  • This paper makes a general description of the results at government project which were peformed for several years. Through this project, the real capacity of 3.3KV/4MW Class 3 Level Voltage Source Inverter System were designed and the characteristics of its proto type were analyzed, moreover the web based IIMS(Inverter Information Management System) and Virtual Operation Simulator was developed. Now, this system is running for field application test.

The Existence of a Putative Regulatory Element in 3'-Untranslated Region of Proto-oncogene HOX11's mRNA

  • HOX11 encodes a homeodomain-containing transcription factor which directs the development of the spleen during embryogenesis. While HOX11 expression is normally silenced through an unknown mechanism in all tissues by adulthood, the deregulation of HOX11 expression is associated with leukemia, such as T-cell acute lymphoblastic leukemia. The elucidation of regulatory elements contributing to the molecular mechanism underlying the regulation of HOX11 gene expression is of great importance. Previous reports of HOX11 regulatory elements mainly focused on the 5'-flanking region of HOX11 on the chromosome related to transcriptional control. To expand the search of putative cis-elements involved in HOX11 regulation at the post-transcriptional level, we analyzed HOX11 mRNA 3'-untranslated region (3'UTR) and found an AU-rich region. To characterize this AU-rich region, in vitro analysis of HOX11 mRNA 3'UTR was performed with human RNA-binding protein HuR, which interacts with AU-rich element (ARE) existing in the 3'UTR of many growth factors' and cytokines' mRNAs. Our results showed that the HOX11 mRNA 3'UTR can specifically bind with human HuR protein in vitro. This specific binding could be competed effectively by typical ARE containing RNA. After the deletion of the AU-rich region present in the HOX11 mRNA 3'UTR, the interaction of HOX11 mRNA 3'UTR with HuR protein was abolished. These findings suggest that HOX11 mRNA 3'UTR contains cis-acting element which shares similarity in the action pattern with RE-HuR interactions and may involve in the post-transcriptional regulation of the HOX11 gene.

  • In this thesis, we presents the component design and retrieval prototype system which can retrieve the most appropriate component for reuse system. If it used only an abject to reuse unit, it can not understand mutual reaction and can not sustain relationship between objects. Therefore, we porposed clustering component for reuse unit. It makes to understand relationship and reaction between object, so User can retrieve the proper component for reuse system. And porposed retrieval prototype system can select the correct things. it provide 3 facet retrieval environment against ambiguous retrieval definition.

  • In this paper, a novel optical measuring system for the measurement of nonuniform electric field was proposed. The electric field distorted by the discharges was detected through proposed optical measuring system based on the Pockets effect and Mach-Zehnder interferometer. In order to produce distorted electric field, corona discharge was generated from needle-plane electrode in air and detected by optical measuring system. This optical measuring system is constructed by He-Ne laser, single mode optical fiber, $2{\times}2$ 50/50 beam splitter, $LiNbO_3$ Pockets cell, photo detector and PC. In this system, output signal of Pockels sensor is measured by digital oscilloscope and transferred to the PC for recording and statistical processing. Through this paper, a promising possibilities of proto-type optical measuring system were evinced.

Optimization of Shoot Induction, Histological Study and Genetic Stability of in vitro Cultured Pisum sativum cv. 'Sparkle'

  • An efficient shoot regeneration condition for pea cv. 'Sparkle' was developed by using optimum explant, plant growth regulator concentrations, and pretreatment of BA onto explant. The average shoot number per explant showed the highest on two kinds of shoot induction media (MSB5 media containing 2 mg/L BA and a combination of 2 mg/L BA and 1 mg/L TDZ) when cotyledonary node explants were cultured. Moreover, the pretreatment of explant in 200 mg/L BA solution was found to be more effective in shoot induction than that of non-pretreatment. By histological study, cell division and proto-meristem were formed near the surface of the sub-epidermal and epidermal cell layers of cotyledonary node in earlier than 3 days after culture. The analysis of genetic stability of regenerants by using thirteen ISSR markers showed that in vitro regenerated plants showed polymorphism with 8.3% compared with their mother plants.

A Multi-Epoch, Simultaneous Water and Methanol Maser Survey Toward Intermediate-Mass Young Stellar Objects

  • We report multi-epoch, simultaneous 22 GHz water and 44 GHz Class I methanol maser line survey towards 180 intermediate-mass young stellar objects, including 14 Class 0, 19 Class I objects, and 147 Herbig Ae/Be stars. We detected $H_2O$ and $CH_3OH$ maser emission towards 16 (9%) and 10 (6%) sources with one new $H_2O$ and six new $CH_3OH$ maser sources. The detection rates of both masers rapidly decrease as the central (proto)stars evolve, which is contrary to the trends in high-mass star-forming regions. This suggests that the excitations of the two masers are closely related to the evolutionary stage of the central (proto)stars and the circumstellar environments. $H_2O$ maser velocities deviate on average 9 km s-1 from the ambient gas velocities whereas $CH_3OH$ maser velocities well match with. For both maser emissions, large velocity difference (${\mid}v_{H2O}-v_{sys}{\mid}\;>\; 10kms^{-1}\; and\;{\mid}v_{CH3OH}-v_{sys}{\mid}\;>\;1kms^{-1}$) is mostly confined to Class 0 objects. The formation and disappearance of $H_2O$ maser lines are frequent and the integrated intensities of them change up to two orders of magnitude. In contrast, $CH_3OH$ maser lines usually show no significant change in the intensity, shape, and velocity. This consistent with the previous suggestion that $H_2O$ maser emission originates from the base of an outflow while 44 GHz Class I $CH_3OH$ maser emission arises from the interaction region of the outflow with the ambient gas. The isotropic maser luminosities are well correlated with the bolometric luminosities of the central the objects. The fitted relations are $L_{H2O}=1.71{\ast}10^{-9}(L_{bol})^{0.97}$ and $L_{CH3OH}=1.71{\ast}10^{-10}(L_{bol})^{1.22}$.

