인간 신장암 Caki세포에서 Par-4에 의한 MMP-2 활성 저해를 통한 세포 이동 조절 (Par-4 Modulates Cell Migration through Inhibition of MMP-2 Activity in Human Renal Carcinoma Caki Cells)

  • Par-4는 다양한 세포사멸 자극에 세포 사멸을 조절하고, 종양 억제기능을 가지고 있다. 그러나, Par-4에 의한 암세포의 이동 및 침윤에 대한 연구는 수행되지 않았다. 본 연구에서 Par-4단백질의 과발현이 인간 신장암 Caki세포에서 MMP-2의 활성화를 억제하지만 MMP-9 활성에는 영향을 주지 않았다. Par-4에 의한 MMP-2의 활성 억제는 leucine zipper domain이 결실된 Par-4 에서는 확인되지 않았다. Par-4 siRNA를 이용한 knock-down 실험에서 PMA 처리 시 세포이동 및 침윤 증가함을 확인하였다. Par-4의 과발현과 knock-dwon에서 MMP-2 mRNA 발현의 변화를 확인 할 수 없었다. 이 점은 Par-4 매개의 MMP-2 활성 억제는 전사 후 조절을 통하여 야기됨을 추측 할 수 있다.

1,3-비스페닐치오 프로판을 배위자로 한 백금 (II)착체의 선택적 세포독성 (Selective Cytotoxicity of New Platinum (II) Complex Containing 1,3-Bis-phenylthiopropane)

  • A new series of highly water soluble platinum(II) complexes {Pt(II)[1,3-bis(phenylthio) propane](trans- -1,2-diaminocyclohexane) (PC-1) and Pt(II)[1,3-bis-(phenythio)propane] cis-1,2-diaminocyclohexane(PC-2)} were synthesized, and characterized by their elemental analysis and by various spectroscopic techniques[infrared(IR), 13C-nuclear magnetic resonance (NMR)]. In vitro antitumor activity of new Pt(II) complexes was tested against P-388 and L-1210 mouse lymphocytic leukemia cell lines, PC-14 / P, PC-14/ADM and PC-14 / CDDP human pulmonary adenocarcinima, DU-145 human prostate carcinoma, HT-1376 human bladder carcinoma, ZR-75-1 human breast carcinoma, MKN-45/P and MKN-45/CDDP human gastric adenocarcinoma cell lines using colorimetric MTT[3-(4,5-dimethyl thiazol-2-yl)-2.5-diphenyltetrazoliumbromide] assay for cell survival and proliferation. PC-1 showed active against L-1210, P-388 leukemia, human lung, stomach, prostate, bladder and breast cancer cell lines, and the antitumor activity of these compounds were comparable or superior to those of PC-2 and displatin. The nephrotoxicities of PC-1 and PC-2 were found quite less than that of cisplatin using MTT and [3H] thymidine uptake in rabbit proximal tubule cells and human kidney cortical cells. Based on these results, this novel platinum (II) complex compound (PC-1) represents a valuable lead in the development of a new anticancer chemotherapeutic agent capable of improving antitumor activity and low nephrotoxicity.

Molecular Mechanism Underlying Hesperetin-induced Apoptosis by in silico Analysis and in Prostate Cancer PC-3 Cells

  • Aim: To investigate the molecular mechanisms underlying triggering of apoptosis by hesperetin using in silico and in vitro methods. Methods: The mechanism of binding of hesperetin with NF-${\kappa}B$ and other apoptotic proteins like BAX, BAD, $BCL_2$ and $BCL_{XL}$ was analysed in silico using Schrodinger suite 2009. In vitro studies were also carried out to evaluate the potency of hesperetin in inducing apoptosis using the human prostate cancer PC-3 cell line. Results: Hesperetin was found to exhibit high-affinity binding resulting from greater intermolecular forces between the ligand and its receptor NF-${\kappa}B$ (-7.48 Glide score). In vitro analysis using MTT assay confirmed that hesperetin reduced cell proliferation ($IC_{50}$ values of 90 and $40{\mu}M$ at 24 and 48h respectively) in PC-3 cells. Hesperetin also downregulated expression of the anti-apoptotic gene $BCL_{XL}$ at both mRNA and protein levels and increased the expression of pro-apoptotic genes like BAD at mRNA level and BAX at mRNA as well as protein levels. Conclusion: The results suggest that hesperetin can induce apoptosis by inhibiting NF-${\kappa}B$.

