We are generally interested in the analysis, detection and prediction of structural motifs in proteins, in order to infer compatibility of amino acid sequence to structure in proteins of known three-dimensional structure available in the Protein Data Bank. In this context, we are analyzing some of the well-characterized structural motifs in proteins. We have analyzed simple structural motifs, such as, ${\beta}$-turns and ${\gamma}$-turns by evaluating the statistically significant type-dependent amino acid positional preferences in enlarged representative protein datasets and revised the amino acid preferences. In doing so, we identified a number of ‘unexpected’ isolated ${\beta}$-turns with a proline amino acid residue at the (i+2) position. We extended our study to the identification of multiple turns, continuous turns and to peptides that correspond to the combinations of individual ${\beta}$ and ${\gamma}$-turns in proteins and examined the hydrogen-bond interactions likely to stabilize these peptides. This led us to develop a database of structural motifs in proteins (DSMP) that would primarily allow us to make queries based on the various fields in the database for some well-characterized structural motifs, such as, helices, ${\beta}$-strands, turns, ${\beta}$-hairpins, ${\beta}$-${\alpha}$-${\beta}$, ${\psi}$-loops, ${\beta}$-sheets, disulphide bridges. We have recently implemented this information for all entries in the current PDB in a relational database called ODSMP using Oracle9i that is easy to update and maintain and added few additional structural motifs. We have also developed another relational database corresponding to amino acid sequences and their associated secondary structure for representative proteins in the PDB called PSSARD. This database allows flexible queries to be made on the compatibility of amino acid sequences in the PDB to ‘user-defined’ super-secondary structure conformation and vice-versa. Currently, we have extended this database to include nearly 23,000 protein crystal structures available in the PDB. Further, we have analyzed the ‘structural plasticity’ associated with the ${\beta}$-propeller structural motif We have developed a method to automatically detect ${\beta}$-propellers from the PDB codes. We evaluated the accuracy and consistency of predicting ${\beta}$ and ${\gamma}$-turns in proteins using the residue-coupled model. I will discuss results of our work and describe databases and software applications that have been developed.
With the use of Agrobacterium and gene-gun, cold regulated gene (BN115) has been injected in Chrysanthemum leaf disc and transgenic plants have been produced successfully on the selection media containing phytohormone. To determine the presence of the transferred cold regulated gene (BN115) in the transgenic Chrysanthemum, PCR-amplification indicated the presence of that gene. Real-Time PCR for confirmation of the putative transgenic plants was established. The copy number of cold regulated gene (BN115) is extrapolated on the basis of a standard curve. Serial dilutions of known number of gene copies were in triplicates. In this diagram, PCR cycles are plotted against the fluorescence intensity. The cycle at which the fluorescence reaches a threshold cycle is inversely proportional to the starting amount of target DNA.
There are three pseudourdine ($Psi$)bases in the E. coli $tRNA^{phe}$ In order to study the function of the pseudouridine bases in the $tRNA^{phe}$, changes of bases $tRNA^{phe}$ gene to other bases were undertaken by the site-specific mutagenesis. Site-specific mutagenesis of T in the pheW gene, a $tRNA^{phe}$ gene of E. coli, corresponding to the baseat the No.32 position to C and also T corresponding to the base at the No.39 position to C were performed using Kunkel's uracil-containing template method. Identification of mutants were undertaken by the KNA sequencing techniques of the mutated pheW genes and activities of the mutated pheW genes complementing to E. coli NP37 mutant($pheS^{-ts}$) using the recombinant plasmid containing the mutated genes. Neither NP37 harboring pheW gene mutated at No.32 position nor NP37 harboring pheW gene mutated at No.39 position can be grown at non-permissive temperature. The result means that both mutated pheW genes can not complement to E. coli NP37, and that the pseudouridine bases are essential to the activity of the E. coli $tRNA^{phe}$ in vivo.
