In vitro Anti-fungal Activity of Various Hydroxylated Fatty Acids Bioconverted by Pseudomonas aeruginosa PR3

  • The in vitro anti-fungal activity of hydroxylated fatty acids obtained from microbial conversion by Psuedomonas aeruginosa PR3 using ricinoleic acid(RA), eicosadienoic acid(EDA) and conjugated linoleic acid(CLA) as substrates, was investigated. Bioconverted hydroxylated fatty acids showed different anti-fungal activities potentials against the range of phytopathogenic fungi such as Botrytis cinerea, Rhizoctonia solani, Fusarium oxysporum, Sclerotonia sclerotiorum, Colletotricum capsici, Fusarium solani and Phytophthora capsici. RA and EDA showed up to 50% fungal mycelial inhibition at the concentration of $5{\mu}l\;ml^{-1}$. RA, EDA and CLA also exhibited anti-fungal activities with minimum inhibitory concentration(MIC), ranging from 500 to $1000{\mu}g\;ml^{-1}$. Screening was also carried out using varied concentrations of bioconverted RA and EDA for determining the anti-fungal effect on the spore germination of different fungi. Bioconverted RA and EDA showed a considerable degree of spore germination inhibition.

TopoisomeraseII and Topoisomerase IV Gene Mutations Fluoroquinolone Resistance of Pseudomonas aeruginosa

  • The Pseudomonas aeruginosa isolated from the clinical specimens has a mutation on the QRDR (quinolone resistance determining region). There were obvious mutations in both gyrA and parC gene which are major targets of quinolone. Simultaneous mutations were found two sites or more on these genes in all of ten strains. GyrB or parE gene had only silent mutation without converted amino acids. We confirmed that P. aeruginosa from clinical specimens exhibited decreased sensitivity to fluroquiolone due to changed Thr-83→lle and Asp-87→Asn types on gyrA and altered Ser-87→Leu type on parC. This is the first finding that a new Met-93→Thr type on parC as well as mutations on gyrB or parE genes differed from existing patterns. This study showed more mutations of gyrA rather than parC, suggesting that change of Type Ⅳ topoisomerase is more serious than that of type Ⅱ (DNA gyrase).

임상검체로부터 분리한 녹농균의 생물학적 및 혈청학적 특성 (Biological and serological characteristics of Pseudomonas aeruginosa from clinical specimens)

  • One hundred eight strains of Pseudomonas aeruginosa isolated from the patients (sputum, urine, burn skin, stool and blood) of Pusan National University hospital were tested for exonezyme production, antimicrobial susceptibility and serotyping. The results obtained were as follow: In exonezyme production test, 50 strains (46.30%) produced both protease and elastase. Thirty three strains (30.55%) did not produce any exoenzyme, 18 strains (16.67%) produced only protease and 7(6.48%) stains only produced elastase. As the result of antimicrobial susceptibility by the disc diffusion method, most strains were resistant to sulfamethoxazole (96.30%). But the resistant rate against gentamicin and ticarcillin were 47.23% and 46.30% respectively. The resistant rate to other antibiotics were less than 40%. All strains could be serologically typed. Most strains were identified as type Ⅲ: among them, 51 strains were belonged to serotype E. The correlation of serotype and exoenzyme production was not found.

  • The water extracts of oriental herbal medicines which have been clinically used to treat bacterial infections in Korea were screened for in vitro antibacterial activity by the paper disc assay method. Two Gram positive bacteria, Staphylococcus aureus SG511, Bacillus subtilis ATCC 6633 and two Gram negative bacteria, Escherichia coli 055, Pseudomonas aeruginosa 9027 were used as test organisms. Among 83 of the extracts tested, 25 were active against Staphylococcus aureus SG511, 9 were active against Bacillus subtilis ATCC 6633, while none showed inhibitory activity against Eschelichia coli 055 and Pseudomonas aeruginosa 9027. Among them, Hwangyonhaedoktang plus hwangyon, Chongwisan, and Ssangbaksan showed remarkably potent antibacterial activity.

아연 또는 비소와 경유로 오염된 토양의 복합정화공법 개발 (Development of Hybrid Remediation Method for Contaminated Soils with Zinc or Arsenic and Diesel)

  • The purpose of this study was to develope the remediation method of contaminated soils with metals and petroleum. The diesel degrading strain was isolated and identified from the soil contaminated by petroleum at industrial sites. Diesel biodegradation experiment was performed by diesel degrading bacteria in both solution and soil slurry. Contaminated soils by Zn or As and diesel were treated consecutively by steam-vapor extraction, biodegradation, and acid washing. The strain was identified as Pseudomonas aeruginosa, and named as Pseudomonas aeruginosa TPH1. The optimal culture conditions of TPH1 were $20^{\circ}C$ and pH 7.0, 3% of diesel concentration. Biodegradation of diesel was performed using the separated strain in liquid medium, and 63% of diesel was degraded in 72 hours. And 52% of diesel was removed in the tested soils. In the treatment of contaminated soils with diesel and Zn or As, 29% ~ 44% of diesel was reduced by steamvapor extraction, 60% ~ 71% of diesel was removed after biodegradation. 47% of Zn and 96% of As were removed after acid(mixture of sulfuric and oxalic acids) washing. It is recommended that consecutive treatment method of steam-vapor extraction, biodegradation and acid washing is effective for remediation of complex contaminated soils with metals and petroleum.

