Change in the composition and enzyme activity of culturable lactic acid bacteria in Nuruk during fermentation at different temperatures (온도를 달리한 누룩 발효 기간별 배양 유산균 변화 및 분리 유산균들의 효소 활성)

  • The microbial composition in Nuruk, a Korean cereal fermentation starter, is a critical factor for the quality and organoleptic properties of traditional alcoholic beverages. This study was aimed at monitoring the compositional change and enzyme activity of culturable lactic acid bacteria (LAB) in two types of Nuruk fermented at different temperatures. All culturable LAB were isolated at various time points (0, 3, 6, 10, 20, and 30 days) and identified by 16S rRNA sequencing. In traditional Nuruk type A (TN-A), which was fermented at $36^{\circ}C$, the population of total culturable LAB during the fermentation period was between $10^4$ and $10^5$ log CFU/mL. On the other hand, the LAB population in traditional Nuruk type B (TN-B) fermented at $45^{\circ}C$ (primary fermentation for 10 days) and $35^{\circ}C$ (secondary fermentation for 20 days) was $10^2$ log CFU/mL; however, these bacteria could not be detected after 6 days. Major LAB strains were identified in both Nuruk types: (1) from the MRS-culture of TN-A, Pediococcus pentosaceus at 3-30 days; (2) from MRS-culture of TN-B, P. pentosaceus at 3 days and Enterococcus hirae at 6 days. The protease activities of the dominant LAB isolated from the TN-A and TN-B cultures were within the ranges of 0.64~1.03 mg/mL and 0.74~0.81 mg/mL (tyrosine content), respectively, whereas the ${\alpha}$-amylase activities were 0.75~0.98 mg/mL and 0.78~0.79 mg/mL (amylose content), respectively.

Increacing profit of dairy farm through improvement of raw milk quality : According to SCC and mastitis (유질개선을 통한 낙농가 소득증대 - SCC와 유방염을 중심으로-)

