Biochemical Assessment of Deer Velvet Antler Extract and its Cytotoxic Effect including Acute Oral Toxicity using an ICR Mice Model (ICR 마우스 모델을 이용한 녹용 추출물의 생화학적 평가 및 급성 경구 독성을 포함한 세포 독성 효과)

  • Velvet antler is widely used as a traditional medicine, and numerous studies have demonstrated its tremendous nutritional and medicinal values including immunity-enhancing effects. This study aimed to investigate different deer velvet extracts (Sample 1: raw extract, Sample 2: dried extract, and Sample 3: freeze-dried extract) for proximate composition, uronic acid, sulfated glycosaminoglycan, sialic acid, collagen levels, and chemical components using ultra-performance liquid chromatography-quadrupole-time-of-light mass spectrometry. In addition, we evaluated the cytotoxic effect of the deer velvet extracts on BV2 microglia, HT22 hippocampal cells, HaCaT keratinocytes, and RAW264.7 macrophages using the cell viability MTT assay. Furthermore, we evaluated acute toxicity of the deer velvet extracts at different doses (0, 500, 1000, and 2000 mg/kg) administered orally to both male and female ICR mice for 14 d (five mice per group). After treatment, we evaluated general toxicity, survival rate, body weight changes, mortality, clinical signs, and necropsy findings in the experimental mice based on OECD guidelines. The results suggested that in vitro treatment with the evaluated extracts had no cytotoxic effect in HaCaT keratinocytes cells, whereas Sample-2 had a cytotoxic effect at 500 and 1000 ㎍/mL on HT22 hippocampal cells and RAW264.7 macrophages. Sample 3 was also cytotoxic at concentrations of 500 and 1000 ㎍/mL to RAW264.7 and BV2 microglial cells. However, the mice treated in vivo with the velvet extracts at doses of 500-2000 mg/kg BW showed no clinical signs, mortality, or necropsy findings, indicating that the LD50 is higher than this dosage. These findings indicate that there were no toxicological abnormalities connected with the deer velvet extract treatment in mice. However, further human and animal studies are needed before sufficient safety information is available to justify its use in humans.

A Safety Survey for Residual Pesticides in Agricultural Products in Meal-kits (밀키트(가정간편식) 중 농산물의 잔류농약 안전성 조사)

  • To investigate residual pesticide levels in agricultural products contained in Meal-kits, 27 Meal-kit products were collected from marts, Meal-kit shops, and online stores in Incheon City, South Korea. Seventy-six vegetable and thirty-seven mushroom products were analyzed for residual levels of 339 pesticides. Residual pesticides were detected in 23 out of 76 vegetables and were not present in the 37 mushroom products. The residual pesticide detection rate was 20.4% (23/113 cases). The pesticides famoxadone 0.034 mg/kg (standard: 0.01 mg/kg or less, PLS) and fenpyroximate 0.302 mg/kg (standard: 0.01 mg/kg or less, PLS) exceeded their maximum residue levels (MRL). This survey revealed that various types of pesticides remain in agricultural products in Meal-kits. Due to the nature of Meal-kit products, there is no separate standard for residual pesticides in agricultural products. Therefore, continuous monitoring of residual pesticides is necessary.

Development of a Rapid Enrichment Broth for Vibrio parahaemolyticus Using a Predictive Model of Microbial Growth with Response Surface Analysis (미생물 생장 예측모델과 반응표면분석법을 이용한 Vibrio parahaemolyticus의 신속 증균배지 개발)

