Standardization of the Emergence Time of a Protaetia brevitarsis Adult (흰점박이꽃무지(Protaetia brevitarsis) 성충의 우화되는 시기의 표준화)

  • This study was conducted to determine and to standardize the time of emergence of Protaetia brevitarsis adults, through four breeding experiments, including one which selected the dominant individuals of Protaetia brevitarsis. The breeding experiments were divided into two groups with hibernating and non-hibernating Protaetia brevitarsis by placing 100, 150, 200, 250 and 300 Protaetia brevitarsis in each breeding box (50 cm × 35 cm × 15 cm) filled with 12 L of fermented sawdust. Breeding tests were with four replicates each for 3 months. The test period was classified into four stages: Exp. 1 (Sep 1, 2019 to Nov 30, 2019), Exp. 2 (Dec 1, 2019 to Feb 29, 2020), Exp. 3 (Mar 1, 2019 to May 31, 2020) and Exp. 4 (Jun 1, 2020 to Aug 31, 2020). The survey items included average dead number, average emergence number, average mortality, and average emergence rate. In Exp. 1 and 2, the changes in Protaetia brevitarsis adults were similar. In Tables 1 and 2, the allegorical changes of adult white-spotted flowers were similar. In addition, the average dead number, average emergence number, average mortality, and average emergence rate showed better results in the hibernation state than in both Exp. 1 and 2. Exp. 3 and 4 showed a pattern of change different from the results of Exp. 1 and 2 and these characteristics were superior in the hibernation states compared to the non-hibernation states. In conclusion, this result was found to be most appropriate as a standardization of the emergence time of 200 Protaetia brevitarsis treatments through four breeding experiments in the hibernating state.

Study on the Application of Protaetia brevitarsis Larva Excrement as Organic Fertilizer

  • This study was performed to assess Protaetia brevitarsis larva excrement as an organic fertilizer for corn cultivation. Furthermore, the study investigated insect communities in each treatment. In 2009, the corn growth rate was worst in soil treated with P. brevitarsis larva excrement and in 2010, the corn growth rate was worst in untreated soil. From the outcome of the study, P. brevitarsis larva excrement could be utilized as organic fertilizer for corn cultivation.

Antithrombotic Efficacy of Protaetia brevitarsis Extract (흰점박이꽃무지(Protaetia brevitarsis) 추출물의 항혈전 효능)

  • We aimed to evaluate antithrombotic efficacy of Protaetia brevitarsis extract during 21 days. Rats (SPF rat, weight 240~260 g) were divided into 16 groups (5 rats per group), they were: control group and Protaetia brevitarsis extract groups with dose of 0.1, 0.5, 1, 2.5, 5, 10, 25, 50, 75, 100, 200, 250, 500, 750, 1,000 mg/kg kg of body weight. Thromboplastin time (PT) and activated partial thromboplastin time (aPPT) as antithrombotic efficacy were tested in this animal experiment (at 7, 14 and 21 days). Overall, the admistration dose of Protaetia brevitarsis extract over 50 mg/kg at 7, 14 and 21 days for PT and over 25 mg/kg at 7, 14 and 21 days for aPPT tented to be longer than that of other groups. In addition, the optimal admistration doses of Protaetia brevitarsis extract to improves antithrombotic efficacy were 75, 100, 200 and 250 mg/kg at 7, 14 and 21 days for PT (p<0.05) and 50 and 100 mg/kg at 7 days, 75 mg/kg at 14 days, or 50, 100, 200 and 250 mg/kg at 21 days for aPPT (p<0.05). It can be concluded that Protaetia brevitarsis extract at optimal levels have antithrombotic efficacy.

The Anti-inflammatory Mechanism of Protaetia brevitarsis Lewis via Suppression the Activation of NF-κB and Caspase-1 in LPS-stimulated RAW264.7 Cells

