Mechanism of Growth Inhibition in Herbicide-Resistant Transgenic Rice Overexpressing Protoporphyrinogen Oxidase (Protox) Gene (Protoporphyrinogen Oxidase (Protox) 유전자 과다발현 제초제 저항성 형질전환 벼의 생육저해 기작)

  • We investigated the levels of resistance and accumulation of terapyrroles, reactive oxygen species, lipid peroxidation, and antioxidative enzymes for reasons of growth reduction in herbicide-transgenic rice overexpressing Myxococcus xanthus, Arabidopsis thaliana, and human protoporphyrinogen oxidase (Protox) genes. The transgenic rice overexpressing M. xanthus (MX, MX1, PX), A. thaliana (AP31, AP36, AP37), and human (H45, H48, H49) Protox genes showed 43~65, 41~72 and 17~70-fold more resistance to oxyfluorfen, respectively, than the wild type. Among transgenic rice lines overexpressing Protox genes, several lines showed normal growth compared with the wild type, but several lines showed in reduction of plant height and shoot fresh weight under different light conditions. However, reduction of plant height of AP37 was much higher than other lines for the experimental period. On the other hand, the reduction of plant height and shoot fresh weight in the transgenic rice was higher in high light condition than in low light condition. Enhanced levels of Proto IX were observed in transgenic lines AP31, AP37, and H48 at 7 days after seeding (DAS) and transgenic lines PX, AP37, and H48 at 14 DAS relative to wild type. There were no differences in Mg-Proto IX of transgenic lines except for H41 and H48 and Mg-Proto IX monomethyl ester of transgenic lines except for MX, MX1, and PX. Although accumulation of tetrapyrrole intermediates was observed in transgenic lines, their tetrapyrrole accumulation levels were not enough to inhibit growth of transgenic rice. There were no differences in reactive oxygen species, MDA, ALA synthesizing capacity, and chlorophyll between transgenic lines and wild type indicating that accumulated tetrapyrrole intermediate were apparently not high enough to inhibit growth of transgenic rice. Therefore, the growth reduction in certain transgenic lines may not be caused by a single factor such as Proto IX, but by interaction of many other factors.

Classification According to Site of Action of Paddy Herbicides Registered in Korea (국내 수도용 제초제의 작용기작별 분류)

  • This review study was conducted to recommend the effective use of herbicide mixtures in Korea. The herbicide ingredients by Herbicide Resistancce Action Committee (HRAC) was classified into 23 groupes according to the mode of action (acetyl CoA carboxylase inhibitors, acetolactate synthase, photosystem I and II inhibitors, protoporphyrinogen oxidase inhibitors, carotenoid biosynthesis inhibitors, enolpyruvyl shikimate-3-phosphate synthase inhibitors, glutamine synthetase inhibitors, dihydropteroate synthetase inhibitors, mitosis inhibitors, cellulose inhibitors, oxidative phosphorylation uncouplers, fatty acid and lipid biosynthesis inhibitors, synthetic auxins, auxin transport inhibitors and potential nucleic acid inhibitors or non-descript mode of action). The rice herbicide mixtures registered in Korea were classified based on the guideline of HRAC. Accordingly, such a classification system for resistance management can help to avoid continuous use of the herbicide having the same mode of action in the same field.

Phytotoxic Effect of 5-Aminolevulinic Acid, a Biodegradable Photodynamic Biomaterial, on Rice and Barnyardgrass

