The Comparative Quantitative Risk Assessment of LNG Tank Designs for the Safety Improvement of Above Ground Membrane Tank (지상식 멤브레인 LNG저장탱크 안전성 향상을 위한 설계형식별 정량적 위험성 비교 평가)

  • The objective of paper is to carry out a comparative Quantitative Risk Assessment (QRA) of two KOGAS tank designs using a fault tree methodology, a standard 'Full Containment' tank and a 'Membrane' tank. For the membrane tank, both the initial KOGAS design and 4 modified KOGAS designs have been assessed, giving six separate cases. In this paper, the frequencies of releases are quantified using a fault tree approach. For clarity in the analysis, and to ensure consistency, all cases have been quantified using the same fault tree. Logic within the fault tree is used to select each of the cases. Full quantification of risks is often difficult, owing to a lack of relevant failure data, but the aim of this study has been to be as quantitative as possible, with full transparency of failure information. The most significant general cause of external LNG leaks is predicted to be a seismic event, which has been quantified nominally. 4modified KOGAS desiens to Prevent damage of bottom membrane panels that was shown in preparatory estimation could quantitively confirm safety improvement. According to result, the predicted frequencies of an external LNG leak for the full containment and modified membrane tanks are very similar, failures due to dropped pumps are predicted to be significantly greater for the membrane tank with thickened plate than for the full containment tank.

Protective Effects of Vitamin C against Genomic DNA Damage Caused by Genotoxicants (유전독성물질의 유전체 손상 작용에 대한 Vitamin C의 방호효과)

  • Although it is popularly believed that vitamin C protects cells from various genotoxicants, the degrees and mechanisms of itsprotective actions are not fully understood. In this study, vitamin C's protective effects against various genotoxicants were quantified, together with subsequent analyses on the mechanisms of these protective effects. Comet assay was employed to measure the degree of DNA damage in Chinese hamster ovary cells (CHO-K1) exposed to five genotoxicants, $H_2O_2$, $HgCl_2$, N-methyl-N-nitro-N-nitrosoguanidine (MNNG), 4-nitroquinoline-1-oxide (4NQO), and UV-irradiation. In cases cells were treated with $H_2O_2$, $HgCl_2$, and 4NQO together with vitamin C, the damage to DNA decreased to the level of the control group. In cases of UV-irradiation, the protective effect of vitamin C appeared, but did not reach the control levels. Interestingly, vitamin C did not have protective effects against the genotoxicity of MNNG. The degrees of DNA damage of cells treated with vitamin C prior to exposure togenotoxicants were 28~49% lower than those of cells treated with vitamin C after being exposed to genotoxicants. In conclusion, vitamin C had strong antioxidanteffects against genotoxicants by being a primary antioxidant blocking genotoxicity reaching the cells, rather than being a secondary antioxidant acting on post-exposure DNA repair processes. However, vitamin C's protective effects appearto be limited, as there are genotoxicants, such as MNNG, whosegenotoxicityis not affected by vitamin C. Therefore, the results of this study warrant furtherstudies on toxic mechanisms of genotoxicants and their interactions with protective mechanisms of vitamin C.

Quantification of Climate Change Vulnerability Index for Extreme Weather - Focused on Typhoon case - (기후변화에 따른 극한기상의 취약성 지수 정량화 연구 - 태풍을 중심으로 -)

  • VRI(Vulnerability-Resilience Index), which is defined as a function of 3 variables: climate exposure, sensitivity, and adaptive capacity, has been quantified for the case of Typhoon which is one of the extreme weathers that will become more serious as climate change proceeds. Because VRI is only indicating the relative importance of vulnerability between regions, the VRI quantification is prerequisite for the effective adaptation policy for climate in Korea. For this purpose, damage statistics such as amount of damage, occurrence frequency, and major damaged districts caused by Typhoon over the past 20 years, has been employed. According to the VRI definition, we first calculated VRI over every district in the case of both with and without weighting factors of climate exposure proxy variables. For the quantitative estimation of weighting factors, we calculated correlation coefficients (R) for each of the proxy variables against damage statistics of Typhoon, and then used R as weighting factors of proxy variables. The results without applying weighting factors indicates some biases between VRI and damage statistics in some regions, but most of biases has been improved by applying weighting factors. Finally, due to the relations between VRI and damage statistics, we are able to quantify VRI expressed as a unit of KRW, showing that VRI=1 is approximately corresponding to 500 hundred million KRW. This methodology of VRI quantification employed in this study, can be also practically applied to the number of future climate scenario studies over Korea.

