Alternanthera mosaic virus (AltMV) is a member of the genus Potexvirus which has been known for less than twenty years, and has been detected in Australasia, Europe, North and South America, and Asia. The natural host range to date includes species in at least twenty-four taxonomically diverse plant families, with species in at least four other families known to be infected experimentally. AltMV has been shown to differ from Potato virus X (PVX), the type member of the genus Potexvirus, in a number of ways, including the subcellular localization of the Triple Gene Block 3 (TGB3) protein and apparent absence of interactions between TGB3 and TGB2. Differences between AltMV variants have allowed identification of viral determinants of pathogenicity, and identification of residues involved in interactions with host proteins. Infectious clones of AltMV differing significantly in symptom severity and efficiency of RNA silencing suppression have been produced, suitable either for high level protein expression (with efficient RNA silencing suppression) or for Virus-Induced Gene Silencing (VIGS; with weaker RNA silencing suppression), demonstrating a range of utility not available with most other plant viral vectors. The difference in silencing suppression efficiency was shown to be due to a single amino acid residue substitution in TGB1, and to differences in subcellular localization of TGB1 to the nucleus and nucleolus. The current state of knowledge of AltMV biology, including host range, strain differentiation, host interactions, and utility as a plant viral vector for both protein expression and VIGS are summarized.

The immobilization of proteins and their molecular interactions on various polymer-modified glass substrates [i.e. 3-aminopropyltriethoxysilane (APTS), 3-glycidoxypropyltrimethoxysilane (GPTS), poly (ethylene glycol) diacrylate (PEG-DA), chitosan (CHI), glutaraldehyde (GA), 3-(trichlorosilyl)propyl methacrylate (TPM), 3'-mercaptopropyltrimethoxysilane (MPTMS), glycidyl methacrylate (GMA) and poly-l-lysine (PL).] for potential applications in a nanoarray protein chip at the single-molecule level was evaluated using prismtype dual-color total internal reflection fluorescence microscopy (dual-color TIRFM). A dual-color TIRF microscope, which contained two individual laser beams and a single high-sensitivity camera, was used for the rapid and simultaneous dual-color detection of the interactions and colocalization of different proteins labeled with different fluorescent dyes such as Alexa Fluor® 488, Qdot® 525 and Alexa Fluor® 633. Most of the polymer-modified glass substrates showed good stability and a relative high signal-to-noise (S/N) ratio over a 40-day period after making the substrates. The GPTS/CHI/GA-modified glass substrate showed a 13.5-56.3% higher relative S/N ratio than the other substrates. 1% Top-Block in 10 mM phosphate buffered saline (pH 7.4) showed a 99.2% increase in the blocking effect of non-specific adsorption. These results show that dual-color TIRFM is a powerful methodology for detecting proteins at the single-molecule level with potential applications in nanoarray chips or nano-biosensors.
As the clinical practice of using more than one drug at a time increase, the clinician is faced with ever-increasing number of potential drug interactions. Although many interactions have little clinical significances, some may interfere with treatment or even be life-threatening. This review provides a better understanding of drug-drug interactions often encountered in pharmacotherapy of depression. Drug interactions can be grouped into two principal subdivisions : pharmacokinetic and pharmacodynamic. These subgroups serve to focus attention on possible sites of interaction as a drug moves from the site of administration and absorption to its site of action. Pharmacokinetic processes are those that include transport to and from the receptor site and consist of absorption, distribution on body tissue, plasma protein binding, metabolism, and excretion. Pharmacodynamic interactions occur at biologically active sites. In this review, emphasis is placed on antidepressant medications, how they are metabolized by the P450 system, and how they alter the metabolism of other drugs. When prescribing antidepressant medications, the clinician must consider the drug-drug interactions that are potentially problematic.

Paramyosin is a myofibrillar protein present in helminth parasites and plays multifunctional roles in host-parasite interactions. In this study, we identified the gene encoding paramyosin of Clonorchis sinensis (CsPmy) and characterized biochemical and immunological properties of its recombinant protein. CsPmy showed a high level of sequence identity with paramyosin from other helminth parasites. Recombinant CsPmy (rCsPmy) expressed in bacteria had an approximate molecular weight of 100 kDa and bound both human collagen and complement 9. The protein was constitutively expressed in various developmental stages of the parasite. Imunofluorescence analysis revealed that CsPmy was mainly localized in the tegument, subtegumental muscles, and the muscle layer surrounding the intestine of the parasite. The rCsPmy showed high levels of positive reactions (74.6%, 56/75) against sera from patients with clonorchiasis. Immunization of experimental rats with rCsPmy evoked high levels of IgG production. These results collectively suggest that CsPmy is a multifunctional protein that not only contributes to the muscle layer structure but also to non-muscular functions in host-parasite interactions. Successful induction of host IgG production also suggests that CsPmy can be applied as a diagnostic antigen and/or vaccine candidate for clonorchiasis.