  • The study for a effect of monitoring on bovine mastitis was conduced for improvement of raw milk from Jan. to Dec. in 1996. Sampling the milk of 367 cows(1, 406 quarters) from 5 herds in Inchon and were carried out California mastitis test(CMT), somatic cell count(SCC), isolation of pathogens and antibiotic sensitivity tests. The results were summarized as follows, 1. The number of bovine mastitis was 177 cows(48.2%) and 371 quarters(26.4%) : clinical mastitis : 25 cows(6.8%), 32 quarters(2.3%) and subclinicsl mastitis : 152 cows(41.4% ), 339 quarters(24.1%). Incidence rate of mastitis by season were Summer 52.0%, Fall and Winter 48% and Spring 41%. Incidence rate of mastitis by quarters were Summer 30%, Fall 28%, Winter 25% and Spring 21%, respectively. 2. In the distribution of CMT degree by quarter, CMT positive(CMT$\pm$) of 1, 406 quarters milk were 50.1% (704 quarters). The ratio of CMT positivity by quarter were left front quarter 55.8%, right front quarter 48.9%, right hind quarter 48.6% and left hind quarter 47% The ratio of CMT positivity by season were Summer 54.1%, Fall 49.7%, Spring 48.5% and Winter 48% 3. The highest mean SCC by season among 5 herds was "A" herd. Mean SCC (cell/ml) of A herd were Summer 2, 032, 000cells/ml, Fall 1, 109, 000cells/ml, Winter 782, 000cells/ml and Spring 577, 000cells/ml. The lowest mean SCC by season among 5 herds was "E" herds. Mean SCC of E herd were Summer 1, 064, 000cells/ml, Spring 795, 000cells/m1, Fall 429, 000cells/ml and Winter 400, 000cells/ml. Mean SCC of the other herds by season were little difference. 4. The milk samples of "A" herd were collected from 10 cows. In 3 seasons, mean SCC of No. 2 and 3 cows were than 1, 000, 000cells/ml. In 1 season, mean SCC of No. 6, 7 and 8 cows were than 1, 000, 000cells/ml. The more than mean SCC 1, 000, 000cells/ml of cows by season were distributed Summer 4 cows, Winter 3 cows, Spring and Fall 1 cow respectively. The milk samples of "B" herd were collected from 14 cows. In 3 seasons, mean SCC of No. 1 cow was more than 1, 000, 000cells/ml. In 2 seasons, mean SCC of No. 5, 9 and 14 cows were more than 1, 000, 000cells/ml. In 1 season, No. 3, 6 and 7 cows were more than 1, 000, 000cells/ml. The more than mean SCC 1, 000, 000cells/ml of cows by season were distributed Fall and Winter 4 cows respectively, Summer 3 cows and Spring 1 cow. The milk samples of "C" herd were collected from 18 cows. In 2 seasons, mean SCC of No. 16 cow was more than 1, 000, 000cells/ml. In 1 season, mean SCC of No. 1, 2, 6, 7, 13, 15 and 18 cows were more than 1, 000, 000cells/ml respectively. The more than mean SCC 1, 000, 000cells/ml of cows by season were distributed Summer 5 cows, Fall 3 cows, Spring 2 cows and Winter 1 COW. The milk sampes of "D" herd were collected 24 cows. In 3 season, mean SCC of No. 14 cow was more than 1, 000, 000cells/ml. In 2 seasons, mean SCC of No. 14 and 18 cows were more than 1, 000, 000cells/ml. In 1 season, mean SCC of No. 1, 2, 3, 8, 12, 17, 19, 20 and 21 cows were more than 1, 000, 000cells/ml. The more than mean SCC 1, 000, 000cells/ml of cows were distributed Fall 15 cows, Spring and Winter 4 cows respectively and Summer 3 cows. The milk samples of "E" herd were collected from 27 cows. In 2 seasons, mean SCC of No. 6, 7 and 21 cows were more than 1, 000, 000cells/ml. In 1 season, mean SCC of No. 2, 4, 7, 11, 14, 16 and 23 cows were more than 1, 000, 000cells/ml. The more than mean SCC 1, 000, 000cells/ml of cows were distributed Spring and Fall 5 cows respectively, Summer and Winter 2 cows, respectively. 5. The rate of isolated pathogenic microorganisms from bovine mastitis were summarized as follows : Staphylococcus sp 168 strains(45.8%), Streptococcus sp 82 strains(22.3%), Gram(-) sp 45 strains(12.3%), Gram(+) sp 51 strains and the other sp 21 strains(5.7%). 6. The highest of antibiotic sensitivity test of each microorganism was summarized as follows : Staphyolcoccus sp - cephalosporin 76%, gentamicin 55%, Streptococcus sp - ampicillin 61%, cephalosporin 63%, Gram(-) sp - gentamicin 58%, Gram(+) sp - cephalosporin 63%, The other sp - cephalosporin 90%. Microorganisms showed the highest sensitivity(68%) to cephalospsorin. Microorganisms showed the highest sensitivity(68%) to cephalospsorin.

Variation of Inflow Density Currents with Different Flood Magnitude in Daecheong Reservoir (홍수 규모별 대청호에 유입하는 하천 밀도류의 특성 변화)

