화피 추출물 첨가 설기떡의 항산화활성 및 품질특성 (Antioxidant Activity and Quality Characteristics of Sulgidduk prepared with Prunus yedoensis Matsumura Extract)

  • This study investigated the quality and antioxidative properties of a Korean steamed-rice cake, Sulgidduk added with Prunus yedoensis Matsumura extract, and vitamin C. Sulgidduk was prepared by adding Prunus yedoensis Matsumura extract at 0, 0.1, 0.2, and 0.3% of rice powder. Antioxidant activities were measured by the scavenging activities of DPPH radicals, $ABTS^+$ radicals, $superoxide^-$ radicals and the reducing power. For analyzing quality characteristics, proximate composition, color, texture profile analysis, and sensory evaluations were measured. The antioxidative effect of the Sulgidduk significantly increased as the addition level increased, compared to the original Sulgidduk (p<0.001). As the content of Prunus yedoensis Matsumura extracts increased, L-values significantly decreased while the a-value and b-value significantly increased (p<0.001). For the texture profile analysis, the control group had a significantly higher value for hardness as compared to the Prunus yedoensis Matsumura extract-added groups (p<0.05). Springiness, chewiness and adhesiveness were not significantly different among the samples. In the sensory evaluation, the samples containing 0.1% and 0.2% Prunus yedoensis Matsumura extracts obtained better results in attribute. From these results, we suggest that Prunus yedoensis Matsumura is a good ingredient for increasing the consumer acceptability and the functionality of Sulgidduk.

  • The origin of cultivated Prunus yedoensis Matsumura has been controvertial among botanists. This plant has been reported grown wild in Mt. Hanla. During the crossing experiment on the P. yedoensis and its putative parent performed from April 7 to 19 in Mt. Hanla, the writer studied morphological and physiological characteristics of various species of Prunus and assumed that the wild P. yedoensis may be a hybrid between P. subhirtella var. pendula form. ascendens and P. donarium.

약용식물 추출물의 항산화 효과 (Antioxidant Activities of Medicinal Plant Extracts)

  • 본 연구에서는 전통적으로 질병치료와 예방의 목적으로 사용되어온 약용식물 10종을 선정하여 총 페놀 및 총 플라보노이드 함량과 항산화 효과를 측정하였다. 약용식물의 총 페놀 함량을 측정한 결과 화피 추출물이 320.10 mg/g, 갈근이 153.93 mg/g으로 높게 나타났으며, 총 플라보노이드 함량은 화피 추출물이 226.25 mg/g, 황금 추출물이 101.98 mg/g으로 높았다. 약용식물 중 가장 높은 DPPH 라디칼 소거능을 나타낸 시료는 화피로 $IC_{50}$ $5.39{\mu}g/mL$를 나타냈으며, 그 다음으로 헛개나무($IC_{50}$ $24.87{\mu}g/mL$), 오가피($IC_{50}$ $27.79{\mu}g/mL$) 순으로 높게 나타났다. 아질산 소거 활성 측정결과 헛개나무가 $IC_{50}$ $173.96{\mu}g/mL$, 화피 추출물이 $IC_{50}$ $167.94{\mu}g/mL$로 비슷한 활성을 나타내었다. $Superoxide^-$ 라디칼 소거능과 $ABTS^+$ 라디칼 소거능에서는 화피와 오가피가 유의적으로(p<0.001) 높은 소거능을 보였다. 환원력에 대한 흡광도 값을 측정한 결과는 10종의 약용식물 중에서 화피 추출물이 2.72의 흡광도를 나타내어, 양성대조군인 비타민C의 3.04과 비교해도 크게 뒤떨어지지 않음을 알 수 있었다. 본 연구의 결과 화피, 헛개나무, 오가피 추출물은 총 페놀 및 플라보노이드 함량이 높고, 항산화 활성도 높게 나타나 식품첨가물 및 식품보존제 등 기능성 소재로써 활용될 수 있을 것으로 사료된다.

  • Horizontal starch gel electrophoresis in three buffers was used to compare the electrophoretic patterns in three cherry species, wild Prunus subhirtella, cultivated P. yedoensis and P. donarium. Electrophoretic patterns of glutamate oxaloacetate, transaminase-2(GOT-2), malate dehydrogenase-2(MDH-2), and phosphoglucose isomerase(PGI) in three species showed strong evidence that P. yedoensis might be originated by hybridization between P. subhirtella and P. donarium.

화피 추출물이 첨가된 개성주악의 품질특성 및 항산화 활성 (Quality Characteristics and Antioxidant Activity of Gaeseong-Juak prepared with Prunus yedoensis Matsumura Extract)

  • This study investigated the quality and antioxidative properties of a Korean fried rice cake, Gaeseong-Juak (doughnut of waxy rice) added with Prunus yedoensis Matsumura (PYM) extract, vitamin C and BHT. Gaeseong-Juak was prepared by adding PYM extract at 0, 0.5, 1, and 2% of waxy rice powder. The moisture contents of Gaeseong-juak added with PYM extract varied between 17.50% and 21.70%. Among the samples, the lowest moisture content was 2% Gaeseong-juak added with PYM extract. The contents of the samples obtained from crude protein, crude fat, crude ash were 4.47~4.73, 11.15~12.50, and 0.73~0.82%, respectively. The as the content of PYM extract increased, L-values significantly decreased while a-value and b-value significantly increased (p<0.001). The texture profile analysis of the Gaeseong-Juak was not significantly different at the samples. In the sensory test, the sample containing 0.5% PYM extract obtained good scores. The antioxidative effect of the Gaeseong-Juak significantly increased as the addition level increased, compared to the original Gaeseong-Juak (p<0.001). From these results, we suggest that PYM is a good ingredient for increasing the consumer acceptability and the functionality of Gaeseong-Juak.