Generation of 1E8 Single Chain Fv-Fc Construct Against Human CD59

  • Background: Therapeutic approaches using monoclonal antibodies (mAbs) against complement regulatory proteins (CRPs:i.e.,CD46,CD55 and CD59) have been reported for adjuvant cancer therapy. In this study, we generated a recombinant 1E8 single-chain anti-CD59 antibody (scFv-Fc) and tested anti-cancer effect.by using complement dependent cytotoxicity (CDC). Methods: We isolated mRNA from 1E8 hybridoma cells and amplified the variable regions of the heavy chain (VH) and light chain (VL) genes using reversetranscriptase polymerase chain reaction (RT-PCR). Using a linker, the amplified sequences for the heavy and light chains were each connected to the sequence for a single polypeptide chain that was designed to be expressed. The VL and VH fragments were cloned into the pOptiVEC-TOPO vector that contained the human CH2-CH3 fragment. Then, 293T cells were transfected with the 1E8 single-chain Fv-Fc (scFv-Fc) constructs. CD59 expression was evaluated in the prostate cancer cell lines using flow cytometry. The enhancement of CDC effect by mouse 1E8 and 1E8 scFv-Fc were evaluated using a cytotoxicity assay. Results: The scFv-Fc constructs were expressed by the transfected 293T cells and secreted into the culture medium. The immunoreactivity of the secreted scFv-Fc construct was similar to that of the mouse 1E8 for CCRF-CEM cells. The molecular masses of 1E8 scFv-Fc were about 120 kDa and 55 kDa under reducing and non-reducing conditions, respectively. The DNA sequence of 1E8 scFv-Fc was obtained and presented. CD59 was highly expressed by the prostate cancer cell line. The recombinant 1E8 scFv-Fc mAb revealed significantly enhanced CDC effect similar with mouse 1E8 for prostate cancer cells. Conclusion: A 1E8 scFv-Fc construct for adjuvant cancer therapy was developed.

효과적인 항암효능측정을 위한 발광 전립선 세포의 개발 및 평가 (A New Bioluminescent Rat Prostate Cancer Cell Line: Rapid and Accurate Monitoring of Tumor Growth)

  • 기존의 동물모델에서 암의 성장은 caliper를 이용하여 고형암 부피를 측정으로써 조사하였으나, 암 조직 속의괴사와 부종으로 인하여 부피측정에 신뢰성이 결여 되어 있다. 이러한 문제점을 해결하기 위해 발광 암세포를 이용하여 광학생체영상적으로 분석하는 방법이 개발 되었다. 본 연구에서는 전립선 발광 암세포를 제조하여 고형암 동물모델에서 B16 발광 암세포와 암 성장을 비교 측정하여 신규발광 암세포를 평가하였다. In vitro에서 세포 수와 발광강도는 높은 상관관계를 보였고($R^2$=0.99), 고형암 동물모델에서 암 성장 측정은 괴사에 의한 오차를 줄였다. 이러한 발광신호를 기반으로 한 측정방법은 caliper의 부피 측정에 비하여 높은 항암효과를 보임으로써 기존의 발광 암세포보다 신규 발광전립선 암세포의 유용성을 증명하였다.

