Pseudomonas sp. K-52 유래의 levanase 에 의해 생산되는 levanoctaose 가 growth factor로서 각종 장내미생물에 미치는 영향을 조사하였다. In vitro 실험에서 0.5% levanoctaose를 탄소원으로 하여 glucose 일 경우와 상호 비교하여 분석한 결과 levanoctaose 는 Escherichia coli, Clostridium perfringens, Eubacterium limosum, Staphylococcus aureus 보다 Bifidobacterium adolescentis, Lactobacillius acidophilus, Bacteroid fragilis 등에 의해 효율적으로 이용되었다. 쥐를 이용한 in vivo 실험에서 탄소원의일부로 levanotaose를 제공한 경우, 장내 Bifidobacteria 수는 약 10배, butyrate 양은 2.3배, $\beta$-fructosidase 활성은 약 1.5배 증가하였다. 따라서 분리균주 Pseudomonas sp. K-2의 levanase로부터 생성되는 levanoctaose는 Bifidobacteria, Lactobacillus 와 같은 장내 유익균주에 선별적으로 이용되는 것이 확인되었다.
본 연구는 농업과 어업, 그리고 생태체험과 같은 인간들의 활동으로 인하여 상당히 영향을 받는 갯벌환경 중의 하나인 순천만을 모델장소로 갈대의 환경정화 기능에 있어 근권에 분포하는 미생물의 역할에 대한 기초 자료를 얻고자 수행하였다. 우선, 순천만의 갈대근권 토양을 시료로하고 anthracene, naphthalene, phenanthrene, pyrene 등이 첨가된 다환성 방향족 화합물(polycyclic aromatic hydrocarbons; PAH)을 탄소원 및 에너지원으로 하는 농화 배양을 통하여 두 개의 consortium을 획득하였다. 두 consortium으로부터 순수 분리된 우수한 PAH분해능을 갖는 4개의 균주(SCB1, SCB2, SCB6,그리고 SCB7)를 형태 및 생리학적 특성과 16S rRNA유전자서열을 기초로 분석한 결과 각 균주는 $99{\%}$ 이상의 신뢰도로 Burkholderia sp., Aicaligenes sp., Achromobacter sp., and Pseudomonas sp.로 동정되었다. 주목할 만한 점은 Burkholderia sp. SCB1과 Alcaligenes sp. SCB2는 naphthalene이나 phenanthrene보다 훨씬 안정되어 있는 구조의 anthracene이나 pyrene에서 더 빠른 성장률과 기질 분해율을 나타내는 것으로 밝혀졌다. 반면,Achromobacter sp. SCB6와 Pseudomonas sp. SCB7은 pyrene을 제외한 다른 시험기질에 대하여 유사한 성장 및 분해패턴을 나타내었다. 이러한 결과는 주요한 염습지 식물중의 하나인 갈대의 근권에서 살아가는 이들 PAH 분해 균주들이 PAH와 같은 물질로 오염된 근권 환경의 정화작용에 중요한 역할을 할 수 있음을 제시해 주었다.
For the production of D-lactic acid from DL-2-chloropropionic acid, about 80 strains of bacteria capable of assimilating DL-2-chloropropionic acid as a sole carbon and energy source were isolated from the soil. JH-007 strain that showed the higest productivity of D-lactic acid and didn't produce L-lactic acid from DL-2-chloropropionic acid was selected from them and identified as Pseudomonas sp. The optimal conditions for the production of D-lactic acid from DL-2-chloropropionic acid were examined. The resting cells of JH-007 cultured in LB medium containing 3 g/l of DL-2-chloropropionic acid were used as an enzyme source. The reaction mixtures for the maximal production of D-lactic acid were consist of 10 g/l of resting cells and 3 g/l of DL-2-chloropropionic acid in 125 mM sodium carbonate buffer. The optimal pH for the reaction was 10.0 and the optimal temperature was 30$\circ$C. When 1 g/l of DL-2-chloropropionic acid was added intermittently to the reaction mixture under the above condition, 5.72 g/l of D-lactic acid was produced after incubation of 5 hrs. This amount of D-lactic acid corresponded to a 98.4% yields and the optical purity was 99.8%.