This research is to compare the infant's temperament and parenting stress perceived by premature infants' mother and full-term infants' mother. It aims at establishing the healthy relationship between mothers and infants. It investigates the basic data of nursing intervention program to help the mother of premature infant. The period of data collection was from May 1, 2005 to May 30, 2005, and the subjects were total 123 mothers: 44 mothers of premature infants and 79 mothers of full-term infants under 6 months who visited general hospitals, individual pediatrics, and health center located in G city. "What My Baby Is Like(WBL)" developed by Pridham, Chang, and Chiu(1994) and translated by Bang Kyeong Sook(1999) was used as a measuring instrument of infants' temperament, and Parenting Stress Index (PSI) developed by Abidin(1990) and revised and complemented by Kim Dong Hee(1997) was used as a measuring instrument of parenting stress. Collected Data were analyzed through $X^2$-test, t-test, Pearson's correlation with SPSS 12.0 window program. The results are as follows: 1. There was a significant difference in the infant's temperament perceived by premature infants' mothers and full-term infants' mothers (t=-4.08, p=.00). In subcategory, there were significant difference between premature infants' mother and full-term infants' mother in geniality(t=-3.62, p=.00), adaptation(t=-3.43, p=.00) and reaction(t=-2.01, p=.05). 2. There was a significant difference in parenting stress between premature infants' mother and full-term infants' mother(t=6.57, p=.00). The parenting stress of premature infants mothers appeared to be higher than full-term infants' mothers. They showed the higher stress in the mother-child relationship area(t=6.27, p=.00) and child area(t=7.38, p=.00) among 3 areas of parenting stress. 3. There were negative correlation between infants' temperament and parenting stress. As mothers perceived the infant's temperament negatively, the parenting stress. Especially, the negative correlation of infants' temperament and parenting stress of premature infants' mothers(r=-.44) was stronger than that of full-term infants' mothers(r=-37). From the research, as mothers of premature infants receive more stress, their stress can cause the serious problem to the relationship of the mother and the infant. Therefore, the nursing intervention should be carried out in order to change the negative perception of mothers towards their infants into the positive perception reduce the parenting stress.

In this thesis, we investigate the psychosocial variables within the family environments of Attention Deficit Hyperactivity Disorder with (ADHD+CD/ODD) or without (ADHD) externalizing symptoms. The subjects in this research were 86 boys and girls(aged range 6 to 14 years) consisted of 20 ADHD, 22 comorbid ADHD(ADHD+CD/ODD) and 44 normal control group(NC). We have collected data on children and their mothers. The psychosocial variables included in the analysis are socioeconomic status, parent's educational level, life stress event, and the rate of psychiatric disorders in relatives. The self-reported questionnaires marital discord(MAS), parenting stress(PSI), and parenting attitute(MBRI) completed by mothers. The results indicated that ADHD+CD/ODD is supposed to have higher level of family adversity suggested by the lower SES, lower parental educational level, higher life stress events, and more psychic disorders in relatives compared with ADHD or normal control group. In MAS, ADHD+CD/ODD group has significantly the lowest scores on each factor of the measure of marital adjustment. Parents of ADHD+CD/ODD are much more likely to have positive parenting stress when compared with the parents of ADHD. Especially, mothers of ADHD+CD/ODD have the lowest tendency in the mean score on affective, accepted attitudd. In an inapropriate parenting attitude perceived by children, father of ADHD+CD/ODD have the most negative, contradictory attitude and mothers of ADHD+CD/ODD have the most restrictive, negative and contradictory attitude.
Optical structure of $V_{2}O_{5}$ thin films were analyzed and confirmed, the films were deposited in oxygen partial pressure 0% and 10% by RF magnetron sputtering system. Measurements of the elliptic constants were made in the range of $0.75{\sim}4.0\;eV$ by using phase modulated spectroscopic ellipsometer. The elliptic constants of the thin films were analyze by Double Amorphous dispersion relation. The calculated n, k spectra of $V_{2}O_{5}$ layer were obtained over the range of $0.75{\sim}4.0\;eV$ photon energy. SEM and XRD measurements were also made to validate the ellipsometric analysis and they give good agreement with the structural properties of the films. It was found that optical structure of the $V_{2}O_{5}$ layer has a 3 phase(roughness/film/substrate) and optical absorption properties are greatly depend on the partial pressure of the oxygen.