  • Stream inflows induced by flood runoffs have a higher density than the ambient reservoir water because of a lower water temperature and elevated suspended sediment(SS) concentration. As the propagation of density currents that formed by density difference between inflow and ambient water affects reservoir water quality and ecosystem, an understanding of reservoir density current is essential for an optimization of filed monitoring, analysis and forecast of SS and nutrient transport, and their proper management and control. This study was aimed to quantify the characteristics of inflow density current including plunge depth($d_p$) and distance($X_p$), separation depth($d_s$), interflow thickness($h_i$), arrival time to dam($t_a$), reduction ratio(${\beta}$) of SS contained stream inflow for different flood magnitude in Daecheong Reservoir with a validated two-dimensional(2D) numerical model. 10 different flood scenarios corresponding to inflow densimetric Froude number($Fr_i$) range from 0.920 to 9.205 were set up based on the hydrograph obtained from June 13 to July 3, 2004. A fully developed stratification condition was assumed as an initial water temperature profile. Higher $Fr_i$(inertia-to-buoyancy ratio) resulted in a greater $d_p,\;X_p,\;d_s,\;h_i$, and faster propagation of interflow, while the effect of reservoir geometry on these characteristics was significant. The Hebbert equation that estimates $d_p$ assuming steady-state flow condition with triangular cross section substantially over-estimated the $d_p$ because it does not consider the spatial variation of reservoir geometry and water surface changes during flood events. The ${\beta}$ values between inflow and dam sites were decreased as $Fr_i$ increased, but reversed after $Fr_i$>9.0 because of turbulent mixing effect. The results provides a practical and effective prediction measures for reservoir operators to first capture the behavior of turbidity inflow.

A Study on the Smog Reduction Strategies in China (중국의 스모그 저감정책에 대한 고찰)

  • Atmospheric environment in Korea is influenced by outside, especially China. The concentrations of air pollutants in China have showed decreasing trends since 2000. However, these concentration levels in China are still higher than other developed countries. The Chinese Government has tried several measures to control the air pollution. In this study, the details of the amendments and smog reduction strategies in China, especially for Beijing are reviewed and the strategies for Korean side to promote cooperation in Northeast Asia are suggested and discussed. The Chinese State Council amended the Environmental Protection Provisions and Clean Air Act and announced The Action Plan for Air Pollution Control (2013-2017), focusing on three key regions, Beijing-Tianjin-Hebei area (Jing-Jin-Ji), Yangtze River Delta (YRD) and Pearl River Delta (PRD). These policy actions and plan are mainly for the reducing coal usage and emissions from vehicles. It is suggested that, Korea should actively promote multi-national cooperation in the region to take an initiative role in environmental areas.

Analysis on the Characteristics of Nonpoint sources during the Precipitation in Residential Area (강우 시 주거지역에서의 비점오염원 유출특성 분석)

  • In this study, divided into small category groups for the residential area it was carried out monitoring for the runoff during precipitation. Based on the results analyzed according to the nonpoint sources Housing leakage characteristics. Analysis of the rainfall runoff and concentration of each type of exclusive detached house with apartments, in the majority of precipitation types runoff concentrations were higher in early. In the case of a difference of two points per runoff rate rainfall it was largely investigation. The average runoff is estimated loadings of BOD $101.1kg/km^2$, SS $232.2kg/km^2$, T-N $18.2kg/km^2$, T-P $2.0kg/km^2$ detached house case, if the apartment was estimated at point BOD $108.82kg/km^2$, SS $329.18kg/km^2$, T-N $57.67kg/km^2$, T-P $4.21kg/km^2$. The average EMCs is BOD BOD 6.6 mg/L, SS 12.8 mg/L, T-N 1.518 mg/L, T-P 0.099 mg/L detached house case, if the apartment was estimated at point BOD 6.3 mg/L, COD 11.2mg/L, SS 14.5 mg/L, T-N 3.1 mg/L, T-P 0.2 mg/L. The initial 30 percentage calculated based on the initial results, the total flow of 30% if the outflow of detached house showed a net percentage difference to T-P 1.04 > T-N 0.97 > BOD 0.90 > SS 0.80. The apartment area showed the percentage difference in the water quality in the order of BOD 1.49 > T-P 1.40 > SS 1.30 > T-N 0.96 per item.