  • ALA (5-aminolevulinic acid) has been proposed as a tetrapyrrole-dependent photodynamic herbicide by the action of the protoporphyrinogen IX oxidase (Protox IX). A study was conducted to determine photodynamic herbicidal effect of ALA on seedling growth of rice (Oryza sativa L.) and barnyard grass (Echinochloa crus-galli Beauv. var. oryzicola Ohwi) under dry and wet conditions. ALA effect on early plant growth of rice and barnyardgrass was greatly concentration dependant, suggesting that it promotes plant growth at very low concentration and inhibits at high concentration. No significant difference in herbicidal activity of biologically and synthetically produced ALAs on plant lengths of test plants was observed ALA exhibited significant photodynamic activity regardless of PSDIP and its duration. Significant shoot growth inhibition by ALA soaking treatment exhibited apparently, indicating that ALA absorbed through root system was translocated into shoot part of plants. ALA reduced plant heights of rice and barnyardgrass seedlings by 6% and 27%, respectively, showing more tolerant to ALA in rice under wet condition. Leaf thickness was reduced markedly by ALA with increasing of ALA concentration, due to mainly membrane destruction and severe loss of turgidity in mesophyll cells, although the epidermal was little affected. It was observed that photodynamic herbicidal activity of ALA applied by pre-and post-emergence application exhibited differently on plant species, and that the activity of ALA against susceptible plants was highly correlated with growing condition.

Herbicidal Activity and Molecular Similarity of 1-(4-chloro-2-fluoro-5-propargyloxyphenyl)-3-thiourea Derivatives (1-(4-chloro-2-fluoro-5-propargyloxyphenyl)-3-thiourea 유도체의 제초활성과 분자 유사성)

  • In the search for third generation herbicidal cyclic imide derivatives, the average values of herbicidal activity ($pI_{50}$) in vivo (pre-emergence) of 40 new peroxidizing herbicides, 1-(4-chloro-2-fluoro-5-propargyloxyphenyl)thiourea derivatives (1-40) against rice plant (Orysa sativa) and barnyard grass (Echinochlor crus-galli) were studied. The molecular similarity between protoporphyrinogen IX (protogen) as the substrate of protox enzyme and Urea derivatives (1-40) was discussed quantitatively. The diallyl (20) and 3-nitro substituent (33) showed the selective herbicidal activity against barnyard grass. Allyl substituent (8) and their molecular similarity in dice (S=0.81) showed the highest levels of herbicidal activity ($pI_{50}$=4.71). Also, similarity indices (S) and superimposed volume (C) of protogen and aryl-substituents (21-40) showed good correlation.

Selection and Validation of an Analytical Method for Trifludimoxazin in Agricultural Products with LC-MS/MS (LC-MS/MS를 이용한 농산물 중 Trifludimoxazin의 시험법 선정 및 검증)

  • Trifludimoxazin is a triazinone herbicide that inhibits the synthesis of protoporphyrinogen oxidase (PPO). The lack of PPO damages the cell membranes, leading to plant cell death. An official analytical method for the safety management of trifludimoxazin is necessary because it is a newly registered herbicide in Korea. Therefore, this study aimed to develop a residual analysis method to detect trifludimoxazin in five representative agricultural products. The EN method was established as the final extraction method by comparing the recovery test and matrix effect with those of the QuEChERS method. Various sorbent agents were used to establish the clean-up method, and no differences were observed among them. MgSO4 and PSA were selected as the final clean-up conditions. We used LC-MS/MS considering the selectivity and sensitivity of the target pesticide and analyzed the samples in the MRM mode. The recovery test results using the established analysis method and inter-laboratory validation showed a valid range of 73.5-100.7%, with a relative standard deviation and coefficient of variation less than 12.6% and 14.5%, respectively. Therefore, the presence of trifludimoxazin can be analyzed using a modified QuEChERS method, which is widely available in Korea to ensure the safety of residual insecticides.

Spectrophotometric microtitre assay for rapid screening of membrane-disrupting herbicides (분광분석을 이용한 막과산화작용 제초제의 신속한 검정법)

  • This study was conducted to establish an efficient screening method of membrane-disrupting herbicides, based on the result that membrane leakage leading to evolution of carbonyl compounds and aldehydes could be colorimetrically assessed by formation of stable chromophores with MBTH and ferric chloride. Assay conditions including plant material, light intensity, irradiation time, concentrations of reagents for colorization, and reaction time were optimized, and activities of different types of known herbicides were investigated with cucumber cotyledon discs. That result indicated that this assay was sensitive to membrane-disrupting herbicides, such as protoporphyrinogen oxidase inhibitors (acifluorfen, oxyfluorfen, oxadiazon) and an electron interceptor from photosystem I(paraquat). With seven compounds newly synthesized, highly positive correlation was observed between herbicidal activities measured by greenhouse bioassay and MBTH method. Because only a single disc(4 mm diameter) from cucumber cotyledon in 60 ${\mu}L$ of incubation solution was required to assay, a large number of herbicides could be tested simultaneously using 96-well microtitre plates. But the assay efficiency is not likely to significantly increase because of the boiling step during the reaction.

