표고버섯 분말을 첨가한 천연 조미료 추출물의 주요성분 및 항산화 효과 (Component Analysis and Antioxidant Activity of Natural Seasoning Using Shiitake(Lentinus edodes) Powder)

  • This study was performed to determine the component analysis and measurements of antioxidant activities from natural seasoning containing Lentinus edodes powder (NSLP) in order to detect the biological activities and develop novel functional resources. It was extracted with 70% ethanol and then further fractionated to hexane, chloroform, ethyl acetate, butanol and water. Methods used in this experiment were measured to examine total polyphenol contents and total flavonoid contents, reducing power and Superoxide dismutase-like activity. Composition analysis were highly composed of carbohydrate as 44.1%. The minerals of different organics were highly composed of sodium as 5,073 mg/100g. There were seventeen total amino acids in NSLP. The glutamic acid content was high up to 16.9 mg/g and aspartic acid, lysine, leucine, alanine were followed. Predominant fatty acid was linoleic acid (62.7%) in NSLP. Contents of total polyphenols in butanol fraction from NSLP were 16.38 mg/100mL. While overall butanol fraction have higher reducing power than ethanol extract, after the addition of 400 ${\mu}g/mL$ butanol fraction, auto oxidation of pyrogallol decreased to 83.62% as a result of superoxide dismutase-like activity. A positive correlation was observed between total polyphenol, flavonoid and antioxidant activities.

한국산 재래종 담배잎에서 정제한 Polyphenol Oxidase의 성상에 관한 연구 (Some Properties of Polyphenol Oxidase Purified from Korean Native Tobacco Variety Leaves)

  • Purification of polyphenol oxidase(PPO) from Korean native tobacco variety leaves was carried out through the procedure of acetone preciptation, ammonium sulfate fractionation, and Sephadex G-150 gel filtration, resulting in a 84-fold increase in specific activity. The enzyme was stable in a range of pH 7.5 to 8.0 with an optimum of pH 7.5. The optimum temperature for the enzymic reaction was about $60^{\circ}$. It was thermostable with a half-life equal to 20 min at $70^{\circ}$. Km values for (+)-catechin and pyrogallol were $1.6{\times}10^{-3}$ and $0.5{\times}10^{-3}M$, respectively. It possesses high catecholase activity but little or no cresolase activity. Lineweaver-Burk analysis of inhibition data revealed that the inhibition of (+)-catechin oxidation by potassium cyanide, 4-nitrocatechol, cystein and 2-mercaptoethanol was competitive with Ki values of $1.1{\times}10^{-6}$, $1.8{\times}10^{-6}$, $8.9{\times}10^{-6}$ and $1.3{\times}10^{-5}$, respectively.

Antioxidant activity and nitrite scavenging ability of each fractions from Phyllostachys bambusoides ethanolic extract

  • This study was conducted to investigate the efficacy of antioxidative activity and nitrite scavenging ability of each fractions from Phyllostachys bambusoides S. et Z. (P. bambusoides) trunk ethanolic extract using reverse-phase flash chromatography. Among the each fractions, fraction 3 $(H_2O\;:\;MeOH\;=\;1:1)$ showed high DPPH free radical scavenging activity (81.33%) at $80\;{\mu}g/mL$ concentrations and strongly inhibited autooxidation of pyrogallol by superoxide dismutase-like activity (45.8 %) at 0.46 mg/mL concentrations compared with different fractions. The fraction 3 was also increased to 76.62% cell viability against hydrogen peroxide-mediated cytotoxicity. Nitrite scavenging ability was the most remarkable under pH 1.2 condition among various pH regions examined and effectively exhibited to 65.6% by treatment of the fraction 3 with a concentration of 0.2 mg/ml. In general, nitrite scavenging ability decreased with higher pH condition. These results suggest that fraction 3 from P. bambusoides ethanolic extract can be used for bioacitve and functional materials.

오가피 메탄올 추출물의 항산화 효과 및 아질산염 소거작용 (Antioxidative Activity and Nitrite Scavenging Ability of Ethyl Acetate Extract from Acanthopanax sessiliflorus)

  • Efficacy of antioxidative activity and nitrite scavenging ability of methanol extract from Acanthopanax sessiliflorus was investigated. Electron-donating ability of extract at $RC_{50}$ was 29.93 ${\mu}g/mL$. After addition of 0.96 mg/mL extract, autooxidation of pyrogallol decreased to 22.85% by superoxide dismutase-like activity. In antioxidative activity of extract against linoleic acid during incubation times of 24, 48, 96 hours at $40^{\circ}C$, lipid peroxidation values significantly decreased by 48.89%, 45.0%, 46.34% with addition of 0.2 mg/mL, respectively. Total phenolic content was determined as gallic acid equivalents (GAE) and values revealed $410.25{\pm}4.74$ GAE ${\mu}g/mg$ of extract. Nitrite scavenging ability showed the most remarkable effect at pH 1.2, exhibited to 88.3% by addition of 0.2 mg/mL. These results suggest that methanol extract from A. sessiliflorus can be used as bioactive and functional material.

