Purification and Cellular Localization of Extracellular Nuclease of Serratia marcescens Expressed in Escherichia coli (대장균에 발현된 Serratia marcescens의 Nuclease의 정제와 세포내 분포)

  • Nuclease was secreted to the environmental media from the Escherichia coli JM107 tranformant harboring the extracellular nuclease gene of Serratia marcescens in the plasmid of pNUC4. Under the growth conditions, the amount of secreted enzyme was increased in parallel with bacterial growth conditions, the amount of secreted enzyme was increased in parallel with bacterial growth. The enzyme was purified using chromatofraphic procedures of Matrex green gel and heparin agarose affinity gel, resulted in 50-fold purification with 15% recovery of the enzyme. The apparent molecular weight of the enzyme was estimated to be 29Kda by sodium dodecylsulfate denaturing gel electrophoresis. Using the purified enzyme, polyclonal antibody was obtained from the rabbit. The specificity of the antibody was confirmed by immunoblotting and immunoprecipitaion. For the investigation of cellular distribution of the enzyme, cells were fractionated into three fractions; cytoplasm, periplasm and extracellular fluid. While more than 80% of the enzymatic activity was detected in the extracellular fluid and periplasm, a little was found in the cytoplasm, indicating that the enzyme was likely to be immediately exported to the membrane for excretion after biosynthesis. These results were confirmed again by immunocytochemistry technique using the antibody.

Characteristics of Tobacco Mosaic Virus Isolated from Wasabi (Eutrema wasabi) in Korea

  • Wasabies showing mosaic symptoms were collected and extracted for virus purification. Tobacco mosaic virus (TMV) was identified as causal agent by electron microscopy and nucleic acid and coat protein analyses. TMV strains were determined by enzyme-linked immunosorbent assay (ELISA). TMV was identified as W and C strain in wasabi. The results of host reaction indicated that this virus induced local lesions on Nicotiana tabacum cv. Bright Yellow and N. glutinosa, leaf spots on Chenopodium amaranticolor and mosaic symptoms on wasabi. Rot shape virus particles were observed and was about 300 nm in length. About 6.5 kb single RNA molecule was observed from extracted viral RNA sample and 26 KDa coat protein was detected in denatured acrylamide gel. Infection ratio of TMV was 8% for the first cultivation year, but was 22% for the second year when TMV-W antiserum was used. The results of this experiment showed that infection ratios of both TMV-W and TMV-C strains were higher compared to that of TMV-P strain.

A Survey of THMs Formation in J Water Purification Plant and Its Reduction by PAC Treatment during Summer (J 정수장의 하절기 THMs 생성현황과 분말활성탄 처리에 의한 저감효과)

  • This study was conducted to survey trihalomethane(THMs) levels in finished water of J water treatment plant and examine its reduction by powder activated carbon(PAC) treatment. Samples were collected weekly based and head-space technique was employed to determine THMs levels by G.C-ECD. THMs levels in finished waters were highest in August and showed close relationship with water temperature. All the samples satisfied the drinking water limit(100 ${\mu}$g/l) for THMs. The individual formation rates of THMs were 64.8% for CHCl$_3$, 28.4% for CHCl$_2$BR, 6.5% for CHClBr$_2$ and 0.3% for CHBr$_3$, respectively and showed little monthly difference. The reduction efficiency of THMs formation by PAC treatment was 67% during July and August. Bromine substituents were more efficiently reduced than CHCl$_3$ by PAC.

A multi-level approach for the optimization of an ultrafiltration plant processing surface water

  • This paper will integrate models at different levels (from filtration, backwashing to chemical cleaning and membrane lifetime) that can be used to minimize overall operating costs of a dead-end ultra filtration process that is used for the purification of surface water. Integration of the models leads to a multi-level optimization problem (at different levels different objectives should be reached). This problem is solved as a MINLP. Systematic modelling and optimization of membrane systems is not extensively discussed in the scientific literature. In this paper the first steps are taken in the formulation of proper models and the use of systems engineering tools to come to real optimal operating conditions. The optimized variables are used to calculate fouling profiles which can subsequently be used as inputs for a control system that actually enforces the profiles to a real pilot plant.