In order to compare the metabolic activities of methanol utilizing bacteria, Pseudomonas sp. grown in different carbon sources, changes in respiratory activities, prinicipal enzyme activities for the energy metabolism, and the macromolecular compositions of the cells grown on methanol or glucose were measured. 1. The respiratory activity of cells grown on methanol was higher than that of cells grown on glucose, while glucose exhibited the highest $O_2-consumption$ rate among the different respiratory substrates. 2. TRhe activity of hydroxy pyruvate reductase which participates in serine pathway was high in the cells grown on methanol. However, activities of NAD-linked alcohol dehydrogenase, formaldehyde dehydrogenase and formate dehydrogenase were slightly lower in the cells grown on glucose thant on methanol. 4. For succinic dehydrogenase and malic dehydrogenase which take part in TCA cycle, the specific activities were higher in the cells grown on methanol than in those grown on glucose. No activity of glucose-6-phosphate dehydrogenase, which participates in pentose monophosphate shunt, was detectable in the cells grown on either carbon sources. 5. Protein contents of the cells grown on methanol increased relatively compared with those of the cells grown on glucose. However, there are no changes in the contents of carbohydrate and nucleic acid.
A bacterial strain, designated T116, degrading terephthalic acid (TPA) was isolated from the soil around Taegu industrial area into which dye works wastewater flow. The isolate was identified as pseudomonas sp. based on its morphological and physiological characteristics. Degradation of TPA by the strain T116 was confirmed with UV scanning and HPLC. About 90% and 98% of TPA were degraded after 36 and 60 hours, respectively, during the culture in a liquid medium containing 0.1% TPA. Addition of KH2PO4 at a final concentration of 100ppm enhanced the chemical oxygen demand (COD) removal rate about 50% from dye works wastewater by Pseudomonas sp. T116. Optimum pH and temperature for COD reduction from wastewater were 7.0 and 3$0^{\circ}C$, respectively. The bacterium was applied to the continuous culture for the treatment of dye works wastewater whose TPA concentration and CODMn were 2,200ppm and 1,620ppm, respectively. It was observed that 90-95% of COD was eliminated after 4 days culture in the continuous culture with a retention time of 37 or 47 hours.
Pseudomonas agarici에 의해 발생하는 세균성회색무늬병은 양송이 재배에서 문제가 되는 대표적인 병해이다. 본 연구에서는 세균성회색무늬병의 생물학적 방제법에 이용할 수 있는 길항미생물의 항균활성과 선발된 길항미생물에 대해 폿트 수준의 생물검정 실험을 실시하였다. 재배중인 양송이 배지에서 세균성회색무늬병 병원균을 강하게 억제하는 길항세균 HC12를 선발하였으며, 생리 생화학적 실험과 유전적 실험결과 HC12균주는 Alcaligenes sp.로 동정되었다. Alcaligenes sp. HC12를 양송이에 처리한 결과 63%의 방제효과를 보였다. 따라서 Alcaligenes sp. HC12가 양송이버섯 세균성회색무늬병 방제를 위해 합성농약을 대체할 수 있는 친환경 방제제가 될 수 있을 것으로 생각된다.
본 연구의 목적은 가축사체 매몰지 토양에서 추출된 미생물을 이용하여, 가축사체 매몰지 오염 토양 유래 유기탄소의 분해 효과를 분석하고, 분해에 관여하는 미생물의 종을 규명하는 것이다. 경기도 안성에 위치한 가축 매몰지 토양 유래 용존 유기탄소를 대상으로 토양 서식 미생물의 생분해율을 평가한 결과(56일 동안), 1번 용액(초기 용존 유기탄소 농도 = 19.88 mg C/L)에서는 13일 이내에 48%의 용존 유기탄소가 감소하였으며, 56일째 용존 유기탄소는 $8.8{\pm}0.4$ mg C/L로 56% 분해되었다. 2번 용액(초기 용존 유기탄소 농도 = 19.80 mg C/L)에서도 초기 13일 이내에 용존 유기탄소 농도가 급격히 감소하였으며, 56일째 용존 유기탄소는 $6.0{\pm}1.6$ mg C/L로 70%가 분해되었다. 생분해 실험과정에서, 전체 유기탄소 물질에서 방향족 탄소구조의 비율을 나타낸 지표인 $SUVA_{254}$값은 용존 유기탄소와는 다르게, 일정기간(21일) 동안 증가하다가 감소하는 경향을 나타냈다. $SUVA_{254}$값은 1번 용액과 2번 용액 모두, 21일째 가장 높은 값을 나타내었다. 생분해 실험 14일째 미생물 분석 결과, 토양에서 쉽게 발견되는 Pseudomonas fluorescens, Achromobacter xylosoxidans, Nocardioides simplex, Pseudomonas mandelii, Bosea sp. 등 미생물이 검출되었다. 그리고, 56일째 미생물 분석 결과, Pseudomonas veronii가 우점종으로 나타났다.