  • The larva of Protaetia brevitarsis Lewis (P. brevitarsis), edible insect, is traditionally consumed as alternative source of nutrients and has various health benefits. However, the exact pharmaceutical effects of P. brevitarsis on inflammatory response are still not well understood. Thus, we investigated the anti-inflammatory effects and mechanisms of P. brevitarsis in lipopolysaccharide (LPS)-stimulated RAW264.7 cells. We investigated the effects of P. brevitarsis on the expression levels of inflammatory-related genes, including inflammatory cytokines, prostaglandin E2 (PGE2), cyclooxygenase (COX)-2 and inducible nitric oxide synthase (iNOS) in LPS-stimulated RAW264.7 cells. To understand the anti-inflammatory mechanism of P. brevitarsis, we explored the regulatory effect of P. brevitarsis on nuclear factor (NF)-κB and caspase-1 activation. The findings of this study demonstrated that P. brevitarsis inhibits the LPS-induced inflammatory cytokine and PGE2 levels, as well as COX-2 and iNOS expression. Moreover, we confirmed that the anti-inflammatory effect of P. brevitarsis occurs via suppression of the activation of NF-κB and caspase-1. Conclusively, these findings provide experimental evidence that P. brevitarsis may be useful candidate for the treatment of inflammatory-related diseases.

Effect of Some Environmental Factors on Oviposition and Developmental Characteristic of Protaetia brevitarsis and Allomyrina dichotoma (흰점박이꽃무지와 장수풍뎅이의 산란과 발육에 미치는 환경요인)

  • This study was carried out to obtain information about a several environmental factors influencing on the bionomical characteristics of Protaetia brevitarsis and Allomyrina dichotoma, which have been used for an important traditional medicinal stuffs. The moisture contents of humus affected the oviposition of P. brevitarsis and A. dichotoma, showing a suitable condition in range of $65{\sim}75%$. Very high or very low moisture condition in humus decreased the egg laying of P. brevitarsis and A. dichotoma, Also, moisture condition of humus was an important factor influencing on the egg and larval survival, egg developmental period, and feeding activity of P. brevitarsis and A. dichotoma.

Analysis of General Composition and Harmful Material of Protaetia brevitarsis (흰점박이꽃무지(Protaetia brevitarsis)의 일반성분 및 유해물질 분석)

  • To evaluate Protaetia brevitarsis as a food material, we investigated its composition of nutritional and harmful components. Analysis of nutritional composition (moisture, crude protein, crude fat, crude ash, crude fiber, and carbohydrates) showed that the content of crude protein, fat, fiber, and ash were $57.86{\pm}0.01$%, $16.57{\pm}1.81$%, $5.31{\pm}0.10$%, and $8.36{\pm}0.10$% in Protaetia brevitarsis powder, respectively. Amino acids were composed of 17.68% essential and 33.97% non-essential in Protaetia brevitarsis powder. Protaetia brevitarsis powder contained 61.10% unsaturated fatty acid with oleic acids. Additionally, Protaetia brevitarsis powder had a large quantity of minerals related to body organization, such as K (1597 mg/100 g), P (724.1 mg/100 g), Mg (366.3 mg/100 g), and so on. We also confirmed that all bacteria and all heavy metals analyzed in this study, except for very small amount of Hg ($0.1{\pm}0.042$ mg/kg), were not detected in the lysophilized Protaetia brevitarsis powder.

Radiation Protection Effect of Protaetia Brevitarsis Larvae Extracts on Blood and Prostate in Male Rats Irradiated with Co-60 Gamma-ray (흰점박이꽃무지 유충 추출물이 Co-60 감마선에 조사된 수컷 흰쥐의 혈구 및 전립선에 미치는 방사선 방호효과)

  • This study is desinged to examine for radiation protection effect of Protaetia Brevitarsis Larvae extracts on the blood and prostate of male rat as a natural radiation protection agent. 5 groups were classified using 90 male rat as experimental animals. Each group was clssified as normal control group (NC Group), the group administered protaetia brevitarsis larvae extracts (PBE Group), irradiated group (IR Group), irradiated group after administration of protaetia brevitarsis larvae extracts (PBE+IR Group), the group administered protaetia brevitarsis larvae extracts after irradiaton (IR+PBE Group). In IR Group, 7 Gy/h of Co-60 gamma ray was irradiated to SD rats. In PBE+IR Group, protaetia brevitarsis larvae extacts wewe injected at 200 mg/kg/day for 14 days before irradiation, In IR+PBE Group, protaetia brevitarsis larvae extract was injeted after irradiation. On the 1, 7 and 21 days after irradiation, the experimental animals were sacrificed to evaluate the changes in blood cell component, superoxide dismutase (SOD) activity, histopathological evaluation of the liver and prostate gland. As a result, the PBE+IR Group and IR+PBE Group showed a significantly recovery of white blood cell (p<0.01, p<0.01), platelet (p<0.01, p<0.01) than the IR Group. It was also confirmed that SOD activity of PBE+IR Group (p<0.01) and IR+PBE Group (p<0.01) was significantly increased than the IR Group. Also PBE+IR Group and IR+PBE Group showed less inflammatory reactions of cystoplasm in the prostate gland than the IR Group. In conclusion, the protaetia brevitarsis larvae have radioprotection effect against blood and prostate gland. It is expected to be useful for research of radiation protection agent.

