PCR과 Southern hybridization을 이용한 구지뽕나무와 무궁화의 클론감별 (Clone Identification of Cudraria Tricuspidata and Hibiscus Syriacus by Using PCR and Southern Hybridization)

  • 본 연구는 polymerase chain reaction (PCR)와 Southern hybridization을 이용하여 서로 붙어있는 나무가 한 나무인지 두 나무인지 규명하였다. 공시된 수종은 구지뽕나무와 무궁화이다. 구지뽕나무는 경상북도 성주군 금수면 봉두리 안새출 금수식당 앞에 있는 나무로 두 사람이 서로 안고 있는 형상인데, 한 나무의 두 가지인지 가까이 자란 두 나무가 붙었는지를 구별하기 어려운 나무였다. 다섯 종류의 PCR primer $(17{\sim}24-mers)$ 중 355 primer의 경우 한가지의 잎 DNA에서만 PCR 산물이 검출되어 이것을 방사선표지한 후 genomic Southern hybridization을 행하였던 바 이 probe와 결합하는 DNA 단편이 동일한 위치에서 검출되었으나 정도는 다르게 나타났다. 아울러 OPERON 10-mer kits A에 있는 20종류의 primer를 이용하여 PCR을 행한 결과 OPA01 primer (CAGGCCCTTC)에 의한 PCR 생성물은 동일한 위치의 band 뿐만 아니라 추가로 4개가 한가지의 잎 DNA에서 더 나타났다. 따라서 구지뽕나무는 두 나무가 연결되어 한 나무를 이루는 것으로 짐작된다. 또한 무궁화는 경상남도 합천군 청덕면 두곡리 청덕초등학교 내에 있는데 수령 50년 이상으로 멀리서 보면 한 나무의 형상을 하고 있으나, 가까이 다가가서 줄기의 아래부분을 보면 세 줄기가 붙어있는 형상을 하고 있다. 따라서 이 무궁화 역시 한 나무의 세 가지인지 세나무가 가까이 자라 붙었는지 PCR과 Southern hybridization 방법을 이용하여 규명하려 하였다. Southern hybridization 결과에 의하면 구지뽕나무의 분석에 사용한 probe와 결합하는 DNA 단편은 검출되지 많았으며, 35S primer에 의한 PCR 생성물은 동일하였으나 OPA04와 OPA13 primer의 경우 약간의 상이성을 보였다. 이러한 결과에 따라 무궁화는 한 나무의 세 가지인 듯하나 추가적인 연구가 필요하다고 판단된다.

옥돔(Branchiostegus japonicus) 비늘 유래 젤라틴의 가수분해 및 가수분해물의 기능성 (Processing and Biological Activity of Gelatin Hydrolysate from Branchiostegus japonicus Scales)

  • The potential utility of fish scales to the functional food industry has been investigated due to its antioxidant and antihypertensive characteristics. In this study, we report on the reactive oxygen species (ROS) scavenging and angiotensin I converting enzyme (ACE) inhibitory activities of gelatin hydrolysates processed from Branchiostegus japonicus scales, which are also high in protein content (about 46.1%). We prepared the enzymatic gelatin hydrolysates with four proteases (${\alpha}$-chymotrypsin, Alcalase, Neutrase and trypsin) from B. japonicus scale gelatin, which was prepared according to different reaction times, substrate/enzyme ratios and substrate concentrations. The enzymatic hydrolytic degrees of the gelatin increased time-dependently up to 6 hrs, while the Alcalase gelatin hydrolysates showed the highest hydrolysis degrees compared to the others. Furthermore, gelatin hydrolysates of Neutrase and ${\alpha}$-chymotrypsin showed the highest DPPH radical and $H_2O_2$ scavenging activities ($IC_{50}$ value; 9.18 mg/mL and 9.74 mg/mL), respectively. However, the activities were not significant (P<0.05). We also observed that the four gelatin hydrolysates significantly increased ACE inhibitory activities from approximately 20% to 60% (P<0.05), Among them, the Alcalase gelatin hydrolysates showed the higher ACE inhibitory activity ($IC_{50}$ value; 0.73 mg/mL) compared to the others. These results suggest that the enzymatic gelatin hydrolysates prepared from B. japonicus scales may possess a potentially useful function as an ACE inhibitory agent. As such, the utility of B. japonicus scales should be given due consideration for application in the functional food industry.