Aromatic hydrocarbons can be utilized as carbon and energy sources by some microorganisms at lower concentrations. However, they can also act as stresses to these organisms at higher concentrations. Pseudomonas sp. DJ-12 is capable of degrading 0.5 mM concentration of benzoate and 4-chlorobenzoate (4CBA). In this study, the exposure of Pseudomonas sp. DJ-12 to the pollutant stresses of benzoate and 4CBA at various concentrations was comparatively studied for its cellular responses, including survival tolerance, degradability of the aromatics, and morphological changes. Pseudomonas sp. DJ-12 utilized 0.5 to 1.0mM benzoate and 4CBA as carbon and energy sources for growth. However, the organism could not degrade the aromatics at concentrations of 3 mM or higher, resulting in reduced cell viability due to the destruction of the cell envelopes. Pseudomonas sp. DJ-12 cells produced stress-shock proteins such as DnaK and GroEL when treated with benzoate and 4CBA at concentrations of 0.5mM, or higher as sublethal dosage; Yet, there were differing responses between the cells treated with either benzoate or 4CBA. 4CBA affected the degradability of the cells more critically than benzoate. The DnaK and GroEL stress-shock proteins were produced either by 1mM benzoate with 5 min treatment or by higher concentrations after 10min. The proteins were also induced by 0.5mM 4CBA, however, it needed at least 20min treatment or longer. These results indicate that the chlorination of benzoate increased the recalcitrance of the pollutant aromatics and changed the conditions to lower concentrations and longer treatment times for the production of stress-shock proteins. of stress-shock proteins produced by the aromatics at sublethal concentrations functioned interactively between the aromatics for survival tolerance to lethal concentrations.
2-Hydroxy-6-oxo-6phenylhexa-2,4-dienoate (HOPDA) hydrolase catalyzes the hydrolytic cleavage of HOPDA to bemzpate and 2-hydroxypenta-2, 4-dienoate (HPD) during microbial catabolism of biphenyl and polychlorinated biphenyls. A HOPDA hydrolase gene (pcbD) was isolated from the genomic library of Pseudomonas sp. P20 and designated as pCNUO1201; a 7.5-kb XbaI DNA fragment from Pseudomonas sp. P20 was inserted into the pBluescript SK(+) XbaI site. E. coli HB101 harboring pCNU1201 exhibited HOPDA hydrolase activity. The open reading frame (ORF) corresponding to the pcbD gene consisted of 855 base pairs with an ATG initiation codon and a TGA termination codon. The ORF was preceded by a rebosome-binding sequence of 5'-TGGAGC-3' and its G+C content was 55 mol%. The pcbD gene of Pseudomonas sp. P20 was located immedeately downstream of the pcbC gene encoding 2,3- dihydroxybiphenyl 1,2-dioxygenase, and approximately 4-kb upstream of the pcbE gene encoding HPD hydratase. The pcbK gene was able to encode a polypeptide with a molecular weight of 31,732 containing 284 amino acid residues. The deduced amino acid sequence of the HOPDA hydrolase of Pseudomonas sp. P20 exhibited high identity (62%) with those of the HOPDA hydrolases of P. putida KF715, P. pseudoalcaligenes KF707, and Burkholderia cepacia LB400, and also significant homology with those of other hydrolytic enzymes including esterase, transferase, and peptidase.