  • The c-myc proto-onco9ene, one of the immediately early genes, is involved in ceflular proliferation and differentiation, and its biologleal function is regulated hy dimerization with a heterodimeric partner, myn. In the present study, gene expression patterns of c-myc and myn during mouse preimplantation embryo development were examined using a semi-quantitative reverse transcription-poiymerase chain reaction (RT-PCR). Myn transcripts were rather constitutively expressed throughout embryonic stages with a slight increase only at biastocyst stage. in contrast, expression of c-myc transcripts wm developmental stage-'pedfic. The c-myc transcripts were detected at 1-cell stage, declined abruptly at 2-cell stage and then increased gradually at blastocyst stage. To examine the possible role of myn during preimpiantation mouse embryo development, two myn antisense oligonucleotides spanning the tail of zipper dognain (myn2; 20-mer) and the second helix domain (myn3; 20-mer) were microinjected into the fertilized 1-cellembryos. Microinjection of myn2 and myn3 resulted in developmental tion at morula/biastocyst transition stage, leading to the fiagentation of embryos. Talien together, these results suggest that c-myc and its heterodimeric partner, myn, are differentially expressed In a developmental stage-dependent manner, and myn may play an important role in mouse preimpiantation embryo development.

FORMATION OF PROTO-GLOBULAR CLUSTER CLOUDS BY THERMAL INSTABILITY

  • Many models of globular cluster formation assume the presence of cold dense clouds in early universe. Here we re-examine the Fall & Rees (1985) model for formation of proto-globular cluster clouds (PGCCs) via thermal instabilities in a protogalactic halo. We first argue, based on the previous study of two-dimensional numerical simulations of thermally unstable clouds in a stratified halo of galaxy clusters by Real et al. (1991), that under the protogalactic environments only nonlinear (${\delta}{\ge}1$) density inhomogeneities can condense into PGCCs without being disrupted by the buoyancy-driven dynamical instabilities. We then carry out numerical simulations of the collapse of overdense douds in one-dimensional spherical geometry, including self-gravity and radiative cooling down to T = $10^4$ K. Since imprinting of Jeans mass at $10^4$ K is essential to this model, here we focus on the cases where external UV background radiation prevents the formation of $H_2$ molecules and so prevent the cloud from cooling below $10^4$ K. The quantitative results from these simulations can be summarized as follows: 1) Perturbations smaller than $M_{min}\~(10^{5.6}\;M{\bigodot})(nh/0.05cm^{-3})^{-2}$ cool isobarically, where nh is the unperturbed halo density, while perturbations larger than $M_{min}\~(10^8\;M{\bigodot})(nh/0.05cm^{-3})^{-2}$ cool isochorically and thermal instabilities do not operate. On the other hand, intermediate size perturbations ($M_{min} < M_{pgcc} < M_{max}$) are compressed supersonically, accompanied by strong accretion shocks. 2) For supersonically collapsing clouds, the density compression factor after they cool to $T_c = 10^4$ K range $10^{2.5} - 10^6$, while the isobaric compression factor is only $10^{2.5}$. 3) Isobarically collapsed clouds ($M < M_{min}$) are too small to be gravitationally bound. For supersonically collapsing clouds, however, the Jeans mass can be reduced to as small as $10^{5.5}\;M_{\bigodot}(nh/0.05cm^{-3})^{-1/2}$ at the maximum compression owing to the increased density compression. 4) The density profile of simulated PGCCs can be approximated by a constant core with a halo of $p{\infty} r^{-2}$ rather than a singular isothermal sphere.

  • The signal transduction pathways through the RAS gene product and adenyl cyclease play a critical role in regulation of the cell cycle in yeast, Saccharomyces cerevisiae. We examined the genetic relationship between the spt3 gene and ras/cAMP pathway. A mutation in the SPT3 gene suppressed cell cycle arrest at the G1 phase caused by either an inactivation of the RAS or CYR1 gene which encodes a yeast homologue of human ras proto-oncogene or adenyl cyclase, respectively. The phenotypes such as sporulation and heat shock resistancy, that resulted from a partial inactivation of the RAS or CYR1 genes, were also suppressed by the spt3 mutation. Expression of the SSA1 gene encoding one of th heat shock proteins (Hsp70) can be induced by heat shock or nitrogen starvation. Expression of this gene is derepressed in cry1-2 and spt3 mutants. The bcy 1 mutation repressed by the bcy1 mutation, but not in spt3 mutants. These results suggest that the SPT gene is involved in expression of genes that are affected by the RAS/cAMP pathway.

Hybrid-Type Active Power Filters for Compensating Harmonic Current and Unbalanced Source Voltages (고조파 전류와 불평형 전원전압 보상을 위한 복합형 능동전력 필터)

  • In this paper, a novel control scheme compensating source voltage unbalance and harmonic currents for the combined system of series active and shunt passive power filter is proposed, where no low/high-pass filters are used in deriving the reference voltage for compensation. The phase angle and the reference voltages compensating for harmonic current and unbalanced voltage are derived from the positive sequence component of the unbalanced voltage set, which is simply obtained by using digital all-pass filters. In order to remove the phase delay in generating the reference voltage for compensation, the reference of 5th and 7th harmonic components is predicted one-sampling ahead. The validity of the proposed scheme has been verified for 3[kVA] proto-type active power filter system.
