The purpose of this study was to investigate the effect of mixing methods on the biodegradation of sorbed naphthalene and phenanthrene in soils. Biodegradation was initiated by inoculating Pseudomonas sp. KM1 into equilibrated soil slurry vials. Four different mixing methods, including no mixing, orbital shaking, rolling and rotating were utilized to enhance the biodegradation of both naphthalene and phenanthrene. The experimental results showed that the sorbed compounds were more effectively biodegraded with rolling and rotating mixing methods. The sorbed naphthalene concentrations were reduced to 0 mg/kg via the rolling and rotating methods. However, with no mixing and the orbital shaking methods, the sorbed naphthalene concentrations were comparatively high, ranging from 2.59 to 20.45 mg/kg. Similar trends were observed for the biodegradation of phenanthrene, but the concentrations remaining were higher than those of naphthalene, due to the limited bioavailability of the sorbed phenanthrene. The rolling and rotating mixing methods are suggested can distribute bacteria uniformly in the slurry system; improve the mass transfer rate and the probability of physical contact between bacteria and the sorbed contaminants, resulting in higher bioavailability of the contaminants.

Bacteriocin-producing lactic acid bacteria was isolated from Kimchi using MRS as selective media and Lactobacillus delbruekii subsp. delbruekii as an indicator strain. Strain JC-3 was tentatively identified as Lactococcus latis subsp. lactis through the API test and the bacteriocin produced by JC-3 showed the inhibitory activity against Grampositive pathogens and other lactic acid bacteria. The antimicrobial substance was inactivated by Protamax, Aroase AP-10, Neutrase, R-AMANO and was confirmed to be heating at $100^{\circ}C$. However, it was lost at high pH values showed the highest bacteriocin activity at a culture temperature of $30^{\circ}C$. The bacteriocin was partially purified by ammonium sulfate precipitation, Sep-pak $C_{18}$ cartridge. The apparent molecular mass of the bacteriocin was about 8 Kda, which was determined through the direct detection of bactericidal activity using SDS -PAGE.
This study was undertaken to find a new formulation of soil amendment, and selection of antogonists and to effectively control brown and large patch of turfgrasses caused by Rhizoctoniz solani AG1-1 and AG 2-2. Fourteen inorgainc chemicals (1%, w/w) were added individually in vitro, and some chemicals showed suppressiveness to R. solani. Alum suppressed effectively mycelial growth of R. solani in the range of 17 to 77% as compared with control. The four chemicals such as Al2(SO4)3, alum, CaO, and NH4NO3 were finally selected. Out of three organic compounds, composted pine bark (CPB) showed prominent suppressive effect as compared with milled alfalfa and pine leaves. After inoculation of R. solani isolates AG-1 and AG2-2 on the turf seedlings, water soaked lesions and blight symptoms were developed on the whole seedlings. According to inhibition zone method, mycelial growth of the fungus were greatly suppressed by culture filterates of the antagonists, Gliocladium virens (Gl1-) and Pseudomonas sp. (P713). CPB soil amendment mixed with antagonists (1% w/w) controlled not only brown and large patch of turfgrasses, but also promote the good growth of the seedlings. In addition, the controlling effect was maintained more than 30 days. Especially, the controlling effect of two antagonists was similar to Cㅖㅠ soil amendment with the antagonists and also stimulated a favorable growth of the seedlings. Therefore, its is expected that continuous control of Rhizoctonia blight of turfgrasses can be obtained in field by subsequent applications of the antagonists.
Mechanol extracts of three cold-tolerant eucalyptus trees-Eucalyptus darlympleana, E. gunnii and E. unigera were screened for antimicrobial activity against twenty two phyto-pathogenic fungi and six food-borne bacterial pathogens. E. unigera showed the antagonistic activity against all the tested pathogens. Among the tested fungal pathogens, Pythium species were highly sensitive to the leaf extracts. Especially, P. vanterpoolii, a causal agent of leaf blight in creeping bentgrass (Agrostis palustris), was completely inhibited by the extracts. The eucalyptus extracts were also effective in inhibiting the fungal growth of Botrytis cinerea and Phomopsis sp. isolated from the lesions of kiwifruit soft rot during post-harvest storage. Escherichia coli O-157 was less sensitive to the inhibition than the other bacterial pathogens tested. It was likely that Gram positive bacteria-Bacillus subtilis and Streptococcus mutans were more sensitive to the eucalyptus extracts than Gram negative bacteria-Escherichia coli, Salmonella enteritidis and Pseudomonas aeruginosa. Our findings suggest that the cold-tolerant eucalyptus species have antimicrobial properties that can serve the development of novel fungitoxic agents or food preservatives.
H$_2$-producing bacteria were isolated from anaerobic granular sludge. Out of 72 colonies (36 grown under aerobic conditions and 36 under anaerobic conditions) arbitrarily chosen from the agar plate cultures of a Suspended sludge, 34 colonies (15 under aerobic conditions and 19 under anaerobic conditions) produced H2 under anaerobic conditions. Based on various biochemical tests and microscopic observations, they were classified into 13 groups and tentatively identified as follows: From aerobic isolates, Aeromonar spp. (7 strains), Pseudomonas spp. (3 strains), and Vibrio spp. (5 strains); from anaerobic isolates, Actinomyces spp. (11 Strains), Clostridium 5pp. (7 strains). and Porphyromonas sp. When glucose was used as the carbon substrate, all isolates showed a similar cell density and a H$_2$ production yield in the batch cultivations after 12 h (2.24-2.74 OD at 600 nm and 1.02-1.22 mol H$_2$/mol glucose, respectively). The major fermentation by-products were ethanol and acetate for the aerobic isolates, and ethanol, acetate and propionate for the anaerobic isolates. This study demonstrated that several H$_2$ producers in an anaerobic granular sludge exist En large proportions and their performance in terms of H$_2$ production is quite similar.
