To investigate the effects of dietary calcium levels on the Ca metabolism in a rat model of ovariectomized osteoporosis, two studies were conducted. In Expt. I, five week-old femalc rats ovariectomized and fed a low Ca diet(0.06% Ca) for four weeks were compared with rats fed a normal (0.53% Ca) or low Ca diet under the sham-operated condition. Ovariectomized rats showed a significant increase in body weight and food intake. In rats fed the low Ca diet, a remarkable decrease was shown regardless of ovariectomy in serum Ca concentration, breaking force of bones, Ca and phopsphrus contents of bones, and apparent absorption and retention of Ca. Furthermore hte decrease of Ca contents of serum and bones in rats ovariectomized and fed the low Ca diet was similar to that in rats model of postmenopausal osteoporosis. In Expt. II, rats ovariectomized and fed on the low Ca diet for four weeks were divided into three groups, those given low Ca diet, normal Ca diet and high Ca diet(1.06%) respectively. The results indicated that supplementations of Ca at the intake level of 0.53% and 1.06% for 4 weeks tend to improve the relative Ca deficiency shown in experimental rat model of ovariectomized osteoporosis.
Biochemical studies such as growth rate, blood cholesterol and ascorbic acid contents in various organs of albino male rats were studied in the Garlic, Vitamin E, high and low protein diet fed groups. The results of this study were summarized as follows : 1) Either single 10% or 30% protein diet fed rats were not shown growth rate properly. In supplementation of Vitamin E or Garlic on 30% protein growth rate was a more increased than that of the 10% protein diet group. 2) The rate of food consumption of rats fed a 10% or 30% protein diet supplemented with Vitamin E and Garlic was more increased than that of the control Group receving 10% or 30% protein diet alone. 3) No essential difference was observed between the cholesterol level in blood of rats fed 10% protein diet supplemented with Vitamin E and Garlic and that of rats Receiving 10% protein diet alone. But the cholesterol level in blood of rats fed 30% protein diet supplemented with Vitamin E and Garlic was lower than that of control receiving 30% protein diet alone. 4) The Vitamin C contents in various organs of rats fed the diet supplemented with Vitamin E and Garlic seems to be a little higher than that of the control group fed the protein diet alone.
Purpose : Neuronal migration disorder(NMD) is a major underlying pathology of patients with intractable epilepsy. The role of NMD on seizure susceptibility or epileptogenecity, however, has not been documented. Methods : External irradiation of total amount of 250 cGY was performed to the fetal rats on days 16(E16) and 17(E17) of gestation. After delivery, the rats of 230-260g were decapitated for the histopathologic study. Epileptog-enecity of the NMD was studied by observing electroclinical events after intraperitoneal kainic acid(KA) injection in the control rats and NMD rats. Results : Histopathologic findings revealed focal and/or diffuse cortical dysplasia consisting of dyslamination of the cerebral cortex and appearance of the cytomegalic neurons, neuronal heterotopia in the periventricular white matter, dispersion of the pyramidal layer and the dentate gyrus of the hippocampus, and agenesis of the corpus callosum. Abnormal expression of neurofilaments protein(NF-M/H) was characteristically observed in the dysplastic neurons of the neocortex and hippocampus. Early ictal onset and prolonged ictal activity on EEG and clinical seizures were observed from the NMD rats unlike with the control rats. Conclusions : Exteranl irradiation on the fetal rats produced NMD. And the rats with NMD were highly susceptible to kainic acid provoked seizures. This animal model would be useful to study the pathophysiology of clinically relevant NMDs.