To characterize cytosolic $Ca^{2+}$ fluctuations under metabolic inhibition, rat ventricular myocytes were exposed to $200{\mu}M$ 2,4-dinitrophenol (DNP), and mitochondrial $Ca^{2+}$, mitochondrial membrane potential (${\Delta}{\Psi}m$), and cytosolic $Ca^{2+}$ were measured, using Rhod-2 AM, TMRE, and Fluo-4 AM fluorescent dyes, respectively, by Laser Scanning Confocal Microscopy (LSCM). Furthermore, the role of sarcolemmal $Na^+$/$Ca^{2+}$ exchange (NCX) in cytosolic $Ca^{2+}$ efflux was studied in KB-R7943 and $Na^+$-free normal Tyrode's solution (143 mM LiCl ). When DNP was applied to cells loaded with Fluo-4 AM, Fluo-4 AM fluorescence intensity initially increased by $70{\pm}10$% within $70{\pm}10$ s, and later by $400{\pm}200$% at $850{\pm}45$ s. Fluorescence intensity of both Rhod-2 AM and TMRE were initially decreased by DNP, coincident with the initial increase of Fluo-4 AM fluorescence intensity. When sarcoplasmic reticulum (SR) $Ca^{2+}$ was depleted by $1{\mu}M thapsigargin plus $10{\mu}M ryanodine, the initial increase of Fluo-4 AM fluorescence intensity was unaffected, however, the subsequent progressive increase was abolished. KB-R7943 delayed both the first and the second phases of cytosolic $Ca^{2+}$ overload, while $Na^+$-free solution accelerated the second. The above results suggest that: 1) the initial rise in cytosolic $Ca^{2+}$ under DNP results from mitochondrial depolarization; 2) the secondary increase is caused by progressive $Ca^{2+}$ release from SR; 3) NCX plays an important role in transient cytosolic $Ca^{2+}$ shifts under metabolic inhibition with DNP.
Purpose: This review article was written to theoretically compare the depressing force (pressure, adhesion) to the cornea between when the spherical lenses were being tightly and flat fitted. Methods: Mathematical equations and their numerical solution programs (model) were formulated to calculate the depressing (adhesion) force to the cornea by both the tightly and flat fitted contact lenses. Based on this proposed model the effects of parameters characterizing a contact lens such as BCs, diameters, edge shape and corneal shape (ratio of long and short corneal axis, p) on the depressing force to the cornea were predicted/analyzed in both tightly and flat fitting regimes. Results: Corneal adhesion increased as the corneal p-value increased. Adhesion increase caused by the increased p-value was much larger in flat fitted case than in tight fitted one. Corneal adhesion reduced abruptly as the BC increased in flat fitting regimes while the adhesion rise was insignificant in tight fitting ones. Reduction in corneal adhesion due to lens-size increase was predicted to be insignificant in both tight and flat fitting regimes. Both the lens edge shape (edge angle) and thickness were relevant only in tight fitting regime. Corneal adhesion increased as the increased with tight-fitted lenses. As the thickness of tight fitted lenses increased, corneal adhesion inversely decreased. Conclusions: The two most significantly affecting the depressing force to cornea were found to be the degree of corneal bending toward the periphery and the BCs of lenses.

