임상검체에서 Pseudomonas spp.의 분리빈도와 항균제 감수성 (Isolation Frequency and Antimicrobial Susceptibility of Pseudomonas Species from Clinical Materials)

  • From the total 121,294 clinical materials submitted to the Department of Laboratory Medicine of "C" hospital from December 1, 2004 to November 30, 2006, 3,408 Pseudomonas spp. were isolated. The isolation frequencies of Pseudomonas spp. were as follows, P. aeruginosa 95.5%, P. putida 2.5%, P. fluorescens 0.8%, along with low frequencies of P. luteola, P. alcaligenes, P. stutzeri, P. oryzihabitants, P. mendocina and unidentified Pseudomonas species. The isolation rates of Pseudomonas spp. according to season and sex were evenly distributed. The isolated frequency of Pseudomonas spp. in male was two times higher than that of in female showing significantly more male patients in surgical areas and more female patients in internal areas (p<0.001). In monthly analysis, Pseudomonas spp. were the most frequently isolated in July (10.4%), but lowest in February (5.6%). Half of Pseudomonas spp. were isolated from sputum (48.2%). In the susceptibility analysis of Pseudomonas spp. by VITEK II AST cards, the Pseudomonas spp showing higher susceptibility against antimicrobial agents were piperacillin/tazobactam (82.7%) in P. aeruginosa; amikacin (84.7%), colistin (83.3%) in P. putida; and amikacin (96.3%), cefepime (87.5%), ceftazidime (87.5%) ciprofloxacin (92.3%), colistin (88.5%) gentamicin (96.2%), isepamicin (96,1%), meropenem (92.3%), netilmicin (96.0%), piperacillin/ tazobactam (95.4%) and tobramycin (92.6%) in P. fluorescens.

Exo-inulinase 생산 균주의 분리ㆍ동정 및 효소 생산의 최적화 (Isolation and identification of Exo-Inulinase Producing Bacterium and Optimization of the Enzyme Production)

  • A bacterium producing exo-inulinase was isolated from soil and identified Pseudomonas sp. and named as Pseudomonas sp. NO5. The optimal culture conditions for the efficient production of exo-inulinase from Pseudomonas sp. NO5 were obtained by cultivating with the medium 1$\%$ sucrose, 0.5$\%$ yeast extract, 0.5$\%$ $(NH_4)_2$$HPO_4$, 0.05$\%$ $MgSO_4$$7H_2$0, 0.001$\%$ and $FeSO_4$$7H_2$0 at $37^{\circ}C$ in initial pH 7.0 for 20 hours. The enzyme was induced maximally in the presence of sucrose or inulin at early stationary phase about 20 hour after cultivation.

아닐린 생분해 균주의 분리 및 특성 (Isolation and Characterization of Aniline Degrading Bacteria)

  • 한강의 지천으로부터 채취한 시료에서 29종의 아닐린 분해 균주를 분리선발하였다. 분리 균주중 10, 균주에 대하여 동정 시험하여 본 결과 9균주는 Pseudomonas 속이었고 한 균주는 Acinetobacter 속이었다. 전체 29균주중 plasmid를 갖고 있는 균주는 모두 5종이었으며 이들 5균주중 4균주는 meta 경로를 통하여 아닐린을 분해 이용하였고 나머지 한 균주는 ortho 경로를 통하여 아닐린을 분해하였다. meta 경로 이용 균주의 하나인 Pseudomonas acidovorans 4A1은 230kb 크기의 plasmid를 갖고 있었으며, 접합실험과 curing 실험을 통하여 이 plasmid에 아닐린 분해 유전 정보가 수록되었을 가능성을 확인할 수 있었다.

  • Four bacterial strains able to degrade dichlorobenzene as the sole source of carbon and energy were isolated from soil by selective enrichment culture, and among them, one isolation was the best in the cell growth and identified as Pseudomonas sp. DCB3 by its morphology and physiological properties. Cell growth dramatically increased in a minimal medium containing 500ppm of dichlorobenzene was not detected any more at 72 hours after cultivation. The optimal temperature and initial pH for the growth of the isolated strain were 30$\circ$C and 7.0, respectively. Cell growth was increased by supplementing $(NH_2)_2CO$ and 50 ppm yeast extract as additional nutrients. Therefore, it was suggested that Pseudomonas sp. DCB3 could be effectively used for the biological treatment of wastewater containing dichlorobenzene.

