Nutrient Utilization, Body Composition and Lactation Performance of First Lactation Bali Cows (Bos sondaicus) on Grass-Legume Based Diets

  • A study on energy and protein utilization, and milk production of Bali cows on grass-legume diets was carried out using 12 first lactation cows (initial BW $263.79{\pm}21.66kg$) during a period of 16 weeks starting immediately post calving. The animals were randomly allotted into 4 dietary treatment groups R1, R2, R3 and R4, receiving from the last 2 months of pregnancy onwards, graded improved rations based on a mixture of locally available grass and legume feed ad libitum. R1 contained on a DM basis 70% elephant grass (PP, Penisetum purpureum) plus 30% Gliricidia sepia leaves (GS), R2 was 30% PP plus 55% GS supplemented with 15% Hibiscus tilliactus leaves (HT, defaunating effect), R3 and R4 were 22.5% PP+41.25% GS+11.25% HT+25% concentrate, where R3 was not and R4 supplemented with zinc di-acetate. TDN, CP and zinc contents of the diets were 58.2%, 12.05% and 18.3 mg/kg respectively for R1, 65.05%, 16.9% and 25.6 mg/kg respectively for R2, 66.03%, 16.71% and 29.02 mg/kg respectively for R3 and 66.03%, 16.71% and 60.47 mg/kg respectively for R4. Milk production and body weight were monitored throughout the experimental period. In vivo body composition by the urea space technique validated by the body density method and supported by carcass data was estimated at the start and termination of the experiment. Nutrient balance and rumen performance characteristics were measured during a balance trial of 7 days during the 3rd and 4th week of the lactation period. Results indicated that quality of ration caused improvement of ruminal total VFA concentration, increments being 52 to 65% for R2, R3 and R4 above R1, with increments of acetate being less (31 to 48%) and propionate being proportionally more in comparison to total VFA increments. Similarly, ammonia concentrations increased to 5.24 to 7.07 mM, equivalent to 7.34 to 9.90 mg $NH_3-N/100ml$ rumen fluid. Results also indicated that feed quality did not affect DE and ME intakes, and heat production (HP), but increased GE, UE, energy in milk and total retained energy (RE total) in body tissues and milk. Intake-, digestible- and catabolized-protein, and retained-protein in body tissues and milk (Rprot) were all elevated increasing the quality of ration. Similar results were obtained for milk yield and components with mean values reaching 2.085 kg/d (R4) versus 0.92 kg/d (R1) for milk yield, and 170.22 g/d (R4) vs 71.69 g/d (R1), 105.74 g/d (R4) vs 45.35 g/d (R1), 101.34 g/d (R4) vs 46.36 g/d (R1) for milk-fat, -protein, and -lactose, respectively. Relatively high yields of milk production was maintained longer for R4 as compared to the other treatment groups. There were no significant effects on body mass and components due to lactation. From the relationship $RE_{total}$ (MJ/d)=12.79-0.373 ME (MJ/d); (r=0.73), it was found that $ME_{m}=0.53MJ/kgW^{0.75}.d$. Requirement of energy to support the production of milk, ranging from 0.5 to 3.0 kg/d, follows the equation: Milk Prod. ($Q_{mp}$, kg/d)=[-2.48+4.31 ME($MJ/kg^{0.75}.d$)]; (r=0.6) or $Q_{mp}$=-3.4+[0.08($ME-RE_{body\;tissue}$)]MJ/d]; (r=0.94). The requirement for protein intake for maintenance ($IP_m$) equals $6.19 g/kg^{0.75}.d$ derived from the relationship RP=-47.4+0.12 IP; (r=0.74, n=9). Equation for protein requirement for lactation is $Q_{nl}$=[($Q_{mp}$)(% protein in milk)($I_{mp}$)]/100, where $Q_{nl}$ is g protein required for lactation, $Q_{mp}$ is daily milk yield, Bali cow's milk-protein content av. 5.04%, and $I_{mp}$ is metabolic increment for milk production ($ME_{lakt}/ME_{m}=1.46$).

Development of Multiplex Polymerase Chain Reaction Assay for Identification of Angelica Species (Multiplex Polymerase Chain Reaction을 이용한 당귀 종 판별)

  • Background: Angelica gigas, A. sinensis, and A. acutiloba are commercially important in the herbal medicine market, and among them, A. gigas has the highest economic value and price. However, their similar morphological traits are often used for fraud. Despite their importance in herbal medicine, recognition of the differences between Angelica species is currently inadequate. Methods and Results: A multiplex polymerase chain reaction (PCR) method was developed for direct detection and identification of A. gigas, A. sinensis, and A. acutiloba. The gene for the distinction of species was targeted at ITS in the nucleus and trnC-petN gene in chloroplasts. The optimized multiplex PCR in the present study utilized each Angelica species-specific primer pairs. Each primer pair yielded products of 229 base pairs (bp) for A. gigas, 53 bp for A. sinensis, 170 bp for A. acutiloba. Additionally non-specific PCR products were not detected in similar species by species-specific primers. Conclusions: In the present study, a multiplex-PCR assay, successfully assessed the authenticity of Angelica species (A. gigas, A. sinensis, and A. acutiloba). and whole genome amplification (WGA) was performed after DNA extraction to identify, the species in the product. The detection method of raw materials developed in the present study could be applied to herbal medicine and health functional food management.

