Acinetobacter baumannii strains are emerging pathogens of the nosocomial infection with an increasing frequency in recent years. The therapeutic difficulty due to the wide spread of multiple resistant strains was major problem in A. baumannii infection. It seems likely that high frequency of A. baumannii infection will be increasing epidemiological importance in the future. However, the current limited understanding of the epidemiology of A. baumannii infections is caused by lack of a rapid and practical method for the molecular characterization of A. baumannii strains. This study was undertaken to determine molecular types and genetic similarity among A. baumannii strains isolated from four hospitals by RAPD analysis. Eighty-five strains, including 40 from Chunnam University Hospital, 27 from Dankook University Hospital, 15 from Yonsei University Hospital, and 3 from Seonam University Hospital, were classified into three molecular types. Molecular type II was the most common pattern and included 72 strains. All strains from Dankook University Hospital and 40 strains from Chunnam University Hospital belonged to molecular type I or II. A. baumannii strains form Yonsei University Hospital were very distant similarity values. The range of genetic similarity values among 85 strains of A. baumannii was 0.26 to 1.00. Although phenotypes including biotype and antimicrobial resistance pattern of A. baumannii strains were same or very similar to each other, their RAPD patterns were quite different. Typing with phenotypes was found to be less reliable than molecular typing by RAPD analysis. These results suggest that RAPD analysis provides rapid and simple typing method of A. baumannii strains for epidemiological studies. This work is the first epidemiological report of A. baumannii infections in Korea and it is hoped that results of this work may contribute to a better understanding of the clinical importance and epidemiology of A. baumannii strains.
The study was carried out for comparison of variants and development of genetic markers using Randomly Amplified Polymorphic D사A (RAPD) analysis method. The ginseng variants used were as follows: Chungkyung-Chong, Hwangskoog-Chong, KG101 selected by the pureline selection method, and 6 kinds of Jakyung-Chong strains Uinjakyung, Jakyung-Chong 81783, Jakyung-Chong 847913, Jaky tong-Chong 79742, Jinjakyung of USSR, and Mimaki of Japan). Four of 10 RAPD primers showed the distinctive polymorphism among 9 ginseng variants and lines, and were selected for more detailed polymorphic analysis. The sequences of 4 selected primers were TGCCGAGCTG (Primer#2), AATCGGGCTG (#4), GAAACGGGTG (U7), and GTGACGTAGG (#8). All primers produced several common bands among the strains. However, when primer # 2 was applied, the electrophoregram showed the specific band at 1.8 kb region in Chungkyung-Chong, Hwangskoog- chong, and KG101, and 1 kb in the Jakyung-Chong 847913. In primer #4, 1.1 kb band was shown in Chungkyung-Chong, Hwangskoog-Chong, KG101, and Jakyung-Chong 79742. In primer # 7, 700 bp band was appeared in Jakyung-Chong 81783 and Jinjakyung of USSR In primer # 8, 800 bp band was observed only in Mimaki, comparing to another strains. When Similarity Index (SI) was calculated, Chungkyung-Chong and Hwngskoog-Chong, and Jakyung- chong 81783 and Jinjakyung of USSR showed the most close SI, 0.11 and 0.08, respectively. The data of KG101, which showed the SI of 0.13 with the group of Chungkyung-Chong and Hwangskoog-Chong, coincided with the fact that it was released from Hwangskoog-Chong by breeding process. The data of Jakyung strains indicated the significant variation among the strains. From these results, RAPD analysis method could be succesively applied to the classification and genetic analysis for breeding of Korean ginseng.
RAPD를 이용하여 국내에 생육하는 4종의 자생 민들레와 2종의 귀화 민들레 종간의 유전적인 유연관계를 분석하였다 Primer Screening을 통해 선발된 30개의 Primer를 이용하여 RAPD를 행한 결과, Polymorphic band 수는 141개로 나타나 민들레 종간의 유전적인 차이 분석이 가능하였다. Primer OPC12, OPD16, OPK16, OPK17, OPK20, OPSI에서는 각 종마다. 특정 밴드가 있는 것으로 조사되었다. 특히 OPS8에서는 564bp에서 귀화종인 서양민들레 종에서만 특이 밴드가 나타났다. RAPD분석결과 자생종과 귀화종 민들레들은 명확히 2군으로 구분되었다. 1군에는 서양민들레. 붉은씨서양민들레 등의 귀화종 그룹이었으며, II군은 민들레, 산민들레, 좀민들레, 횐민들레 등의 자생종 민들레 그룹으로 나뉘어졌다. Bootstrap 방법으로 6종의 유연관계를 분석한 결과도 비유사계수를 통한 방법으로 분석한 결과와 매우 유사하였다 우리 나라에서 자생하는 민들레속 6종의 주요형질을 조사한 결과 I군에 속하는 민들레류는 II군에 속한 종들에 비해 개화일수가 길고, 외총포편의 방향이 다르며 털의 유무 또한 다른 특징이 있었다. ll군에 속하는 횐민들레는 다른 민들레종과는 화색에서 확연한 차이를 보였으며 자생종들 속에서도 잎의 방향과 발아의 생리적 특성 등 유전적으로 여러 다른 점이 복합적으로 작용함으로서 II군내에서도 유전적 거리가 가장 먼 것으로 나타났다.