Isolation of a Pseudomonas sp. Capable of Utilizing 4-Nonylphenol in the Presence of Phenol

  • Enrichment techniques led to the isolation of a Pseudomonas sp. strain P2 from municipal waste-contaminated soil sample, which could utilize different isomers of a commercial mixture of 4-nonylphenol when grown in the presence of phenol. The isolate was identified as Pseudomonas sp., based on the morphological, nutritional, and biochemical characteristics and 16S rDNA sequence analysis. The ${\beta}$-ketoadipate pathway was found to be involved in the degradation of phenol by Pseudomonas sp. strain P2. Gas chromatography-mass spectrometric analysis of the culture media indicated degradation of various major isomers of 4-nonylphenol in the range of 29-50%. However, the selected ion monitoring mode of analysis of biodegraded products of 4-nonylphenol indicated the absence of any aromatic compounds other than those of the isomers of 4-nonylphenol. Moreover, Pseudomonas sp. strain P2 was incapable of utilizing various alkanes individually as sole carbon source, whereas the degradation of 4-nonylphenol was observed only when the test organism was induced with phenol, suggesting that the degradation of 4-nonylphenol was possibly initiated from the phenolic moiety of the molecule, but not from the alkyl side-chain.

  • 경기도 일대의 가금류 공장부근 토양로부터 단백질 분해활성이 높은 균주를 분리한 후 이것들 가운데 Keratinolytic protease 생산성이 우수한 균주는 KP-364를 선별하여 효소생산을 위한 최적 배양조건을 검토하였다. 선별된 KP-364 는 형태학적 생화학적 특성을 종합한 결과 Pseudomonas sp 로 동정되었으며 편의상 Pseudomonas sp. KP-364로 명명하였다. 효소생산을 위한 최적 배지조성은 Keratin 0.1% glucose 2.0% soybeam meal 0.5% NaNO3 0.5% KCI 0.2%이었고 초기 pH 6.5 배양온도 $37^{\circ}C$, 48시간진탕 배양할때 효소생산이 가장 우수하였으며 이때의 효소의 역가는 1,270 U/ml/hr이었다.

Ethylene Glycol 분해균주의 순수분리.동전 및 배양특성 (Pure Isolation, Identification and Culture Characteristics of Ethylene Glycol Degrading Bacteria)

  • A large quantity of ethylene glycol(EG) is remained in the effluent after pretreating polyester weight-loss wastewater physicochemically in the fist stage and must be treated biologically in the second stage. Therefore, an excellent EG-utilizing bacteria strain was isolated from the natural system and the optimal culture conditions of the strain were investigated. The optimal culture conditions of temperature, pH, and nitrogen source were found to be 35$^{\circ}C$, 7.5 and ammonium chloride, respectively, when CODCr removal efficiency was more than 90%. The growth of stains and EG removal efficiency was slightly improved by adding elements such as niacin and biotin. With increasing inoculation size in a batch culture, the removal efficiency of EG was conspicuously increased. Growth rate was inhibited when the initial concentration of EG was more then 30g/L. The strain was identified as Pseudomonas sp. based on morphological and biological characteristics and named as Pseudomonas sp. EG1.

한국환자유래의 S형 Pyocin 생성균주 Pseudomonas aeruginosa 90-2-2205의 분리 및 Pyocin 생산 (Isolation of Pseudomonas aeruginosa 90-2-2205 Producing the S-type Pyocin from Korean Patients and the Pyocin Production)

  • The s-type pyocin, one kind of bacteriocins, is a bactericidal substabce of protein nature produced by certain Pseudomonas aeruginosa and is active against some other strains of the same or closely related species. Among many Pseudomonas aeruginosa strains collected from the patients at the Hospitals in Seoul and Taegu cities, some Pseudomonas aeruginosa pyocinogeny were determined by pyocin typing. As a result, Pseudomonas aeruginosa 90-2-2205 was selected as the S-type pyocin producing microorganism due to its highest antimicrobial and protease sensitivity.

Biphenyl 분해 미생물 Pseudomonas sp. DS-94의 분리 및 동정 (Isolation and Identification of a Biphenyl-degrading Bacterium, Pseudomonas sp. DS-94)

  • Three biphenyl-degrading microorganisms were isolated from polluted soil samples in Sasang-gu, Busan. Among them, isolate DS-94 showing the strong degrading activity was selected. The morphological, physiological, and biochemical characteristics of DS-94 were investigated by API 20NE and other tests. This bacterium was identified as the genus Pseudomonas by 16S rDNA sequencing and designated as Pseudomonas sp. DS-94. The optimum temperature and pH for the growth of Pseudomonas sp. DS-94 were $25^{\circ}C$ and pH 7.0, respectively. This isolate could utilize biphenyl as sole source of carbon and energy. Biphenyl-degrading efficiency of this isolate was measured by HPLC analysis. As a result of biological biphenyl-degradation at high biphenyl concentration (500 mg/L), biphenyl-removal efficiency by this isolate was 73.5% for 7 days.