Organogenesis from Callus Derived from In Vitro Root Tissues of Wild Prunus yedoensis Matsumura

  • In vitro organogenesis system of the valuable ornamental species, Prunus yedoensis which is native to Korea, was established through callus culture derived from root tissues. Callus were induced on the medium supplemented with 2,4-D and BA or NAA and kinetin. Organogenesis was differ from the callus type, and NAA and kinetin combination was effective to induce organogenic callus. Growth of callus was influenced by sucrose concentrations. High level of sucrose (over 5%) had adverse effects such as decreased fresh weight and increased mortality of callus. Shoots developed from the callus when $GA_3$ was treated with BA in the medium. Results showed that $GA_3$ is essential for shoot development and elongation from callus in this species.

왕벚나무 껍질의 에탄올 추출물과 용매 분획물의 항산화활성 비교 (Comparison of Prunus yedoensis Matsumura bark ethanol extract and solvent fraction antioxidant activities)

  • 본 연구에서는 왕벚나무 껍질 에탄올 추출물과 분획물의 항산화물질 및 항산화활성을 측정하였다. 왕벚나무 껍질 에탄올 추출물 및 분획물의 총 페놀 함량은 ethyl acetate 분획물이 496.80 mg GAE/g으로 가장 높았고, 총 플라보노이드 함량은 7.26-265.52 mg RE/g으로 ethyl acetate 분획물이 가장 높게 나타났다. 프로안토시아니딘 함량은 ethyl acetate, n-buthanol, ethanol, dichloromethane, water, n-hexane 순으로 높았다. DPPH, ABTS+, superoxide- 라디칼 소거능 및 환원력은 ethyl acetate 분획물의 활성이 가장 좋았고, 아질산염 소거능에서는 에탄올 추출물이 가장 높은 활성을 나타냈다. 반면에 n-hexane과 water 분획물은 항산화물질의 함량이 낮고 항산화활성이 다른 시료에 비해 낮게 측정되었다. 따라서 왕벚나무 껍질 ethyl acetate 분획물은 항산화물질이 풍부하고 높은 항산화활성을 나타내어 천연 항산화제로 활용될 수 있을 것으로 기대된다.

왕벚나무 꽃잎 추출물에 대한 견직물의 염색성과 색채특성 (Dyeing Properties and Color Characteristics of Silk Fabrics Dyed with Prunus yedoensis Matsumura Flower Extract)

  • This study was carried out to identify the optimum dyeing conditions for maximum dye uptake of Prunus yedoensis Matsumura flower extract on silk fabric and to investigate hue/tone characteristics and fastness properties of the dyed fabrics. As results, the flower extract was found to contain flavonoids by FT-IR and to show yellowish shade by UV-vis spectrum. Silk fabric was optimized for maximum dye uptake to five repetitions of dyeing at $80^{\circ}C$ for 60 min with an extract concentration of 600% owf in terms of both K/S and color differences. Pre-mordanting was more effective to dye uptake than post-mordanting for all of the mordants used. Un-mordanted and pre-mordanted fabric showed YR(Yellow Red) while most of post-mordanted ones did Y(Yellow). Main tones of dyed fabrics were sf(soft) and lt(light) in the case of unmordanting or Al-mordanting whereas they were d(dull) and g(grayish) by post-mordanting with Cu, Cr, and Fe, respectively. The fastness grades to rub and dry cleaning were reasonably good for most dyeing conditions while those to light were poor. Finally dyeing condition for each hue/tone of the dyed silk with the flower extract was optimized considering fastness properties.

왕벚나무 화외밀선의 당액 분비에 관한 미세구조적 연구 (Ultrasturctural Study on Nectar Secretion from Extrafloral Nectary of Prunus yedoensis Matsumura)

  • 한국산 왕벚나무(Prunus yedoensis Matsumura) 화외밀선의 미세구조 및 당액 분비기작을 규명하기 위하여 광학현미경, 주사형 및 투과형 전자현미경을 이용하여 관찰하였다. 밀선은 secretory layer와 subsecretory layer로 구성되어 있었으며 엽병의 유관속은 subsecretory layer에 있는 유관속과 연결되어 있었다. Secretory cell에서는 cristae가 잘 발달되지 않은 다수의 mitochonidria가 관찰되었으나 소포체는 소수 나타났고 골지체가 전혀 관찰되지 않았다. 색소체는 thylakoid가 거의 발달되지 않았으며, 소수가 관찰되었고 $0.2-0.3\;\mu\textrm{m}$ 크기의 소포를 특징적으로 갖고 있었다. subsecretory cell에서는 원형질 연락사가 잘 발달해 있었고 다수의 Calcium oxalate 결정이 관찰되었는데 이는 당투과에 간접적으로 관여하는 것 같다. 당액 전구물질은 사부에서 subsecretory layer를 거쳐 secretory layer까지 이동하는 것을 원형질 연락사를 통한 symplastic transport에 의하여 일어나는 것으로 생각되며 secretory layer에서 당액으로 재합성되어 외부로 배출되는 것 같다. Secertory cell의 맨 바깥쪽 세포벽은 cuticle 에 의하여 덮여 있었으며 secretory cell에서 당액은 원형질막, 세포벽, cuticle을 차례로 통과하는 누출상 분비(eccrine secretion)에 의하여 분비되는 것으로 사료된다.