형태적으로 매우 유사한 적조생물 C. polykrikoides, G. impudicum, G. catenatum을 RAPD 방법을 이용하여 유전적 유연관계를 조사했다. 12개의 primer 중 4종류만 선택되었고 증폭된 밴드수는 59개이며 그 크기는 0.2에서 3.0 kb까지였다. C. polykrikoides, G. impudicum, G. catenatum의 다형화된 밴드수는 16개, 8개, 16개로 각각 나타났다. 반면에, 17개의 밴드만 동일하였다. C. polykrikoides, G. impudicum, G. catenatum의 종 특이적인 밴드수는 26개, 34개, 26개로 각각 보였다. C. polykrikoides와 G. impudicum/G. catenatum의 유전적 유사성은 0.83이며, G. impudicum과 G. catenatum은 0.78로 나타났다. 이러한 결과로 볼 때 형태적으로는 유사하게 보이지만, RAPD 분석에 의하면 C. polykrikoides, G. impudicum, G. catenatum은 현저하게 상이한 적조생물이다. 앞으로 RAPD 기법을 이용하면 이러한 와편모조류의 유전적 변이를 탐색하는데 유용할 것으로 보인다.
Primer 20가지로 Listeria spp.에 대해 screening을 하여 병원균인 L. monocytogenes를 구별하게 하는 RAPD-PCR의 band pattern을 나타내는 10-mer random primer가 OPG-13이 라는 것을 알았다. OPG-13은 GC%가 70%이므로 계산상으로는 annealing 온도가 34$^{\circ}C$이다. 32~36$^{\circ}C$까지 다섯 가지의 온도로 annealing한 결과 L. monocytogenes만이 갖는 특정한 크기의 band가 역시 34$^{\circ}C$에서 형성됨을 알아냈고, 34$^{\circ}C$를 annealing 온도로 정하였다. Line 1부터 4까지 1. monocytogenes ATCC15313, 19111, 19112, 19113은 2개의 700 bp와 1500 bp band를 형성하였고 그 밖의 Listeria spp.들 Line 6부터 11까지 L. ivanovii ATCC 19119, L. grayi ATCC19120, L. murrayi ATCC25401, L. innocua ATCC33090, L. welshimeri ATCC35897, L. seeligeri ATCC35967은 약 2,000 ~ 2,300 bp크기 한 개의 band를 보여 병원성 균과 비병원성 균이 매우 확실하게 구분되는 band pattern이 나타나는 이러한 결과로 10-mer random primer인 OPG-13을 이용한 RAPD-PCR 방법이 병윈균인 L. monocytogenes를 검색하는데 이용될 수 있음을 확인할 수 있었다.

In this study, we investigated genetic diversity of 22 microsporidian isolates infecting tropical tasar silkworm, Antheraea mylitta collected from various geographical forest locations in the state of Jharkhand, India, using polymerase chain reaction (PCR)-based marker assay: random amplified polymorphic DNA (RAPD). A type species, NIK-1s_mys was used as control for comparison. The shape of mature microsporidians was found to be oval to elongate, measuring 3.80 to $5.10{\mu}m$ in length and 2.56 to $3.30{\mu}m$ in width. Of the 20 RAPD primers screened, 16 primers generated reproducible profiles with 298 polymorphic fragments displaying high degree of polymorphism (97%). A total of 14 RAPD primers produced 45 unique putative genetic markers, which were used to differentiate the microsporidians. Calculation of genetic distance coefficients based on dice coefficient method and clustering with un-weighted pair group method using arithmetic average (UPGMA) analysis was conducted to unravel the genetic diversity of microsporidians infecting tasar silkworm. The similarity coefficients varied from 0.059 to 0.980. UPGMA analysis generated a dendrogram with four microsporidian groups, which appear to be different from each other as well as from NIK-1s_mys. Two-dimensional distribution based on Euclidean distance matrix also revealed considerable variability among different microsporidians identified from the tasar silkworms. Clustering of few microsporidian isolates was in accordance with the geographic origin. The results indicate that the RAPD profiles and specific/unique genetic markers can be used for differentiating as well as to identify different microsporidians with considerable accuracy.
The genetic diversity among the genus Viola was evaluated using the random amplified polymorphic DNA (RAPD) method. A total of 142 distinct amplification fragments by 18 random primers were scored to perform the cluster analysis with UPGMA. Viola species from the subsection Patellares were clustered into group I to IV. The groups from I to IV were consistent with its morphological taxonomy, series Pinnatae, Chinensis, Variegatae, and Patellares in the subsection Patellares, respectively. Even though V. albida and V. albida var. takahasii were classified in Chinensis, they were assigned into group I. The cluster analysis separated other subsections from Patellares in the section Nomimium. Interestingly, V. verecunda and V. grypoceras in subsections Biobatae and Trigonocarpae, respectively, were clustered into group C with a high similarity coefficient. Therefore, RAPD analysis can be used for providing an alternative classification system to identify genotypes and morphological characters of Viola species.