  • Selection of high producer strain, optimization of production medium and cultivation in bioreactor system were carried out in order to produce an antifungal substance, PAFS in large amounts which sources and 41 kinds of nitrogen sources, a synthetic medium consisting of fructose(70 g/1) and ammonium sulfate (10g/l) and a complex medium including galactose(30g/l), fructose(20g/l) and cottonseed flour(35g/l) were determined as opti-mized media for PAFS production. In bioreactor studies examining physiological characteristics of the pro- ducer microorganism with the complex medium, typical pattern of diauxic growth was observed as demonstrated by the result that fructose was not used before almost exhaustion on readily utilizable carbon source, galactose. When galactose was supplemented additionally during the fermentation period. PAFS pro-ductivity did no increases any more, indicating that large portion of the added galactose was used for cell growth instead of biosynthesis of the secondary metabolite. It was deduced that PAFS production could be enhananced by employing fed-batch operation in order to overcome the apparent phenomenon of catabolite repression and /or inhibition.

  • The antibiotic-producing Streptomyces strains were isolated from the soils of various localities in Yeang-Kwang and tested for the activity against some of gram positive and negative microorganisms, especially against Pseudomonas aeruginosa. The results are summarized as follows. 1. A total of 1464 Streptomyces strains were isolated from 336 soil samples. Of all the strains tested, 636 strains of the isolates showed antibiotic activity against either gram positive or negative test organisms. And 408 of the 636 strains were strongly active against one or all of those test­organisms. 2. Ninety-three of Streptomyces strains among the active isolates showed the antibiotic activity against Pseudomonas aeruginosa, and 24 of 93 strains showed the strong antibiotic activity against all of the test organisms i.e. Staphylococcus aureus, Bacillus subtilis, Escherichia coli, Salmonella typhi and Shigella jlexneri. 3. Antibiotic-producing Streptomyces strains active against Pseudomonas aeruginosa were isolated from the soil samples taken from vegetable field, paddy field, sandy ground and roadway, but not isolated from the soil taken from dwelling area, grass field and piers.

Recombinant Azurin from Pseudomonas aeruginosa Induces Apoptotic Cell Death in Oral Squamous Carcinoma Cells

  • The use of bacteria in the treatment of cancer has a long and interesting history. The use of live bacteria in this way however has a number of potential problems including toxicity. Purified low molecular weight bacterial proteins have therefore been tested as anticancer agents to avoid such complications. Oral cancer is a widely occurring disease around the world and these lesions are typically very resistant to anticancer agents. In our present study we investigated the effects of purified recombinant azurin from Pseudomonas (P.) aeruginosa against YD-9 (p53-positive) human oral squamous carcinoma cells. Azurin showed cytotoxic effects against these cells in a dose dependent manner. The cell death accompanied by this treatment was found to be characterized by chromatin condensation and apoptotic bodies. Azurin treatment was further found to increase the expression of p53 The stabilization of p53 and induction of apoptosis in YD-9 cells by azurin suggests that it has potentially very strong anticancer properties in oral squamous carcinoma.

녹농균과 진균류의 생물학적 상호작용에 의한 Aflatoxin 류의 생성능과 성질 변화에 대한 연구 (Studies on Changes of Aflatoxin Productivity and Properties by a Pseudomonad)

  • Investigation on modification of aflatoxin structures by Pseudomonads was attempted as a biological detoxifiying process of mycotoxins. Firstly, when any variation of aflatoxin yield of Aspergillus parasiticus in a mixed culture with Pseudomonas aeruginosa was examined, there was no noticible effect by growth of Pseudomonads on aflatoxin yield of Asperillus sp. Secondly, when capacity of Pseudomonas aeruginosa utilizing aflatoxin as a carbon source for its growth was tested, there was some indication that aflatoxin might be used for growth of Pseudomonads. It was also noticed that the residual aflatoxin showed different migrating pattern compared with that of the intact aflatoxin by thin layer chromatography. Thirdly, the cell-free extract prepared from Pseudomonas aeruginosa grown in a glucose minimal medium supplemented with aflatoxin and the intact aflatoxins were incubated in the presence of $Mg^{++}$. After a certain length of incubation period, the reaction mixtures were applied on TLC plates. As a result, aflatoxins in the reaction mixture migrated differently as the control did. Such results may indicate that some changes of aflatoxin properties were induced by Pseudomonads.

Fungal-Sporulation Suppressing Substances Produced by Pseudomonas aeruginosa KMCS-1

  • Among the bacteria isolated from compost piles of cattle excretion in a pasture located at the suburbs of Chunchon city, Pseudomonas aeruginosa KMCS-1 was selected for the test of antifungal substances produced. Six fractions were separated by silica gel column chromatography, and then the antifungal activity of each fraction was assayed against Escherichia coli, Bacillus subtilis, Candida albicans, Rhizopus sp., Aspergillus nidulans, Coprinus cinereus, and Pyricularia oryzae by paper disc method. Two fractions showed significant suppressive activities against A. nidulans, C. cinereus, and P. oryzae however, their mycelial growth was not affected by neither of these fractions. Inhibitory activities of these fractions to sporulation was assayed at the concentration of 50. 25, 12. 5, and 6.25 $\mu$g/ml and the average inhibition rates against sporulation of A. nidulans, C. cinereus, and P. oryzae were 94.0, 98.3, and 77.9%, respectively. Further purification and analysis of active substances are now being conducted.