노인 당뇨환자 교육용 기능성 게임 콘텐츠 개발을 위한 예비연구 (A Pilot Study for Development of the Serious Game Contents for Education in the Elderly Diabetes)

  • 본 연구는 노인 당뇨환자를 위한 교육용 기능성 게임 콘텐츠(롤리폴리 160)를 개발하고 적용가능성을 평가하기 위한 예비연구이다. 롤리폴리 160은 문헌고찰, 요구도 조사 및 자문의뢰, 게임을 활용한 기능성 콘텐츠 추출, 롤리폴리 160 개발, 사용자 사전 만족도 조사, 보건교육자 대상 워크샵, 롤리폴리 160 수정, 롤리폴리 160 사용자 만족도 조사와 같은 8단계 연구절차를 걸쳐서 개발되었다. 롤리폴리 160은 노인 당뇨환자가 자가관리를 할 수 있는 3가지 모듈(식사자가관리, 카드게임, 퀴즈게임)로 직관적으로 구성되었으며, 6가지 원칙에 입각하여 설계되었다. 첫째, 사용자의 연령을 고려한 직관적인 인터페이스를 구축하였다. 둘째, 음식은 한국인이 주로 섭취하는 음식위주로 배열하고 음식선택시 칼로리 학습이 동시에 이루어지도록 하였다. 셋째, 사용자 선택한 음식의 칼로리와 영양소(탄수화물, 지방, 단백질, 칼슘, 나트륨)가 분석되고, 문진표에 기록한 모든 검사자료는 년, 월, 주 별로 그래프로 도식화하여 변화추이가 한눈에 파악되도록 설계하였다. 넷째, 필요한 자료는 저장하여 출력하여 교육 자료로 활용 가능하도록 하였다. 다섯째, 사용자의 자료는 호환 및 합산이 가능하며 1억 명까지 회원가입이 가능하도록 하였다. 여섯째, 필요 시 모바일 App으로 개발이 가능하도록 설계하였다. 롤리폴리 160을 활용하여 119명의 노인 당뇨환자에게 당뇨교육을 실시한 후 만족도 점수는 5점 만점 중 4.26점으로 나타났다. 이는 롤리폴리 160이 당뇨환자 자가간호 도구로 적절하며, 향후 보건소나 병 의원에서 당뇨환자를 교육하는 프로그램으로 활용가능성이 있음을 시사해준다.

Effects of aged garlic extract and endurance exercise on skeletal muscle FNDC-5 and circulating irisin in high-fat-diet rat models

  • BACKGROUND/OBJECTIVES: Irisin, a newly identified hormone, is associated with energy homeostasis. We investigated whether aged garlic extract (AGE) and exercise training intervention could improve body weight, insulin sensitivity, skeletal muscle fibronectin domain containing protein 5 (FNDC-5) levels, and plasma irisin in high-fat diet (HFD). MATERIALS/METHODS: Male Sprague Dawley rats were fed a ND (normal diet, n=5) or HFD (n=28) for 6 weeks. After 6 weeks, all rats were divided into 5 groups for the next 4 weeks: ND, (normal diet, n=5), HFD (high-fat diet, n=7), HFDA (high-fat diet + aged garlic extract, n=7), HFDE (high-fat diet + exercise, n=7), and HFDEA (high-fat diet + exercise + aged garlic extract, n=7). Exercise groups performed treadmill exercises for 15-60 min, 5 days/week, and AGE groups received AGE (2.86 g/kg, orally injected) for 4 weeks. RESULTS: Significant decreases in body weight were observed in the ND, HFDE, and HFDEA groups, as compared with the HFD group. Neither intervention affected the masses of the gastrocnemius muscle or liver. There were no significant differences in glucose levels across the groups. The homeostatic model assessments of insulin resistance were significantly higher in the HFD group, as compared with the ND, HFDA, HFDE, and HFDEA groups. However, skeletal muscle FNDC-5 levels and plasma irisin concentrations were unaffected by AGE or exercise in obese rats. AGE supplementation and exercise training did not affect skeletal muscle FNDC-5 or plasma irisin, which are associated with insulin sensitivity in obese rats. CONCLUSION: Our results suggest that the protection against HFD-induced increases in body fat/weight and insulin resistance that are provided by AGE supplementation and exercise training may not be mediated by the regulation of FNDC-5 or irisin.

