The Experimental Study of Efficacy in Using Fel Ursi, Bovis Calculus & Moschus Pharmacopuncture Solution as Eye Drop (점안용 웅담.우황.사향약침액의 유효성 평가)

  • Objectives : This experimental study was performed to investigate the efficacy of Fel Ursi, Bovis Calculus & Moschus pharmacopuncture solution manufactured with freezing dryness method to use eye drop. Methods : After administering Fel Ursi, Bovis Calculus & Moschus pharmacopuncture solution on bacterial species(Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum, Candida albicans) which cause Keratitis, MIC(Minimum Inhibition Concentration) and the size of inhibition zone were measured. Anti-bacterial potency was also measured using the size of inhibition zone. Results : There was no response to MIC on bacterial species(Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum, Candida albicans) after Fel Ursi, Bovis Calculus & Moschus pharmacopuncture solution was medicated. Conclusions : The present study suggests that Fel Ursi, Bovis Calculus & Moschus pharmacopuncture solution dosen't have anti-bacterial effects on bacterial species(Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum, Candida albicans) which cause Keratitis. These study result recommends that more research on other herbal medicines of eye drop for Keratitis are required.

Antibacterial Activity of Poncirus trifoliata Juice against Pathogenic Bacteria (병원성 세균에 대한 탱자즙의 항균효과)

  • For development of functional food, antibacterial effect of Poncirus trifoliata juice was examined. Strong antibacterial activities of Poncirus trifoliata juice were observed aginst Gram positive and negative pathogenic bacteria such as Baillus cereus, Bacillus subtilis, Corynebacterium zerosis, Listeria monocytogenes, Micrococcus luteus, Rhodococcus equi, Klebsiella pneumoniae, Pseudomonas aeruginosa, Vibrio alginolyticus, Vibrio cholerae, Vibrio parahaemolyticus, Vibrio vnlnificus and Yersinia enterocolitica. The minimum inhibitory concentration(MIC) of Poncirus trifoliata juice against Bacillus cereus. Listeria monocytogenes, Micrococcus luteus, Rhodococcus equi, Staphylococcus epidermidis, Citrobacter freundil and Pseudomonas aeruginosa was 2.5% and the MIC against Vibrio cholerae, Vibrio parahemolyticus, Vibrio vulnificus and Yersinia enterocolitica was 1.25%. Also, antibacterial activities of Poncirus trifoliata juice treated for 15 min at 121$^{\circ}C$ were confirmed to be stable.

Biochemical and Drug Susceptibility Test of Pseudomonas aeruginosa Isolated from Diseased Chicken (병계(病鷄)로부터 분리(分離)한 Pseudomonas aeruginosa의 생화학적(生化學的) 성상(性狀)과 항균성(抗菌性) 약제내성(藥劑耐性))

  • Biochemical and antimicrobial susceptibility tests were conducted on 40 strains of Pseudomonas aeruginosa originated from diseased chicken submitted for diagnosis to this Institute during 1978-80. An extensive study of the biochemical properties revealed that the tested strains can be identified with Pseudomonas aeruginosa. Antibiogram showed that all the strains were susceptible to gentamicin, colistin and amikacin but resist to nitrofurantoin, trimethoprim+sulfamethoxazole, ampicillin, methicillin and kanamycin, and had varing degreed of resistance to other antimicrobials including carbenicillin, sulfomamides, neomycin, streptomycin, tetracycline and chloramphenicol. Three of the most frequent resitance patterns observed were FM SXT AM ME KM CM TC SM NE SSS Pattern, FM SXT AM ME KM CM TC SM NE SSS CB Pattern and FM SXT AM EKM CM TC SM Pattern, and these resistance patterns contained 72.5% of the tested strains.

Serobiological Characteristics and Antibiotic Sensitivity of Pseudomonas spp. Isolated from Spring Waters in Seoul Area (서울근교 약수터에서 분리한 Pseudomonas속균의 혈청생화학적 특성 및 항균제 감수성)