Research Trends in Hormetic Stimulation Effects of Herbicides in Plants (식물에서 제초제의 양면성 촉진반응 연구동향)

  • Hormesis is a dose-response phenomenon that is characterized by low-dose stimulation and high-dose inhibition. This biphasic dose-responses have had a long and extensive history in the fields of chemical toxicology, radiation biology and pharmacology. Hormesis has been found from bacteria, fungi, plants and animals, but hormesis in plants has received relatively little attention. Thus principles, occurrence, factors affecting the expression of hormetic responses, and their mechanisms in plants induced by herbicides are reviewed to provide the potentials for crop enhancement. Bromacil, bromoxynil, chloramben, propachlor, terbacil, EPTC, MSMA, and glyphosate at low doses showed stimulatory response in growth. Subtoxic dose of glyphosate increased sucrose content in sugarcane that is used worldwide in sugarcane production. Low dose of protoporphyrinogen-inhibiting herbicides induced increased pathogen defence, and low dose of triazine herbicides improved nitrogen metabolism and increased protein content in some crops. Further researches on potential benefits and risks of hormesis and its mechanism are needed for application of crop enhancement in agriculture.

Weed Control and Safety of Transgenic Rice Event, CPPO06 in Direct-Seeding Flooded Rice Field (담수직파에서 형질전환 CPPO06이벤트 벼의 안전성 및 잡초방제효과)

  • This study was conducted to evaluate the efficacy of weed control and phytotoxicity of oxyfluorfen using a transgenic rice line (CPPO06 event) resistant to protoporphyrinogen oxidase (Protox) inhibiting herbicides in the direct-seeding flooded rice. Five annual weeds including Echinochloa oryzoides and two perennial weeds were occurred in the test field. Oxyfluorfen at 120 g a.i. $ha^{-1}$ in the application timing of two days before sowing and 0 and five days after sowing showed more than 90% weed control value except for Juglans mandshurica. Total weed control value was more than 95% in any application timing indicating a highly effective herbicide in the direct-seeding flooded rice. When compared with untreated control, no visual injuries were detected at single and double dosage of oxyfluorfen. The agronomic characteristics and yield components of CPPO06 event was reasonable in any time of application in this study. Based on these data, oxyfluorfen application before and after sowing can be applied to provide effective weed management in the direct-seeding flooded field.

Quantitative structure-activity relationships and molecular shape similarity of the herbicidal N-substituted phenyl-3,4-dimethylmaleimide Derivatives (제초성 N-치환 phenyl-3,4-dimethylmaleimide 유도체의 정량적인 구조-활성관계와 분자 유사성)

  • To improve the growth inhibitory activity against the shoot and root of rice plant (Oryza sativa L) and barnyard grass (Echinochloa crus-galli), a series of N-substituted phenyl-3,4-dimethylmaleimdes derivatives as substrates were synthesized and then their the inhibitory activities of protoporphyrinogen oxidase (1.3.3.4), protox were measured. The quantitative structure-activity relationships (QSAR) between structures and the inhibitory activities were studied quantitatively using the 2D-QSAR method. And also, molecular sharp similarity between the substrate derivatives and protogen, substrare of protox enzyme were studied. The activities of the two plants indicated that barnyard grass had a higher activity than the rice plant and their correlation relationships have shown in proportion for each. Accordingly, the results of SARs suggest that the electron donating groups as $R_2=Sub.X$ group will bind to phenyl ring because the bigger surface area of negative charged atoms in the substrate molecule derivatives may increase to the higher the activity against barnyard grass. Based on the molecular shape similarity, when the derivatives and protogen, subsbrate of protox enzyme were superimposed by atom fitting, the similarity indices (S) were above 0.8 level but the correlation coefficients (r) between S values and the activities showed not good.