DNA Damage by X-ray and Low Energy Electron Beam Irradiation (X선과 저에너지 전자선에 의한 DNA 손상)

  • We observed DNA damages as a function of mean absorbed dose to identify the indirect effect of high-energy radiation such as x-ray. Monolayer films of lyophilized pGEM-3Zf(-) plasmid DNA deposited on tantalum foils were exposed to Al $K{\alpha}$ X-ray (1.5 keV) for 0, 3, 7 and 10 min, respectively, in a condition of ultrahigh vacuum state. We compared DNA damages by X-ray irradiation with those by 3 eV electron irradiation. X-ray photons produced low-energy electrons (mainly below 20 eV) from the tantalum foils and DNA damage was induced chiefly by these electrons. For electron beam irradiation, DNA damage was directly caused by 3 eV electrons. Irradiated DNA was analyzed by agarose gel electrophoresis and quantified by ImagaQuant program. The quantities of remained supercoiled DNA after irradiation were linearly decreased as a function of mean absorbed dose. On the other hand, the yields of nicked circular (single strand break, SSB) and interduplex crosslinked form 1 DNA were linearly increased as a function of mean absorbed dose. From this study, it was confirmed that DNA damage was also induced by low energy electrons ($0{\sim}10\;eV$) even below threshold energies for the ionization of DNA.

  • As the types and usage of chemical increase, modern countries should protect their health and environment from the risk of hazardous chemical. Chemical accidents not only affect humans but also cause huge losses to the environment. Moreover, since its effects do not end in a short period of time, it is necessary to identify the extent of the damage and establish a prevention and response system in advance. In 2015, the Chemical Substances Management Act provided a system for assessing the impact on the people and the environment around the workplace. However, it is difficult to quantitatively evaluate the impact on environmental factors such as vegetation and aquatic, with the current hazard assessment methods. The purpose of this study is to analyze the quantitative risk of environmental receptors. This study improved the existing risk assessment formula by using the environmental vulnerability index and established the end point concentration criterion which can estimate the damage range to environmental media. To verify the results of the study, a virtual accident scenario was selected and a case study was conducted. As a result, the extent of impact on the environmental medium can be calculated, and the degree of environmental risk of the zone can be quantified through the risk analysis considering the environmental vulnerability. This study is expected to increase the reliability of the reliability of the existing risk anaylsis method beacause it is a risk analysis method that can be applied when the environmental factors are absolutely necessary and when the residents and environment are complex.

Evaluation of the impact of typhoon on daily maximum precipitation (태풍이 일 최대강수량에 미치는 영향 평가)

  • Typhoons are accompanied by strong wind and heavy rains. It causes casualties and property damage on the Korean peninsula every year. The effect of typhoon to daily precipitation should be quantified to prevent the damage of typhoon. Daily precipitation, maximum wind speed and, mean wind speed data was collected from 60 weather stations between 1976 and 2016. The parameters of the generalized extreme value distribution were estimated through the maximum likelihood estimation and the L-moment estimation. The impact of a typhoon can be obtained through a comparison of return levels between the whole data and typhoon excluded data. We conclude that the eastern and southern coastline are exposed to the risk of heavy rainfall which is caused by typhoon.

Study on the Effect of Toxic Gas Filter Effect of Wet Towel during Fire (화재 시 젖은 수건의 유독가스 필터 효과에 관한 연구)

  • The damage caused by fires is fatal as opposed to the damage caused by heat, and toxic gas generated by fires can be protected against by using a wet towel. In this study, we quantified the filtration effect of gas generated by an actual fire using a wet towel. In order to confirm the filtration effect of the wet towel on three harmful gases ($CO_2$, HCl, HCN), gases passed through a filter using FT-IR were analyzed in realtime. HCl and HCN, which are gases, were filtered by a wet towel, and the detection time of each gas was delayed. Therefore, it was confirmed that evacuation time can be secured by using a wet towel in the case of toxic gas, especially water-soluble gases in an actual fire.