Effect of Psychological Characteristics on Job Performances of Workers at Residential Homes for the Disabled

  • The purpose of this research was to analyze the effect of psychological characteristics on job performances of workers at residential homes for the disabled. For the study, a survey was conducted on 170 workers at residential homes for the disabled in Gyeongsangbuk-do (Sangjoo-si, Andong-si and Yecheon-gun) from the 7th of October to 11th of November, 2016. For the analysis, SPSS WIN 18.0 and Amos 21.0 programs were used to conduct t-test, one-way analysis of variance, correlation analysis and multiple regression analysis at a significance level of 5%. The research results were as follows. First, it was found that the psychological characteristics of workers at residential homes for the disabled vary significantly upon their work loads. Second, the differences in job performances of workers at the residential homes for the disabled with their general characteristics were analyzed: it was revealed that the job performance of the workers varies significantly with their education levels, positions and monthly incomes. Third, the job performance of workers at the residential homes for the disabled was found to be having positive correlation with their psychological characteristics as follows: self-esteem (r=.363, p<.001) and self-efficacy (r=.275, p<.001), while having negative correlation with depression (r=-.267, p<.001). This paper is required to be used as a primary source for political development on the enhancement of the psychological characteristics of workers at residential homes for the disabled.

Molecular Identification of Sarcocystis grueneri in Wild Korean Water Deer (Hydropotes inermis argyropus)

  • The cysts of Sarcocystis grueneri were detected and characterized from the cardiac muscles of the Korean water deer (Hydropotes inermis argyropus). Of the 38 heart muscle samples examined by light microscopy, 10 were found infected with the cysts of Sarcocystis sp. The cysts appeared oval to spherical shape and measured $110-380{\mu}m$ in length and $90-170{\mu}m$ in width. A phylogenetic tree of the 18S rRNA sequences (1.5 kb) revealed a close relationship of the infected cysts to genus Sarcocystis. The 18S rRNA sequence of the infected cysts showed 100% identity to S. grueneri and 97% to S. capracanis. Here, we first report the S. grueneri infections in the Korean water deer.

Experimental Study on Particle and Soluble Gas Removal Efficiency of Water-cyclone (Water-cyclone을 이용한 미세입자 및 수용성가스 제거효율에 대한 실험적 연구)

  • The fine-particles, moisture and variety of hazardous gases are produced during electronic manufacture process. Most of the fine-particles are 0.1~10 ${\mu}m$ in size and the hazardous gases such as HF, $SiH_4$, CO, $NH_3$, etc. seriously affect environment, human's body and manufacturing process. To remove these characterized gases and fine-particles, Water-Cyclone designed and tested for removal efficiency on fine-particles and $NH_3$ under -980Pa negative pressure condition. As a result, under 0.1~1.0 $m^3/min$ flow condition, the efficiency on 5 ${\mu}m$ particles was 80~96%, 10 ${\mu}m$ particles was 86~96%, and 20 ${\mu}m$ particles was 91~99%. Besides, the removal efficiency on soluble gas $NH_3$ was 56.5% at 0.5m3/min and 79.1% at 1.0m3/min under 500 ppm flow concentration and 70.0% at 1.0 $m^3/min$ under 1,000 ppm flow concentration. Therefore, on particles, as the flow rate and particle size increased, the collection efficiency rate was increased. On soluble gas, as the flow rate increased, the removal efficiency was increased under the same concentration.

The microbial diversity analysis of the Korea traditional post-fermented tea (Chungtaejeon) (한국 전통 미생물발효차(청태전)의 미생물 군집분석)

  • Tea is the most popular beverage in the world. In fact, there are mainly three different kinds of tea (Green tea, black tea, and post-fermented tea). Post-fermented tea is produced by the microbial fermentation process using sun-dried green tea leaves (Camellia sinensis) as the raw material. Chungtaejeon was a traditional tea introduced in the age of the ancient three states and is the only "Ddeok-cha or Don-cha" culture in the world that survived on the southwestern shore of Republic of Korea. In this study, the structures of the bacterial community involved in the production of oriental traditional post-fermented tea (Chungtaejeon) were investigated using 16S rRNA gene analysis. The 16S rRNA gene analysis of dominant microbial bacteria in post-fermented tea confirmed the presence of Pantoea sp., and Klebsiella oxytoca. Phylogenetic analysis suggested that the taxonomic affiliation of the dominant species in the post-fermented tea was ${\gamma}$-proteobacteria. As a result of the microbial community size analysis, it was confirmed that the size of the microbial communities of Chungtaejeon was the largest compared to other teas

Design and Evaluation of Aluminum Casting Alloys for Thermal Managing Application (방열소재용 알루미늄 주조합금 설계 및 특성평가)