  • Expression of gonadotropin releasing hormone(GnRH) has been described in the rat ovary. It remains, however, unkown whether GnRH is synthesized as a prohormone. Therefore, this study was performed to verify the expression of pro-GnRH by in situ hybridization and further to investigate the effect of gonadotropin on GnRH or GnRH mRNA in rat ovary by immunohistochemical and in situ hybridization techniques. Adult female Sprague-Dawely rats were used and the estrous cycle was synchronized by intraperitoneal injection of pregnant mare's serum gonadotropin(PMSG). Ovaries were fixed with 4% paraformaldehyde and embedded with G.C.T. compound and cut by cryostat. For immunohistochemistry, avidin-biotin peroxidase complex(ABS) method was employed and for in situ hybridization, $^{35}S$-end labeled oligonucleotide was used and followed by autoradiography. By in situ hybridization using GnRH oligomer and GAP(GnRH associated protein) oligomer, GnRH mRNA and GAP mRNA were co-localized in the fullicular cells, luteal cells, interstitial cells and theca cells. GnRH or GnRH mRNA signals in the ovary increased by human chorionic gonadotropin(hCG) injection. At the 3 and 6 hrs after hCG injection, the number of GnRH and GnRH mRNA containing cells increased rapidly and the density of GnRH and GnRH mRHA culminated at 9 hrs after heG injection. With the follicular development, the high expression of GnRH and GnRH mRNA was also observed within the follicles. After ovulation, the density of GnRH or GnRH mRNA decreased in the follicles but increased in the corpus lutea.

Effects of a Dietary Chitosan-Alginate-Fe(II) Complex on Meat Quality of Pig Longissimus Muscle during Ageing

  • The current study was conducted to investigate the effects of dietary chitosan-alginate-Fe(II) complex (CAFC) supplementation on carcass and meat qualities of pig m. longissimus during chiller ageing. One hundred and twenty-two LYD (Landrace${\times}$Yorkshire${\times}$Duroc) pigs were sampled from an industrial population. Seventy-four pigs (32 gilts and 42 barrows) were administered 3 ml of dietary supplementation of CAFC per day from 25 to 70 days of age, while the remaining 48 pigs (20 gilts and 28 barrows) were fed the same commercial feeding regime without the supplementation. For assessing the dietary effects on pH, objective meat color, cooking loss, water-holding capacity (WHC), thiobarbituric acid reactive substances (TBARS), volatile basic nitrogen (VBN) and fatty acid composition during ageing, 20 barrows (10 of each treatment) were randomly sampled, and aged for 3, 7, 12, 16, 20 and 25 days in a $1^{\circ}C$ chiller. The results showed that CAFC-fed pigs required approximately 10 fewer feeding days than the control group. Furthermore, the treatment resulted in greatly higher carcass grade whereby the grade A was increased by approximately 35% and 7% for gilts and barrows, respectively. The treatment had no significant effect (p>0.05) on pH, meat color and WHC during ageing. On the other hand, the CAFC-fed pigs showed significantly (p<0.05) lower TBARS values from 20 days of storage. In addition, the sum of unsaturated fatty acids for the treated group was significantly (p<0.05) higher than that for the control group after the storage time. This implied that CAFC supplementation could reduce the formation of free radicals in fatty acids (i.e., lipid oxidation). The treatment also significantly (p<0.05) retarded VBN formation during ageing, indicating a significant reduction in protein degradation. However, as there was no difference in pH between the two groups, the result raised a possibility that antibacterial activity of the CAFC alone could cause reduction in the formation of TBARS and VBN. In this regard, although the treatment effectively slowed down the formation of TBARS and TBA during chiller ageing, it was not resolved whether that was associated with the direct effect of the antioxidant function of chitosan and/or alginate, or a consequence of their antibacterial functions.