  • In this study, we developed Rapid Enrichment Broth for Vibrio parahaemolyticus (REB-V), a broth capable enriching V. parahaemolyticus from 100 CFU/mL to 106 CFU/mL within 6 hours, which greatly facilitates the rapid detection of V. parahaemolyticus. Using a modified Gompertz model and response surface methodology, we optimized supplement sources to rapidly enrich V. parahaemolyticus. The addition of 0.003 g/10 mL of D-(+)-mannose, 0.002 g/10 mL of L-valine, and 0.002 g/10 mL of magnesium sulfate to 2% (w/v) NaCl BPW was the most effective combination of V. parahaemolyticus enrichment. Optimal V. parahaemolyticus culture conditions using REB-V were at pH 7.84 and 37℃. To confirm REB-V culture efficiency compared to 2% (w/v) NaCl BPW, we assessed the amount of enrichment achieved in 7 hours in each medium and extracted DNA samples from each culture every hour. Real-time PCR was performed using the extracted DNA to verify the applicability of this REB-V culture method to molecular diagnosis. V. parahaemolyticus was enriched to 5.452±0.151 Log CFU/mL in 2% (w/v) NaCl BPW in 7 hours, while in REB-V, it reached 7.831±0.323 Log CFU/mL. This confirmed that REB-V enriched V. parahaemolyticus to more than 106 CFU/mL within 6 hours. The enrichment rate of REB-V was faster than that of 2% (w/v) NaCl BPW, and the amount of enrichment within the same time was greater than that of 2% (w/v) NaCl BPW, indicating that REB-V exhibits excellent enrichment efficiency.

Sorghum Field Segmentation with U-Net from UAV RGB (무인기 기반 RGB 영상 활용 U-Net을 이용한 수수 재배지 분할)

  • When converting rice fields into fields,sorghum (sorghum bicolor L. Moench) has excellent moisture resistance, enabling stable production along with soybeans. Therefore, it is a crop that is expected to improve the self-sufficiency rate of domestic food crops and solve the rice supply-demand imbalance problem. However, there is a lack of fundamental statistics,such as cultivation fields required for estimating yields, due to the traditional survey method, which takes a long time even with a large manpower. In this study, U-Net was applied to RGB images based on unmanned aerial vehicle to confirm the possibility of non-destructive segmentation of sorghum cultivation fields. RGB images were acquired on July 28, August 13, and August 25, 2022. On each image acquisition date, datasets were divided into 6,000 training datasets and 1,000 validation datasets with a size of 512 × 512 images. Classification models were developed based on three classes consisting of Sorghum fields(sorghum), rice and soybean fields(others), and non-agricultural fields(background), and two classes consisting of sorghum and non-sorghum (others+background). The classification accuracy of sorghum cultivation fields was higher than 0.91 in the three class-based models at all acquisition dates, but learning confusion occurred in the other classes in the August dataset. In contrast, the two-class-based model showed an accuracy of 0.95 or better in all classes, with stable learning on the August dataset. As a result, two class-based models in August will be advantageous for calculating the cultivation fields of sorghum.

Anti-tumor and Anti-inflammatory Effects of Ecklonia cava in CT26 Tumor-bearing BALB/cKorl Syngeneic Mice (CT26 고형암을 내포하는 BALB/cKorl Syngeneic 마우스에서 Ecklonia cava의 항암효과 및 항염증효과)

  • The inflammatory response have been considered as one of important targets for cancer treatment because they play a key role during all steps of tumor development including initiation, promotion, malignant conversion and progression. To investigate the anti-inflammatory response during anti-tumor activity of an aqueous extracts of Ecklonia cava (AEC), alterations on the distribution of mast cells and the expression of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2), nuclear factor (NF)-κB, inflammasome compositional protein and inflammatory cytokines were examined in CT26 colon tumor-bearing BALB/cKorl syngeneic mice after administrating AEC for five weeks. After treatment of AEC, total weight of tumor and necrotic region of tumor section were significantly decreased compared to vehicle treated group. The number of infiltered mast cells was higher in AEC treated group than vehicle treated group, while the expression levels of COX-2 and iNOS were decreased in AEC treated group. Also, similar decrease pattern were detected in the expression levels of NF-κB, NLR family pyrin domain containing 3 (NLRP3), apoptosis-associated speck-like protein containing a caspase recruitment domain (ASC) and caspase-1 (Cas-1) after AEC treatment although the decrease rate was varied. Furthermore, the mRNA expressions of three inflammatory cytokines including tumor necrosis factor-α (TNF-α), interleukin-1α (IL-1α) and interleukin-6 (IL-6) were remarkably decreased in AEC treated group compared to vehicle treated group. These results suggest that inhibition of inflammatory response may be tightly associated with anti-tumor activity of AEC in CT26 colon tumor-bearing BALB/cKorl syngeneic mice.