  • In order to investigate the species, serobiological characteristics and antibiotic sensitivity of Pseudomonas spp, we isolated Pseudomonas spp from 57 spring waters around Seoul area for spring, summer and autumn and identified Pseudomonas spp by biochemical characteristics and serological method. And also we tested the antibiotic sensitivity test by discdiffusion method. Of 57 spring waters tested, Pseudomonas spp were isolated from 33 spring waters(57.9%). Isolation rate of Pseudomonas spp in spring season was 28.1%, summer 21.1% and autumn 28.1%. Only 1 spring water was detected Pseudomonas spp in all seasons and 9 (15.8%) were detected for 2 seasons and 13 (22.8%) were for only 1 season. Isolation rate of Pseudomonas spp at Mt. Cheonggye was 50% and followed by Mt. Bookhan 35.7%, Mt. Daemo 33.3%, Mt. Dobong 29.6%, Mt. Surak 25.9%, Mt. Woomyun 22.2% and Mt. Bulam 7.4%. Of 44 Pseudomonas spp, 22 strains (50%) were identified by Ps. putida, Ps. aeruginosa, Ps. fluorescens and Ps. mendocina were identified 6 strains (13.6%), respectively. 4 strains (9.1%) were identified by Ps. aureofaciens. Of 6 Ps. aeruginosa, serotype A was 2 strains, B, E, G, and K was 1 strain, respectively. Of 44 Pseudomonas spp, resistance rate to amoxicillin was 90.9% and followed by chloramphenicol 84.1%, tetracycline 84.1%, carbenicillin 81.8%, nalidixic acid 68.2%, neomycin 38.6%, streptomycin 31.8%, gentamicin 4.6%, kanamycin 4.6% and colistin 2.3%. Ps. aeruginosa was more sensitive to carbenicillin than other Pseudomonas spp isolated from spring waters in Seoul area but more resistant to kanamycin, and Ps. aureofaciens was no resistant to streptomycin. Among multiple drug resistance, resistance to 5 drugs was 31.8%, 4 drugs 15.9%, 7 drugs 13.6%, 1 drug and 2 drugs 4.6%, and 8 drugs 2.3%, respectively. The multiple resistance patterns detected highestly were NA-CB-C-TE-AMC (18.2%), NA-CB-N-C-TE-AMC (13.6%), CBC-TE-AMC (11.4%) and NA-CB-N-C-TE-AMC-S (9.1%).

Analysis of Pseudomonas aeruginosa Motility in Microchannels (미세유로 내에서 Pseudomonas aeruginosa의 유영 운동 분석)

  • This study presents the effects of micro-geometries on the swimming behavior of Pseudomonas aeruginosa. First, we have measured parameters of single-cell motility including cell speed, run duration time, and tumble angle under two dimensional space. The results are used to calculate motility coefficients in the width of microchannels ranging from 10 to $100{\mu}m$. Since the single-cell motility parameters measured depend on the interaction of flagella with the microchannel wall, the duration time of the running cell in restricted geometries is distinctively different. Therefore, the motility of bacteria is decreased by restricted geometries. This study suggests that microfluidic approach is useful tool for the analysis of bacterial motility under the restricted space and rapid analytical tool.

  • Objectives : This experimental study was performed to investigate the safety and efficacy of Bovis Calculus pharmacopuncture solution manufactured with freezing dryness method to use eye drop. To identify the use of it as eye drop, the eye irritation test of rabbits and the antibacterial test of Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum, and Candida albicans were performed. Methods : 1. The eye irritation test of this material was performed according to the Regulation of Korea Food & Drug Administration(2005. 10. 21, KFDA 2005-60). After Bovis Calculus pharmacopuncture solution was administered in the left eye of the rabbits, eye irritation of the cornea, iris and conjunctiva was observed at 1, 2, 3, 4 & 7day. 2. After administering Bovis Calculus pharmacopuncture solution on bacterial species(Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum, Candida albicans) which cause Keratitis, MIC(Minimum Inhibition Concentration) and the size of inhibition zone were measured. Anti-bacterial potency was also measured using the size of inhibition zone. Results : 1. After Bovis Calculus pharmacopuncture solution was administered in the left eye of the rabbits, it was found that none of nine rabbits have abnormal signs and weight changes. 2. After Bovis Calculus pharmacopuncture solution was medicated in the left eye of the rabbits, no eye irritation of the cornea, iris and conjunctiva was observed at 1, 2, 3, 4 & 7day. 3. There was no response to MIC on bacterial species(Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum, Candida albicans) after Bovis Calculus pharmacopuncture solution was medicated. Conclusions : The present study suggests that Bovis Calculus pharmacopuncture solution is a nontoxic and non-irritant medicine, which does not cause eye irritation in rabbits, but dosen't have antibacterial effects on bacterial species(Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum, Candida albicans) which cause Keratitis. These study result recommends that more research on other herbal medicines of eye drop for Keratitis are required.

A Case Report of a Stroke Patient Treated with Korean Medicine Diagnosed with a Catheter-associated Urinary Tract Infection Caused by Multidrug-resistant Pseudomonas Aeruginosa (뇌졸중 환자에게 유치도뇨관 삽입 이후 발생한 다제내성 녹농균 요로감염 한방치험 1례)

  • This case study reports on the effect of Korean medicine on a catheter-associated urinary tract infection (CAUTI) caused by multidrug-resistant Pseudomonas aeruginosa. An 83-year-old man diagnosed with stroke had dysuria, and it was found that an indwelling urinary catheter led to CAUTI. From laboratory tests, we identified multidrug-resistant Pseudomonas aeruginosa and applied Korean medicine to him. After herbal medication with acupuncture and moxibustion, we studied a urinalysis and urine culture again for follow-up. We found meaningful improvement in bacteriuria and bacterial identification. This case suggests that Korean medicine could have a beneficial effect on urinary tract infections caused by multidrug-resistant Pseudomonas aeruginosa.

