Identification of Novel Non-Metal Haloperoxidases from the Marine Metagenome

  • Haloperoxidase (HPO, E.C.1.11.1.7) is a metal-containing enzyme oxidizing halonium species, which can be used in the synthesis of halogenated organic compounds, for instance in the production of antimicrobial agents, cosmetics, etc., in the presence of halides and $H_2O_2$. To isolate and evaluate a novel non-metal HPO using a culture-independent method, a cassette PCR library was constructed from marine seawater in Japan. We first isolated a novel HPO gene from Pseudomonas putida ATCC11172 by PCR for constructing the chimeric HPO library (HPO11172). HPO11172 showed each single open-reading frame of 828 base pairs coding for 276 amino acids, respectively, and showed 87% similarity with P. putida IF-3 sequences. Approximately 600 transformants screened for chimeric genes between P. putida ATCC11173 and HPO central fragments were able to identify 113 active clones. Among them, we finally isolated 20 novel HPO genes. Sequence analyses of the obtained 20 clones showed higher homology genes with P. putida or Sinorhizobium or Streptomyces strains. Although the HPO A9 clone showed the lowest homology with HPO11172, clones in group B, including CS19, showed a relatively higher homology of 80%, with 70% identy. E. coli cells expressing these HPO chimeric genes were able to successfully bioconvert chlorodimedone with KBr or KCl as substrate.

당뇨족 궤양의 세균 역학과 항생제 감수성 (Bacteriology and Antibiotic Sensitivity for Diabetic Foot Ulcer)

  • Polymicrobial nature of diabetic foot infection has been well documented in the literature. Initial antibiotic therapy of diabetic foot infection is usually empiric until reliable culture data is shown. This study was carried out to determine the common bacteriological flora of diabetic foot infection and antimicrobial sensitivity pattern in order to enhance possible empiric treatment. The specimens were obtained from wounds of 207 cases of diabetic foot ulcer, and the bacteriological isolation, and antimicrobial susceptibility tests of the isolates were carried out by standard microbiological methods. Staphylococcus aureus was the most common isolate, with 46.2% of recover rate among total bacterial isolated cases. Among gram-negative organisms, Pseudomonas aeruginosa was most common. Gram-positive organisms showed significant susceptibility to clindamycin, trimethoprim/sulfamethoxazole, and levofloxacin, besides vancomycin. Cefoperazone, piperacillin/tazobactam, and amikacin in addition to imipenem were most effective agents compared to gram-negative organisms. Diabetic foot infection requires use of combined antimicrobial therapy for initial management. Our results indicate that the most effective antibiotic combination for diabetic foot infection of Korean patients is clindamycin plus cefoperazone.

Study of antibacterial and antifungal activity of traditional Cedrus deodara and Pinus roxburghii Sarg

  • In the present study, the volatile oil, chloroform extract and methanol extract of the woods of the plants Cedrus deodara and Pinus roxburghii were screened for their antibacterial and antifungal activities against Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus, Bacillus subtilis, Streptococcus pyogenes, Candida albicans, Aspergillus niger, and Aspergillus clavatus using the Agar diffusion method. The susceptibilities of the microorganisms to the extracts were compared with each other and with a selected standard antibiotic. It was observed that the volatile oil and chloroform extracts showed the significant antibacterial activities while the least antibacterial activity was recorded with the methanolic extracts. The higher Minimum Inhibitory Concentration value of the extracts and oils against fungus suggested that the plants may possess less antifungal activity. Phytochemical analysis and thin-layer chromatography profiling revealed the presence of flavonoids and terpenoids in the oil and chloroform extracts, which could explain the antimicrobial activity. The findings suggest that the Cedrus deodara and Pinus roxburghii have antimicrobial properties and they can be used in the treatment of infectious diseases. However, further work is required in order to isolate the active constituents of the plants responsible for the antibacterial activity.

연안 갯벌에서 분리한 Chloroaniline 화합물 분해 미생물의 특징 (Characterization of Chloroanilines-degrading Bacteria Isolated from Seaside Sediment)

  • Chloroanilines are aromatic amines used as intermediate products in the synthesis of herbicides, azo-dyes, and pharmaceuticals. 3,4-dichloroaniline (DCA) is the degradation product of some herbicides (diuron, propanil, and linuron) and of trichlorocarbanilide, a chemical used as an active agent in the cosmetic industry. The compound, however, is considered a potential pollutant due to its toxicity and recalcitrant property to humans and other species. With the increasing necessity for bioremediation, we sought to isolate bacteria that degraded 3,4-DCA. A bacterium capable of growth on 3,4-DCA as the sole carbon source was isolated from seaside sediment using a dilution method with a culture enriched in 3,4-DCA. The isolated strain, YM-7 was identified to be Pseudomonas sp. The isolated strain was also able to degrade other chloroaniline compounds. The isolated strain showed a high level of catechol 2,3-dioxygenase activity on exposure to 3,4-DCA, suggesting that this enzyme is an important factor in 3,4-DCA degradation. The activity toward 4-methylcatechol was 53.1% that of catechol, while the activity toward 3-methylcatechol, 4-chlorocatechol and 4,5-chlorocatechol was 18.1, 33.1, and 6.9%, respectively.