This study established the genetic characterisation of 10 microsporidian isolates infecting non-mulberry silkworms (Antheraea assamensis and Samia cynthia ricini) collected from biogeographical forest locations in the State of Assam, India, using PCR-based markers assays: inter simple sequence repeat (ISSR) and random amplified polymorphic DNA (RAPD). A Nosema type species (NIK-1s_mys) was used as control for comparison. The shape of mature microsporidian spores were observed oval to elongated, measuring 3.80 to $4.90{\mu}m$ in length and 2.60 to $3.05{\mu}m$ in width. Fourteen ISSR primers generated reproducible profiles and yielded 178 fragments, of which 175 were polymorphic (98%), while 16 RAPD primers generated reproducible profiles with 198 amplified fragments displaying 95% of polymorphism. Estimation of genetic distance coefficients based on dice coefficients method and clustering with un-weighted pair group method using arithmetic average (UPGMA) analysis was done to unravel the genetic diversity of microsporidians infecting Indian muga and eri silkworm. The similarity coefficients varied from 0.385 to 0.941 in ISSR and 0.083 to 0.938 in RAPD data. UPGMA analysis generated dendrograms with two microsporidian groups, which appear to be different from each other. Based on Euclidean distance matrix method, 2-dimensional distribution also revealed considerable variability among different identified microsporidians. Clustering of these microsporidian isolates was in accordance with their host and biogeographic origin. Both techniques represent a useful and efficient tool for taxonomical grouping as well as for phylogenetic classification of different microsporidians in general and genotyping of these pathogens in particular.
ITS 염기서열과 RAPD를 이용하여 F. velutipes 29개의 팽이버섯 품종 간의 유전적 변이를 분석하였다. ITS 부위에서 720 bp의 염기서열을 확인 하였으나 29개의 팽이품종간에 유의적인 차이가 없었다. RAPD 분석 결과 40개의 random primer 중 다형성을 나타내는 primer는 16개였으며, 그 중 뚜렸한 다형성을 띄는 primer는 OPA-2,4,3,9,10,20 이었다. 이들 29개 품종에서 primer에 의해 증폭된 밴드는 모두 3,030개 였으며, DNA 단편의 크기는 200~2,000 bp 사이에 위치하였다. 또한 3,030개의 scrabble RAPD band들을 marker로 하여 Nei-Li's의 방법을 이용한 비유사도 지수행렬을 조사한 결과 전체 29개 품종의 종내 유전적 변이는 3.3~45%였고, 특히 한국 야생팽이의 종내 유전적 변이도는 17~38.6%로 품종 간 다형성을 확인하였다. RAPD 변이에 기초하여 neighbor-joining tree (NJ) 분석에서는 5개의 cluster로 구분되었으며, 각각의 cluster는 품종, 지역 적 특성을 나타내었다. 본 연구 결과 RAPD와 실험을 통해 확인된 OPA, OPB primer의 경우 미확인 팽이품종들을 검색 하는데 분자 유전적 표지 maker로써 이용 할 수 있는 것으로 생각된다.
Oenanthe javanica와 O. javanica var. japonica 한국 내 식용미나리이다. Cicuta virosa는 독미나리로 시큐톡신을 함유하고 있다. 미나리에 대한 RAPD 마크에 의한 분자적 변이를 조사하였다. RAPD 분석을 위해 10개 올리고프라이머(오페론, OPB)로 6개 집단을 분석하였다. 6개 집단에서 72개 DNA 분절(밴드)을 찾았다. 72 밴드 중 61개(84.7%) 밴드는 다형현상을 나타내었다. 한국 내 독미나리 자연 집단은 작고 격리되어 패치 분포를 이루고 있지만 높은 유전적 다양도를 가지고 있었다. 반면에, 재배종 미나리(Oenanthe javanica var. japonica) 집단은 채소용으로 논에서 광범위한 분포를 나타내지만 독미나리와 야생종 미나리에 비해 낮은 유전적 다양도를 나타내었다. 비록 야생집단이 재배집단에 비해 다양도가 높지만 유의한 차이는 없었다. 전체 유전적 다양도(HT)는 0.342였다. 집단 내 유전적 다양도(HS)는 0.201이였다. 유전자 좌위에 근거한 집단 간 분화에서 전체 유전적 다양도의 비율(GST)은 0.414이므로 전체 변이의 41.4%는 집단 간에 있었다. 결론으로 RAPD기법은 독미나리와 식용미나리의 동정에 유용하였다. 또, RAPD의 OPB 마크는 미나리의 다른 자원과 식품 자원을 식별하는 데 도움이 될 수 있는 분자 마크임을 보여주었다.
