Quantitative Analysis of Cyanidin-3-O-rutinoside and Quercetin-3-O-rutinoside from Extracts of Morus alba Fruit (오디 추출물로부터 Cyanidin-3-O-rutinoside와 Quercetin-3-O-rutinoside의 함량분석)

  • Morus alba fruit contains several anthocyanins and flavonoids like cyanidin-3-O-rutinoside and quercetin-3-O-rutinoside. Extractions from Morus alba fruit were performed with five different compositions of ethanol/water [v/v]. High-Performance Liquid Chromatography (HPLC) was used for the validation and content determination of cyanidin-3-O-rutinoside and quercetin-3-O-rutinoside. The contents of cyanidin-3-O-rutinoside in ethanol extracts (100%, 70%, 40%, 20% and 0%, ethanol/water [v/v]) were 1.150%, 0.822%, 0.749%, 0.663% and 0.597%, respectively. Quercetin-3-O-rutinoside contents of ethanol extracts (100%, 70%, 40%, 20% and 0%, ethanol/water [v/v]) were 0.167%, 0.161%, 0.159%, 0.155% and 0.096%, respectively. The highest contents of cyanidin-3-O-rutinoside and quercetin-3-O-rutinoside were in the ethanol extract with 100% ethanol/water [v/v]. These HPLC analysis method could be used as basic data for standardization of functional food from Morus alba fruit.

  • A new flavonol glycoside, quercetin 3-rhamnosyl $(1{\rightarrow}3)$ galactoside [5] was isolated from the leaves of M. africana. The known compounds kaempferol 3-rutinoside [1], 3'-O-methylquercetin 3-rutinoside [2], quercetin 3-rutinoside [3], and quercetin 3-rhamnosyl $(1{\rightarrow}6)$ galactoside [4] were also isolated for the first time from this plant. Their structures were determined by chemical and spectroscopic methods.

  • The antioxidative constituents isolated from Hedyotis diffusa were identified as quercetin 3-O-${\beta}$-rutinoside (1) and quercetin 3-O-${\beta}$-glucoside (2). We also isolated asperuloside (3) from this plant. Identification was done based on spectroscopic analysis. Quercetin 3-O-${\beta}$-rutinoside was the stronger antioxidant than quercetin 3-O-${\beta}$-glycoside while asperuloside was inactive.

Flavonoids analysis in leaves and fruits of Korean mulberry cultivar, Baekokwang having white fruits

  • Morus alba has white and/or purple fruits with a very sweet taste and low acidity. Most Korean mulberry trees have purple fruits. However, Baekokwang is a unique mulberry genetic resource in Korea with white fruits. In this study, flavonoids contents of Baekokwang mulberry leaf and fruit were analyzed using ultrahigh performance liquid chromatography coupled with diode array detection and quadrupole time-of-flight mass spectrometry (UPLC-DAD-QTOF/MS) technique. UPLC-DAD-QTOF/MS chromatogram showed that 15 flavonoids and 9 flavonoids were isolated and identified from the mulberry leaf and fruit. Total flavonoids contents of Baekokwang leaves and fruits were 812.7 mg and 35.0 mg, respectively. Baekokwang leaves had 4 major flavonoids including quercetin 3-O-(6"-O-malonyl) glucoside, 235.3 ppm, kaempferol 3-O-(6"-O-malonyl) glucoside, 132.3 ppm, kaempferol 3-O-rutinoside (nicotiflorin), 108.1 ppm, and quercetin 3-O-rutinoside (rutin), 103.8 ppm. Baekokwang fruits had 3 major flavonoids including quercetin 3-O-(6"-O-malonyl) glucoside, 13.0 ppm, quercetin 3-O-rutinoside (rutin), 7.8 ppm, and kaempferol 3-O-rutinoside (nicotiflorin), 5.7 ppm. From the above results, mulberry leaves have rich flavonoids compared to its fruits.