인터넷 상에 단백질 및 관련 데이터의 축적에 따라, 도메인에 기반하여 단백질의 상호작용을 계산적으로 예측하는 많은 기법들이 제안되었다. 그러나, 대부분의 기법들이 예측에서 낮은 정확도와 복수개의 단백질 쌍에 대한 상호작용 가능성들 간에 순위 정보를 제공하지 못하는 등의 한계로 인하여 실무 적용에 한계를 가지고 있다. 본 논문에서는 도메인 조합 기반 단백질 상호작용 예측 기법을 재평가하고 상호작용하는 것으로 예측되는 복수개의 단백질 쌍들에서 이들의 상호작용 가능성들 간에 순위를 부여하는 방법을 제시한다. 순위 부여 방법은 도메인 조합에 기반한 단백질 상호작용 예측 방법의 틀 내에서 확률 식을 고안하여 제시한다. 제시된 순위 부여 기법을 사용함으로써, 상호작용을 하는 것으로 예측된 단백질 쌍들간에 상호작용 가능성이 좀 더 높은 것을 구별해 낼 수 있다. 또한 순위 부여 기법의 검증 과정에서 학습에 사용된 단백질 집단의 PIP(Primary Interaction Probability)값과 일치된 PIP값을 가지는 단백질 쌍 그룹의 경우에는, 상호작용 확률과 예측 정확도 사이에 상관관계가 존재함을 확인할 수 있었다.
Nitrogenase 촉매에서 Fe-단백질을 포함하는 [4Fe-4S] 클라스터의 기능은 기질의 결합과 환원 자리를 포함하는 MoFe-단백질로 핵산 의존 전자 주개로 작용하는 것이다. 이러한 방법의 Fe-단백질의 기능은 Mofe-단백질과 상호작용을 위해 적합한 구조를 갖추며 전자 전달을 위한 추진력을 제공하기 위해 산화 환원 퍼텐셜을 변화시키는 능력에 의존한다. Nitrogenase Fe-단백질에 MgADP가 결합한 (혹은 떨어진) 구조적 정보는 핵산 결합 자리로부터 MoFe-단백질과의 결합력을 조절하기 위한 장거리 상호작용 메커니즘을 제시한다. 스위치 I과 II의 두 가지 경로가 뉴클레오티드의 신호전달 메커니즘을 담당한다. 계산을 통한 단백질 상호작용 예측 기법의 중요성이 제기되면서 많은 단백질 상호 작용 예측 기법이 제안되고 있다. 하지만 이러한 기법들이 일반 사용자가 손쉽게 사용할 수 있는 서비스 형태로 제공되고 있는 경우는 드물다. 본 논문에서는 현재까지 알려진 단백질 상호작용 예측 기법 중 예측 기법의 완성도가 높고 상대적으로 예측 정확도가 높은 것으로 알려진 도메인 조합 기반 단백질 상호 작용 예측 기법을 이용하여 서비스 시스템으로 설계하고 구현하였다. 효모(Yeast)의 단백질 집합에 대하여 학습한 후, 학습된 단백질 집합과 공통된 도메인을 가지지만 학습 집합에 존재하지 않는 단백질 쌍들에 예측 기법을 적용하여 매우 높은 $77\%$의 민감도(sensitivity)와 $95\%$의 특이도(specificity)를 보였다. 더불어 DIP CORE, HMS-PCI, TAP 데이타의 테스트를 통해서 이 기법의 안정성을 확인하였다. 시스템의 기능들은 핵심 기능, 부가 기능 그리고 일반 서비스 기능으로 분류하였다. 시스템 설계의 주요 목표인 성능, 개방성 그리고 확장성에 따라, 개별 서비스들은 병렬화, 웹 서비스 표준 준수 및 계층화된 구조화를 지원하도록 구현하였다. 본 논문에서는 몇 가지 대표적인 사용자 인터페이스와 상세한 사용 지침도 소개한다.
The binding interactions between human serum albumin (HSA) and the edible food dyes amaranth, tartrazine and sunset yellow have been studied. Intrinsic association constants and the free energy changes associated with dye-protein binding at physiological pH for amaranth and tartrazine, and at two different pH values for sunset yellow have been calculated from ultrafiltration data. The temperature dependence $(20-40^{\circ}C)$ of the intrinsic association constants at pH 7.4 for amaranth-HSA and tartrazine-HSA mixtures have been measured, from which a plot of the van't Hoff isochore exhibits a marked change in slope around $30^{\circ}C$ indicating a possible change in protein conformation. The number of dye binding sites on HSA is reported for all the above conditions. HSA-ligand binding enthalpies have been used in conjunction with the N-B transitional binding enthalpy for HSA, to calculate the enthalpy for the N-B transition when ligands are bound with the protein.
In this paper, we have fabricated on extended-gate field effect transistor (EGFET)-type protein sensor for the application to a CRP detection. We used the self-assembled monolayer (SAM) to adhere or entrap biomolecules, namely CRP antibodies. The experimental result shows that the proposed SAM is well immobilized on the gold gate surface. So the drain current was varied by antigen-antibody interactions on the gate surface because of the CRP charge. Experimental results related to the formation of SAM, antibody, antigen were obtained by measuring the electrical characteristics of the EGFET device.