Early Responses of Planted Quercus serrata Seedlings and Understory Vegetation to Artificial Gap Treatments in Black Locust Plantation (아까시나무림에서 인공 숲틈 처리에 대한 졸참나무 식재목 및 하층식생의 초기 반응)

  • Black locust (Robinia pseudoacacia) stand is representative lowland exotic plantation with low ecological quality and arrested succession in South Korea. To facilitate succession and restore natural vegetation, small canopy gaps (${\sim}57m^2$), which can modify minimally structural variables and reduce restoration related disturbances on stand, was established in the black locust stand, and oak (Quercus serrata) seedlings were introduced in the gap. Two types of varying levels were introduced for gap creation; cutting (C) and girdling (G) on canopies. Understory removal (CU and GU) treatment was applied as subtypes of structural modification. Growth (diameter, height and leaf area) of target species and responses (species composition, diversity and coverage) of understory community were monitored during study years (2007~2008). Canopy openness was different significantly among treatments but not for light availability. Based on the result of logistic regression, growth of height and leaf area of seedlings were significant variables on seedling survival. Height and leaf area of seedlings were increased during study years, although radial growth was reduced. During study years, there were no significant differences in species composition and diversity, and total coverage increased about 20%. Increase of resources by gap creation and understory removal likely affect growth of target species. Small gap creation was effective to reduce understory responses in composition and diverstiy. Synthesized, growth of target species and responses of understory community to small canopy gap creation exhibited, in short term, possibility of utilization in alternative forest restoration and management option. Long-term monitoring is necessary to certificate effect of artificial gap creation on forest restoration.

Characteristics of PM2.5 and PM10 Concentration in Pusan Area (부산지역 PM2.5와 PM10의 농도 특성)

  • It is necessary to improve the ambient air quality through the proper treatment and control of pollutants by designating air pollutants to regulatory ones. Especially, human took a concern for particulate matters which raised visibility reduction, public health effects and injury of property for urban areas. In order to reduce the effect of particulate matters, we need to establish proper control strategies based on the concentration characteristics of particulate matters. In this study. to evaluate the characteristics of $PM_{2.5}$ and $PM_{10}$. thirty-eight samples of $PM_{2.5}$ and $PM_{10}$ were collected at Nam-Gu sampling site where continuous air monitoring system has been operated, from May, 1999 to November, 1999, and their concentrations for the mass and anion components($Cl^-$, $NO_3{^-}$, $SO_4{^{2-}}$) were analyzed. The important conclusions obtained in this study were as followings. Total average mass concentrations of $PM_{2.5}$ and $PM_{10}$ were 35.016 and $50.293{\mu}g/m^3$ respectively. and $PM_{2.5}/PM_{10}$ ratio was calculated 0.692. Total average concentrations of anion components in $PM_{2.5}$ were $1.581(Cl^-)$, $3.690(NO_3{^-})$ and $12.825(SO_4{^{2-}}){\mu}g/m^3$ and those in $PM_{10}$ were $2.471(Cl^-)$, $5.819(NO_3{^-})$ and $14.414(SO_4{^{2-}}){\mu}g/m^3$ respectively. From the correlations analysis. the correlation coefficient between mass concentration of $PM_{2.5}$ and $PM_{10}$ was calculated as 0.945. The correlation coefficients between $PM_{2.5}$ and anion components were analyzed as $Cl^-$(0.025), $NO_3{^-}$(0.788) and $SO_4{^{2-}}$(0.500) respectively, and the correlation coefficients between $PM_{10}$ and anion components were analyzed as $Cl^-$(-0.019), $NO_3{^-}$(0.806) and $SO_4{^{2-}}$)(0.535) respectively.

Characteristics of Aquatic Environment and Algal Bloom in a Small-scaled Agricultural Reservoir (Jundae Reservoir) (소규모 농업용 전대저수지의 수환경 변화와 조류발생 특성)