아주까리 메탄을 추출물의 항산화 효과 및 아질산염 소거작용 (Antioxidative Activity and Nitrite Scavenging Ability of Methanol Extract from Ricinus communis)

  • Efficacy of antioxidative activity and nitrite scavenging ability of methanol extract from Ricinus communis. was investigated. Electron-donating ability of extract at RC$_{50}$ was 114.02 ${\mu}g$/mL. After addition of 0.46 mg/mL extract, autoxidation of pyrogallol decreased to 32.99% by superoxide dismutase-like activity. In antioxidative activity of extract against linoleic acid during incubation times of 24, 48, 96 hours at 40$^{\circ}C$, lipid peroxidation values significantly decreased by 85.50%, 87.77%, 90.95% with addition of 0.2 mg/mL, respectively. Total phenolic content was determined as gallic acid equivalents (GAE) and values revealed 83.98 ${\pm}$ 5.66 GAE ${\mu}g$/mg of extract. Nitrite scavenging ability showed the most remarkable effect at pH 1.2, decreasing to 47.24% by addition of 0.2 mg/mL. These results suggest that methanol extract from Ricinus communis. can be used as bioactive and functional material.

고려인삼의 토끼 음경 해면체 평활근에 대한 작용 (Effect of Korean Ginseng on the Isolated Rabbit Corpus Cavernosal Smooth Muscle)

  • Rabbit corpus cavernosal smooth muscle strips submaximally precontracted with phenylephrine (5$\times$$10^6$ M) and treated with increasing concentrations of Korean ginseng (1, 5, 10, 20, 30, 40 mg/ml) showed tension decrease concentration-dependently (1 mg/ml: 1.7%, 5 mg/ml: 10.2%, 10 mg/ml: 22.7%, 20 mg/ml: 44.0%, 30 mg/ml: 65.2%, 40 mg/ml: 95.6%). Relaxations to Korean ginseng were inhibited significantly by endothelial disruption, by pretreatment with methylene blue, pyrogallol, L-NNA and atropine. Pretreatment of the muscle strips with ginseng caused concentration-related inhibition of a phenylephrine induced contraction, and in calcium-free high potassium depolarizing solution, decreased basal tension as well as inhibited contraction induced by Caclr. Korean ginseng also produced the reduction of responses to depolarizing medium(50, 40, 60 mM KCI). With these results we can confirm the relaxation effect of ginseng at a dose dependent on the cavernosal smooth muscle and suggest that its action is mediated by multiple action mechanisms that include increasing the release of nitric oxide from the corporal sinusoids. Increasing intracellular calcium sequestration, and a hyperpolarizating action.

Bidirectional Interactions between Green Tea (GT) Polyphenols and Human Gut Bacteria

  • Green tea (GT) polyphenols undergo extensive metabolism within gastrointestinal tract (GIT), where their derivatives compounds potentially modulate the gut microbiome. This biotransformation process involves a cascade of exclusive gut microbial enzymes which chemically modify the GT polyphenols influencing both their bioactivity and bioavailability in host. Herein, we examined the in vitro interactions between 37 different human gut microbiota and the GT polyphenols. UHPLC-LTQ-Orbitrap-MS/MS analysis of the culture broth extracts unravel that genera Adlercreutzia, Eggerthella and Lactiplantibacillus plantarum KACC11451 promoted C-ring opening reaction in GT catechins. In addition, L. plantarum also hydrolyzed catechin galloyl esters to produce gallic acid and pyrogallol, and also converted flavonoid glycosides to their aglycone derivatives. Biotransformation of GT polyphenols into derivative compounds enhanced their antioxidant bioactivities in culture broth extracts. Considering the effects of GT polyphenols on specific growth rates of gut bacteria, we noted that GT polyphenols and their derivate compounds inhibited most species in phylum Actinobacteria, Bacteroides, and Firmicutes except genus Lactobacillus. The present study delineates the likely mechanisms involved in the metabolism and bioavailability of GT polyphenols upon exposure to gut microbiota. Further, widening this workflow to understand the metabolism of various other dietary polyphenols can unravel their biotransformation mechanisms and associated functions in human GIT.

