This study was conducted to investigate the effects of quiescent treatment of donor cells and activation treatment time of recipient cytoplasm on nuclear remodeling and in vitro development of somatic cell-cloned bovine embryos. Serum starved, confluent and nonquiescent cycling adult skin cells were teansferred into enucleated oocytes. Nuclear transfer oocytes were activated at 30 min, 1 and 2 hrs after electrofusion. Some nuclear transfer embryos(23% to 35%) extruded a polar body, which was not affected by quiescent treatment of donor cells and activiation time of recipient cytoplasm. About 68% of nuclear transfer embryos fused with a serum starved cells has a chromatin clump, but which was not different from embryos fused with confluent(51%) and nonquiescent(47%) cells. The proportion of embryos with a single chromatin clump was sightly increased when nuclear transfer embryos were activated within 30 min after fusion(69%) compared to those were activated at 1 and 2 hrs after fusion, but there was not significantly different. Development rates to the blastocyst stage were 8.6% and 15.9% when serum starved and confluent cells were transferred, which were higher than that of control group. Developmental rate to the blastocyst stage was higher in embryos were activated within 30 min after fusion (17.3%) compared to those of embryos were activated at 1 and 2 hrs after fusion (P<0.05). From the present result, it is suggested that quiescent treatment of donor cells and activation time of recipient cytoplasm can affect the in vitro development. Quiescent plasm activation within 30 min after fusion could increase the number of embryos with a normal chromation structure, which results in increased in vitro development.
본 연구는 세포의 휴면처리가 소 태아섬유아세포 유래 핵이식란의 핵형변화와 체외발육에 미치는 영향을 검토하였다. 임신 3∼4 개월령 한우 웅성 태아의 피부세포를 채취하여 계대배양 후 동결하였다가 핵이식 전에 혈청기아처리 또는 confluency방법으로 휴면처리를 하여 미수정란의 탈핵세포질에 이식하였다. 전기융합과 활성화처리 후 7∼9 일간 체외배양하여 발육능을 검토하였으며, 일부는 whole-mount 법으로 고정하여 염색질 구조를 관찰하였다. 복제란의 극체방출율은 혈청기아처리구와 confluence 구에서 각각 24.5% 와 20.3% 로 무처리구(36.0%) 에 비해 낮은 경향을 보였다. 활성화 후 1개의 염색질괴를 갖는 복제란은 혈청기아처리구(50.9%)와 confluence구 (49.2%)가 무처리구 (40.0%) 보다 높은 경향을 보였다. 극체방출에 따른 염색질의 구조는 극체 미방출구에서 정상적인 1개의 염색질괴를 갖는 핵이식란이 60.5% 로, 극체 방출구 (4.7%) 에 비하여 유의적으로 높았다 (P<0.01). 배반포기 발육율은 혈청기아처리구 (21.7%) 와 confluence (20.9%) 가 무처리구 (14.1%) 에 비하여 비교적 높게 나타났다. 본 연구의 결과 혈청기아처리나 confluency 방법에 의한 donor 세포의 휴면처리는 태아섬유아세포 유래 핵이식란의 비정상적인 핵형변화를 감소시켜 체외발육능을 향상시키는 것으로 사료된다.
The quantity of residuals generated from water treatment plants depends upon the raw water quality, dosage of chemicals used, performance of the treatment process, method of sludge removal, efficiency of sedimentation, and backwashing frequency. Sludge production by the physical separation of SS occurs under quiescent conditions in the primary clarifier, where suspended solids are allowed to settle and to consolidate on the clarifier bottom. Raw primary sludge results when the settled solids are hydraulically removed from the tank. The relative solid and liquid fractions of a slurry are most commonly described by the solids concentration, expressed as mg/L or percent solids. The purpose of the present investigation is to estimate a suitability on the design capacities of residuals treatment facilities by the quantity of dewatered sludge generated from water treatment plants.

At present, it is difficult to solve problems related with thickening and dewatering of WTP sludge, and discharging waste water to river. The glial fibrillary acidic protein (GFAP) is traditionally used as a marker for astrocytes of the brain, and more recently for the hepatic stellate cells (HSCs) of the liver. Several GFAP splice variants have been previously reported in the astrocytes of the CNS and in the non-myelinating Schwann cells of the PNS. In this study, we investigate whether GFAP splice variants are present in the HSCs and their expression as a function of HSCs activation. Furthermore, the regulation of these transcripts upon treatment with interferon gamma ($IFN-{\gamma}$) will be explored. Using semi-quan-titative RT-PCR and real-time PCR, we examine the expression and regulation of GFAP splice variants in HSCs as well as their respective half-life. We discover that most of the GFAP splice variants ($GFAP{\alpha}$, ${\beta}$, ${\delta}$, ${\varepsilon}$ and $\kappa$) found in the neural system are also expressed in quiescent and culture-activated primary HSCs. Interestingly, $GFAP{\alpha}$ is the predominant form in quiescent and culture-activated primary HSCs, while $GFAP{\beta}$, predominates in the SV40-immortalized activated HSC-T6. $GFAP{\delta}$, ${\varepsilon}$ and ${\kappa}$ have similar half-lives of 10 hours, while $GFAP{\beta}$ has a half-life of 17 hours. Treatment of HSC-T6 with $IFN-{\gamma}$ results in a significant 1.29-fold up-regulation of $GFAP{\alpha}$ whereas the level of the other transcripts remains unchanged. In summary, $GFAP{\alpha}$, ${\beta}$, ${\delta}$, ${\varepsilon}$ and $\kappa$ are present in HSCs. They are differentially regulated on the transcription level, implying a role of the 5' and 3' untranslated regions.