  • This study was conducted to identify the relationship between environmental factors and algal bloom, and provide information for efficient management based on the results of monitoring the environmental parameters and algal diversity in the Jundai reservoir from March 2011 to October 2013. Little change in the weather conditions was observed during the study period except for a slight decrease in rainfall. Concentration of TN and TP in the reservoir exceeded water quality standards for agriculture and significant correlation between algal growth and environmental factors was observed. Phytoplankton in Jundai reservoir included 6 classes, 40 genus, 62 species, and the phytoplankton abundance was in the range of $1.3{\times}10^4{\sim}2.8{\times}10^6$ cells $mL^{-1}$. The annual average of phytoplankton abundance and Chl-a gradually decreased as TN and TP concentrations decreased. Overall Anabaena sp., Oscillatoria sp., and Microcystis sp. were the dominant species in Jundai reservoir. As the water temperature increased, the dominant species were Anabaena sp., Microcystis sp. and Oscillatoria sp., in that order. Anabaena sp. was dominant from spring to early summer with increase in water temperature and pollutant concentrations, and high correlation with environmental factors was observed. Microcystis sp. was dominant depending on changes in the nutrient levels. In the case of Oscillatoria sp., there was no significant correlation between phytoplankton biomess and Chl-a. However, efficient management of water environment and practical control of algal bloom in small scale reservoir polluted by livestock and farm irrigation should be achieved by identification of the relationship between algal growth and environmental factors.

Characteristics of Pollution Loading from Kyongan Stream Watershed by BASINS/SWAT. (BASINS/SWAT 모델을 이용한 경안천 유역의 오염부하 배출 특성)

  • A mathematical modeling program called Soil and Water Assessment Tool (SWAT) developed by USDA was applied to Kyongan stream watershed. It was run under BASINS (Better Assessment Science for Integrating point and Non-point Sources) program, and the model was calibrated and validated using KTMDL monitoring data of 2004${\sim}$2008. The model efficiency of flow ranged from very good to fair in comparison between simulated and observed data and it was good in the water quality parameters like flow range. The model reliability and performance were within the expectation considering complexity of the watershed and pollutant sources. The results of pollutant loads estimation as yearly (2004${\sim}$2008), pollutant loadings from 2006 were higher than rest of year caused by high precipitation and flow. Average non-point source (NPS) pollution rates were 30.4%, 45.3%, 28.1% for SS, TN and TP respectably. The NPS pollutant loading for SS, TN and TP during the monsoon rainy season (June to September) was about 61.8${\sim}$88.7% of total NPS pollutant loading, and flow volume was also in a similar range. SS concentration depended on precipitation and pollution loading patterns, but TN and TP concentration was not necessarily high during the rainy season, and showed a decreasing trend with increasing water flow. SWAT based on BASINS was applied to the Kyongan stream watershed successfully without difficulty, and it was found that the model could be used conveniently to assess watershed characteristics and to estimate pollutant loading including point and non-point sources in watershed scale.

Characteristics of Seepage Water and Groundwater in a Coastal LPG Storage Cavern of Jeonnam (전남 해안 LPG 저장공동 유출수와 주변 지하수의 수질특성)

  • Water curtain of an underground LPG storage cavern is a facility to prevent leakage of high pressure gases, for which groundwater should flow freely towards the cavern and groundwater level also must be stably maintained. In this study, in order to evaluate qualities of seepage water and surrounding groundwater of an underground LPG storage cavern in Yeosu, 4 rounds of samplings, field measurements and laboratory analyses (February, May, August, October of 2007) were conducted. According to field measurements, pH was weak acidic to neutral but it gradually increased with time. Electrical conductivity (EC) of groundwater near a salt stack showed very high values between 10.47 and 38.50 mS/cm. Dissolved oxygen (DO) showed a very wide range of 0.20~8.74 mg/L and a mean of oxidation-reduction potential (ORP) was 159 mV, which indicated an oxidized condition. Levels of $Fe^{2+}$ and $Mn^{2+}$ were mostly less than 3 mg/L. All of seepage waters showed a Na-Cl type while only groundwater near the salt stack showed a Na-Cl type with a high total dissolved solid. The other groundwaters exhibited typical $Ca-HCO_3$ types. Levels of aerobic bacteria were mostly very high (573-39,520 CFU/mL). Based on the analyses of these hydrochemistry and biological characteristics, it is concluded that there are no particular problems in groundwater and seepage water, which not causing a trouble in the cavern operation. However, both for control of bio-clogging and for sustainable operation of the water curtain system, a regular hydrochemical and microbiological monitoring is required for the seepage water and surrounding groundwater.