  • A methodology to evaluate the performance analysis for small hydropower at existing water treatment facilities has been studied. It consists of two main parts; flow duration function which can describe existing water treatment facilities and performance analysis to estimate the output characteristics of small hydro power plants. The output performance characteristics for Mi-ho reservoir, Sum-kang low dam, Sun-cheon sewage treatment plant and Nam-dong purification plant were analyzed, using developed model. According to the simulation results, the predicted data show that the data were in good agreement with measured results. Also, it was found that the developed model in this study can be used to analyze the output characteristics for small hydro power at existing water treatment facilities.

Radical Scavenging Activity of Gallic Acid from Woodfordia Fruticosa Flowers

  • Woodfordia fruticosa Kurz (Lythraceae) is used in the treatment of various ailments in traditional medicines. DPPH activity guided fractionation and purification process was used to identify the free radical-scavenging components from the flowers of this plant. The methanolic extract of the plant was first fractionated into four extracts; namely, n-hexane, chloroform, ethyl acetate and water fractions. Among them, the ethyl acetate fraction was found to be the most effective and was further subjected to activity guided-fractionation and isolation procedures. After successive column chromatography on silica gel and Sephadex LH-20, gallic acid, which is responsible for the radical scavenging activity, was isolated and its structure was elucidated by spectral methods ($^1H$ NMR, $^{13}C$ NMR) and by comparison with literature.

해녀콩 유식물 자엽내 BApNAase의 성질

  • 해녀콩(Canavalia lineata) 유식물 자엽에서 N-$\alpha$-benzoyl-DL-arginine p-nitroanilide hydrolase(BApNAase)를 부분정제하여 그 성질을 밝혔다. 부분정제한 BApNAase는 purification fold가 77.5이었고 회수율은 7%이었으며 비활성도는 7.75 unit/mg이었다. BApNAase의 분자량은 200 kD이고 젤라틴분해효소 P3인 것으로 밝혀졌으며 최적 pH는 9.5이었다. BApNAase의 Vmax와 Km은 각각 15.5 unit/mg와 1.6 mM로 최대반응속도가 동물의 트립신보다 7배 가량 낮은 반면에 N-$\alpha$-benzoyl-DL-arginine p-nitroanilide(BApNA)에 대한 기질 친화성은 4배 가량 높았다. 또한, BApNAse는 1 mM의 phenylmethanesulfonyl fluoride(PMSF)에 의해 90%나 크게 억제된 반면 aprotinin에 의해서는 크게 억제되지 않아 트립신과는 다른 serine proteinase로 판단되었으며, 효소활성은 Ca2+과 Mg2+에 의해 다소 증가하나 Mn2+, Hg2+, Zn2+에 의해 크게 억제되었다.

Studies on Some Properties of Cellulase Isolated from Pirieularia oryzae (Piricularia oryzae로부터 추출한 cellulase의 몇가지 성질에 대한 연구)

  • Studies on some properties of cellulase isolated from Piricularia oryzae. Crude cellulases were prepared from dried rice plant powder (Tong-il, Pal-dal) culture of P. oryzae(N-2, C-8, T-2). The best yield of enzyme was obtained from the medium using Tong-il rice plant powder for P. oryzae cav. N-2 and 2%-sucrose concentration in preculture media. Two units of the enzyme were incubated at $60^{\circ}C$ for 1 hour with 1.0ml, 0.6% Na-CMC. The optimum temperature for the enzyme activity was at $60^{\circ}C$ and the optimum pH was at pH4.0. When Na-CMC was used as substrate the $K_m$ values of crude enzyme were calculated to be $1.05{\times}10^{-4}\;mM\;and\;V_{max}$ was 2.8 mmole/hour. A 10-fold partial purification was achieved by $(NH_4)_2SO_4$ precipitation followed by column chromatography on DEAE Sephadex A-25.

  • With architectural technology, a building has been a far dense and close. So the thermal environment of the building has become pleasant, but the quality of indoor air has been degraded. Using synthetic products for construction materials and furniture indoors escalates the concentration of volatile organic compounds(VOCs) at indoor air, threatening the health of the residents. To reduce the concentration of volatile organic compounds at indoor air, many methods are designed, and of late, concern has been increased about the effect of air purification using air purifying plants. Field measurements were performed using Aglaonema brevispathum, Pachira aquatica and Ficus benjamiana, which were verified as air-purifying plants by NASA. The effect of reducing the concentration of air contaminants by plant studied in a full scale mock-up model. The variations of concentration of Benzene, Toluene, Ethylbenzene and Formaldehyde were monitored. In most cases, the effect was excellent in Toluene and formaldehyde in summer.