Changes of DNA fragmentation by Irradiation Doses and Storage in Gamma-irradiated Meats and Poultry (감마선 조사 육류, 가금류에서 저장전과 후의 조사선량에 따른 DNA fragmentation의 변화)

  • The changes in DNA damage were investigated during storage after irradiation. Beef, pork and chicken were irradiated at 1.0, 3.0 and 5.0 kGy and stored for 6 months at $-20^{\circ}C$. The comet assay was applied to the sample muscles at the beginning of irradiation and at the end of storage. Muscles were isolated, sliced, and the suspended cells were embedded in an agarose layer. After lysis of the cells, they were electrophoresed for 2 min. and then stained. DNA fragmentation in tissues caused by irradiation was quantified as tail length and tail moment (tail length ${\times}$ % DNA in tail) by comet image analyzing system. Right after irradiation, the differences in tail length between unirradiated and irradiated muscles were significant(p<0.05) in beef, pork and chicken. With increasing the increasing doses, statistically significant longer extension of the DNA from the nucleus toward anode was observed. Similarly even 6 months after irradiation, all the irradiated muscles significantly showed longer tail length than the unirradiated controls. The results represented as tail moment showed similar tendency to those of tail length, but the latter parameter was more sensitive than the former. These results indicate that the comet assay could be one of the simple methods of detecting irradiated muscles. Moreover, this method suggest that using comet assay, we were able to detect DNA damage differences even after 6 months after irradiation.

Circ_UBE2D2 Attenuates the Progression of Septic Acute Kidney Injury in Rats by Targeting miR-370-3p/NR4A3 Axis

  • As circ_UBE2D2 has been confirmed to have targeted binding sites with multiple miRNAs involved in septic acute kidney injury (SAKI), efforts in this study are directed to unveiling the specific role and relevant mechanism of circ_UBE2D2 in SAKI. HK-2 cells were treated with lipopolysaccharide (LPS) to construct SAKI model in vitro. After sh-circ_UBE2D2 was transfected into cells, the transfection efficiency was detected by qRT-PCR, cell viability and apoptosis were determined by MTT assay and flow cytometry, and expressions of Bcl-2, Bax and Cleaved-caspase 3 were quantified by western blot. Target genes associated with circ_UBE2D2 were predicted using bioinformatics analysis. After the establishment of SAKI rat model, HE staining and TUNEL staining were exploited to observe the effect of circ_UBE2D2 on tissue damage and cell apoptosis. The expression of circ_UBE2D2 was overtly elevated in LPS-induced HK-2 cells. Sh-circ_UBE2D2 can offset the inhibition of cell viability and the promotion of cell apoptosis induced by LPS. Circ_UBE2D2 and miR-370-3p as well as miR-370-3p and NR4A3 have targeted binding sites. MiR-370-3p inhibitor reversed the promoting effect of circ_UB2D2 silencing on viability of LPS-treated cells, but shNR4A3 neutralized the above inhibitory effect of miR-370-3p inhibitor. MiR-370-3p inhibitor weakened the down-regulation of NR4A3, Bax and Cleaved caspase-3 and the up-regulation of Bcl-2 induced by circ_UB2D2 silencing, but these trends were reversed by shNR4A3. In addition, sh-circ_UBE2D2 could alleviate the damage of rat kidney tissue. Circ_UBE2D2 mitigates the progression of SAKI in rats by targeting miR-370-3p/NR4A3 axis.

Study on the Effect of Pile Tip Shape on Driven Pile Behavior Using 3D Printers (3D 프린터를 이용한 선단 모양 변화에 따른 타입말뚝 거동 연구)

  • In this study, the impact of pile tip geometry, including shape, size, and angle, on the drivability and stress concentration during pile driving was investigated using 3D printing technology and finite element numerical analysis. A series of field loading tests were conducted on a test pile with various pile tip conditions, including width, angle, and shape. The changes in settlement were quantified as a ratio to the settlement of a conventional pile tip case and large deformation finite element analysis was used to investigate the maximum stress on a pile tip and the location of possible damage during pile driving. The results showed that by modifying the shape, size, and angle of the pile tip, the drivability of the pile could be improved and the maximum stress concentration around the pile tip could be significantly reduced, thereby ensuring the structural integrity of the pile during pile driving.