  • In order to develop an aluminum alloy, that can combine high thermal conductivity and good castability and anodizability, aluminum alloys with low Si content, such as Al-(0.5~1.5)Mg-1Fe-0.5Si and Al-(1.0~1.5)Si-1Fe-1Zn, were designed. The developed aluminum alloys exhibited 170~190% thermal conductivity (160~180 W/mK), 60~85% fluidity, and equal or higher ultimate tensile strength compared with those of the ADC12 alloy. In each developed alloy system, the thermal conductivity decreased and the strength increased with the increment of Mg and Si, which are the significant alloying elements. The fluidity was in reverse proportion to the Mg content and in proportion to the Si content. The Al-(0.5~1.5)Mg-1Fe-0.5Si alloys exhibited better fluidity in thick-wall castings, while the Al-(1.0~1.5)Si-1Fe-1Zn alloys were better in thin-wall castability due to their lower surface energies. The fluidity behavior was complexly affected by the heat release for the solidification, viscosity, solidification range, and the type, quantity, and formation juncture of the main secondary phase.

Dependence of Barredness of Late-Type Galaxies on Galaxy Properties and Environment

  • We investigate the dependence of occurrence of bar in galaxies on galaxy properties and environment. The environmental conditions considered include the large-scale background density and distance to the nearest neighbor galaxy. We use a volume-limited sample of 33,296 galaxies brighter than $M_r$=-19.5+5logh at $0.02{\leqq}z{\leqq}0.05489$, drawn from the Sloan Digital Sky Survey Data Release 7. We classify the galaxies into early and late types, and identify bars by visual inspection. We find that the fraction of barred galaxies ($f_{bar}$) is 18.2% on average in the case of late-type galaxies, and depends on both u-r color and central velocity dispersion $(\sigma);f_{bar}$ is a monotonically increasing function of u-r color, and has a maximum value at intermediate velocity dispersion (${\sigma}{\simeq}170km\;s^{-1}$). This trend suggests that bars are dominantly hosted by systems having intermediate-mass with no recent interaction or merger history. We also find that $f_{bar}$ does not directly depend on the large-scale background density as its dependence disappears when other physical parameters are fixed. We discover the bar fraction decreases as the separation to the nearest neighbor galaxy becomes smaller than 0.1 times the virial radius of the neighbor regardless of neighbor's morphology. These results imply that it is difficult for bars to be maintained during strong tidal interactions, and that the source for this phenomenon is gravitational and not hydrodynamical.

Screening of a Potent, Raw Naked Barley Saccharifying Enzyme Producer and Its Application on the Uncooked Alcohol Fermentation (쌀보리 전분 당화효소 생산균의 분리 동정 및 무증자 알코올 발효에의 이용)

  • Microorganisms capable of degrading the raw naked barley were isolated from soil, and the amylase productivity of each strain was examined on plate contained 2% raw naked barley. Of the fungi and actinomycetes tested, 71 strains were subjected to subsequent testing for amylase production, and 4 strains were selected as potent amylase producers. Among them, Strain No. 281 produced the most potent raw naked barley saccharifying enzyme, and was identified as genus Rhizopus from morphological and physiological studies. The ratio of raw starch saccharifying activity (RDA) of the crude enzyme derived from the Rhizopus sp. No. 281 was showed 2-3 fold higher than that of commercial enzyme when the raw naked barley was used as the substrate. In the case of uncooked alcohol fermentation using Rhizopus sp. No. 281 glucoamylase preparation, the alcohol yield of the broth was 2% higher than that of the commercial enzyme.

Effects of 8-Fluorociprofloxacin And Ciprofloxacin on the Central Nervous System

  • The objectives of this sutdy were to investigate the effects of 8-fluorociprofloxacin(8-FCP) on the central nervous system (CNS) and to compare with those of ciprofloxacin(CP). The $LD_{50}$ values of intravenous 8-FCP were similar or slightly lower in rat (M;203.6mg/kg, F;186.1mg/kg)and markedly lower in mice (M;126.5mg/kg, F;163.1mg/kg), as compared to those of CP. However, no recognizable differences in the clinical signs and recovery were found between 8-FCP and CP in both species. In combination with fenbufen, the convulsive liability of 8-FCP was higher than that of CP. At an intravenous dose of 10mg/kg, 8-FCP provoked convulsive signs and subsequent death in mice, whereas CP produced convulsion at a dose of 40mg/kg. The hexobabital -induced sleeping time was markedly lengthened by the oral administration of 8-FCP, but slightly increased by CP. In addition, the two quinolone derivatives had analgesic effects. The analgesic activity of 8-FCP was approximately two times higher than that at CP. However, both 8-FCP and CP had little effects of pentylenetetrazole-or strychnine-induced convulsion and muscle relaxation. Our finding that 8-FCP had more remarkable CNS effects than CP strongly suggests that there should be differences in the pharmacokinetic characteristics and/or in the binding affinity for specific biologic targets, or receptors, in the CNS.