인삼의 적변을 유발하는 세균에 대하여 항균활성을 가지는 방선균 선발 및 동정 (Screening and Identification of Antibacterial Actinomycetes against Bacteria Causing Rusty Root on Ginseng)

  • Rusty root, the browning disease on ginseng, decreases quality and value. Recent studies indicated that endophytic bacteria could be a possible cause of rusty root. Actinomycetes antagonistic to the rusty-root-causing bacteria were isolated from soil. Twenty nine out of 932-isolates of Actinomycetes from soil showed antibacterial activity against Agrobacterium tumefaciens and Pseudomonas veronii an endophytic isolate in ginseng. The strongest antibacterial strain(ATO4O104) was classified based on 16S rDNA sequence. The Actinomycetes strain, ATO4O104, isolated in soil of USA volcano national park was identified as Streptomyces adephospholyticus. To test plant toxicity, radish seeds were sprouted with the culture of S. adephospholyticus and it did not show any harmful effect. The butanol partition out of n-hexane, ethyl acetate, butanol, and water partions showed the highest antibacterial activity.

산업체 작업환경의 실내 공기에서 미생물 오염도 (Microbiologic Pollution of Indoor Air in Industrial Work-Places)

  • This study was investigated to isolate identify the total bacteria and fungi from the indoor air of work-place of the shoes, paint, stainless steel, and plastic industries. The number of bacterial colonies on the nutrient agar plates were calculated by the open petridish method for 30 minutes in indoor air of work-places at the autumn and winter. The isolated bacteria were identified by Gram stain and biochemical test using API Staph and API 20E kits. The isolated fungal colonies were identified by gross appearance of the giant colonies and microscopic examination of their spore and hyphal characteristics on the slide culture method. The minimum inhibitory concentration (MIC) of several antibiotics against isolated bacteria was determined by the microdilution method with Mueller-Hinton broth. The 70-400 colonies in autumn and 54-236 colonies in winter were isolated from the indoor air of work-places of several industry. The isolation rates of Gram positive cocci, Gram positive bacilli, Gram negative bacilli, and Gram negative cocci were 46.3%, 19.8%, 17.3%, and 16.1%, respectively. In Gram positive cocci, the most strains were identified as Aerococcus spp, Micrococcus spp, and Staphylococcus spp. In Gram positive and negative bacilli, and Gram negative cocci were identified as Bacillus spp, Pseudomonas spp, and Neisseria spp, respectively. The frequently isolated fungi were Aspergillus spp, Penicillium spp and Rhizopus spp, respectively. The frequently isolated Aerococcus spp, Micrococcus spp, and Staphylococus spp were highly resistance against ampicillin, erythromycin, methicillin, and tetracycline. These results arouse our attention to microbiologic pollution in the indoor air of work-places of industries.

CMT 양성 유즙에서 유방염 원인균 분리 및 분리균의 항균제 감수성 (Isolation of causative agents from CMT-positive mastitic milk and antimicrobial susceptibility of isolates)

  • This study was carried out to isolate of causative agents from CMT-positive and mean somatic cell count(SCC) $\geq$500,000 cells/ml mastitic milk, and evaluate to antimicrobial susceptibility of isolates in Iksan branch area from January to November, 1996. 1. The CMT-positivity(SCC 500,000 cells/ml) of 610 heads was 36.2% (221), and of 2,373 quarter milks was 16.1% (383). 2. The Gram-positive isolates were 153 strains which was Staphylococcus sp (115), Micrococcus sp (18), Streptococcus sp (10), Listeria monocytogenes (5) and Enterococcus faecalis(5). 3. The Gram-negative isolates were 66 strains including E coli(14), Yersinia sp (13), Shigella sp(8), Enterobacillus sp(8), Cedecea sp(5), Pseudomonas aeruginosa(5), Proteus sp(5), Klebsiella sp(4), Salmonella sp(2), kluyvera ascorbate(1) and Tatumella ptyseos (1). 4. The Gram positive strains of isolates were moderately susceptible to T/s, Cp, Fd, Imp, Aug, Rif, Cft and Va. And the Gram negative strains of Isolates were moderately susceptible to T/s, Cp, Imp, Pi and Ti, In order. 5. Multiple antimicrobial resistant patterns were encountered 62 and 36 from Gram positive and negative isolates, respectively.