Isolation and Characterization of Oil Degrading Bacteria from Southern Sea of Korea (남해안 해수로부터 원유 분해 세균의 분리 및 특성)

  • A marine bacterium having a high oil-degrading activity was isolated form the oil-polluted southern sea of Korea, and was identified as Pseudomonas aeruginosa and was named Pseudomonas aeruginosa BYK-2. The optimal tmeperatur, culture time, pH and NaCl concentration for biosurfactant production and cell growth showed $25^{\circ}C$, 48h, 7.0 and 0%(w/v), respectively. After cultivation at $25^{\circ}C$, 180 rpm in 250 mL erlenmeyer flask for 7days, 1%(w/v) arabian light crude oil and bunker C oil which are considered to be hardly degradable compounds were degraded 92.1%(w/w) and 76%(w/w) respectively. And then, cell adherence was measured on various carbon sources. The cell adherence indicated over 80% on hydrocarbons(arabian light crude oil, kuwait curde oil, bunker C oil, n-paraffine, n-hexadecane, n-tetradecane) as carbon sources. Lecithin among fatty acids(oleic acid, olive oil, lecithin) showed highest cell adherence of 91.5%. The cell adherence of sugars(arabinose, trehalose, dextrose, galactose, lactose, fructose, maltose, sorbitol, sucrose) observed to be less than 70% except for arabinose, galactose, sorbitol and sucrose.

Comparison on prevalence and antimicrobial resistance of Pseudomonas spp. isolated from bovine mastitis milk in South Korea (젖소 유방염에서 분리한 Pseudomonas spp.의 분포 및 항생제 내성 비교)

  • This study was aimed to investigate the prevalence and antimicrobial resistance of Pseudomonas spp. isolated from bovine mastitis milk samples. A total of 50 (4.9%) Pseudomonas spp. was isolated from 1,023 samples, those collected between 2018 and 2021, derived from 110 dairy farms. The prevalence of the identified species of Pseudomonas isolates was as follows; P. aeruginosa (70.0%), P. fluorescens (14.0%), P. putida (10.0%), P. fragi (4.0%), and P. chlororaphis (2.0%). Most of somatic cell counts in the quarter milk carrying Pseudomonas spp. were less than 3,000,000 cell/ml (90.0%). The isolates of Pseudomonas spp. showed high susceptibility to cefepime (98.0%), ciprofloxacin (98.0%), ceftazidime (96.0%), and colistin (96.0%). The rate of antibiotic resistance in the isolates was highest to ceftiofur (92.0%), followed by the resistance rate to chloramphenicol (86.0%) and trimethoprim/sulphamethoxazole (80.0%). In addition, there is a remarkable difference in antimicrobial resistance pattern among Pseudomonas species. P. aeruginosa and P. putida showed a similar resistance pattern, whereas P. fluorescens showed exceptionally lower resistance to trimethoprim/sulphamethoxazole and chloramphenicol than that of the other species. This study showed that prevalence of Pseudomonas spp. other than P. aeruginosa were 30.0% in bovine mastitis milk, and the occurrence rate of antibiotic resistance were similar or higher level, compared with the previous reports on the mastitisderived Pseudomonas spp. isolated in Korea.

Protection of Rabbits from Experimental Pseudomonas Endophthalmitis by Human Anti-P. aeruginosa Outer Membrane Proteins IgG

  • In order to develop an effective means to treat P. aeruginosa infections, we have purified P. aeruginosa outer membrane proteins (OMPs)-specific human IgG antibody. In this study, we investigated the protective activity of the purified anti-OMPs IgC against P. aeruginosa infection in a rabbit endophthalmitis model. Rabbits were inoculated by an intravitreal injection with P. aeruginosa, and treated with a single dose of 1 mg anti-P. aeruginosa OMPs IgG. All the control rabbits predominantly developed edematous responses and opacity in the eyes, but the rabbits treated with the antibody showed only very limited degree of edema. Aliquots of the vitreous humor were extracted and analyzed for the number of viable bacteria and endotoxin level. The results showed that the anti-OMPs IgC significantly reduced the bacterial count compared with the control group, and that the endotoxin level of the vitreous from the IgG-treated rabbits was more than 70-fold lower 6 h after the administration than the control animals. These data suggested that the anti-P. aeruginosa OMPs IgG is effective in inhibiting the bacterial growth and thereby in reducing endotoxin levels in the vitreous, warranting further development of the anti-P. aeruginosa OMPs IgG as a therapeutic means for treating Pseudomonas endophthalmitis.
