Objective : This experimental study was performed to investigate the effect of Coptidis rhizoma extract compared with quantity on Pseudomonas aeruginosa that induce keratitis. Methods : MIC was measured by dropping to 50 ${\mu}l$ according to density Coptidis rhizoma extract(100%, 10%, 1%, 0.1%) compared with quantity(40g, 80g, 120g, 160g). Anti-bacterial potency was measured by the size of inhibition zone with change of volume. Results : MIC on Pseudomonas aeruginosa in Coptidis rhizoma extract compared with quantity(40g, 80g, 120g, 160g) was not showed all samples(100%, 10%, 1%, 0.1%). Conclusions : Coptidis rhizoma extract compared with quantity was not showed anti-bacterial potency on Pseudomonas aeruginosa. The results differ that Coptidis rhizoma extract was showed anti-bacterial potency in 1999 year. Anti-bacterial Potency of Coptidis rhizoma extract compared with quantity on Pseudomonas aeruginosa must be studied.

Pseudomonas sp. P20 isolated from the polluted environment is capable of degrading biphenyl and 4-chlorobiphenyl. The pcbABCD genes responsible for degradation of biphenyl and 4-chlorobiphenyl were cloned using pBluescript SK(+) from the chromosomal DNA of Pseudomonas sp. P20 to construct pCK1 and pCK102, harbouring pcbABCD and pcbCD, respectively. The 2, 3-DHBP dioxygenase gene, pcbC, was cloned again from pCK102 by using pKT230 which is known as a shuttle vector and pKK1 hybrid plasmid was constructed. The E. coli KK1 transformant obtained by transforming the pKK1 into E. coli XL1-Blue showed 2, 3-DHBP dioxygenase activity. The specific 2, 3-DHBP dioxygenase activity of E. coli KK1 was similar to that of the E. coli CK102, but much higher than those of the natural isolates, Pseudomonas sp. DJ-12 and Pseudomonas sp. P20.
Three biphenyl-degrading microorganisms were isolated from polluted soil samples in Sasang-gu, Busan. Among them, isolate DS-94 showing the strong degrading activity was selected. The morphological, physiological, and biochemical characteristics of DS-94 were investigated by API 20NE and other tests. This bacterium was identified as the genus Pseudomonas by 16S rDNA sequencing and designated as Pseudomonas sp. DS-94. The optimum temperature and pH for the growth of Pseudomonas sp. DS-94 were $25^{\circ}C$ and pH 7.0, respectively. This isolate could utilize biphenyl as sole source of carbon and energy. Biphenyl-degrading efficiency of this isolate was measured by HPLC analysis. As a result of biological biphenyl-degradation at high biphenyl concentration (500 mg/L), biphenyl-removal efficiency by this isolate was 73.5% for 7 days.

Anti-Pseudomonas aeruginosa activities for ethanol extract of 34 medicinal plants widely used in the folk medicine were evaluated to screening of anti-Pseudomonas aeruginosa agents. The ethanol extracts of Gardenia jasminoides, Arctium lappa, Citrus unshiu, and Phellodendron amurense showed antimicrobial activities against P. aeruginosa ATCC 27853. The ethanol extracts of Gardenia jasminoides among these medicinal plants showed significant antimicrobial activity against P. aeruginosa. Therefore, we expect that these medicinal plants will be useful for nature antimicrobial agent against P. aeruginosa in future.
The ability of Pseudomonas putida to degrade toluene was studied in toluene-containing vials. The strain grows anaerobically in toluene as a sole source of carbon. When the initial toluene concentrations injected in the vial are varied, the changes of headspace toluene concentration and cell density are observed. We set a model for this vial and simulated the vial reactor using Matlab. With a variation of model parameters, simulated results were compared with the experiment.