Development Strengths of High Strength Headed Bars of RC and SFRC Exterior Beam-Column Joint (RC 및 SFRC 외부 보-기둥 접합부에 대한 고강도 확대머리 철근의 정착강도)

  • In this study, the development performance of the head bars, which is SD700, was experimentally evaluated at the RC (reinforced concrete) or SFRC (steel fiber reinforced concrete external beam-column joint. A total of 10 specimens were tested, and variables such as steel fibers, length of settlement, effective depth of the beam, and stirrups of the column were planned. As a result of the experiment, the specimens showed side-face blowout, concrete breakout, and shear failure depending on the experimental variables. In the RC series experiments with development length as a variable, it was confirmed that the development strength increased by 26.5~42.2% as the development length increased by 25-80%, which was not proportional to the development length. JD-based experiments with twice the effective depth of beams showed concrete breakout failure, reducing the maximum strength by 31.5% to 62% compared to the reference experiment. The S-series experiment, in which the spacing of the shear reinforcement around the enlarged head reinforcement was 1/2 times that of the reference experiment, increased the maximum strength by 8.4 to 9.7%. The concrete compressive strength of SFRC was evaluated to be 29.3% smaller than the concrete compressive strength of RC, but the development strength of SFRC specimens increased by 7.3% to 12.2%. Accordingly it was confirmed that the development performance of the head bar was greatly improved by reinforcing the steel fiber. Considering the results of 92% and 99% of the experimental maximum strength of the experiment arranged with 92% and 110% of the KDS-based settlement length, it is judged that the safety rate needs to be considered even more. In addition, it is required to present a design formula that considers the effective depth of the beam compared to the development length.

A Study on the Perception of Policy Targets to Improve the Effectiveness of Child Safety Measure - Focusing on Children, Guardians, and Workers in Children's Facilities - (어린이 안전대책 실효성 향상을 위한 정책대상자 인식조사 연구 - 어린이, 보호자, 어린이이용시설 종사자 중심으로 -)

  • Purpose: This study aims to come up with improvement measures to improve the effectiveness of child safety measures. Method: The current status of child safety accidents was investigated and implications were deduced by analyzing major child safety measures by government department in the past. In addition, a perception survey was conducted on 1,000 people including children, guardians, and children's facility workers who are subject to child safety policies. Result: Regarding the safety of children's living space(environment), 35.3% of guardians answered that more than 1/3 of them were not safe. Both guardians(95.3%) and children's facility workers(89%) answered that there was the highest risk of 'traffic accidents', and the second risk factor was parents(carelessness of workers at children's facilities) and children's facility workers(careless of guardians at home). Looking at the risks by place, "road and sidewalk" was the most dangerous place and for child safety, guardians(64.3%) and workers (78.3%) both said that the role of "parent" is the most important. For improvements to prevent child safety accidents, the response rate of "strengthening safety management of road traffic facilities" is the most necessary with 75.8% for guardians and 65% for child use facilities. Conclusion: The reinforcement measures to strengthen the effectiveness of child safety measures are as follows. First, in order to ensure the continuity of child safety measures, it should be operated effectively so that those subject to the establishment of the Comprehensive Plan for Child Safety, which took effect in August 2022, can feel it. Second, in order to improve the sensitivity of children's policy targets, promotion measures that take into account the characteristics of each child safety field should be continuously strengthened. Third, it is necessary to expand safety infrastructure for each field to secure child safety. Fourth, it is necessary to strengthen safety education that can ensure safety for children themselves and to come up with detailed measures to make safety education for parents(guardians) mandatory.

  • Today, when innovation and creativity become increasingly important, management of human resources is a key factor for corporate performance and competitive advantage. Corporate are implementing and introducing various types of support methods for members to achieve goals and improve organizational performance. Organizational culture and organizational trust affect the cognitive and emotional state of members. Furthermore, it can bring about changes in organizational performance such as job stress and job satisfaction. From an institutional point of view, work-life balance is also a major factor affecting organizational performance. The imbalance between work and life leads to a decline in organizational performance, such as decreased morale and dissatisfaction with work. In relation to work-life balance, the low birth rate problem intensified and the importance began to emerge. Therefore, the government has implemented various policy support for workers' work-life balance, and the "52-hour workweek" is a representative example. This study analyzed the effect of organizational culture applying the competitive value model on workers' job stress. In addition, the mediating effects of overtime work per week and organizational trust were analyzed. Job stress corresponds to a prerequisite stage that affects job commitment, job satisfaction, and turnover intention. However, research measuring job stress by organizational performance is insufficient. In addition, there are few studies analyzing the relationship between overtime and organizational performance. Considering this, it is necessary to understand the influence relationship. The results of the study are as follows. First, a hierarchical culture increases the job stress of workers. On the other hand, innovation-oriented, relationship-oriented, and competition-oriented corporate culture reduce job stress. Second, a hierarchical culture has reduced trust in the organization, and other organizational cultures have increased trust in the organization. Third, relationship-oriented and competition-oriented organizational culture reduced overtime. Innovation-oriented, hierarchical-oriented culture increased overtime Fourth, organizational trust and overtime have the effect of mediating organizational culture and job stress. Based on these analysis results, this study presented academic and political implications.

