Use of Dietary Salt to Rainbow Trout (Oncorhynchus mykiss) for Increasing Seawater Adaptability (무지개송어(Onchorhynchus mykiss)의 해수 적응 능력 개선을 위한 식염사료의 적용)

  • One 30-day feeding trial was conducted to examine the effects of dietary salt on seawater adaptability of rainbow trout (Onchorhynchus mykiss) fed three experimental diets containing 0% (control), 4% and 8% salt. The experimetal period included 30 days of feeding trial in freshwater, 3 days of the step by step seawater acclimation with-out feeding diets, and 21 more days of seawater adaptation period (not with all experimental fish) with feeding the basal diet. Growth rates from triplicate groups were determined fur 30 days of feeding trial. Blood samples were taken at the begining and at the end of feeding trial, and 3 times (on 1st, 4th and 8th day) of the seawater adaptation period. Daily survival rates of duplicate groups from three experimetal treatments were recorded for 21 days of the seawater adaptation period. Total average initial and final fish weight were $149.5{\pm}7.6\;and\;187.1{\pm}7.6g$. Feed efficiency of fish fed diets containing 4% and 8% salt were significantly better than those of fish fed the control diet. Average cumulative survival rates were 72, 80 and 88% from the control, 4% and 8% salt diets, respectively. Pulse rate per minutes decreased with dietary salt level. Serum $Na^+\;and\;Cl^-$ concentrations of fish fed 4% and 8% salt diets were significantly higher than those of fish fed the control diet (P<0.05), however, the concentrations were stabilized after 8 days of seawater adaptation. Serum cortisol, glucose, cholesterol and tryglyceride concentrations, and the osmorality of fish decreased with dietary salt level, these values were significantly lower than those of fish fed the control diet. These results indicated that the dietary supplementation of salt could have advantages for seawater adaptability of rainbow trout.

The growth effect of agglomerated and nonagglomerated particles in Condensation Nuclei Counter (CNC) (응집입자와 비응집 입자의 CNC 내에서 응축성장에 관한 실험적 연구)

  • Agglomerated and nonagglomerated SiO2 particles are synthesized in furnace by the electrohydrodynamic spraying method and the vapor feeding method for the test particle generator this study. These polydispersed particles are classified with DMA to extract equal mobility particles. Then these particles are introduced into CNC (Condensation Nuclei Counter) to see the pulse height using Multi-channel Analyzer. The response characteristics of these two kinds of particles in CNCs (TSI CNC 3022 and 3025A) have been studied as a function of particle size using mono disperse particles classified by DMA. The results show that the higher drag resistance particles, so called agglomerated particles have generated a lower CNC pulse height than the spherical particles for these two different CNCs, which means the nonagglomerated particles may start to grow larger than the agglomerated particles.

  • Traction Motor is being transferred to the squirrel cage induction motor from D.C motor, by the improved power semiconductors like GTO, IGBT, etc. In the Induction Motor, inverter system must be used for delivering variable voltage variable frequency. But, by pulsating in the system the harmonics would be produced, and that cause the torque ripple and enfeeble the dynamic characteristics of the motor. So, to use the inverter system, we should take the torque ripple into consideration. To minimize the torque ripple in the VSI fed Traction Motor, the optimal pulsating was presented in this paper. By using the SPWM(Sinusoidal Pulse Width Modulation) method, feeding the appropriate pulse, we can minimize the torque ripple and improve the transient response.

  • In case that electronic ballast employing a valley-fill passive power factor correction (PFC) circuit is used for feeding fluorescent lamps, a new method to reduce crest factor of the lamp current is studied in this paper. In order to reduce crest factor to lower value, a pulse frequency modulation technique based on the waveform of the dc-link voltage which is predetermined by the passive PFC circuit, is taken into the switching control action of the electronic ballast. An equation-based analysis between the crest factor of lamp current and the effect of varying the inverter switching frequency is comprehensively performed.