Strain M-18 was isolated from the rhizosphere soil of sweet melon, using 1-aminocyclopropane-1-carboxylate (ACC) as a sole nitrogen source. Its phenotypic characteristics, metabolic tests, and 16S rDNA sequence were analyzed. The antibiotics secreted by strain M-18 were determined to be phenazine 1-carboxylic acid and pyoluteorin. These data showed that strain M-18 was a new fluorescent Pseudomonas strain that produced both phenazine 1-carboxylic acid and pyoluteorin, some features being similar to Pseudomonas aeruginosa and Pseudomonas fluorescens. Therefore, the strain M-18 appears to be the first pseudomonad described to date that is capable of producing both phenazine 1-carboxylic acid and pyoluteorin.
Purpose: Pseudomonas aeruginosa is an etiologic agent in serious wound infection. Pseudomonas aeruginosa infection is problematic because this organism is resistant to many antimicrobial drugs. The purpose of this study was to compare the bactericidal effect of commonly used topical agents and their effect on wound healing. Methods: Pseudomonas aeruginosa-infected full-thickness skin defect was developed on the mouse to compare 3 commonly used topical agents-Betadine, 2% Gentamicin solution and 0.3% Acetic acid with the control group. Wound size change, bacterial colony counts and histologic findings of each groups were analyzed. Results: The wound size decreased in all treated groups as compared with the control group. However, there was no statistical difference. Gentamicin solution group was showed the lowest bacterial colony count and statistically significant difference compared with the control group(p=0.032). Other treated groups were also effective against Pseudomonas aeruginosa, but not different statistically. Histologic findings revealed that epithelialization, granulation tissue formation and microvessel proliferation were increased and necrosis and inflammation were decreased in all treated groups compared to the control group, but not different statistically. Betadine group significantly increased granulation tissue formation compared to the control group (p= 0.041). Conclusion: There is no universal topical agent that enhances most aspects of wound healing while simultaneously decreasing the bacterial concentration. However, Gentamicin solution may be an optimal topical agent for Pseudomonas aeruginosa infected wound. Further study should experiment on human with Gentamicin solution to confirm a effect on Pseudomonas aeruginosa infected wound for clinical applications.

The degradation of non-volatile-amines by microorganisms were investigated. The degra-ding activity could be noted in four strains isolated from fermented sardine sauce, and those were Pseudomonas aeruginosa, Pseudomonas fluorescens P-2, Pseudomonas fluorescens P-3 and Enterobacter aerogenes. The strongest degrading activity of non-volatile-amines was showed in Pseudomonas fluorescens P-3 among the four strains isolated. The optimum temperature for degradation by Pseudomonas fluorescens P-3 was $35^{\circ}C$, corresponding to the optimum temperature for growth of this strain, pH between 7.0 and 7.5 could gave effective degradation and the optimum concentration of NaCl was 0 and/or $1\%$.
From several sites of petroleum storage basement in South Coasts in Korea, various petroleum degrading bacteria have been isolated and characterized as Pseudomonas fluorescens, Acinetobacter baumanii, Pseudomonas maltophila and Pseudomonas aeruginosa, respectively. They show the ability of petroleum degradation on minimal media which contains petroleum as sole carbon source and loose the ability at high concentration of NaCl as increasing the concentration of NaCl from 0.5% to 6%. It has been confirmed that such bacteria have utilized the simple saturate hydrocarbon; n-decane, n-hexane, n-octane and n-decane because petroleum consists of various kinds of organic compounds. It has been also identified that petroleum degrading bacteria habor the plasmid and show the antibiotic resistance against ampicillin, tetracycline and chloramphenicol. These results strongly suggest that the petroleum degrading gene and antibiotic resistance gene might be located on the high molecular weight plasmid.
Eighty one bacterial strains of parathion degrading bacteria were isolated from soil samples that were contaminated with pesticide in Daejeon area. Among them, one bacterial strain was finally selected in media containing parathion as the sole source of carbon and energy, and this strain was identified as Pseudomonas rhodesiae H5 through physiological and biochemical tests, and analysis of its 16S rRNA sequence. Pseudomonas rhodesiae H5 was able to utilize various carbohydrates but did not utilize sorbose as sole carbon source. Pseudomonas rhodesiae H5 was resistance to ampicillin, spectinomycin, and mitomycin C but sensitive to kanamycin and chloramphenicol. And this strain showed high resistance up to several milligrams of heavy metals such as $BaCl_2$, LiCl, and $MnSO_4$. Optimal growth condition for temperature and pH of P. rhodesiae H5 was 3$0^{\circ}C$, and pH 7.0, respectively. It can be presumed that P. rhodesiae H5 hydrolyzed an organophosphate bond of parathion, forming p-nitrophenol, and then metabolized via ortho-ring cleavage mechanism.