Development of Antimicrobial Edible Film Incorporated with Green Tea Extract

  • This study investigated the antimicrobial activity of soy protein isolate (SPI) film containing green tea extract (GTE, 1-4%, w/w) on dental caries-inducing bacterium (Streptococcus mutans), food pathogenic (Staphylococcus aureus, Escherichia coli 0157, Salmonella typhimurium), and spoilage (Pseudomonas aeruginosa) bacteria. The physical and mechanical properties of the SPI film containing GTE were also studied. The SPI film containing GTE (4%, w/w) exhibited good antimicrobial activity against S. mutans and S. aureus. The antimicrobial activity of the SPI film containing GTE increased against S. mutans as the concentration of GTE increased up to 4%(w/w). SPI films containing GTE showed lower tensile strength and elongation, and higher total soluble matter than those of control SPI film. Therefore, GTE can be used as one of antimicrobial agents for anti-dental caries and food packaging films.

길항세균을 이용한 상추 균핵병의 생물학적 방제 (Biological Control of Sclerotinia sclerotiorum in Lettuce Using Antagonistic Bacteria)

  • 상추 균핵병균에 대하여 길항력을 나타내는 세균을 분리하기 위해 2007년부터 2008년에 걸쳐 초봄에 경기도 남양주시 관내 상추 시설재배지에서 균핵병이 발병한 포장의 토양을 수집하였다. 수집한 토양을 희석평판법을 이용하여 총 196개의 세균을 분리하였고, 분리한 세균을 실험실에서 균핵병균(Sclerotinia sclerotiorum)과의 대치배양을 통해 균사생장억제율이 80% 내외로 우수한 길항력을 나타내는 26개 균주를 선발하였다. 선발된 26개 균주를 16S rDNA염기서열분석으로 동정한 결과 Bacillus megaterium, B. cereus, B. muralis, B. velezensis, Arthrobacter nicotianae, A. oryzae, Pseudomonas fuscovaginae, P. flavescens, Stenotrophomonas maltophilia, Sphingobacterium faecium으로 동정되었다. 길항세균의 균사생장억제율이 높게 나타낸 10개 균주를 대상으로 여름철 포트실험을 통해 균핵 생존에 미치는 영향을 조사한 결과 B. cereus(C210)와 B. megaterium(DK6)이 각각 20%와 35%의 균핵 생존율을 나타냈다. 겨울철에 온실 내에서 포트실험을 통해 상추 균핵병에 대해 방제효과를 조사한 결과 대조구가 80%의 발병율을 나타냈으나, 26개 균주 중 9개 균주가 20% 정도의 발병율을 나타내 향후 상추 균핵병의 생물적 방제를 위한 길항세균으로서의 활용가치가 기대된다.

항MRSA 활성을 보이는 Paenibacillus incheonensis YK5의 분리 및 특성 (Isolation and Characterization of a Paenibacillus incheonensis YK5 with Antimicrobial Activity aginst MRSA)

  • 국내 토양으로부터 MRSA에 대해 항균활성을 보이는 세균들을 분리하였다. 이들 가운데 가장 높은 항MRSA 균 활성을 보인 YK5 균주는 호기성 간균으로 운동성이 있고, 내생포자를 형성하였으며, 인돌을 형성하지 않았고, mannitol을 분해하여 산을 생성시키지 않았다. 이 균주의 16S rRNA 유전자 염기서열은 Paenibacillus 속 세균들의 유전자와 $95{\sim}98%$의 상동성을 나타냈는데 특히 P. elgii와 가장 높은 상동성을 보였으나, 기타 생리 생화학적 특성에서 다른 점들이 관찰되어 P. incheonensis YK5로 명명되었다. P. incheonensis YK5 를 SST 배지에 접종하여 $37^{\circ}C$에서 96 시간 배양한 다음 그 상등액으로 부터 부탄올로 추출된 항균물질은 그람 양성균인 MRSA 20 균주들과 Streptococcus pneumoniae, 그람 음성균인 Pseudomonas aeruginosa 10균주들, Salmonella spp., Shigella spp., Escherichia coli와 Klebsiella pneumoniae 및 인체병원성 진균인 Cryptococcus neoformans와 Trichophyton spp.에 대하여 광범위한 항균효과를 나타냈다. 또한 이 항균물질은 $20{\sim}100^{\circ}C$ 사이의 온도구간에서 안정한 활성을 유지하였을 뿐만 아니라, pH $3.0{\sim}7.0$의 넓은 pH범위에서 활성을 나타내 MRSA를 비롯한 다양한 병원균에 대한 항생물질로서의 가능성을 보였다.