To examine the pro-apoptotic role of the human ortholog (YPEL5) of the Drosophila Yippee protein, the cell viability of Saccharomyces cerevisiae mutant strain with deleted MOH1, the yeast ortholog, was compared with that of the wild-type (WT)-MOH1 strain after exposure to different apoptogenic stimulants, including UV irradiation, methyl methanesulfonate (MMS), camptothecin (CPT), heat shock, and hyperosmotic shock. The $moh1{\Delta}$ mutant exhibited enhanced cell viability compared with the WT-MOH1 strain when treated with lethal UV irradiation, 1.8 mM MMS, $100{\mu}M$ CPT, heat shock at $50^{\circ}C$, or 1.2 M KCl. At the same time, the level of Moh1 protein was commonly up-regulated in the WT-MOH1 strain as was that of Ynk1 protein, which is known as a marker for DNA damage. Although the enhanced UV resistance of the $moh1{\Delta}$ mutant largely disappeared following transformation with the yeast MOH1 gene or one of the human YPEL1-YPEL5 genes, the transformant bearing pYES2-YPEL5 was more sensitive to lethal UV irradiation and its UV sensitivity was similar to that of the WT-MOH1 strain. Under these conditions, the UV irradiation-induced apoptotic events, such as FITC-Annexin V stainability, mitochondrial membrane potential (${\Delta}{\psi}m$) loss, and metacaspase activation, occurred to a much lesser extent in the $moh1{\Delta}$ mutant compared with the WT-MOH1 strain and the mutant strain bearing pYES2-MOH1 or pYES2-YPEL5. These results demonstrate the functional conservation between yeast Moh1 and human YPEL5, and their involvement in mitochondria-dependent apoptosis induced by DNA damage.

The power water flowed out from the multipurpose darn influences the ecosystem approximately because of the low water temperature. An appropriate counter measure to the rising water temperature is needed for growing crops especially when the temperature is below 18˚C in the source of the irrigation water This observational study is practiced in Yong-Doo water warming canal and pond in the down stream of Choong-Ju multipurpose dam and is practiced for analyse and compare the rising effects in actural water temperature by actual measurement with the rising effects of planned water temperatuer by the basic theoritical method and for the help to present the direction in plan establishment through investigate the results afterwards. The results are as follows. 1.The degree of the rise of the water temperature can be decided by $\theta$x=$\theta$o +K L--v.h (T-$\theta$˚)Then, K values of a factor representing the characteristics of the water warming canal were 0.00002043 for the type I. and 0.0000173 for the type II. respectively. 2.A variation of water temperature which produced by the difference effective temperature and water temperature in the water warming canal was $\theta$x1 = 16.5 + 15.9(1-e -0.00018x), $\theta$x2 =18.8 + 8.4( 1-e -0.000298x)for the type I. and $\theta$x, = 19.6 + 12.8 ( 1-e -0.00041x) for the type II. 3.It was shown that the effects of the rise of water temperature for the type I. water warming canal were greater than that of type II. as a resultes of broadening the surface of the canal compared with the depth of water, coloring the surface of water canal and installing the resistance block. 4.In case of the type I. water warming canal, the equation between the air temperature and the degree of the rise of water temprature could be made ;Y= 0.4134X + 7.728 In addition, in case of the type II. water warming canal, the correlation was very low. 5.A monthly variation of the water temperature in the water warming canal was the highest in August during the irrigation period and the water temperature rose with the air temperature until August. However, it was blunted after then. 6.A rising degree of water temperature of the practical value in the water warming pond was higher than that of the theoritical equation by 69% for the type I. and 57% for the type II. Accordingly, it was possible to acquire the result near the practical value.$\theta$w-$\theta$o=[1-exp{ -h(1+2$\psi$) . X($\theta$w-$\theta$0)XC Here, C values are 1.69 for the type I. and 1.57 for the type II. 7.It was shown that the effect of the rise of water temperature was favorable when the thermal absorption was to be good by coloring the surface of the water warming pond and removing the bottom osmosis. 8.By enlarging the surface of water in comparison with the depth, and by having dead area of water in the water warming pond, this structure in the water warming pond is helpful for the rise of water temperature.
