakai (Aj) and Aralia continentalis Kitagawa (Ac) (ratios of 1 : 2, 1 : 3, 1 : 5, 2 : 1, 3 : 1 and 5 : 1) on RAW264.7 macrophages. Cell toxicity was determined using a cell counting kit (CCK) assay. We evaluated anti-inflammatory effects of the mixed extracts of Aj and Ac by measuring interleukin $(IL)-1{\beta}$, IL-6, and tumor necrosis factor $(TNF){\alpha}$ using an enzyme-linked immunosorbent assay (ELISA) kit assay. The mixed extracts of Aj and Ac inhibited lipopolysaccharide (LPS)-induced $IL-1{\beta}$ and $TNF{\alpha}$ in LPS-stimulated macrophages. Comparing different ratios of the mixed extracts, the 2 : 1 ratio of Aj and Ac has much more potency and inhibited the production of $TNF{\alpha}$ in LPS-induced RAW264.7 cells. The results of the present study showed that the mixed extracts of Aj and Ac have potential anti-inflammatory effects on RAW264.7 macrophages. Therefore, these extracts may be used as a good source of functional foods for the protection against inflammatory diseases.