갈근(葛根) 추출물이 항산화에 미치는 영향 (Effects of Puerariae Radix extract on the activity of antioxidant)

  • Objective The objective of this study was to investigate the antioxidative effects of Puerariae Radix extract. Method Total antioxidant capacity (TAC), Total antioxidant response (TAR), Total phenolic content, Reactive oxygen species (ROS), 1,1-Diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging activities, lipid peroxidation were examined. Result Total antioxidant status was examined by total antioxidant capacity(TAC) and total antioxidant response(TAR) against potent free radical reactions. TAC and TAR of Puerariae Radix extract at the concentration of 5 mg/ml were 2.02 and 1.50 mM Trolox equivalents, respectively. Total phenolic content of Puerariae Radix extract at the concentration of 5 mg/ml was 2.29 mM gallic acid equivalent. Concentration of Puerariae Radix extract at which DPPH radical scavenging activity was inhibited by 50% was 5.91 mg/ml as compared to 100% by pyrogallol solution as a reference. The inhibitory effect of the extract on lipid peroxidation was examined using rat liver mitochondria induced by FeSO4/ascorbic acid. Puerariae Radix extract at the concentration of 1 mg/ml slightly but significantly decreased TBARS concentration. The extract further prevented lipid peroxidation in a dose-dependent manner. The effect of Puerariae Radix extract on reactive oxygen species (ROS) generation was examined using cell-free system induced by hydrogen peroxide/FeSO4. Addition of 1 mg/ml of Puerariae Radix extract significantly reduced dichloroflurescein (DCF) fluorescence. The extract caused concentration-dependent attenuation of the increase in DCF fluorescence, indicating that the extract significantly prevented ROS generation in vitro. Thus antioxidant effects of Puerariae Radix extract seem to be due to, at least in part, the prevention from free radicals-induced oxidation, followed by inhibition of lipid peroxidation. Conclusion As a result, Puerariae Radix seems to have antioxitative effect and antioxidant compount.

Lactobacillus plantarum 발효에 의한 갈근탕의 생물 전환 성분 연구 (Bioconversion Constituents of Galgeun-tang Fermented by Lactobacillus plantarum)

  • Galgeun-tang (GGT) is a traditional medicinal formula that is widely prescribed to treat cold, asthma, and hives in Korea. Fermented herbal medicines can be made more effective than normal herbal medicines by increasing the absorption and bioavailability of the active compounds. In this study, we fermented Galgeun-tang to produce bioconversion constituents using Lactobacillus plantarum (GGT144), and found that four peaks were decreased, three peaks were increased and two new peaks appeared in the HPLC-DAD chromatogram. After HPLC-DAD-guided fractionation of the newly-appearing compounds (1 and 5) and the increased (6, 7, and 9) compounds, the structure of the compounds was determined using NMR and MS. Using this approach the compounds were identified to be pyrogallol (1), daidzein (5), liquiritigenin (6), cinnamyl alcohol (7), and formononetin (9), respectively. In addition, the decreased compounds were identified to be daidzin (2), liquiritin (3), ononin (4), and cinnam aldehyde (8) using HPLC-DAD analysis with standard compounds. The high performance liquid chromatography method was used to quantify the nine constituents in GGT and GGT144. All calibration curves of the standard compounds displayed excellent linearity with a $R^2$ > 0.9968.

박태기나무의 잎으로부터 피부멜라닌 색소생성 억제성분의 분리 (The Isolation of the Inhibitory Constitutents on Melanin Polymer Formation from the Leaves of Cercis chinensis)

  • Tyrosinase plays an important role in the process of melanin polymer biosynthesis. Therefore, the enzyme inhibitors have been of great concern as cosmetics to have skin-whitening effects on the local hyperpigmentation. During the search for new inhibitory compounds on melanin polymer biosynthesis from natural sources, MeOH extracts of 589 higher plants were tested for the inhibitory effect on tyrosinase activity by the muschroom tyrosinase assay in vitro. Among plants tested, the leaves of Cercis chinensis exhibited potent inhibitory effect on mushroom tyrosinase activity. Subsequently seven active compounds were isolated from the ethyl acetate soluble part of acetone extract of the leaves of C. chinensis by the activity guided fractionation monitoring the inhibitory effect on tyrosinase activity. Their chemical structures were identified as $kaempferol-3-0-{\alpha}-L-rhamnoside$, quercitrin, $myricetin-3-0-{\alpha}-L-rhamnoside$, myricetin-3-0-(2'-O-galloyl)- ${\alpha}$ -L-rhamopyranoside (desmanthin), (-)-epicatechin-3-0-gallate, (-)-epigallocatechin-3-0-gallate, and methyl gallate on the basis of the speculation of spectral data and chemical reaction. Among the flavonol rhamnosides, myricetin-3-0-(2'-O-galloyl)- -L-rhamnoside(desmanthin) showed most potent inhibitory effect on tyrosinase activity and the structure of B-ring in flavonol moiety was related to the activity. (-)-Epigallocatechin-3-O-gallate having pyrogallol group in flavan-3-ol moiety exhibited more potent inhibitory effect than (-)-epicatechin-3-0-gallate having catechol group in flavan-3-ol moiety on mushroom tyrosinase activity.