Seasonal Variations of the Flavonol Glycoside Content from Ginkgo biloba Leaves (은행잎중 Flavonol Glycoside 성분의 계절별 함량 변화에 관한 연구)

  • The seasonal variations of the major six flavonol glycosides(kaempferol 2,6-dirhamnosyl glucoside, quercetin 3-O-rutinoside, kaempferol 3-O-rutinoside, isorhamnetin 3-O-rutinoside, quercetin 3-O-coumaroyl glucorhamnoside and kaempferol 3-O-coumaroyl glucorhamnoside) in Ginkgo biloba leaves were investigated. The contents were determined by HPLC on reversed phase $C_{18}$ column. This result showed that the percentage of six flavonol glycosides decreased during the season from 1.57% in May to 0.39% in November. The content of each flavonol glycoside indicated a similar tendency to decrease. However, the contents of rutinosides of kaempferol, quercetin and isorhamnetin fluctuated markedly than those of coumaroyl glucorhamnosides of kaempferol and quercetin and kaempferol 2,6-dirhamnosyl glucoside.

  • From the herbs of this plant, six compounds were isolated and their structures were characterized as dieicosyl ether (1), scopoletin (2), ${\alpha}-spinasterol-3-O-{\beta}-D-glucopyranoside$ (3), kaempferol (4), quercetin (5), and quercetin 7, 3', 4'-trimethylether-3-O-rutinoside (6) by chemical and spectroscopic evidences. These compounds were isolated for the first time from this plant and compound 6 was first isolated from nature.

  • The dried neeldes(1.5 kg) of Juniperus rigida were ground, extracted with acetone-H2O(7:3, v/v), concentrated, and fractionated with a series of hexane, CH2Cl2, EtOAc and water on a separatory funnel. Each fraction was freeze dried to give dark-brown powder and EtOAc and water soluble fractions were chromatographed on a Sephadex LH-20 column using a series of aqueous methanol and ethanol-hexane mixture as eluent. Spectrometric analysis such as NMR and FAB or EI-MS including TLC were performed to characterize the structures of the isolated compounds. The needles of Juniperus rigida contained a large amount of (+)-catechin, quercetin-3-O-α-L-rhamnopyranoside and isoconiferin, in addition to a small amount of umbelliferone and quercetin-3-O-β-D-rutinoside. The antioxidative activities of each fraction and isolated compounds were tested by DPPH radical scavenging method, and EtOAc soluble fraction, (+)-catechin and quercetin-3-O-α-L-rhamnopyranoside were effective.

DPPH Radical Scavenging Effect of the Aerial Parts of Fagopyrum esculentum and Isolation of Bioactive Flavonoids (메밀 지상부의 DPPH 라디칼 소거작용과 활성 플라보노이드의 분리)

  • The inhibitory effect of the aerial parts of Fagopyrum esculentum on the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical was examined. The n-butanol fraction from the methanol extract of title plant showed stronger inhibitory effect than other fractions on DPPH radical. Two flavonoids were isolated from n-butanol fraction having the potent activity and elucidated as quercetin-3-O-${\alpha}$-L-rhamnoside and quercetin-3-O-rutinoside on the basis of spectral evidence. The $IC_{50}$ values of these compounds on DPPH radical were 6.56 ${\mu}M$ and 8.37 ${\mu}M$, respectively.

  • Four flavonoids were isolated from the aerial parts of Euphorbia ebracteolata. On the basis of chemical and spectroscopic evidence, the structures of these compounds were established as $quercetin-3-O-{\beta}-D-glucoside$(isoquercitrin)(I), quercetin-3-O-rutinoside(rutin)(II), kaempferol-3-O-rutinoside(III) and $quercetin-3-O-(2'-O-galloyl)-{\beta}-D-glucoside$(IV) which was the main flavonoidal component in this plant. The isolation of flavonoids from E. ebracteolata is the first example.

  • A chemical examination of the aerial parts of Euphorbia pekinensis $R_{UPRECHT}$. (Euphorbiaceae) has led to the isolation of seven hydrolyzable tannins and ten fl avonoid glycosides. The former ones have been identified as gallic acid, methylgallate, 3-O-galloyl shikimic acid, 1,3,4,6-tetra-O-galloyl-${\beta}-_D$-glucose, 1,2,3,4,6-penta-O-galloyl-${\beta}-_D$-glucose, corilagin, geraniin and the latter ones as isoquercitrin, quercitrin, astragalin, afzelin, prunin, rutin, kaempferol-3-O-rutinoside, quercetin-3-O-(2"-O-galloyl)-${\beta}-_D$-glucoside and quercetin-3-O-(2"-O-galloyl)-${\alpha}-_L$-rhamnoside on the basis of chemical and spectroscopic evidence.
