A new cell-direct quantitative PCR based method to monitor viable genetically modified Escherichia coli

  • The development and commercialization of industrial genetically modified (GM) organisms is actively progressing worldwide, highlighting an increased need for improved safety management protocols. We sought to establish an environmental monitoring method, using real-time polymerase chain reaction (PCR) and propidium monoazide (PMA) treatment to develop a quantitative detection protocol for living GM microorganisms. We developed a duplex TaqMan quantitative PCR (qPCR) assay to simultaneously detect the selectable antibiotic gene, ampicillin (AmpR), and the single-copy Escherichia coli taxon-specific gene, D-1-deoxyxylulose 5-phosphate synthase (dxs), using a direct cell suspension culture. We identified viable engineered E. coli cells by performing qPCR on PMA-treated cells. The theoretical cell density (true copy numbers) calculated from mean quantification cycle (Cq) values of PMA-qPCR showed a bias of 7.71% from the colony-forming unit (CFU), which was within ±25% of the acceptance criteria of the European Network of GMO Laboratories (ENGL). PMA-qPCR to detect AmpR and dxs was highly sensitive and was able to detect target genes from a 10,000-fold (10-4) diluted cell suspension, with a limit of detection at 95% confidence (LOD95%) of 134 viable E. coli cells. Compared to DNA-based qPCR methods, the cell suspension direct PMA-qPCR analysis provides reliable results and is a quick and accurate method to monitor living GM E. coli cells that can potentially be released into the environment.

In accordance with this social trend, there are diversified lifestyles which influence consumer behavior, The purpose of this study was to develop an instrument that is able to measure more efficiently current housing lifestyle pattern. Eighty questioning items were analyzed and reclassified into same characteristics by using Q-sorting method.

  • In this paper, we report our successful implementation of Q tensor model in threedimensional finite element method (FEM) simulator. The 3D-FEM Q tensor-model-based simulation revealed that the spaly-to-bend transition occurs only at 4 V while the vector-model based FEM solver provides an erroneous transition voltage of 8 V.

  • This paper utilizes the method of Q-parameterization control to design a controller which solves the problem of imbalance in magnetic bearing systems. There are two methods to solve this problem using feedback controal. The first method is to compensate for the imbalance forces by generating opposing forces on the bearing surface (imbalance compensation). The second method is to make the rotor rotate around its axis of inertia (automatic balancing);in this case no imbalance forces will be generated. In this paper we deal with only imbalance compensation. The free parameter of the Q-parameterization controller is chosen such that these goals are achieved. After the introduction of a model of the magnetic bearing system, we explain the Q-parameterization controller design of the magnetic bearing system with emphasis on the rejection of sinusoidal disturbance for imbalance compensation design. The design objectives are formulated as a linear equations in the controller free paramete Q. Finally, simulation and experimental results are presented and showed the robustness and effectiveness of the proposed controllers.

Analysis of quantitative high throughput screening data using a robust method for nonlinear mixed effects models

  • Quantitative high throughput screening (qHTS) assays are used to assess toxicity for many chemicals in a short period by collectively analyzing them at several concentrations. Data are routinely analyzed using nonlinear regression models; however, we propose a new method to analyze qHTS data using a nonlinear mixed effects model. qHTS data are generated by repeating the same experiment several times for each chemical; therefor, they can be viewed as if they are repeated measures data and hence analyzed using a nonlinear mixed effects model which accounts for both intra- and inter-individual variabilities. Furthermore, we apply a one-step approach incorporating robust estimation methods to estimate fixed effect parameters and the variance-covariance structure since outliers or influential observations are not uncommon in qHTS data. The toxicity of chemicals from a qHTS assay is classified based on the significance of a parameter related to the efficacy of the chemicals using the proposed method. We evaluate the performance of the proposed method in terms of power and false discovery rate using simulation studies comparing with one existing method. The proposed method is illustrated using a dataset obtained from the National Toxicology Program.