Spontaneous hypertensive rats (SHR) are an established model of genetic hypertension. Vascular smooth muscle cells (VSMC) from SHR proliferate faster than those of control rats (Wistar-Kyoto rats; WKY). We tested the hypothesis that induction of heme oxygenase (HO)-1 induced by aprotinin inhibits VSMC proliferation through cell cycle arrest in hypertensive rats. Aprotinin treatment inhibited VSMC proliferation in SHR more than in normotensive rats. These inhibitory effects were associated with cell cycle arrest in the G1 phase. Tin protoporphyrin IX (SnPPIX) reversed the anti-proliferative effect of aprotinin in VSMC from SHR. The level of cyclin D was higher in VSMC of SHR than those of WKY. Aprotinin treatment downregulated the cell cycle regulator, cyclin D, but upregulated the cyclin-dependent kinase inhibitor, p21, in VSMC of SHR. Aprotinin induced HO-1 in VSMC of SHR, but not in those of control rats. Furthermore, aprotinin-induced HO-1 inhibited VSMC proliferation of SHR. Consistently, VSMC proliferation in SHR was significantly inhibited by transfection with the HO-1 gene. These results indicate that induction of HO-1 by aprotinin inhibits VSMC proliferation through cell cycle arrest in hypertensive rats.
Thirty mael Sprague Dawley rats of 3 weeks of age were adopted to investigate changes in nutritive quality of the traditional, fermented soybean foods, Meju and Chungkugjang prepared by culturing with Aspergillus oryzae and the rice straw, respectively. The levels of each dietary protein were set at 10% . The effect of supplementation of fermented soybean on the rat diet was evaluated by measuring growth rate, feed efficiency. weight of the pancreas and biological values, such as NPU, PER and NPR on day 10. After incubation of soybean with Aspergilllus oryzae and the rice straw, the contents of crude protein, crude fat and crude ash were increased, but the levels of crude fiber and nitrogen-free extract were reduced. The amounts of feed intake, body weight gain an feed efficiency were greater for the rats fed dietscontaining heated unfermented soybeans or Meju compared to rats fed the diet containing raw soybeans. Pancreating enlargement was observed in the rats fed diets containing raw soybeans. The values of NPR and PER in rats fed diets containing Meju and Chungkugjang were significantly higher than those of rats fed diets containing raw soybeans. The values of NPU in rats fed diets containing raw soybans, heated unfermeted soybeans. Meju and Chungkugjang were 39.40%, 40.60%, 45.00% and 46.205, respectively, demonstrating no significant differences.
To study the effect of caffeine intake levels on iron metabolism, the iron utilization, iron contents of serum, liver, spleen, kidney, hemoglobin and hematocrit were compared in rats of different sex fed various levels of caffeine (3.5 and 7.0mg/100g body weight) for three weeks. There were no significant caffeine induced differences in feed intake, body weight gain but feed intake of male rats were significantly lower than that of female rats. Hemoglobin, hematocrit and iron contents of the serum were not significantly different between caffeine free and caffeine groups or male and female rats. Iron contents of kidney were decreased by elevation of injected caffeine levels rather than those of liver and spleen. caffeine male groups showed more increased uine volume, urinary and fecal excretions of iron than caffeine free or caffeine female groups. Apparent digestibility and retention of iron were significantly decreased by increment of injected caffeine levels. Male rate rats were more susceptible to injected caffeine on iron excretion than female rats. Current findings suggest that excessive caffeine consumption can affect iron excretion via urine and feces thereby decrease the utilization of iron, and have more significant effect on male than female rats.
This study was undertaken to determine the bioavailability of isoflavones in weanling Sprague-Dawley rats by providing diets containing different levels of soy isoflavones for 6 weeks: 0.025% (low isoflavone intake; LI), 0.125% (medium isoflavone intake; MI), and 0.25% (high isoflavone intake; HI). The subsequent fecal and urinary excretion of daidzein and genistein was then measured. As the levels of dietary isoflavones increased, the amount of food intakes significantly decreased, and weight gain was slower in female rats. In male rats, there was no significant difference in weight gains related to dietary intakes. Urinary excretion of daidzein and genistein was significantly higher in the MI and HI groups in both male and female rats than the control and LI groups. The recovery % of daidzein and genistein in the urine was significantly lower in the MI and HI groups. Fecal daidzein increased as dietary isoflavone intakes increased in female rats; however, in male rats the increase was significant only in the HI group. The recovery % of daidzein and genistein in the feces of female rats was not significantly different among the four groups. When dietary isoflavones were increased from 0.025% to 0.25%, the amounts of daidzein and genistein excreted in the urine and feces increased; however, the low recovery rate of both daidzein and genistein in the urine implies an increased bioavailability of isoflavones. We also observed sex-related differences in the urinary and fecal recovery of isoflavone intakes.