Microarray를 이용한 pipernonaline의 인간 전립선 암세포에 대한 기능 조절 분석 (Regulation of Pipernonaline on Biological Functions of Human Prostate Cancer Cells Based on Microarray Analysis)

  • Pipernonaline은 후추나무과에 속하는 필발(Piper longum Linn.)의 유도체로서 전립선 암세포에 대한 항암활성이 보고되고 있다. 하지만 실제 암세포 내에서 생물학적 정보를 가진 수 많은 유전자들에 대한 발현이 어떻게 이루어지고 있는지 알려진 바가 없다. 본 연구에 사용된 microarray 분석은 동시에 수 만개 이상의 유전자 발현양상을 한번에 관찰할 수 있는 기술로서 특정 질병의 유전학적 특성과 기전 연구를 더 광범위하게 연구 할 수 있는 기술이다. 본 연구에서는 전립선 암세포인 PC-3 세포에 pipernonaline을 처리하여 cDNA microarray를 실시하였다. 이후, DAVID database를 이용하여 gene ontology의 Biological Process를 분석하여 세포사멸과 세포주기, 세포성장 및 증식에 관련된 유전자들을 우선적으로 분석하였다. 그 결과, 세포주기관련 256개, 세포사멸관련 197개, 세포성장 및 증식관련에 154개의 유전자가 확인 되었다. 이러한 결과는 pipernonaline은 전립선 암세포 내에 존재하는 생물학적 신호전달체계에 관련된 유전자 발현을 조절함으로써 항암활성을 나타내 것을 알 수 있었고, 이후 이러한 microarray의 추가적인 분석은 암세포 내 새로운 유전자의 탐색 및 메커니즘을 규명하는데 유용하게 사용할 수 있을 것으로 사료된다.

Calcium Sensing Receptor Modulation for Cancer Therapy

  • The calcium sensing receptor (CaSR) is a member of the largest family of cell surface receptors, the G protein-coupled receptors involved in calcium homeostasis. The role of the CaSR in neoplasia appears to be homeostatic; loss of normal CaSR-induced response to extracellular calcium is observed in cancers of the colon and ovary, while increased release of PTHrP is observed in cancers of the breast, prostate and Leydig cells. Currently CaSR can be considered as a molecule that can either promote or prevent tumor growth depending on the type of cancer. Therefore, recognition of the multifaceted role of CaSR in gliomas and other malignant tumors in general is fundamental to elucidating the mechanisms of tumor progression and the development of novel therapeutic agents. Emphasis should be placed on development of drug-targeting methods to modulate CaSR activity in cancer cells.

인체 전립선 암세포에서 Methyl Gallate의 항암효과 (Anticancer Activity of Methyl Gallate in RC-58T/h/SA#4 Primary Human Prostate Cancer Cells)

  • 본 연구에서는 MG의 RC-58T/h/SA#4 인체 전립선 암세포에 대한 증식 억제 및 apoptosis 유도효과에 대하여 확인하였다. MG는 RC-58T/h/SA#4 세포의 증식을 농도 의존적으로 억제하였다. 이러한 MG에 의한 RC-58T/h/SA#4 세포의 사멸이 apoptosis에 의해 일어나는지를 sub-G1 함량 측정 및 Hoechst 33258 염색을 이용하여 확인하였다. 그 결과 MG를 처리한 군의 sub-G1의 함량이 대조군에 비하여 증가하였으며, methyl gallate를 처리한 군에서 핵의 응축과 apoptotic body 형성을 Hoechst 33258 염색을 통하여 관찰할 수 있었다. 또한 RC-58T/h/SA#4 세포에서 MG가 유도하는 apoptosis의 기전을 확인하기 위하여 다양한 실험을 실시하였다. MG는 DNA의 분절량 증가 및 caspase 활성을 유도하였으며, 활성화된 caspase-8, -9 및 -3에 의해 PARP와 Bid 단백질의 분절을 발현시켰으며, Bcl-2 family 단백질의 발현에 영향을 미쳐 apoptosis를 유도하였음을 확인하였다. 한편 환경호르몬인 dioxin과 bisphenol A를 다양한 농도로 처리한 결과 각각 1 nM, $0.1{\mu}M$ 농도에서 가장 높은 전립선 암세포 과다증식을 유도하였으며, 이를 바탕으로 MG를 농도별로 처리한 결과 환경호르몬에 의해 유도된 인체 전립선 암세포의 증식을 농도 의존적으로 억제시켰다. 본 연구 결과 MG는 RC-58T/h/SA#4 전립선 암세포에서 apoptosis 유도를 통한 암세포 성장 억제효과를 가지고 있음을 확인하였으며, 환경호르몬에 의해 유발될 수 있는 암에 대해서도 유사한 보호효과를 가지고 있음을 증명하였다.

