Study of the Coverage of Nutrition Labeling System on the Nutrient Intake of Koreans - using the 2013 Korea National Health and Nutrition Examination Survey (KNHANES) Data (현 영양표시제도로 파악할 수 있는 한국인의 영양소 섭취 정보의 범위: 2013년 국민건강영양조사 자료를 이용하여)

  • Objectives: The purpose of this study was to examine the coverage of the current mandatory nutrition labeling system on the nutrient intake of Koreans. Methods: KNHANES dietary intake data (2013) of 7,242 subjects were used in the analysis. KNHANES dietary intake data were collected by a 24-hour recall method by trained dietitians. For analysis, all food items consumed by the subjects were classified into two groups (foods with mandatory labeling and other foods). In the next step, all food items were reclassified into four groups according to the food type and nutrition labeling regulations: raw material food, processed food of raw material characteristics, processed foods without mandatory labeling, and processed foods with mandatory labeling. The intake of energy and five nutrients (carbohydrate, protein, fat, saturated fat, and sodium) of subjects from each food group were analyzed to determine the coverage of the mandatory nutrition labeling system among the total nutrient intake of Koreans. Results: The average intake of foods with mandatory labeling were 384g/day, which was approximately one quarter of the total daily food intake (1,544 g/day). The proportion of energy and five nutrients intake from foods with mandatory labeling was 18.1%~47.4%. The average food intake from the 4 food groups were 745 g/day (48.3%) for the raw food materials, 54 g/day (3.5%) for the processed food of raw material characteristics, 391 g/day (25.3%) for the processed foods without mandatory labeling, and 354 g/day (22.9%) for the processed foods with mandatory labeling. Conclusions: Although nutrition labeling is a useful tool for providing nutritional information to consumers, the coverage of current mandatory nutrition labeling system on daily nutrient intake of the Korean population is not high. To encourage informed choices and improve healthy eating habits of the Korean population, the nutrition labeling system should be expanded to include more food items and foodservice menus.

  • The incorporation of ³H-proline by epithelial and connective tissue elements of rat palatal mucosae was studied in order to investigate the relative levels of protein synthesis by the epithelium and underlying connective tissue cells. Following a sixty minutes incorporation of the radioactive tracer in vitro, it was found that the suprabasal cells had most grains per unit area. Furthermore, the grains were more concentrated over the cytoplasm than the nucleus. This was in contrast with the labeling of basal cells which had twice as many grains over the nucleoplasm than that over the cytoplasm. In intermediate cells; i.e., the spinous layer, the number of silver grains per unit area was decreased from that of the suprabasal cells. In areas where desmosomes were more prominent, many grains were in touch with such desmosomes. However, the labeling appeared to be reduced as soon as the cells became flattened. Moreover, the epidermal keratohyalin granules were relatively free of grains. Except for certain intercellular surfaces the keratinized cells were generally free of grains. On the connective tissue side, silver grains were primarily localized over the fibroblasts with occasional grains being found over palatal muscle cells, neural elements and so on. Most grains over collagenous fibers were found in relation to mature collagen fibrils. Thus, protein synthesis in isolated mucosae of the rat palate appeared to take place both in epithelial and connective elements. There were no apparent tissue alterations caused by the in vitro incorporation procedure utilized under conditions of this study.

Biochemical Properties and Localization of the β-Expansin OsEXPB3 in Rice (Oryza sativa L.)

  • ${\alpha}$-Expansins are bound to the cell wall of plants and can be solubilized with an extraction buffer containing 1 M NaCl. Localization of ${\alpha}$-expansins in the cell wall was confirmed by immunogold labeling and electron microscopy. The subcellular localization of vegetative ${\beta}$-expansins has not yet been studied. Using antibodies specific for OsEXPB3, a vegetative ${\beta}$-expansin of rice (Oryza sativa L.), we found that OsEXPB3 is tightly bound to the cell wall and, unlike ${\alpha}$-expansins, cannot be solubilized with extraction buffer containing 1 M NaCl. OsEXPB3 protein could only be extracted with buffer containing SDS. The subcellular localization of the OsEXPB3 protein was confirmed by immunogold labeling and electron microscopy. Gold particles were mainly distributed over the primary cell walls. Immunohistochemistry showed that OsEXPB3 is present in all regions of the coleoptile and root tissues tested.

Synthesis of 18F-labeled 2-cyanobenzothiazole derivative for efficient radiolabeling of N-terminal cysteine-bearing biomolecules

  • This article provides an efficient 18F-labeling protocol based on a rapid condensation reaction between 2-cyanobenzothiazole (CBT) and N-terminal cysteine-containing biomolecules. The 18F-labeled CBT (18F-1) was prepared by radiofluorination of the tosylated precursor 4 with 18-crown-6/K+/[18F]F- complex. Using the purified 18F-1, 18F-labeled peptide (18F-7) and protein (18F-8) could be synthesized efficiently under mild conditions. This strategy would provide a convenient approach for rapid and site-specific 18F-labeling of various peptides and proteins for in vivo imaging and biomedical applications.