Effects of Startup Motivation, Competency, and Passion on Startup Satisfaction : A Comparative Study between Early Startups and General Startups (창업동기, 역량 및 열정이 창업 만족도에 미치는 영향 : 일반 창업기업과 스타트업의 차이를 중심으로)

  • Although the importance of starting a business is being emphasized, it is not easy for a start-up to survive. Therefore, it is necessary to come up with a plan to strengthen startup satisfaction and improve the survival rate of startup companies. In this study, we examined the impact of start-up motivation, start-up capabilities, and start-up passion of start-up companies on start-up satisfaction, as well as the moderating effect according to the founding type of general start-up companies and startups. Entrepreneurship motivation was divided into self-actualization motivation and pro-social motivation. Entrepreneurship capabilities were divided into marketing capabilities and experience capabilities. Entrepreneurship passion was divided into harmonious passion and obsessive passion. Empirical analysis was conducted by receiving surveys from 361 managers of general and startup companies within 7 years of founding their businesses online and offline. As a result, self-actualization motivation, pro-social motivation, marketing competency, experiential competency, and harmonious passion had a significant positive (+) effect on startup satisfaction. However, obsessive passion did not have a significant effect. In addition, the magnitude of influence on startup satisfaction appeared in the following order: harmonious passion, marketing competency, self-actualization motivation, pro-social motivation, and experiential competency. The moderating effect according to the type of founding of general start-ups and startups was found in pro-social motivation, experience competency, and passion for harmony. However, there was no significant moderating effect between self-actualization motivation, marketing competency, obsessive passion, and entrepreneurship satisfaction. This study presented academic implications that harmonious passion and marketing capabilities are important factors in improving startup satisfaction. In addition, practical and policy implications were presented, including the need for programs to support the psychological stability of startup business owners, strengthening startup capabilities through experts, and strengthening startup education programs that can foster self-actualization motivation and pro-social motivation.

Constructing a Conceptual Framework of Smart Ageing Bridging Sustainability and Demographic Transformation (인구감소 시대와 초고령 사회의 지속가능한 삶으로서 스마트 에이징의 개념과 모형에 관한 탐색적 연구)

  • As population ageing and shrinking accompanied by dramatically expanded individual life expectancy and declining fertility rate is a global phenomenon, ageing becomes its broader perspective of ageing well embedded into sustained health and well-being, and also the fourth industrial revolution speeds up a more robust and inclusive view of smart ageing. While the latest paradigm of SA has gained considerable attention in the midst of sharply surging demand for health and social services and rapidly declining labor force, the definition has been widely and constantly discussed. This research is to constitute a conceptual framework of smart ageing (SA) from systematic literature review and the use of a series of secondary data and Geographical Information Systems(GIS), and to explore its components. The findings indicate that SA is considered to be an innovative approach to ensuring quality of life and protecting dignity, and identifies its constituents. Indeed, the construct of SA elaborates the multidimensional nature of independent living, encompassing three spheres - Aging in Place (AP), Well Aging (WA), and Active Ageing (AA). AP aims at maintaining independence and autonomy, entails safety, comfort, familiarity and emotional attachment, and it values social supports and services. WA assures physical, psycho-social and economic domains of well-being, and it concerns subjective happiness. AA focuses on both social engagement and economic participation. Moreover, the three constructs of SA are underpinned by specific elements (right to housing, income adequacy, health security, social care, and civic engagement) which are interrelated and interconnected.