  • In this paper, a new control scheme of Class-E inverter for reducing the crest factor of electronic ballasts for fluorescent lamps using Pulse-Frequency-Modulation (PFM) is Introduced The lifetime of the lamps is guaranteed by decreasing the lamp crest factor and also voltage stress of the switch is significantly decreased by a new scheme although conventional Class-E Inverter is used In this paper. The proposed PFM control scheme didn't use any auxiliary circuit. The proposed control strategy is executed by feeding back the Input voltage, and the zero-voltage-switching (ZVS) is ensured by maintaining constant turn-off time of the switch Therefore, the control principles of proposed method are explained in detail and its validity is verified through several simulations and experimental results.

  • A pulse driven atmospheric plasma jet controlled by external ballast capacitor is developed. Unlike the most commonly use DBD sources, the proposed device utilizes bare metal electrode. The discharge energy per pulse can precisely be determined by changing voltage and capacitance of the ballast capacitor. It is shown that the device can provide wide range of plasma, from stable glow mode to near arc state. Current-voltage waveforms, optical emission spectra and discharge images are investigated as a function of an injection energy. The OES shows that He and oxygen lines are increased as a function of the external ballast capacitor. Ozone and rotational temperature have similar tendency with a power consumption. The feeding gas is He and the applied DC voltage is from 400V to 800V when the gap distance is $500{\mu}m$.

  • The color of textile-wastewater hindered spectrometric measurements of $H_2O_2$ and enzyme activity during enzymatic decoloring. By using ABTS, we developed a new method for measuring peroxidase activity and $H_2O_2$ concentration. The ratio of enzyme and $H_2O_2$ was optimized as 1:150 by investigating the effects of $H_2O_2$ on enzymatic decoloring. Pulse feeding of $H_2O_2$, upon depletion, significantly increased the decoloring of Congo Red.

  • Induction motors are widely used as a source of driving force in electric vehicles and pulse width modulation (PWM) inverters are applied to their operation. The issue is that insulation of inverter-fed induction motors (IFMs) are more stressed than in line-powered motors by transient voltages. This paper dealt with experimental results on transient voltage produced by the PWM operation of an induction motor. The peak and the dv/dt of transient voltage depending on the length of power feeding cable and operating frequency were investigated. In the experiment, transient voltages up to 3.3PU of the rated-inverter voltage were recorded for the cable length of 50m. As the cable length is increased, the peak voltage appeared at the motor terminals increases. This phenomenon can be explained by the reflection and the transmission of travelling wave. Consequently, special care for the cable length between the motor and the inverter should be taken in the use of IFM to ensure the full life of insulation system.

Improved Production of Medium-Chain-Length Polyhydroxyalkanoates in Glucose-Based Fed-Batch Cultivations of Metabolically Engineered Pseudomonas putida Strains

  • One of the major challenges in metabolic engineering for enhanced synthesis of value-added chemicals is to design and develop new strains that can be translated into well-controlled fermentation processes using bioreactors. The aim of this study was to assess the influence of various fed-batch strategies in the performance of metabolically engineered Pseudomonas putida strains, ${\Delta}gcd$ and ${\Delta}gcd-pgl$, for improving production of medium-chain-length polyhydroxyalkanoates (mcl-PHAs) using glucose as the only carbon source. First we developed a fed-batch process that comprised an initial phase of biomass accumulation based on an exponential feeding carbon-limited strategy. For the mcl-PHA accumulation stage, three induction techniques were tested under nitrogen limitation. The substrate-pulse feeding was more efficient than the constant-feeding approach to promote the accumulation of the desirable product. Nonetheless, the most efficient approach for maximum PHA synthesis was the application of a dissolved-oxygen-stat feeding strategy (DO-stat), where P. putida ${\Delta}gcd$ mutant strain showed a final PHA content and specific PHA productivity of 67% and $0.83g{\cdot}l^{-1}{\cdot}h^{-1}$, respectively. To our knowledge, this mcl-PHA titer is the highest value that has been ever reported using glucose as the sole carbon and energy source. Our results also highlighted the effect of different fed-batch strategies upon the extent of realization of the intended metabolic modification of the mutant strains.