  • The differential quadrature method (D.Q.M.) is applied to computation of eigenvalues of small-amplitude free vibration for horizontally curved beams including a warping contribution. Fundamental frequencies are calculated for a single-span, curved, wide-flange beam with both ends simply supported or clamped, or simply supported-clamped end conditions. The results are compared with existing exact solutions and numerical solutions by other methods for cases in which they are available. The differential quadrature method gives good accuracy even when only a limited number of grid points is used.

A Q-Methodological Study on the Community Nursing Practice of Nursing Students (간호 학생의 지역사회간호 실습 경험에 대한 유형 분석 -Q방법론적 접근-)

  • This study measures the subjectivity of nursing students' experience in community fields through community nursing practice. The purpose of this study is as follows: 1) to find out typologies based on opinions and attitudes toward community nursing practice. 2) to describe the characteristic of each type. 3) to provide alternative strategies for solving community nursing practice problems. A Q-Methodological method was used for that purpose. As a research method, Q-statements were collected through indepth interviews and review of the current literature. For this study 34 Q-statements were selected. 24 nursing students were subjects for the research. The 24 nursing students sorted 34 Q-statements using the principle of Forced Normal Distribution. The principle of Forced Normal Distribution, which has nine scales to measure individual opinions, was called, a Q- Factor Analysis by using a PC Quanl Program to supply the material. According to the results of this study, there were three categories of opinion concerning community nursing practice. The first type is the realistic problem-oriented approach: the second type is the self-responsibility or pursuit of life meaning approach: the third type is the group approach for problem solving. As a result, we need to develop and revise a more realistic way of community nursing practice for nursing students. Finally, the result of this study will provide to the educational program alternative strategies for community nursing practice for nursing students.

A Variable-Q Digital Graphic Equalizer with Opposite Filters (Opposite 필터를 적용한 가변 Q 디지털 그래픽 이퀄라이저)

  • This paper proposes a variable-Q digital graphic equalizer with the opposite filters. A method for designing the proposed equalizer is also presented. In the proposed variable-Q equalizer, we adjust the Q-factor of the equalizer filter depending on the gain, yielding an improved equalizer performance. Also, by increasing the Q-factor of the opposite filters gracefully as the gain becomes greater, the inter-band interference can be removed effectively. We shall show that the frequency response of the proposed equalizer can reproduce the user's gain setting faithfully.

  • The space vector control based voltage control method for a three-phase PWM rectifier requires a lot of effort to design an optimal switching pattern since a switching pattern for the switching section must be designed. In this study, a D-Q control based SPWM output voltage control algorithm was studied for the three-phase six-pulse CVS type rectifier. In the output voltage control algorithm, three-phase reference signals are obtained from the D-Q transformation based on the space vector representation method, instead of the switching pattern, SPWM method is used to generate rectifier switching control signals. Next, a three-phase six-pulse CVS PWM rectifier based on D-Q transformation and SPWM was modeled using EMTP-RV. Finally, the validity of the D-Q control-based SPWM voltage control algorithm was confirmed by comparing the output voltage waveform obtained through EMTP-RV simulation works with a reference value and confirming that the output voltage accurately follows the reference voltage.

Comparative Study on the Attenuation of P and S Waves in the Crust of the Southeastern Korea (한국 남동부 지각의 P파와 5파 감쇠구조 비교연구)

  • The Yangsan fault in the southeastern Korea has been receiving increasing attention in its seismic activity. In this fault region, by using the extended coda-normalization method for 707 seismograms of local earthquakes, were obtained 0.009f$^{-1.05}$ and 0.004f$^{-0.70}$ for fitting values of Q$_p^{-1}$ and Q$_s^{-1}$, respectively. These results indicate that Q$_p^{-1}$ and Q$_s^{-1}$ in the southeastern Korea is the lowest level in the world although the exponent values agree well with those in the other areas. The low Q-1 is not related to the movement of the Yangsan fault but to the tectonically inactive status like a shield area.
