Development of Dual Reporter System of Mutant Dopamine 2 Receptor ($D_2R$) and Sodium Iodide Symporter (NIS) Transgenes (변이 도파민 2 수용체와 나트륨 옥소 공동 수송체 이입유전자의 이중 리포터시스템 개발)

  • Purpose: Both human NIS and mutant $D_2R$ transgenes are proposed as reporting system in transplanted cell tracking. Using hepatoma cell lines, we constructed a dual reporter system containing human sodium-iodide symporter (hNIS) and dopamine 2 receptor ($D_2R$) and compared its characteristics. Materials and Methods: The recombinant plasmid ($pIRES-hNIS/D_2R$) was constructed with IRES (internal ribosome entry site) under control of the CMV promoter $pIRES-hNIS/D_2R$ was transfected to human hepatoma SK-Hep1 cell line with lipofectamine. HEP-ND ($SK-Hep1-hNIS/D_2R$) cells stably expressing hNIS and $D_2R$ was established by selection with G418 for two weeks. RT-PCR was performed to investigate the expression of both hNIS and $D_2R$ genes. The expressions of hNIS and $D_2R$ were measured by $^{125}I$ uptake assays and receptor binding assays. Specific binding of $D_2R$ to $[^3H]spiperone$ was verified by Scatchard plot with (+) butaclamol as a specific inhibitor. $K_d\;and\;B_{max}$ values were estimated. The correlation between hNIS and $D_2R$ expression was compared by using each clone. Results: Similar quantities of hNIS and $D_2R$ genes were expressed on HEP-ND as RT-PCR assays. HEP-ND cells showed 30 to 40 fold higher radioiodine uptakes than those of parental SK-Hep1 cells. $^{125}I$ uptake in HEP-ND cells was completely inhibited by $KClO_4$, a NIS inhibitor Specific binding to HEP-ND cells was saturable and the $K_d\;and\;B_{max}$ values for HEP-ND cells were 2.92 nM, 745.25 fmol/mg protein and 2.91nM, 1323 fmole/mg protein in two clones, respectively. The radioiodine uptake by hNIS activity and $D_2R$ binding was highly correlated. Conclusion: We developed a dual positron and gamma imaging reporter system of hNIS and $D_2R$ in a stably transfected cell line. We expect that $D_2R$ and hNIS genes can complement mutually as a nuclear reporting system or that $D_2R$ can be used as reporter gene when hNIS gene were used as a treatment gene.

  • The NMR chemical shift arising from 4d electron orbital angular momentum and 4d electron spin dipolar-nuclear Spin angular momentum interactions for a $4d^2$ system in a strong crystal field environment of octahedral symmetry has been investigated when the four fold axis is taken as the quantization axis. The NMR results are comparted with the multipolar shift at various R-values and we find that the exact results are in agreement with the multipolar shift when $R{\geqslant}0.20 nm.$ We also separate the NMR shift into the contribution of the $1/R^5$ and $1/R^7$ terms. It is found that the contribution of the $1/R^5$term to the NMR shift is dominant than the contribution of the $1/R^7$ term. Temperature dependence analysis shows that the $1/T^2$ term is the dominant contribution to the NMR shift for a $4d^2$ system but the contribution of the 1/T term may not negligible. The similar results are obtained for a $4d^1$ system from the temperature dependence analysis.

International R&D Contest with IPR Coordination and Cost Externality

  • Purpose - This paper examines the international R&D contest in which the extent of intellectual property right (IPR) affects both the size of prize for the winning firm and the extent of positive spillover through cost of firms. Recognizing the possibility of incomplete protection of IPR, the present paper analyzes the effect of changes in the extent of IPR on payoffs to firms and social welfare. Design/methodology - This paper examines coordination of IPRs by countries in economic integration. The paper then develops a general model of international R&D contest with incomplete protection of IPR. An increase in the extent of IPR augments the share of the prize the winning firm can appropriate, while decreasing the positive cost externality. To derive sharper results, the paper considers the cases of linear and fixed spillovers. Findings - Under plausible assumptions, an increase in the IPR augments the payoff to each firm and the aggregate payoffs as well. The paper also shows that the number of firms participating in the R&D contest can be endogenously determined in the two-stage R&D contest. The higher the extent of cost spillover, and the larger the effective prize, the more firms participate in the international R&D contest. Originality/value - Existing studies assume that firms winning the R&D contest enjoy perfect IPR to the output of their R&D activities. This is a very restrictive assumption in that other firms can copy the new products or processes. By allowing for the incompleteness of the IPR, the present paper develops a more realistic model of R&D contest. The novelty of the present paper is to allow for the possibility that the higher extent of IPR increases the prize and decreases positive cost externality at the same time. The findings of the present paper can serve as a basis for government policy toward R&D activities of firms and protection of IPRs.

  • In the course of a search for tumor necrosis factor $(TNF)-{\alpha}$ inhibitory compounds from medicinal plants, we identified neolignans, honokiol and magnolol, from the alcoholic extract of Magnolia offcinalis as active inhibitory principles. These compounds dose-dependently inhibited $TNF-{\alpha}$ production without displaying cytotoxicity and their inhibitory activities measured by $IC_{50}$ values were 53.7 and $61.4\;{\mu}M$, respectively.

Performance Evaluation of Private R&D Projects using BSC/DEA (BSC/DEA를 활용한 기업 연구개발 프로젝트 성과평가)

  • This paper proposes a R&D project performance measurement model for private firms combining balanced scorecard (BSC) and data envelopment analysis (DEA). The efficiency of R&D projects is measured in terms of each of the three perspectives of BSC by using DEA : the internal process perspective (DEA-P), the customer perspective (DEA-C), and the financial perspective (DEA-F). The performance indicators of the three perspectives of BSC are considered as outputs of the corresponding DEA models. To provide strategic implications for R&D planning, we also propose the R&D project performance matrices composed of two different types of efficiency dimensions. The proposed model is expected to be fruitfully utilized for R&D performance measurement of private firms.

  • In this study, as the result of analyzing the relationship and influence between economic outcome and R&D supporting investment, the number of supporting subjects among the technological innovational areas of SMEs, it is as below. First, as the economic result of analyzing companies from the investment in R&D of technological innovational areas of minor companies, the number of supporting subjects and amount of R&D have relationship with increase of sales and export amount, employee reduction & the effect of new job creation shows positive correlation with the effect of import replacement. Second, as analyzing the influence of the investment in R&D has economic effect from of technological innovational minor companies. This is thought that the financial and R&D support increase a significant effect on economical, technical against SMEs.

Impact of COVID-19 on R&D Cost Stickiness in IT industry

  • This study aims to examine whether there are some differences in the cost stickiness of R&D expenses of IT companies before and after the COVID-19 crisis. Before COVID-19, IT companies did not reduce R&D expenses even if sales decreased, resulting in cost stickiness. As a result, it appears that during the COVID-19, IT companies adjust R&D expenses in proportion to changes in sales. This is interpreted as a reduction in R&D investment, which takes a long time to create results, in case of a decrease in sales due to uncertainty in future management performance due to the COVID-19 pandemic. In other words, during the COVID-19 risk period, IT companies, like other companies, reduced R&D expenses as sales decreased, resulting in reduced cost stickiness. This study differs from existing literature in that it examines the impact of the COVID-19 pandemic on the R&D expenditure behavior of companies in the IT industry.

Thus it is important to develop differentiated technology for secure competitiveness. In this paper, We analyze patents for R&D strategy about glassless 3D display. Through Company-Technology matrix analysis and Patent trend analysis, We extract promising technology field and core technology. Lastly We suggest R&D strategy by using patent road map.
