Improvement of Cellobiose Dehydrogenase(CDH) and $\beta$-Glucosidase Activity by Phanerochaete chrysosporium Mutant (Phanerochaete chrysosporium 변이주에서의 Cellobiose Dehydrogenase(CDH)와 $\beta$-Glucosidase 활성 향상)

  • Cellobiose dehydrogenase(CDH) as a hemoflavoenzyme is secreted out of cell in the cellulose degradation. As CDH strongly bound to amorphous cellulose, it helps cellulose hydrolysis by cellulase. CDH may have an important role of saccharification process for bioethanol production. In this study, Phanerochaete chrysosporium ATCC 32629 was selected for the production of CDH among other strains tested. The optimal temperature and pH of CDH produced by P. chrysosporium ATCC 32629 were ${55^{\circ}C}$ and 4, respectively. To improve the activity of CDH, the mutation of P. chrysosporium was performed using proton beam that has high energy level partially. As a result, P. chrysosporium mutant with the high activity was selected at 1.2 kGy in a range of 99.9% lethal rate. The CDH and $\beta$-glucosidase activities of mutant were 1.4 fold and 20 fold higher than those of wild strain. Therefore, P. chrysosporium mutant with the high activities of CDH and $\beta$-glucosidase was obtained from mutation by proton beam irradiation.

  • The design of a vacuum pumping system for the KOMAC (Korea Multipurpose Accelerator Complex) RFQ(Radio-Frequency Quadrupole) linac is described. [Fig] Resulted from the lost proton beam, gas streaming from the LEBT (Low Energy Beam Transport) and out-gassing from the surfaces of the RFQ cavity and vacuum plumbing, the total gas load will be on the order of 7.2$\times$10-4 Torr-liters/sec, consisting mainly of hydrogen. The system designed to pump on a continual basis with redundancy to ensure that the minimal operating vacuum level of 1.0$\times$10-6 Torr is maintained even under abnormal conditions. Details of the design, performance analysis and the preliminary test results of the cryogenic pumps are presented.

  • 사이클로트론에서 가속된 70 MeV 양성자 빔은 stripper에서 인출된 후 ISOL 표적에 조사하게 된다. 이 때 희귀동위원소를 생산하기 위한 ISOL 표적의 온도는 평균 $2,000^{\circ}C$ 5%로 유지하여야 한다. 사용하고자 하는 원판형태의 우라늄 표적은 직경이 45 mm 이므로 50 mm 이상의 균일한 양성자 빔이 필요하다. 본 연구에서는 stripper에서부터 ISOL 표적 까지 빔전송에 필요한 최적 조건을 연구하였다. 총 길이는 15 m 이며 두 개의 dipole을 사용하여 빔을 90도 전환시키며 3개의 4중극전자석을 사용하였다. ISOL 표적에 균일한 빔을 조사하기 위해 wobbling 방법을 활용하여 직경 50 mm 의 균일빔을 발생하고자 한다. 빔 전송 프로그램 TRANSPORT을 이용하여 stripper에서 wobbler 전까지의 빔 광학을 계산한 결과를 보고자 하고 TRANSPORT의 결과를 TURTLE 프로그램에 적용시켜 ISOL 표적에서의 균일도를 5% 미만으로 유지하는 최적조건을 발표한다.

A Synthesis of Iron Oxide Based and Gadolinium Oxide Based Radiosensitizer for the Therapeutic Enhancement of Proton Beam Cancer (양성자 빔 암치료효과 개선을 위한 산화철 및 산화가돌리늄 나노입자 기반의 방사선증감제 합성)

  • Metallic nanoparticles have attractive properties in biomedical applications such as diagnostics and therapeutics. Cross linked dextran coated iron oxide nanoparticles (SPIONs) and silica coated gadolinium oxide nanoparticles (SPGONs) have been synthesized as a radiosensitizer in the proton beam cancer therapy. The dextran and silicaused for the protective moieties on the SPIONs and SPGONs respectively. Size distributions of synthesized nanoparticles were confirmed 3~5 nm for SPIONs and 30~100 nm for SPGONs by transmission electron microscope (TEM). Cell survival fraction measurement and Western blot assay were performed to evaluate the radiosensitization effects of synthesized radiosensitizer. The calculated radiosensitization of SPIONs and SPGONs at 90 % cell death from the measured cell survival curves were 1.23 and 1.03 respectively. Western blotting results also show the same consistent results that the amount of released cytochrome c from mitochondria was considerably increased for the cancer cells taken up SPIONs.

  • In the International Code of Practice for dosimetry TRS-398 published by International Atomic Energy Agency(IAEA), water equivalency plastic phantom may be used under certain circumstances for electron beam dosimetry for beam quality E0${\leq}$ 10 MeV. In this study, Palstic Water$^{TM}$ and Virtual Water$^{TM}$ were evaluated in order to determine fluence scaling factor hpl. Plastic phantom was evaluated for five electron energy from 6 MeV to 20 MeV. From the measured data of Palstic Water$^{TM}$, the fluence scaling factor hpl was found to be average 0.9964 and Virtual Water$^{TM}$ fluence scaling factor was 1.0156.

  • Radioactive isotopes which are manufactured using a cyclotron in a radioisotope used for radiation diagnosis is affected by the production yield according to size and shape of the beam and beam uniform degree from irradiated location when the proton beam investigated the target by cyclotron. Therefore, in this paper developed the BPM(Beam Profile Monitor) device capable of measuring the beam cross-section at the cyclotron beam line. It was configured so as to be able to remote control the BPM device in LabView and used the BPM program it was to be able to easily monitor and display to analyze the graph of two-dimensional graph and a three-dimensional beam distribution numerical information of the beam obtained while scanning the tungsten wire to the X and Y axis. The time it takes to measure the beam can be confirmed 37seconds when step motor driving speed was 2000pps. Through a beam readjusted based on the measured beam distribution information by optimizing the beam distribution it can be made to maximize the RI production yield and contribute supply stabilization.