강황이 수종의 암세포에 미치는 영향 (Effects of Curcuma longa L. on Some Kinds of Cancer Cells)

  • Objectives : The Purpose of this study was to identify anti-tumor effects of Curcuma longa L. on some kinds of cancer cells through molecular biologic methods. Materials & Methods : We used 4 kinds of cancer cell lines such as glioma cells(A172), cervical cancer cells(HeLa), Prostate cancer cells(PC3), lung cancer cells(A549). We injected the boiled extract of Curcuma longa L. $5{\mu}g,\;10{\mu}g$ to culture media(ml) for 24 hours. We measured the cytotoxic effect on 4 kinds of cancer cells through trypan blue exclusion test and the suppressive effect on viability of 4 kinds of cancer cells via MTT assay. We measured the change of mitochondria membrane potential via flow cytometry. The quantitative RT-PCR was used to examine the effect on the revelation of Bcl-2 and Bax which genes are related to apoptosis. We examined the effect on the revelation of Bcl-2 protein and Bax protein by western blot analysis. Results: 1. Extract of Curcuma longa L. showed significant cytotoxic effect on A172, HeLa, PC3 compared to the control group with density dependent manner. 2. Extract of Curcuma longs L. showed significant suppressive effect on viability of A172, HeLa, PC3 compared to the control group with density dependent manner. 3. Curcuma longs L. induced apoptosis by decreasing the membrane potential of mitochondria in A172, HeLa, PC3. 4. In the test about the revelation of genes related to apoptosis, the revelation of Bcl-2 decreased and the revelation of Bax increased in A172. HeLa, PC3 treated with Curcuma longa L. with density dependent manner. 5. In the test about the revelation of protein related to apoptosis, the protein levels of Bcl-2 decreased and the protein levels of Bax increased in A172, HeLa, PC3 treated with Curcuma longa L. Conclusions: This experiment shewed that Curcuma longs L. has anti-tumor effect on glioma, cervical, Prostate cancer cells except on lung cancer. We hope that anti-tumor effects of Curcuma longa L. will be more Practically identified.

복어독의 종양 관련 기존연구 논문에 대한 분석 (Analysis for cancer-related studies using Puffer fish poison)

  • Objective: The aim of this study is to analyze the previously published research articles related to puffer fish toxin focusing on tumor. Method: Literatures were searched in PubMed database, published since 2000, using the keyword; Puffer fish, Fugu and tetrodotoxin (TTX) with cancer or tumor. Research papers were classified by year, country, study model, used material, kind of tumor and study subject. Finally, a total of 41 studies were analyzed in this study. Results: From 2000 to 2018, the most abundant papers were published in 2009 (6 studies) and almost half of the papers were studied in United Kingdom (20 studies). The 39 studies used TTX purified from puffer fish while 2 studies used crude extract of skin and gonad of puffer fish. The most used target cell line was prostate cancer (15 studies), and the next was breast cancer (14 studies). The study methods were classified into 4 clinical studies, 2 animal studies and 35 cell-based studies. Conclusions: Our results show that the overview of cancer-related studies using puffer fish poison. This information would be helpful for the puffer fish-derived drug researches in the future.