  • Growing and finishing Korean native bulls were used to investigate the effects of different dietary energy levels on performance and energy intake. Experimental observations were made over three weight categories (200 to 250 kg, 250 to 350 kg and 350 to 450 kg). Three diets (2.4, 2.6 and 2.8 Mcal ME/kg DM) were used for each weight category. Crude protein levels of the diets were 12, 11 and 10% for the respective weight groups. Dietary energy level did not significantly affect daily body gain within a weight range but daily body gain during the entire experiment was higher (P<.05) in bulls receiving 2.6 Mcal energy diet than in those fed 2.4 and 2.8 Mcal energy diets. The following equation was developed to predict daily body gain(Y) from energy levels(X) of ration. $Y\;=\;1.3.475X\;-\;2.5949X^{2}\;-\;16.355$ Increasing energy levels significantly (P<.05) decreased daily feed intake. The following equation was developed to predict daily feed intake(Y) from energy levels(X) of ration. $Y\;=\;-30.013X\;+\;5.4401X^{2}\;+\;49.119$ Feed intake per metabolic body size during the entire feeding period ranged from 100.9 to 110.8 g and was lower in bulls fed 2.6 and 2.8 Mcal energy diets than in those fed 2.4 Mcal energy diet. Increasing energy levels significantly (P<.05) improved feed efficiency. The following equation was developed to predict feed efficiency(Y) from energy levels(X) of ration. $Y\;=\;-118.34X\;+\;22.448X^{2}\;+\;162.85$ Daily energy intake during the entire experiment ranged from 18.90 to 19.99 Mcal and there was no significant difference among energy levels. Daily energy intake per metabolic body size during the feeding period ranged from 248.6 to 260.8 kcal and was slightly higher in bulls receiving 2.8 Mcal than in those fed 2.4 and 2.6 Mcal energy diets. Energy required per kg body gain ranged from 17.25 to 19.11 Mcal and was slightly lower in bulls receiving 2.6 Mcal energy diet than in those fed 2.4 and 2.8 Mcal energy diets.

The Effects of Restricted Feeding and Feed Form on Growth, Carcass Characteristics and Days to First Egg of Japanese Quail (Coturnix coturnix japonica)

  • A study was conducted to determine the effects of restricted feeding and feed form on the growth performance, characteristics of carcass and digestive tract, and days to first egg of Japanese quail (JQ, Coturnix coturnix japonica). A total of 240 oneweek-old JQ chicks were allocated randomly into 4 experimental groups that consisted of 3 replicates according to a 2${\times}$2 factorial arrangement for two feeding methods (ad libitum, AF and restricted feeding, RF) and two diet forms (mash, MD and crumble, CD). The JQ chicks were placed in a room with floor battery brooders and fed a commercial starter diet from 7 to 14 d of age. According to the experimental design, four treatments (1: ad libitum MD, 2: restricted MD, 3: ad libitum CD, and 4: restricted CD) were applied. Feed restriction was applied by 30% reduction of ad libitum feed intake for both MD and CD from 15 to 28 d of age. All birds were fed ad libitum with treatment diets from 29 d of age until the first laid egg seen (45 d of age). The commercial starter diet, MD and CD were in the same nutrient content (240 g crude protein with 13.4 MJ ME per kg diet). The body weight and overall feed conversion ratio (g feed/g gain) were higher (p<0.05) for the AF quails than the RF at 42 d of age. Carcass weights, dressing percentage and percentage yields of breast and back were similar for AF and RF groups at 42 d of age. The RF delayed (p<0.05) onset of egg production 2 days compared to the AF. Quail fed with the CD showed higher value (p<0.05) for carcass weight and dressing percentage at 42 d of age compared to birds fed with the MD. The interaction effect of feeding method${\times}$feed form on any of the studied parameters was not significant. The results suggest that feed restriction as in the present study can achieve a better feed conversion without reduction in carcass weight, and a significant benefit of feeding the crumble diet over the mash diet was obtained in terms of carcass weight in the JQ.