A rigorous study of single droplet vaporization under quiescent high pressure atmosphere is attempted adopting method of flash evaporation calculation for vapor-liquid equilibrium. Results due to flash method shows excellent agreement with measurement. Also shown is the present model fairly capable of depicting transients of droplet vaporization under high pressure environment, such as ambient gas solubility, property variation, and multicomponent transports. Systematic treatment of these effects with emphasis on vapor-liquid phase equilibrium revealed; conventional treatment for subcritical droplet vaporization, such as $d^2$-law, leads to erroneous prediction of droplet history, augmented gas solubility is significant under supercritical pressure, and vaporization rate proportionally increase with pressure.

Microbial biofilms are resilient, immune-evasive, often antibiotic-resistant health challenges, and increasingly the target for research into novel therapeutic strategies. We evaluated the effects of a nutraceutical enzyme and botanical blend (NEBB) on established biofilm. Five microbial strains with known implications in chronic human illnesses were tested: Candida albicans, Staphylococcus aureus, Staphylococcus simulans (coagulase-negative, penicillin-resistant), Borrelia burgdorferi, and Pseudomonas aeruginosa. The strains were allowed to form biofilm in vitro. Biofilm cultures were treated with NEBB containing enzymes targeted at lipids, proteins, and sugars, also containing the mucolytic compound N-acetyl cysteine, along with antimicrobial extracts from cranberry, berberine, rosemary, and peppermint. The post-treatment biofilm mass was evaluated by crystal-violet staining, and metabolic activity was measured using the MTT assay. Average biofilm mass and metabolic activity for NEBB-treated biofilms were compared to the average of untreated control cultures. Treatment of established biofilm with NEBB resulted in biofilm-disruption, involving significant reductions in biofilm mass and metabolic activity for Candida and both Staphylococcus species. For B. burgdorferi, we observed reduced biofilm mass, but the remaining residual biofilm showed a mild increase in metabolic activity, suggesting a shift from metabolically quiescent, treatment-resistant persister forms of B. burgdorferi to a more active form, potentially more recognizable by the host immune system. For P. aeruginosa, low doses of NEBB significantly reduced biofilm mass and metabolic activity while higher doses of NEBB increased biofilm mass and metabolic activity. The results suggest that targeted nutraceutical support may help disrupt biofilm communities, offering new facets for integrative combinational treatment strategies.
The evolution of conformational constraints in bisphenol-A polycarbonate (BAPC) upon quiescent bulk crystallization was quantitatively analyzed from calorimetric study employing a rigid amorphous fraction (RAF) as an indicator of the level of conformational constraints. From the correlation between corrected crystallinity (X$\sub$c/) and total rigid fraction (f$\sub$r/), it was found that, regardless of molar mass distribution and thermal treatment conditions, semicrystalline BAPC always exhibits greater f$\sub$r/ than X$\sub$c/ maintaining a quantitative relationship of f$\sub$r/〓2X$\sub$c/ in the range of 0.0 $\sub$c/< 0.4. This directly indicates the evolution of approximately the same amount of RAF as X$\sub$c/, (i.e., RAF〓X$\sub$c/) upon bulk crystallization of BAPC. It was also found that T$\sub$g/ per se and T$\sub$g/ broadening enhance as RAF increases, and there appears to be a critical level of RAF (>0.2) needed to initiate significant changes in both quantities.
Manassantin B, a dilignan isolated from Saururus chinensis, significantly inhibited proliferation in HSC-T6 cells in concentration- and time-dependent manners. In addition, treatment of HSC-T6 cells with manassantin B changed cell morphology from flattened myofibroblastic membranous morphology, representing activation state, to slender shape, representing quiescent state. Furthermore, manassantin B effectively reduced collagen content in HSC-T6 cells. These results suggested that manassantin B exerted antifibrotic activity in HSCT6 cells, in part, via inhibition of cell proliferation and decrease of collagen production.
Angiogenesis refers to the formation of new capillary blood vessels, or neovascularization occurring under various physical conditions, such as development of the embryo, formation of corpus luteum, wound healing and pathological conditions including tumor growth and metastases, hemangiomas, diabetic retinopathy, rheumatoid arthritis. There are many evidences that angiogenesis is important for the progressive growth of solid tumors and also permits the shedding of metastatic tumors from the primary site. Thus, treatment of angiogenesis inhibitors might be a novel strategy for tumor growth inhibition. Normal vascular endothelial cells are in a state of differentiation and angiogenic endothelial cells migrate and proliferate, and they subsequently differentiate into vessel-forming quiescent phenotype cells, Therfore, it was speculated that a modifier of cell differentiation could also affect angiogenesis. In order to identify new antiangiogenic factors, the research was conducted to estimate the inhibitory activities of cell differentiation agents by means of chick embryo chorioallantoic membrane(CAM) assay. Hence, we have established the CAM assay for the screening of antiangiogenic agents. Using the CAM assay, we found that ursolic acid, a tumor cell differentiation-inducing agent, showed a markedly inhibitory effect on chick embryonic angiogenesis.