In order to rapidly identify and differentiate Salmonella typhimurium from the intestinal gram-negative bacteria, randomly amplified polymorphic DNA (RAPD) fingerprinting of Salmonella typhimurium was carried out using random primers designated OPA-13 (5'-CAGCACCCAC-3'), OPB-10 (5'-CGTCTGGGAC-3'), OPB-18 (5'-CCACAGCAGT-3'), and OPJ-10 (5'-AAGCCCGAGG-3'), and its patterns compared with 6 representive intestinal, gram-negative bacterial strains, Vibrio parahaemolyticus, V. vulnificus, Enterobacter cloacae, Escherichia coli O157:H7, Pseudomonas aeruginosa, and Proteus sp., which are often found in foods. S. typhimurium had unique and distinct fingerprinting patterns. RAPD fingerprinting is thus concluded to be a rapid and sensitive method for the identification of S. typhimurium compared to conventional culturing procedures or immunoassays.
The most common cause of death is ostrich chick fading syndrome(OCFS), which is due to bacterial infection during artificial incubation and hatching. Six farmed ostrich chicks aged 3 and 10 days in Chonbuk province, were submitted to Chonbuk Livestock Development and Research Institute for necropsy, Clinically, birds showed hair loss, ocular exudate, lethargy, diarrhea, and subsequently died 3-5 days after onset of clinical signs. Grossly, umbilicus was enlarged. White-yellowish purulent nodules were scattered on the lung and the membrane of air-sac was thickened and had inflamed exudate on the surface in two chicks that died 3 days after hatching. In 10 days-old chick, intestine was shown rodding segmentally. Yolk sac was still retarded and its surface was partially hemorrahgic. The synovial fluid of the leg was yellowish. Microscopically, multifocal purulent exudates were scattered on the lung. Capillary microthrombi in the glomerulus were prominent and tubular epithelia were necrotic. Necrotic hepatocytes were scattered and intestine were congested. Microbiologically, Pseudomonas sp and/or E coli were isolated from air-sac, lung and/or liver. This case suggests that poor hygiene during artificial incubation, hatching or in the first week after hatching may cause high mortality of the ostrich chicks.
Influence factors and efficiency characteristics for treatment of wastewater containing phenol were studied with using Pseudomonas sp. B3. It took 130 hours to remove phenol, when only activated sludge of terminal disposal palnt of sewage was innoculated in batch culture, but it was required just 36 hours, when bacteria degrading phenol and activated sludge were simultaneously innoculated. If only phenol an carbon source was used, it necessary 36 hours for biodegradation of phenol, while glucose was added to medium, it took 73 hours. It was revealed as excellent effluent and SVI, when the F/M ratio, COD and phenol concentration were 53mg/l and 1.2mg/l, respectively, and optimum F/M ratio was revealed 0.31. The reactor were seriously shocked as reducing hydraulic retention time at constant phenol concentration more than increasing phenol concentration at constant hydraulic retention time, when volumetric loading rate was increased to $0.8kg\;phenol/m^3{\codt}d$ from $1.6kg\;phenol/m^3{\codt}d$. And also the effluent phenol concentration was 34mg/l after starting 12 hours of shocking and reactor was recovered as steady state after 65 hours of changing in the former test. Although the effluent phenol concentration was maximum value with 12mg/l after starting 20 hours of shocking and reactor was recovered as steady state after 54 hours of changing in the later test.
A novel bacteriocin produced by Streptococcus parauberis Z49 strain was characterized and efficiently extracted from fermented cultures by use of aqueous two-phase systems. The nisin-like bacteriocin, which was active even after a heat treatment at $121^{\circ}C$ for 15 min and in the broad pH range from 2 to 12, showed inhibition of bacterial growth of Micrococcus luteus, Lactobacillus spp., Lactobacillus fermentum, Enterococcus faecium, Listereia monocytogenes, and Pseudomonas fluorescens. Optimal conditions of PEG 600/$Na_2SO_4$ aqueous two-phase systems for the simple and rapid extraction of a novel bacteriocin were determined to be PEG 600 15%, $Na_2SO_4$ 30%, and NaCl 8%, where the bacteriocin was concentrated in PEG layer.
High substrate costs decrease the profitability of polyhydroxyalkanoates (PHAs) production, and thus low-cost carbon substrates coming from agricultural and industrial residuals are tested for the production of these biopolymers. Among them, crude glycerol, formed as a by-product during biodiesel production, seems to be the most promising source of carbon. The object of this study was to characterize the mixed population responsible for the conversion of crude glycerol into PHAs by cultivation-dependent and -independent methods. Enrichment of the microbial community was monitored by applying the Ribosomal Intergenic Spacer Analysis (RISA), and the identification of community members was based on 16S rRNA gene sequencing of cultivable species. Molecular analysis revealed that mixed populations consisted of microorganisms affiliated with four bacterial lineages: ${\alpha}$, ${\gamma}$-Proteobacteria, Actinobacteria, and Bacteroides. Among these, three Pseudomonas strains and Rhodobacter sp. possessed genes coding for polyhydroxyalkanoates synthase. Comparative analysis revealed that most of the microorganisms detected by direct molecular analysis were obtained by the traditional culturing method.