Effects of Ruminal Infusion of Garlic Oil on Fermentation Dynamics, Fatty Acid Profile and Abundance of Bacteria Involved in Biohydrogenation in Rumen of Goats

  • This study aimed to investigate the effects of ruminal infusion of garlic oil (GO) on fermentation dynamics, fatty acid (FA) profile, and abundance of bacteria involved in biohydrogenation in the rumen. Six wethers fitted with ruminal fistula were assigned to two groups for cross-over design with a 14-d interval. Each 30-d experimental period consisted of a 27-d adaptation and a 3-d sample collection. Goats were fed a basal diet without (control) or with GO ruminal infusion (0.8 g/d). Ruminal contents collected before (0 h) and at 2, 4, 6, 8, and 10 h after morning feeding were used for fermentation analysis, and 0 h samples were further used for FA determination and DNA extraction. Garlic oil had no influence on dry matter intakes of concentrate and hay. During ruminal fermentation, GO had no effects on total VFA concentration and individual VFA molar proportions, whereas GO increased the concentrations of ammonia nitrogen and microbial crude protein (p<0.05). Compared with control, GO group took a longer time for total VFA concentration and propionate molar proportion to reach their respective maxima after morning feeding. The ratio of acetate to propionate in control reduced sharply after morning feeding, whereas it remained relatively stable in GO group. Fatty acid analysis showed that GO reduced saturated FA proportion (p<0.05), while increasing the proportions of C18, t11-18:1 (TVA), c9,t11-conjugated linoleic acid (c9,t11-CLA), t10,c12-CLA, and polyunsaturated FA (p<0.05). The values of TVA/(c9,t11-CLA+TVA) and C18:0/(TVA+C18:0) were reduced by GO (p<0.05). Real-time PCR showed that GO tended to reduce Butyrivibrio proteoclasticus abundance (p = 0.058), whereas GO had no effect on total abundance of the Butyrivibrio group bacteria. A low correlation was found between B. proteoclasticus abundance and C18:0/(TVA+C18:0) (p = 0.910). The changes of fermentation over time suggested a role of GO in delaying the fermentation process and maintaining a relatively modest change of ruminal environment. The inhibitory effects of GO on the final step of biohydrogenation may be related to its antibacterial activity against B. proteoclasticus and other unknown bacteria involved.

Growth Performance and Meat Quality of Broiler Chickens Supplemented with Bacillus licheniformis in Drinking Water

  • A feeding trial was conducted to investigate effects of Bacillus licheniformis on growth performance and meat quality of broilers. Nine hundred one-d-old broiler chicks were randomly assigned to 3 experimental groups with three replicate pens of 100 broiler chicks. Three treatments were i) control, ii) basal diets supplemented with 1 ml of B. licheniformis for each in feed water per day iii) basal diets supplemented with 2 ml of B. licheniformis per chick in feed water per day. The supplementation of B. licheniformis significantly increased body weight in grower chickens (p<0.05), and significantly improved the feed conversion in 3 to 6 and 0 to 6 wk feeding period compared with the control group (p<0.05). Additionally, the supplement also resulted in increased protein and free amino acid contents, and decreased fat content in chicken breast fillet (p<0.05). Furthermore, improvement in sensory attributes was observed in broilers fed with the probiotic. In conclusion, B. licheniformis treatments resulted in a significant increase (p<0.05) in broiler productivity based on an index taking into account daily weight gain and feed conversion rate. Meanwhile, the probiotic contributed towards an improvement of the chemical, nutritional and sensorial characteristics of breast fillet. Overall, the study indicates that B. licheniformis can be used as a growth promoter and meat quality enhancer in broiler poultry.