It has been known that activation of tyrosine kinases is involved in signal transduction. Role of the tyrosine kinase in vascular smooth muscle contraction was examined in deoxycorticosterone acetate (DOCA)-salt hypertensive rats. Male Sprague-Dawley rats underwent uninephrectomy, one week after which they were subcutaneously implanted with DOCA (200 mg/kg) and supplied with 1% NaCl and 0.2% KCl drinking water for $4{\sim}6$ weeks. Control rats were treated the same except for that no DOCA was implanted. Helical strips of carotid arteries were mounted in organ baths for measurement of isometric force development. Genistein was used as a tyrosine kinase inhibitor. Concentration-response curves to 5-hydroxytryptamine (5-HT) shifted to the right by genistein in both DOCA-salt hypertensive and control rats. Although the sensitivity to genistein was similar between the two groups, the maximum force generation by 5-HT was less inhibited by genistein in arteries from DOCA-salt hypertensive rats than in those from controls. Genistein-induced relaxations were attenuated in arteries from DOCA-salt rats. Genistein affected the contraction to phorbol 12, 13-dibutyrate (PDBu) neither in DOCA-salt nor in control arteries. These observations suggest that tyrosine kinase is involved in 5-HT-induced vascular contraction, of which role is reduced in DOCA-salt hypertension.
In the present study, the comparison of liver damage in carbon tetrachloride (CCl$_4$)-treated rats with that those pretreated with ethanol and an effect of liver injury on the serum and liver xanthine oxidase (XOD) activity were evaluated. The increasing rate of liver weight per body wt., the levels of serum alanine aminotransferase, and the decreasing rate of hepatic glucose-6-phosphatase activity and the protein contents in the liver cell were higher in carbon tetrachloride-treated animals pretreated with ethanol than the carbon tetrachloride-treated group. Especially, the histopathological findings also showed more severe liver damage in the ethanol-pretreated rats than the rats treated with carbon tetrachloride only. In such a experimental condition the xanthine oxidase activity of serum and liver both of carbon tetrachloride-treated rats and those pretreated with ethanol were higher than that of each control group. And the increasing rate of xanthine oxidase enzyme activity to the control group was higher in carbon tetrachloride-treated group pretreated with ethanol than those treated with CCl$_4$. In addition, the heptic uricase activity and the serum levels of uric acid were more increased in carbon tetrachloride-treated group pretreated with ethanol than those in the CCl$_4$-treated rats. On the other hand, there were no statistical differences in hepatic catalase and glutathione S-transferase activities between the CCl$_4$-treated rats and those pretreated with ethanol. In conclusion, it is assumed that the more severe liver damage in ethanol pretreated rats would be due to oxygen free radical produced by the xanthine oxidase system.
The repeated toxicity of Gleditschia-saponin produced and provided by S.S. Bio-Tech Bench Co. was evaluated in Sprague-Dawley rats. Gleditschia-saponin was administered to rats by oral route at dose levels of high (180 mg/kg/day), medium (90 mg/kg/day) and low (45 mg/kg/day) once a day for 14 days. Saline was administered to another group of rats as control. Each group was consisted of 5 male and female rats. There were no dose-related changes in clinical findings, food and water consumption, organ weights, urine analysis, biochemical examination and hematological findings in all groups of animals treated with Gleditschia.- saponin, except body weights. Body weighs in male and female rats were increased significantly (p < 0.05) from day 4 to 14 in low, middle and high dose groups than control group. Body weight in high dose group was increased higher than control or low, middle dose groups on day 14. Gross and histopathological findings revealed no evidence of specific toxicity to Gleditschia.-saponin. Therefore, it was concluded that Gleditschia-saponin had no toxic or side effects in Sprague-Dawley rats in an repeated oral toxicity tests.