Preventive Effect of A Chafer, Protaetia brevitarsis Extract on Carbon Tetrachloride-Induced Liver Injuries in Rats (사염화탄소로 유발된 랫드의 간 손상에 대한 흰점박이꽃무지 (Protaetia brevitarsis) 추출물의 예방효과)

  • The hepato-protective effects of the extract from Protaetia brevitarsis against hepatotoxicity by carbon tetrachloride ($CCI_4$) were studied in rats. The rats were orally treated with $CCI_4$ (50% in corn oil) at initial dose of $1\;m{\ell}/kg$ followed by $0.5m{\ell}/kg$ four times during 2-week period. The extract of P. brevitarsis (50, 100 or 200 mg/kg) or its vehicle was administered day after day from 1 week before $CCI_4$ Injection during five weeks. $CCI_4$ induced hepato-celluar degeneration and necrosis induced to increase in serum aspartate amintransferase (AST) and alanine aminotransferase (ALT) levels. In biochemical analyses, thiobarbituric acid-reactive substances (TBARS) and antioxidant enzymes such as superoxide dismutase (SOD) and catalase in hepatic tissues were remarkably increased by $CCI_4$ treatment. Not only increases in serum AST and ALT, but also induction of lipid peroxidation and antioxidant enzymes in hepatic tissues caused by $CCI_4$ were significantly attenuated by the P. brevitarsis extract in a dose-dependent manner. Such hepato-protective effects of P. brevitarsis extract were confirmed by histopathological examinations, wherein only mild hepatocytic vacuolations were observed in the liver of rats treated with a high dose (100 mg/kg) of P. brevitarsis extract in comparison with severe hepatocytic degenerations administered with $CCI_4$ alone. From these results, it is suggested that the extract of Protaetia brevitarsis could be a promising candidate for the protection of liver injury, based on the preventive effects against morphological cellular injuries, lipid peroxidation and serum biochemical parameters.

Investigation of the Proper Density for Rearing Protaetia brevitarsis Larvae (흰점박이꽃무지(Protaetia brevitarsis)의 유충사육 시 적정밀도 규명)

  • This study aimed to provide information and data for the management of insect breeding farms by identifying the appropriate density when rearing Protaetia brevitarsis larvae. The breeding box of the insects was filled with 2 L of fermented sawdust on a 50 × 35 × 15 cm sheet of plastic, and the density of the treatments was 200, 300 and 400 P. brevitarsis in the first, second, and third larval stages. Each treatment was repeated five times, and the sawdust medium was replaced three times (10 intervals). The experiments were conducted for a total of 30 days from March 1, 2020. Overall, 200 P.brevitarsis in the first, second, and third larval stages reared in the breeding box had a higher average survival rate and lower average mortality rate, thereby contributing to efficient production. In conclusion, this result suggests a way to increase production efficiency through the environmental management of insect breeding farms.

Investigation of Oviposition Characteristics of Protaetia brevitarsis seulensis (1) - Zone Breeding throughout the Year - (흰점박이꽃무지 산란 특성 조사(1) - 연중 누대사육을 중심으로 -)

  • We investigated the oviposition characteristics of Protaetia brevitarsis seulensis, focusing on zone breeding throughout the year in terms of the development and utilization of insect resources. To select Protaetia brevitarsis seulensis individuals laying a large number of eggs, the number of eggs laid for each individual was measured for each individual after emergence as adults from larvae grown at room temperature in five farmhouses. The five study zone included Gongju (Tancheon), Gyeonggi (Himsen), Chungnam (Gyeryong), Taean (Wonbuk), and Siheung (Baekse). The average oviposition tendency during a 12-week period obtained from the five regions was somewhat different; however, there was no remarkable difference in overall tendency. The maximum oviposition in Protaetia brevitarsisseulensis in the five regions occurred between 6 and 7 weeks. Compared toother regions, the average cumulative number of eggs laid during the 12-week period was higher (40%-60%) in Gongju (Tancheon) and Gyeonggi (Himsen). These results indicate that in two regions, zone breeding is selected as a system for increasing the demand for insects.