Preparation and characterization of Ga-68-deferoxamine to test the feasibility as a bifunctional chelating agent or a renal imaging radiopharmaceutical

  • Chelating agents 1,4,7-triazacyclononanetriacetic acid (NOTA), 1,4,7,10-tetraazacyclododecane-1,4,7,10-tetraacetic acid (DOTA) and 30-amino-3,14,25-trihydroxy-3,9,14,20,25-penta-azatriacontane-2,10,13,21,24-pentaone (desferrioxamine, DFO) were labeled with $^{68}Ga$ and tested in vitro properties to check the feasibility of using DFO as a bifunctional chelating agent or renal imaging agent. The chelating agents of concentration $2{\mu}M$ were labeled with $^{68}Ga$ in 0.1 M HCl at pH 1.7-10.3 at room temperature and $80^{\circ}C$ and the optimal pH for labeling each chelating agent was found. And then, the chelating agents were labeled with $^{68}Ga$ in various concentration of chelating agents at optimal pH. The labeled chelating agents were subject to stability test in human serum and to binding studies to human red blood cell (RBC) and plasma protein. The optimal pH's of NOTA, DOTA and DFO for $^{68}Ga$-labeling were 4.4, 3.6 and 5.6, respectively. DFO ($10{\mu}M$) showed high labeling efficiency (>97%) at pH 5.6. All the labeled chelating agents showed high stability in human serum. $^{68}Ga$-DFO showed low RBC binding but significant amount was bound to plasma protein. The results demonstrated that $^{68}Ga$-DFO can be used as a bifunctional chelating agent but not as a renal imaging agent.

Understanding of Nutrition Labelling Use and Related Factors among Korean Adults

  • This study was conducted to investigate that the nutrition labeling use is associated with demographic and psychosocial factors according to each nutrition information on the nutrition labeling in Korean adults. The study subjects (N=1,140) were individuals who were aged 20 years and more and answered on the question of nutrition label use and who participated in the Korean National Health Examination and Nutrition Survey (KNHANES) in 2010.As age older, there was more interest in information such as sugar, protein, fat, cholesterol than calories. In contrast, as age younger, there was more interest intrans-fat, sodium as well as calories. As higher education level, there were more aware of trans-fat, sodium and calories. From the result that the most interested nutrition items were significantly different by democratic factors, we could understand interested nutrient information on the nutrition labels could change according to individual specific education. Therefore, this can also provide basic data for systematic education program by nutrition label use.

Nanomechanical Protein Detectors Using Electrothermal Nano-gap Actuators (나노간극 구동기를 이용한 나노기계적 단백질 검출기)

  • This paper presents a new method and an associated device, capable of detecting protein presence and size from the shift of the mechanical stiffness changing points due to the presence and size of proteins in a nano-gap actuator. Compared to the conventional resonant detection method, the present nanomechanical stiffness detection method shows higher precision for protein detection. The present method also offers simple and inexpensive protein detection devices by removing labeling process and optical components. We design and fabricate the nanomechanical protein detector using an electrothermal actuator with a nano-gap. In the experimental study, we measure the stiffness changing points and their coordinate shift from the devices with and without target proteins. The fabricated device detects the protein presence and the protein size of 14.0$\pm$7.4nm based on the coordinate shift of stiffness changing points. We experimentally verify the protein presence and size detection capability of the nanomechanical protein detector for applications to high-precision biomolecule detection.

A Study on the Awareness of Female-Consumers for Nutrition Labeling System (영양표시 제도에 대한 여성소비자 인식에 관한 연구)

  • This study had carried out to investigate the usage status of customer, the positive effects and problems, and the most important items of the nutrition labeling at purchasing the food etc. on the current nutrition labeling system in order to establish the customer-centric nutrition labeling system. Survey was carried out by questionnaire method that is targeted on adult female above 20 years old in Seoul and Kyeongnam area from May to June, 2004. For the experience of checking the nutrition label of the processed domestic and imported processed food, 82% and 75.4% of the respondents were replied 'have checked' respectively. For the positive effects due to enforcement of the nutrition labeling system, the respondents agreed highly with 'easy to compare with other products' and 'improve the products quality'. For the problems of the nutrition labeling system, the respondents agreed highly with 'different criteria for each product' and 'incendiary purchasing due to false or exaggerated labeling', and gave the higher scale for the positive effects than the problems relatively. For the necessity of the nutrition labeling system, 96.2% of the respondents were replied 'necessary', and it was indicated a significant difference on age and marital status(p<.01). For the price rising due to enforcement of the nutrition labeling system, 55.2% of respondents agreed, and it was indicated a very significant difference on age and monthly income(p<.001). For the most important nutrition labeling items at purchasing the food, the respondents were replied 'total calorie' on most of the food, and in addition, they checked carefully the lipid, cholesterol, protein, Ca, and Fe.
